Effects of PEG Priming Treatment on Germination and Seedling Growth of Onion Seed(Allium cepa L.)

  • These experiments were conducted to evaluate the variability of seed germination and seedling growth with different levels of polyethylene glycol(PEG 6,000) solution in onion seed. Average germination percentage of seed primed in PEG solution with 1.00 and 0.75 MPa was higher than control, and that of seed primed in 1.50MPa was lower than unprimed control. Germination percentage(GP) of seed primed for 5 days was highest, and as the primed days become long, the GP was decreased. The GP of airation seed during the primed was higher than that of unairation seed, about 5% , respectively. The GP of washed seed after primed was higher than that of unwashed seed, but that of redried seed after primed was lower than that of the others. The highest GP cultivar was Chunjoogoohyung and the lowest GP cultivar was Seouldego in unredried seed after primed, but Chunjoojoonggo was highest and Jungpoonwhang was the lowest cultivar in redried seed after primed. As the PEG concentration increased, the seedling length(SL) was shortened, and seed primed for 15 days was longer than other treatments. The SL of primed seed was similar to GP. The SL of washed seed after primed was longer than that of others, but that of redried seed after primed was shortest among the others. The SL of Chunjoojoonggo and Nongwoodego was longest and Seouldego was shortest among the cultivars in unredried seed after primed, but that of Chunjoogoohyung and Chunjoojoonggo was longest and Seouldego was the shortest cultivars in redried seed after primed.

Antitumor Activity of Crude Sesaminol in Sesame Seed

  • Sesaminol in sesame seed was postulated to have antitumor activity. The present study was performed to characterize the role of crude sesaminol extracted from sesame seed (Sesame Crude Sesaminol; SCS) on inhibiting the in vitro growth of human leukemia HL-60 cells. SCS inhibited the growth of human leukemia HL 60 cells in culture and macromolecular synthesis in a dose and time dependent manner. The cytostatic range of SCS concentration was found to be 60 to 100 $\mu\textrm{g}$/ml. SCS concentration greater than 200 $\mu\textrm{g}$/mlwere cytocidal to HL-60 cells. When SCS concentraction was 6 $\mu\textrm{g}$/mland 50 $\mu\textrm{g}$/ml the synthesis of HL-60 cells was inhibited by 35% for DNA, 6% for RNA and 5% for protein and 83% for DNA, 76% for RNA and 60% for protein. Of specific interest was the irreversible effect of SCS in inhibiting DNA synthesis of HL-60 cells. This was evidenced from the fact that, even after washed with PBS three times, preincubated HL-60 cells still showed the inhibited DNA synthesis.

Enhancement of seed germination and microbial disinfection on ginseng by cold plasma treatment

  • Background: This study aimed to investigate the effect of cold plasma treatment on the improvement of seed germination and surface sterilization of ginseng seeds. Methods: Dehisced ginseng (Panax ginseng) seeds were exposed to dielectric barrier discharge (DBD) plasma operated in argon (Ar) or an argon/oxygen mixture (Ar/O2), and the resulting germination and surface sterilization were compared with those of an untreated control group. Bacterial and fungal detection assays were performed for plasma-treated ginseng seeds after serial dilution of surface-washed suspensions. The microbial colonies (fungi and bacteria) were classified according to their phenotypical morphologies and identified by molecular analysis. Furthermore, the effect of cold plasma treatment on the in vitro antifungal activity and suppression of Cylindrocarpon destructans in 4-year-old ginseng root discs was investigated. Results: Seeds treated with plasma in Ar or Ar/O2 exhibited a higher germination rate (%) compared with the untreated controls. Furthermore, the plasma treatment exhibited bactericidal and fungicidal effects on the seed surface, and the latter effect was stronger than the former. In addition, plasma treatment exhibited in vitro antifungal activity against C. destructans and reduced the disease severity (%) of root rot in 4-year-old ginseng root discs. The results demonstrate the stimulatory effect of plasma treatment on seed germination, surface sterilization, and root rot disease suppression in ginseng. Conclusion: The results of this study indicate that the cold plasma treatment can suppress the microbial community on the seed surface root rot in ginseng.

Antifertility Effect of Neem (Azadrichta indica) Seed Kernel Meal in Chickens

  • The reproductive performance of forty two male broilers divided into three similar groups and fed on isocaloric and isonitrogneous diets containing 0, 10 or 20% water washed neem seed kernel meal (WWNSKM) was investigated from 20 to 32-wks of age. Results on semen characteristics revealed that feeding of WWNSKM led to significant (p < 0.05) reduction in semen volume, sperm concentration associated with increased incidences of morphological abnormalities in the spermatozoa when compared to that of the control birds. A drastic reduction in the fertilizing ability of spermatozoa was observed, the adverse effects being more at higher inclusion level of the cake. Hatchability of eggs also declined in the WWNSKM fed group. Histological examination of testes revealed a higher number of degenerating cells and poor spermatogenesis along with multinucleated giant cells in the seminiferous tubules of the testes of birds receiving the high dose of WWNSKM in diet. It may be concluded that the feeding of WWNSKM by incorporating in isocaloric and isonitrogneous diets to cockerels is associated with adverse effect on their fertility.

