CCD 카메라를 사용한 물벼룩의 실시간 활동량 측정 하드웨어와 원격 모니터링 시스템 구현 (Implementation of Real-time Measurement Hardware for Activity of Water Flea and Remote Monitoring System using CCD Camera)

  • 수질 감시를 위한 물벼룩 활동량 측정을 실시간으로 측정하는 하드웨어를 구현 하였다. 물벼룩은 수질 감시에 빈번히 시용하는 살아있는 생물학적 센서라고 할 수 있다. 물벼룩은 수질이 오염이 되면 활동량의 변화가 나타나기 때문에 물벼룩의 활동량을 감시함으로서 해당 수질의 독성 물질의 유입을 즉각 확인할 수 있다. 물벼룩의 활동량 조사는 기존의 방법은 LED를 이용한 발광부와 Photo TR.을 이용한 수광부를 사용하는 방법이 있으나 물벼룩이 빛에 민감하게 반응하고 측정 대상인 물벼룩을 기준에 두고 2 개의 장치(발광부와 수광부)를 두는 불편한 점이 있었다. 본 논문은 기존의 방식을 대체하기 위해 CCD 카메라를 사용한다. CCD 카메라의 영상처리를 위해 이미지 버퍼에 저장한 후 컴퓨터나 DSP를 사용하여 소프트웨어로 일괄 처리를 하는 것이 아니라 실시간으로 하드웨어로 처리하는 시스템을 구현하였다. 그리고 구현된 하드웨어로 수질 경보를 위한 원격 모니터링 임베디드 시스템을 구축하였다.

수분 및 Glycerol의 섭취가 운동시 renin activity, 직장온도, 혈중 전해질 농도에 미치는 효과 (The Effects of Ingestion of Water or Glycerol on Renin Activity, Rectum Temperature, Blood Electrolytes Concentration in Exercise)

  • 본 연구는 수분 및 글리세를 섭취가 renin activity, 직장온도, 혈중 전해질 농도에 미치는 효과에 대하여 알아보기 위해 수분 섭취 그룹과 글리세롤 섭취 그룹으로 나누어 측정한 결과는 다음과 같다. renin activity의 경우, 안정시와 비교하여 운동 후 40분에 수분 섭취 그룹과 글리세를 섭취 그룹에서 유의한 증가를 보였다(p<0.01). Osmolality의 경우, 수분 섭취 그룹에서 차이를 보이지 않았으나, 글리세를 섭취그룹에서는 안정 시와 비교하여 운동 후 40분에 차이를 보였다(p<0.05). 직장온도의 경우, 수분 섭취 그룹과 글리세를 섭취 그룹 모두 운동 시간에 따라 점진적으로 증가하는 것을 볼 수 있었다. Na과 K의 경우, 수분 섭취 그룹에서 차이를 보이지 않았으나 글리세를 섭취 그룹에서 차이를 보였다(p<0.05). Ca과 Mg의 경우, 두 그룹 모두 그룹과 운동시기 간에 유의한 차이를 보이지 않았다.

분심기음가미방(分心氣飮加味方)의 항산화능과 serotonin 대사에 미치는 영향 (The Effects of BunSimGiEumGami-Bang(Fenxinqiyinjiameifang) on Serotonin of P815 cell)

  • Objectives : This experiment was designed to investigate the effect of the BSGE hot water extract on serotonin biosynthesis of depression model. Methods : The cytotoxicity of the BSGE hot water extract was analyzed by MTT assay on P815 cell. The antioxidant activity was measured by DPPH free radical-scavenging activity and SOD activity on P815 cell. The quantity of 5-HT and expression of TPH-1, AAADC and MAOa mRNA was measured by of HPLC profle Analysis on P815 cell. Results : 1. The BSGE hot water extract increased DPPH free radical-scavenging activity and SOD activity on P815 cell. 2. The BSGE hot water extract increased significantly the quantity of 5-HT. 3. The BSGE hot water extract increased the expression of TPH-1 mRNA. Conclusions : This experiment shows that the BSGE hot water extract might be effective for the prevention and treatment of depression. Investigation into the clinical use of the BSGE hot water extract for depression is suggested for future research.

육두구의 생리활성에 관한 연구 (Functional Properties of Nutmeg)

  • This study investigated the antioxidative and antimicrobial activities of nutmeg (water, ethanol extract and essential oil). The total phenol contents of water, ethanol extract and essential oil were 3.4%, 16.9%, and 3.2%, respectively. Hydrogen donating abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.9%, 41.8% and 6.8%, respectively. The antioxidative activities in linoleic acid substrates were in the order of BHT > ethanol > extract > ${\alpha}$-tocopherol > essential oil > water extract. The antioxidative activities in linoleic acid emulsion substrates were in the order of BHT > water extract > essential oil > ethanol extract > ${\alpha}$-tocopherol. In antimicrobial activity, ethanol extract showed growth inhibition effect against Micrococcus luteus, Bacillus cereus and Salmonella enteritidis, and the essential oil showed growth inhibition effect against Micrococcus luteus. However, no antimicrobial activity of water extract was observed. The nitrite-scavenging abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.5%, 28.8% and 98.8%, respectively, and the ACE inhibitory activities were 0.2%, 11.0% and 10.0%, respectively.

