Cell Proliferation and Antioxidative Effects of Ultrasonic Coffee Extracts

  • Recently, coffee is the most popular beverage for modern people. A great number of substances are found in coffee beans and have been studied for many years such as aliphatic and aromatic compounds. However, studies on the physiological activity of coffee extracts are insufficient. This study was performed to determine the contents of caffeine and chlorogenic acid in coffee extracts according to the solvent and to investigate the physiological activity of coffee extracts. Coffee extracts were extracted by ultrasonication method with various types of solvents including distilled water, ethanol, and other organic solvents under $50^{\circ}C$ and $80^{\circ}C$. The contents of caffeine and chlorogenic acid in coffee extracts were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Also, cytotoxic and antioxidative effects of coffee extracts were evaluated with MTT and DPPH assays to analyze the physiological activity. As a result, it was confirmed that caffeine and chlorogenic acid contents were extracted in distilled water with the highest rate. Antioxidative activity was observed below 10-fold dilute of coffee extracts, however cytotoxicity was not observed. In conclusion, distilled water was the best solvent for extracting caffeine and chlorogenic acids from coffee bean with ultrasonication and these coffee extracts are less cytotoxic in human skin cell lines and have antioxidant effect.

쑥 추출액을 이용한 Microcystis aeruginosa 제거 및 성장억제 연구 (Removal and Regrowth Inhibition of Microcystis aeruginosa using Artemisia asiatica Extracts)

  • Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of Artemisia asiatica extracts to control M. aeruginosa inhibition will be environmentally friendly and promising. Artemisia asiatica extracts removed successfully upto 88% of M. aeruginosa pH 8 at $25^{\circ}C$ of temperature. These results was indicated that the amount of 2.24 g/L Artemisia asiatica extracts was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 2.5 g/L of initial concentration of Artemisia asiatica extracts. In the various growth control models, Luong model showed the highest correlation coefficient of 0.9916. Therefore, the Luong model was the most suitable control model for the growth control of M. aruginosa using Artemisia asiatica extracts. In conclusion, the growth control of M. aruginosa using Artemisia asiatica extracts can be applied in the field without controlling the temperature and pH of rivers and streams, and it is possible to control the growth of M. aruginosa efficiently in a short time. The natural extract, Artemisia asiatica extracts, can be a promising inhibition due to its high efficiency and low dose requirements.

Antioxidant and ACE Inhibitory Activity of Cultivated and Wild Angelica gigas Nakai Extracts Prepared Using Different Extraction Conditions

  • The purpose of this study was to investigate the biological activities of cultivated Angelica gigas Nakai (CAG) and wild Angelica gigas Nakai (WAG) extracts prepared by extraction with water, 30% ethanol, 60% ethanol, or 90% ethanol. The electron donating ability of the WAG extracts was higher than that of the CAG extracts and 0.1% and 1.0% solutions of the comparative substance, L-ascorbic acid. The superoxide dismutase-like activity of the CAG extracts was higher than that of WAG extracts. Superoxide dismutase-like activity was highest (33.95%) in the CAG water extract. The total polyphenol content was highest in the 60% ethanol extracts of WAG. The nitrite scavenging ability of the CAG and WAG extracts was highest at a pH of 1.2. The tyrosinase inhibitory effect was highest (43.72%) in the water extract of WAG. The angiotensin converting enzyme inhibitory activity was highest (83.84%) in the 60% ethanol extract of WAG. The results of the present study will be useful for understanding the antioxidant and angiotensin-converting enzyme inhibitory activities of Angelica gigas Nakai extracts.

용매에 따른 양송이 추출물의 생리활성 효과 (Physiological Activities of Agaricus bisporus Extracts as Affected by Solvents)

  • 갈색과 흰색 양송이를 열수, 50% 에탄올 및 100% 에탄올 등의 추출용매를 사용하여 건물 중량의 10배에 해당되는 부피(w/v)일 때, 추출물의 생리활성을 탐색하였다 전자공여 작용의 경우 갈색양송이의 50% 에탄을 추출물에서 66.54%의 높은 전자공여능을 나타내었으며, SOD 유사활성을 측정한 결과 갈색양송이의 열수 추출물이 55.42%로 다른 추출물에 비해 높은 활성을 보여주었다. Tyrosinase저해활성의 경우 두 종류 모두 100% 에탄을 추출물에서 높은 저해활성을 보여주었으며, 갈색양송이가 흰색양송이보다 다소 높은 활성을 나타내었다. 특히 비교물질인 0.1% L-ascorbate와 유사한 활성을 보여주었다. 총 폴리 페놀 함량의 경우 두 종류 모두 열수 추출물보다 50%, 100% 에탄을 추출물에서 높은 함량을 나타내었다. 아질산염 소거작용을 측정한 결과 pH 1.2일 때 갈색양송이와 흰색양송이 모두 소거능이 높게 나타났다.