절임 배추의 세척 조건에 따른 김치의 숙성중 품질 변화 (Effect of Washing Conditions in Salted Chinese Cabbage on the Quality of Kimchi)

  • 김치의 저장성을 연장하기 위한 시도의 하나로 1,000 ppm 농도의 자몽종자추출물(grapefruit seed extrtact, GFSE)과 구연산 용액에 절임 배추를 각각 씻어서 김치를 담근 다음 숙성시키면서 품질 특성의 변화를 측정하였다. GFSE 및 구연산에 세척한 배추로 담근 김치는 대조구에 비하여 pH 저하 및 적정산도의 증가가 늦어지는 것으로 나타나 숙성 기간의 연장 효과가 있는 것으로 나타났다. 또한 환원장은 GFSE로 처리한 시험구가 숙성 20일까지 가장 높은 값을 나타내어 발효가 억제되는 것으로 나타났다. GFSE를 처리한 시험구는 숙성 10일까지 대조구에 비하여 총균수 및 유산균수가 낮은 것으로 나타났으나 구연산 처리구는 대조구와 큰 차이를 나타내지는 않았다.

식품가공폐유를 이용한 비누의 계면활성과 세척성에 관한 연구 (A Study on the Surface Activity and Detergency of the Soap Made from the Waste Oil from Food Manufacturing Proces)

  • We have gathered a fatty acid to recycle the waste oil of food manufacturing process, and then made a soap from the waste oil by alkali saponification. Effects of the washing elements such as the concentration of the soap, temperature and time were evaluated to find out the optimum washing conditions, and results are as follows. 1. We could find soaps made from the soybean oil (A), corn oil (B), rape seed salad oil (C), cotton seed oil (D) and a soap consisting of the each oil 25% respectively (I) had the lowest surface tension at the concentration of 0.225% -0.25%. Compared with the single fatty-acid soap, the multi-component soap I showed the lower surface tension at the cmc. 2. All the samples of A-I showed the lowest contact angle for the solid material at the concentration of 0.25%. The multi-component soap of I showed higher contact angle than the single.component soaps of A, B, C and D. 3. The soap G, made from the waste oil, show lower surface tension than 5, made from the complex raw fats of the eatable fatty oil acid and H, commercial soap. 4. The washing efficiency depends on the washing time. Especially the 25-minute was found to be the optimum washing time. 5. The highest washing efficiency was found at the 0.25% cont. reagardless of the washing temperature. At 0.15% concentration level the washing efficiency reduces as washing tem- perature increases. At 0.3% concentration level the highest washing efficiency was found between $50^{\circ}$-$60^{\circ}$. 6. The soap made from the waste oil showed the highest washing efficiency in terms of concentration, temperature, and time. 7. The soap made from the waste oil showed the similiar concentration of hydrogen ion to the synthetic detergent. 8. The hand value of the fabric washed by the soap made from the waste oil was a little lower value than those washed by the synthetic detergent.

The effect of osmotic potentials on the temperature response for germination of pepper seed

  • Primed seeds germinate faster and uniformly, since these have already started imbibition and metabolism requiring to complete germination. Several factors such as solution composition, osmotic potential, and treatment duration affect seed priming response. Water potentials of priming solution and germination response of primed seed were investigated to clarify the effects different inorganic salt types and the duration. Pepper seeds were primed in osmotic solutions that were osmotic potential ranged -1.54 to -0.45 MPa in an aerated solution of polyethylene glycol 8000 (PEG; 17%, 22%, 27%), and inorganic salt solution of $KNO_3$, $K_2SO_4$, and $Na_2SO_4$ (100, 200, 300mM) in aerated condition. The seeds were treated at $20^{\circ}C$ for 4, 6 day (D) and stepwise duration combined concentration of 100mM (2 or 4 days) and 300mM(2 or 4days). After soaking treatment, seeds were washed with distilled water and then were dried to approximately 6% moisture content in dry chamber at $25^{\circ}C$. The germination of seed was characterized by ISAT rules at $20^{\circ}C/30^{\circ}C$ and $15^{\circ}C$. Total germination percentage (GP), mean germination time (MGT), germination uniformity (GU), germination rate (GR), and health seedling percent (HS) were calculated on the germinated seed in a 14-day period. Seed water potential (${\psi}$) was correlated with water potential of priming solution ($r^2=0.84$). The effect of seed priming on germination varies with inorganic salt and organic agents (PEG). Germination percentage (GP, 77 and 73%), GR (21.3 and $19.4 %{\cdot}day^{-1}$), and HS (58 and 52 %) was greatest and lowest MGT (4.2 and 4.8 day) when they were primed in 100mM $K_2SO_4$ (${\psi}=-0.45MPa$), and 100mM $KNO_3$ (${\psi}=-0.52MPa$) for 6 days compared to untreated control (67% GP, $19.7%{\cdot}day^{-1}\;GR$, 18% HS, and 10.8 day MGT) in $15^{\circ}C$ chamber. Water potential less then -1.2MPa of osmotic solution resulted lower GP, GR and HS and in $K_2SO_4$ and $KNO_3$. However, stepwise treatment of water potential that changed solution as 100mM for 4 D + 300mM for 2 D or 300mM for 4 D + 100mM for 2 D) were not better than single concentration alone in germination characteristics.

