Antioxidant and antimicrobial efficiency were investigated for: i) water extract of green tea(Camellia sinensis), ii) water extract of middle drying megmoondong(Liriope platyphylla Wang et Tang) and iii) water extract of drying megmoondong. The electron donating ability of the green tea(Camellia sinensis) and drying megmoondong(Liriope platyphylla Wang et Tang) extract were 88 and 79, respectively, while that of the middle drying megmoondong extract showed low levels. The relative inhibitory abilities against lipid peroxidase were: green tea(Camellia sinensis) extract > drying megmoondong(Liriope platyphylla Wang et Tang) extract > middle drying megmoondong extract. The green tea(Camellia sinensis) extract showed the strongest antimicrobial effect against Staphylococcus aureus. The middle drying(Liriope platyphylla Wang et Tang) and drying megmoondong extracts showed no antimicrobial effect against Staphylococcus aureus. The green tea(Camellia sinensis) extract had the strongest antimicrobial effect against Escherichia coli. The middle drying megmoondong(Liriope platyphylla Wang et Tang) and drying megmoondong extracts showed no antimicrobial effect against Escherichia coli.
The aim of this study is to investigate the effects of hot water soluble extract from green tea on the components of serum and the liver and the antioxidative effects in accordance with the different protein types. For this purpose, four experimental groups were set up. As for the protein source casein, isolated soy protein was supplemented to the rats, together with hot water soluble extract from green tea. Four experimental groups kept eight Sprague-Dawley rats respectively. The CP group was supplemented with casein only, the CG group was supplemented with casein and hot water soluble extract from green tea, the ISP group was supplemented with isolated soy protein only, and the ISG group was supplemented with the isolated soy protein with hot water soluble extract from green tea. After 4 weeks of feeding with experimental diet, the levels of serum and liver lipid and antioxidant enzyme activity and TBARS in the liver were measured. The results are; 1. Weight gain and FER were higher in the casein group than in the isolated soy protein group in general. In the casein group, the weight gain and FER were reduced significantly when hot water soluble extract from green tea was supplemented (P < 0.05). There was no significant difference in feed intake. 2. In general, total cholesterol and LDL-cholesterol in the serum were higher in the casein group than in the isolated soy protein group, however just the concentration of LDL-cholesterol in the casein group was significantly lower when the hot water soluble extract from green tea was supplemented (P < 0.05). 3. Triglycerides in the liver were higher in the casein group than in the isolated soy protein group general, however when hot water soluble extract from green teas was supplemented only in the isolated soy protein group, the content of triglyceride in liver was significantly lower (P < 0.05). 4. There was no significant difference in antioxidant enzyme activity in the liver in all the groups, however the content of TBARS was low only in the casein group when hot water soluble extract from green tea was supplemented (P < 0.05).

Effect of green tea extract, on duodenal ulceration was studied in male Sprague Dawley rats treated with cysteamine, a drug, which causes duodenal ulcers in experimental animal. As a result, in the proximal duodenum, a significant decrease of ulceration was detected twenty four hours after cysteamine injection in rats raised in green tea extract for 63days. Special reference to duodenal alkaline phosphatase activity was measured in mucosal homogenates. In control rats raised in tap water Riven saline, significant decrease was observed in proximal duodenal alkaline phosphataes activity. The decrease effect seems site specific, since the enzyme in the distal duodenum remains. Moreover the effect cysteamine in control rats alkaline phosphatase is specific, because, in rats raised in green tea extracts did not show significant change in activity. It is suggested that green tea extract acts in ideal properties as an anti-duodenal ulcer agent.
The sensitivity of various pathogenic bacteria(Listeria monocytogenes, Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli O157:H7 and Salmonella typhimurium) to the pine needle and green tea extracts was tested. Water extract of pine needle(PNW), 70% ethanol extract of pine needle(PNE), water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activty. Tryptic soy broth(TSB) containing 0∼2%(w/v) of pine needle and green tea extracts were inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacterium and incubated at 35$\^{C}$ for 24 hours. The standard plate count method was used to measure the inhibitory effect of the extracts. Minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were derived from the survival curves of pathogenic bacteria. Antibacterial activities of the pine needle and green tea extracts were compared with that of sodium benzoate, a preservative, by clear zone test. L. monocytogenes, S. aureus and A. hydrophila were completely inhibited at 0.4∼1.6% level while E. coli and S. typhimurium were very resistant to the pine needle extracts. Green tea extracts completely inhibited all strains tested at 0.2∼1.0% level and bactercidal to all strains except L. monocytogenes at 0.5∼2.0% level. Antibacterial activities of pine needle and green tea extracts were stronger than that of sodium benzoate. The order of antibacterial activities of pine needle and green tea extracts to the pathogenic bacteria was GTE > GTW > PNE > PNW. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.
Water extract of green tea(GTW) and 70% ethanol extract of green tea(GTE) were prepared for the test of antibacterial activity. The sensitivity of Staphylococcus aureus and Salmonella typhimurium to the green tea extracts in various pH of culture broth was tested. Tryptic soy broth(TSB) containing 0∼2%(w/v) of green tea extracts was adjusted to pH 5.0∼7.0 and inoculated with 10$\^$5/∼10$\^$6/ cells/ml of each bacteria. The plate counting method and clear zone test were used to determine inhibitory effect of green tea extracts. Green tea extracts completely inhibited the growth of S. aureus at 0.5% level and bactercidal at 0.5∼1.0% level of GTW and GTE at pH 5.0∼7.0. Green tea extracts were bactercidal to S. typhimurium at 1.5∼2.0% level of GTW and 1.0∼2.0% level of GTE at pH 7.0. Sal. typhimurium was more resistant than S. aureus. in same concentration of green tea extracts at same pH. The resistance of S. aureus and Sal. typhimurium was increased with decreasing pH of culture broth. The morphology of S. aureus cells treated with green tea extracts showed damage of cell wall and cytoplasmic membrane. Severely damaged cells of S. aureus lost electron dense material and cytoplasm. Green tea extracts stimulated autolysis and cell death of S. aureus. This result suggests that green tea extracts can be used as an effective natural antibacterial agent in food.