Anti-Inflammatory Activity of Pinus koraiensis Cone Bark Extracts Prepared by Micro-Wave Assisted Extraction

  • In this study, we compared the anti-inflammatory activity of Pinus koraiensis cone bark extracts prepared by conventional extraction and microwave-assisted extraction (MAE). Water extracts and 50% ethanol extracts prepared using MAE were applied to RAW 264.7 cell at 5, 10, 25, and $50{\mu}g/mL$ of concentrations, and tested for cytoxicity. The group treated with $50{\mu}g/mL$ of 50% ethanol extracts showed toxicity. In order to investigate the inhibition of nitric oxide (NO) production in RAW 264.7 cells, extracts of water and ethanol were treated with 5, 10, and $25{\mu}g/mL$ concentrations. The inhibitory activity of water and 50% ethanol extracts groups were determined as 40% and 60% at $25{\mu}g/mL$ concentration, respectively. We found concentration dependent decreases on inducible NO synthase. The inhibitory effect against forming inflammatory cytokines, prostaglandin $E_2$, tumor necrosis factor-${\alpha}$, interleukin (IL)-6, and IL-$1{\beta}$, was also superior in the $25{\mu}g/mL$ treated group than the control group. According to these results, the water extracts and 50% ethanol extracts both inhibited inflammatory mediators by reducing the inflammatory response. Therefore, The MAE extracts of P. koraiensis cone bark can be developed as a functional ingredient with anti-inflammatory activity.

생약복방제의 에탄을 농도별 엑스중 인삼 사포닌의 이행량 조사 (Extraction Yields of Ginseng Saponins in the Extracts of Crude Drug Preparations with Various Concentrations of Ethanol)

  • As a part of studios on the Quality control of index components in crude drug preparations, extraction yields of ginseng saponins from crude drug extracts were identified by TLC and quantified by HPLC. So-Shi-Ho-Tang(小柴胡湯), Sa-Kun-Ja-Tang(四君子湯), Yook-Kun-Ja-Tang(六君子湯) and In-sam-Tang(人蔘湯) were extracted with water, 30%-ethanol, 50%-ethanol, 80%-ethanol and absolute ethanol to analyze ginseng saponins in the crude drug extracts prepared with various concentrations of ethanol. Ginseng saponins were extracted considerably more from the extracts with higher concentrations of ethanol than those with water or lower concentrations of ethanol. Extraction yields of ginseng-side-Rb$_1$, -Rb$_2$ and -R$_c$ from four crude drug preparations were the lowest as 4.9~45.9%, 5.0~40.1, and 6.3~43.7% in water extract and the highest as 29.5~62.6%, 26.7~61.4% and 31.4~62.0% in absolute ethanol extract, compared with those of 80%-methanol extracts.

Evaluation of the Biological Activity of Extracts from Star-Anise (Illicium verum)

  • This study was carried out to investigate the biological activities of star anise extracts obtained from four different solvents (water, methanol, ethanol and chloroform) by measuring total polyphenol and flavonoid contents, DPPH radical scavenging, hydroxyl radical scavenging, nitrite scavenging activity and antimicrobial activity. The methanol extract showed the highest extraction yield, followed by ethanol, water and chloroform. The properties of the extracting solvents significantly affected the total polyphenol content. Methanol extracts contained more total polyphenols than any other extracts. The highest DPPH radical scavenging activity was found in methanol extract. The hydroxyl radical and nitrite scavenging activity were highest in methanol and ethanol extracts. In antimicrobial activity, water extract showed stronger activities than methanol and ethanol extract against Micrococcus luteus and Bacillus subtilis, and no inhibitory effects on Gram negative bacteria were found in all extracts at the concentration used.

홍경천(紅景天)의 면역증진(免疫增進) 효능(效能)에 대한 연구(硏究) (A Study on the Effects of Rhodiola rosea Root on the Immune System)

  • Objectives: Effects of Rhodiola rosea root on immunity were examined. Methods: Water extracts and methanol extracts of Rhodiola rosea root were treated on the increase of immunity cells and the activation of cytokines were examined. Results: 1. Water extracts and methanol extracts of Rhodiola rosea root had positive effects on the increase of B-cells, T-cells, and NK cells, and on the facilitation of cytokines such as IL-6 and TNF-${\alpha}$. 2. Water extracts and methanol extracts of Rhodiola rosea root activated NK cells, and induced the generation of NO- of NK cells and macrophages. Conclusions: Rhodiola rosea root can be used to increase immunity.

백부자의 추출물의 흰쥐 건성피부에 대한 생리적 효능 (Physiological Effects of Aconitum koreanum Extracts on the Dry Skin)

  • Water and ethanol extracts of A. koreanum were prepared and tested for their improving effects on the rat's dry skin induced by household detergent. Rats treated with ethanol extracts of A. koreanum had higher serum HDL-cholesterol and lower LDL-cholesterol levels, when compared to the ones treated with water extracts. Animals treated with ethanol extracts showed that improved protein recovery patterns over time, when it compared to the ones treated with water extracts.

  • This study was conducted to investigate the effects of Korean ginseng extracts and their fractions on the growth of Saccharomyces cerevsiae and Saccharomyces uvamm, their glucose consumption and alcohol production. The growth of both yeasts were stimulated by ginseng extracts and their water soluble fractions, but were supressed by ether extracts and an n-butanol extracts. Their growth were enhanced considerably by low molecular weight fractions (< 1,000) in water solubles. Similar results were also obtained with glucose consumption by yeasts. Substances increasing the growth and glucose consumption by yeasts proved to be a low molecular weight fractions (<1,000) in water solubles not saponins. The production of n-propyl alcohol by yeast was enhanced by adding ginseng extracts into the media, but that of ism-butyl alcohol was suppressed at same condition.