Effects of Provenances, Storage Temperature and Duration on Seed Germination of Bombax costatum Pellegr & Vuillet

  • Seed morphology, physiology and environmental conditions have influence on germination of any tropical seeds and their appropriate handling, processing and handling enhances seeds emergent. This study therefore investigated effects of storage durations under different temperatures on germination of Bombax costatum seeds from different provenances. Fresh 25 seeds of B. costatum in four replicates were collected from four different provenances (Aponmu, Oluwa, Ibadan and Oyo). They were surface sterilized, thoroughly washed in distilled water and stored at four different temperature regimes: 28℃, 21℃, 5℃ and -17℃. Samples were taken every 2 months for germination test for 18 months. Germination assessment was carried out daily and recorded. The experimental design was 4×4×10 factorial in 4 replicates. Data were subjected to percentages and analysis of variance (ANOVA). Results showed that there were significant differences among storage temperatures, storage durations and provenances on germination percentage. Freshly collected seeds from Aponmu and Oyo had the highest mean germination of 100% followed by seeds from Ibadan (89.3%). Seeds from Oyo provenance stored 5℃ had germination of 94.0%, 88.70% and 78.7% at 2nd, 4th and 6th month respectively. Seeds from Ibadan and Oyo stored at 28℃, 21℃ recorded 0.0% starting from 8 months of storage. Germination of B. costatum seeds from the four provenances decreased with increase in storage duration at different temperatures.

A Simple Method for the Preparation of Highly Pure Conjugated Linoleic Acid(CLA) Synthesized from Safflower Seed Oil

  • A simple and rapid method was developed to prepare a large quantity of highly pure conjugated linoleic acid (CLA) chemically-synthesized from safflower seed oil (SSO). CLA-SSO(74.9% in purity) was synthesized from fresh SSO(79.9% of linoleic acid) by alkaline isomerization at 18$0^{\circ}C$. Urea(50g) and CLA-SSO (25g) were completely dissolved in ethanol (750ml) using a water bath(5$0^{\circ}C$) and followed by refluxing for 60 min. The resultant was cooled to room temperature and stored in a cold room (4$^{\circ}C$) for 24hrs. After removing the urea adduct by filtration, the filtrate was rotoevaporated under 4$0^{\circ}C$ and the residue was dissolved in hexane (200ml). The hexane extract was washed with distilled water (100ml$\times$3) and dried over sodium sulfate anhydrous. This urea treatment procedure was repeated three times. The purity of CLA recovered from the hexane extract was 95.0%. This method can be applied to prepare a large quantity of highly pure chemically-synthesized CLA (>0.5kg/a batch) from any plant oils containing high percentages (>70%) of linoleic acid.

녹차씨 오일이 염색 및 탈색된 모발의 재손상 및 탈색 방지에 미치는 영향 (Damage Prevention Effect of Green Tea Seed Oil on Colored and Decolored Hair)

  • 염색 및 탈색된 헤어는 일상생활 중의 자외선, 열기구 사용수세 (샴푸)에 의해 재손상되며, 이의 해결을 위해 최근 피부와 두피에 안전한 각종 천연물질의 연구가 활발하다. 이 연구는 녹차씨 오일의 광퇴색 방지와 모발 트리트먼트 효과를 확인하고자 수행되었다. 그러나 녹차의 기능은 이미 많이 알려져 있으나 녹차씨 오일을 이용한 모발 트리트먼트 및 퇴색 방지의 연구는 미비하다. 염색 및 탈색된 헤어에 녹차씨 오일을 이용한 전처리 트리트먼트 효과를 알아보기 위해 인장 강도 및 신장율을 측정하고 모발표면을 SEM으로 분석하였으며 색차계를 이용한 모발 광퇴색을 비교하였다. 실험결과 모발의 인장강도와 신장율의 측정에서 녹차씨 오일 전처리 시료는 버진헤어, 염색헤어, 탈색헤어에서 모두 증가하였고 신장율은 염색헤어와 탈색헤어에서 낮아져 염색헤어와 탈색헤어에서 트리트먼트 효과가 있었다. 모발표면을 촬영한 결과 모든 군에서 전처리된 모발 큐티클의 문리가 더 선명하고 코팅효과가 확인되었다. 모발의 광퇴색 값은 열과 수세의 반복 실험에서 전처리된 각 시료 모두 $L^*$, $a^*$, $b^*$값이 감소하였고 자외선 조사 실험에서는 염색과 탈색헤어에서 $L^*$, $a^*$, $b^*$값이 각각 감소하였다. 위의 실험결과에 의해 녹차씨 오일은 염색 및 탈색모발의 재손상 방지와 열과 자외선에 의한 색상의 퇴색방지 효과를 확인하였으며 앞으로 미용 실무현장에서 고객의 피부와 두피에 적합한 천연화장품 오일로 사용이 가능하리라 사료된다.