담수어와 해수어의 근원섬유단백질의 특성 비교 (Comparison of Biochemical Characteristics of Myofibrillar Protein from Fresh Water Fish and Sea Water Fish)

  • Myofibril and actomyosin were prepared from red muscle and white muscle of fresh water fish and sea water fish, and their biochemical characteristics and SDS PAGE patterns of myofibril were compared. SDS PAGE analysis showed that electrophoretic patterns of myofibril were similar be tween white muscle and red muscle, while difference of 30kDa component of myofibril was detected between fresh water fish and sea water fish. When myofibril were treated with trypsin, difference in hydrolysis of heavy chain was observed between white muscle and red muscle. In activities of Ca ATPase, Mg ATPase, EDTA ATPase and ATPase activity pH curve, myofibrillar protein from fresh water fish showed higher specific activity than those from sea water fish.

Comparison of Antimicrobial Activity of Electrolyzed Water Using Various Electrodes against Biofilm of Oral Pathogens

  • Biofilms of oral microbes can cause various diseases in the oral cavity, such as dental caries, periodontitis and mucosal disease. Electrolyzed water generated by an electric current passed via water using a metal electrode has an antimicrobial effect on pathogenic bacteria which cause food poisoning. This study investigated the antimicrobial activity of electrolyzed waters using various metal electrodes on the floatage and biofilms of oral microbes. The electrolyzed water was generated by passing electric current using copper, silver and platinum electrodes. The electrolyzed water has a neutral pH. Streptococcus mutans, Porphyromonas gingivalis and Tannerella forsythia were cultured, and were used to form a biofilm using specific media. The floatage and biofilm of the microbes were then treated with the electrolyzed water. The electrolyzed water using platinum electrode (EWP) exhibited strong antimicrobial activity against the floatage and biofilm of the oral microbes. However, the electrolyzed water using copper and silver electrodes had no effect. The EWP disrupted the biofilm of oral microbes, except the S. mutans biofilm. Comparing the different electrolyzed waters that we created the platinum electrode generated water may be an ideal candidate for prevention of dental caries and periodontitis.

토양 수분 Stress에 따른 상추의 엽중 상대수분 함량과 아스코브산 관련 효소 활성도 (Relationship Between Relative Water Content and Ascorbate Redox Enzymes Activity in Lettuce Leaves Subjected to Soil Water Stress)

  • The relationship between relative water contents of lettuce leaves and biochemical activities in lettuce was examined in this study to explore an adaptation response of lettuce to water stress from soils. Soil water contents and relative water contents of leaves were positively related to show $R^2$=0.8728. Hydrogen peroxide contents of leaves rapidly increased with reduction of soil water content, whereas soluble protein contents and dry matters rapidly decreased. And chlorophyll a and b contents of leaves decreased with increase in carotenoid content. Furthermore, the activities of ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and dehydroascorbate reductase (DHAR) increased dramatically, and mRNA transcript levels of APX, MDHAR and DHAR also increased. Relationship of relative water content of lettuce leaves to hydrogen peroxide, to ascorbate peroxidase activity, to dehydroascorbate reductase activity, and to monodehydroascorbate reductase activity was shown to be positively correlated. It is highly plausible from this study that these enzyme activities could be developed as an indicator of water states in soils.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

녹차 물 추출액으로 재배한 콩나물의 항산화성 및 효소 활성도 (Antioxidative and Amylase Activity of Soybean Sprouts by Treatment of Green Tea Water Extract)

  • This study examined the growth and sensory characteristics of soybean sprout cultured at 25$\pm$1$^{\circ}C$ for 4 days with distilled water control and green tea extracted water (0.03% and 0.05%). The proximate composition of soybean sprout in the green-tea water was better than that of the control in ash, protein and fat, while the soybean sprout was especially higher in 0.05% green-tea water. The contents of vitamin C and $\beta$-carotene were higher in soybean sprout in green-tea water than the control. The total free amino acids composition of soybean sprout in green-tea water was better than that of the control, with the highest being obtained in soybean sprout in 0.03% green-tea water. Antioxidative activity was assayed by DPPH radical scavenging ability with spectrophotometer at 514 nm. The soybean sprout in green-tea water was higher than control. The amylase activity of the soybean sprout increased steadily during the first 4 days and that of the control was higher than soybean sprout in green-tea water. The proteinase of soybean sprout steadily increased during 4 day culture. Furthermore, the proteinase activity of soybean sprout in green-tea water was higher than that of the control up to 2 days. Whereas after 3 days, it was the highest in 0.03% green-tea water and then decreased remarkably in 0.05% green-tea water.

  • Ginseng leaf/stem extract produced by subcritical water extraction at high temperature ($190^{\circ}C$) posses higher cytotoxic activity against human cancer cell lines than ethanol extract. Subcritical water extraction can be a great candidate for extraction of functional substance from ginseng leaves/stems.