This experiment extracted the natural green-tea using ethanol and obtained the refined oil component after filterated green-tea extract. This study tested the antimicrobial effect as characteristics experiment, and analyzed refined oil component with pH-meter and GC/MS. In the result of this experiment, it obtained the next conclusions. In the first result of extraction experiment, it could know that extraction ratio of refined oil component appeared about 9.0%. In the second result of characteristics experiment, it could certificate that in case of increasing the refined oil component in concentration of 100ppm and above, and according to passage of cultivation time, the number of S-aureus and E-coli in microbe decreased less and less. But in case of blank test not adding the refined oil component, the number of microbe increased more and more. In these phenomena, it could certificate that refined oil component of green-tea appeared antimicrobial effect against microbe. In the third result of instrumental analysis, refined oil component of green-tea appeared about 7.6 in 1% distilled water solution with pH-meter, and the aromatic components of benzene, bonyl acetate, campene, ${\alpha},{\beta},{\gamma}$-pinnene etcs from refined oil component of green-tea was detected with GC/MS.
The sensitivity of various pathogenic bacteria(Escherichia coli O157:H7, Listeria monocytogenes, Staphylococcus aureus 196E, Salmonella typhimurium) to the water extract of green tea was tested. Tryptic soy broth was inoculated with 10$\^$5/CPU/ml of pathogenic bacteria and incubated at 35$^{\circ}C$ for 30 hours. The extract was added at a final concentration of 0-2%(w/v) into culture broth at the mid or late exponential phase of bacteria. The growth of pathogenic bacteria was inhibited with increasing concentrations of the extract in culture broth and the late exponential phase cells were more resistant than the mid exponential phase cells. Cram positive bacteria(L. monocytogenes and S. aureus 196E) were more sensitive than Cram negative bacteria(E. coli O157:H7 and S. typhimurium). S. aureus had the highest sensitivity, followed by L monocytogenes, E. coli O157:H7. S. typhimurium was the most resistant to the water extract of green tea.
Desmutagenic effects of water-soluble and ethanol-soluble extracts of dried green tea toward the mutagenicity of 4-nitroquinoline-N-oxide(4-NQO) and 3-Amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-P-1) in streptomycin-dependent SD510 strains of Salmonella typhimurium TA98 were investigated. Inhibition effects toward the mutagenicity of 4-NQO and Trp-P-1 of water-soluble and ethanol-soluble extracts from green tea were high as increase of concentration of extracts. Desmutagenic effects toward 4-NQO of water-soluble and ethanol-soluble extracts from green tea harvested in May and August were up to 93% in 1,000 $\mu\textrm{g}$ of extract/plate. Desmutagenic effects toward Trp-P-1 of ethanol-soluble extracts from green tea were 53.3∼92.1%, but the effects of water-soluble extracts decreased as increase of concentration of extracts.
The beneficial effects of green and fermented tea are generally attributed to some antioxidant activities including superoxide dismutase (SOD)-like ability and scavenging activity originated from their phenolic compounds and flavonoids. Content of total flavonoid of green tea $(413.3\;{\mu}g/g)$ was similar to those of fermented tea $(405.7\;{\mu}g/g)$. Content of total phenol of green tea $(46.8\;{\mu}g/g)$ was higher than those of fermented tea $(23.5\;{\mu}g/g)$. Major catechin compounds of hot water extract in green tea was EGCG, including EGC, Gc, catechin and catechol. EGCG was not detected .in fermented tea. SOD-like ability was increased in proportional to added concentration of hot water extract. The scavenging activities of hydroxyl radical at $3000\;{\mu}g/ml$ of green and fermented teas were found up to $60\%$. Hot water extract of green tea was more effective in scavenging activity than that of fermented tea.
This study was designed to observe the effect of green tea on colon tumor incidence and biomarkers of colon carcinogenesis in 1, 2-dimethlhydrazine-treated rats. Male Sprague Dawley rats at 7 weeks of age were divided into two groups: control and green tea(GT) groups. Control rats had distilled water as drinking water but GT group received green tea extracts(2.5%, w/v water) as drinking water throughout the experiment periods. All rats were fed the experimental diet containing 15% fat by weight for 20 weeks. and were i.m. injected with DMH for 6 weeks to give total dose of 180mg/kg body weight. Tumor incidence was reduced in GT group (39%) compared with control group (56%) Green tea significantly reduced cell proliferation (total cells per crypt, crypt length and proliferative zone) in colonic mucosa and also significantly reduced the levels of preformed prostalandin E2(PGE2) and thromboxance B2(TXB2) in colonic mucosa but the fatty acid profile of total lipid in colonic mucosa was not significantly influenced by green tea. However the relative percent of C20:4 and the levels f preformed PGE2 and TXB2. were significantly higher in tumor tissue compared with normal surrounding mucosa.Green tea increased the fecal excretion of total bile acid but not scondary bile acid which is known as one of promoters for colon cancer,. These results suggest that green tea could have preventive effect against colon cancer when consumed daily by influencing on antioxidant effect and the metabolism of arachidonic acid.
