Effects of Vitamin E and Selenium on the Antioxidative Defense System in Streptozotocin-induced Diabetic Rats (Vitamin E와 Selenium이 Streptozotocin 유발 당뇨쥐의 항산화계에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E and selenium on the antioxidative defense mechanism in the liver of streptozotocin(STZ)-induced diabetic rats. Sprague-Dawley male rats(120$\pm$10gm) were randomly assigned to one control and five STZ-diabetic groups. Diabetic groups were classified to STZ-0E (vitamin E free diet), STZ-40E(40mg vitamin E/kg of diet), STZ-400E(400mg vitamin E/kg of diet), STZ-S(0.5ppm Se/kg of diet) and STZ-400ES(400mg vitamin E and 0.5ppm Se/kg of diet) according to the level of vitamin E and selenium supplementation. Diabetes was experimentally induced by intravenous adminstration of 55mg/kg of STZ in citrate buffer(pH 4.3) after 4-weeks feedng of six experimental diets. Animals were sacrificed at the 4th day of diabetic states. Activities of the serum glutamic oxaloacetate transaminase(GOT) and the glutaminc pyruvate transaminase(GPT) in STZ-0E, STZ-40E and STZ-S rats were higher than those of control. Liver xanthine oxidase activities were similar to serum GOT and GPT. Liver superoxide dismutase(SOD) activities were higher in STZ-0E and STZ-40E groups by 33%, 22%, respectively than that of control. Glutathione S-transferase(GST) activities of liver were similar to GSH-Px activities. The contents of vitamin E in liver tissue were significantly lower STZ-0E, STZ-40E and STZ-S groups by 50%, 36%, 45% than that of control. Reduced glutathione(GSH) contents of liver were lower STZ-0E, STZ-40E, STZ-400E, STZ-S and STZ-400ES groups by 57%, 51%, 19%, 18%, 12% than that of control. Lipid peroxide values (LPO) in liver were higher 5.6, 2.3 and 2.3 times in STZ-0E, STZ-40E and STZ-S group than that of control. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stress, leading to the acceleration of lipid peroxidation process, which can be more accelerated by feeding the low level of dietary vitamin E. In the coincident supplementation of high dietary vitamin E and selenium antioxidative enzymes activities and physiolosical antioxidants were increased more than those of the separate supplementation of vitamin E or selenium. Therefore, dietary vitamin E and selenium reduced peroxidative damage of tissue, promoting antioxidative defense mechanism against lipid peroxidation by diabetes.

Vitamin A Intake of Exclusively Breast-Fed Infants in Cheongju and Anseong areas (청주ㆍ안성지역 모유영양아의 수유기간별 비타민 A 섭취량)

  • This study was longitudinally conducted to evaluate vitamin A intake of exclusively breast-fed infants compared with the Recommended Dietary Allowances (RDA) for Korean infants. Twenty-seven Korean lactating women and their infants during the first 6 months of lactation in Cheongju and Anseong areas were participated. Retinol and $\beta$-carotene contents in the milk were determined using HPLC and also the milk consumption of the infants was measured by the test-weighing methods. Vitamin A (retinol and $\beta$-carotene) contents of the milk were 65.7, 57.2, 48.1, 43.9, 38.2, 38.7 and 44.0 R.E./100 ml, and vitamin A intakes of the breast-fed infants were 361, 402, 348, 331, 304, 305 and 322 R.E./day at 0.5, 1, 2, 3, 4, 5 and 6 month of lactation, respectively. The average intake of vitamin A was 339.1 R.E./day and the percentage to RDA was 96.9% during 6 months. Vitamin A intakes per body weight of the breast-fed infants were 96.7, 88.3, 62.1, 50.0, 41.8, 39.3 and 39.6 R.E./kg/day at 0.5, 1, 2, 3, 4, 5 and 6 month. The body weight increased normally from 3.4 $\pm$ 0.5 kg at birth during lactation. It is suggested that the breast-fed infants in Cheongju and Anseong areas consumed adequately vitamin A from the milk compared with RDA for Korean infants.

Effects of Vitamin E supplement and Perilla oil on the Cytochrome P-450 contents and Fatty acid composition in Rat Hepatocarcinogenesis

  • The effects of vitamin E supplement on 15%(w/w diet) perilla or corn oils were studied in rat hepatocellular chemical carcinogenesis induced by modified Solt & Farber model, which consists of 20mg/kg body weight diethylintrosamine(DEN) injection, 3 weeks feeding of 0.02%2-acetylaminofluorene(2-AAF) and partial hepatectomy. The area of placental glutathione S-transferase(GST-P) positive foci tended to be smaller in perilla oil group had lower thiobarbituric acid reactive substances(TBARS) CONTENT. Fatty acid compositions in microsomal membrane were reflected by dietary fatty acid compositions, and not affected by carcinogen treatment or vitamin E supplement. By vitamin E supplement, linolenic acid contents of perilla oil group were much increased. By carcinogen treatment, membrane stability decreased significantly in corn oil, but maintained in perilla oil groups Vitamin E supplemental effect was noticed only in the corn-carcinogen group. Perilla oil may prevent hepatocarcinogenesis by maintaining membrane stability and by reducing cytochrome P-450 content. Vitamin E supplement did not seem to have the effect on hepatocarcinogenesis, but prevented lipid peroxidation, reduced cytochrome P-450 content and maintained membrane stability.

  • In this study, lipid composition of milk obtained in colostrum from 22 mothers delivering preterm infants and 17 mothers delivering term infants was analyzed. Fatty acid composition and vitamin E content were analyzed as well as the concentrations of total lipids in preterm and fullterm milk. Lipid and cholestol concentrations were higher in the fulltrm milk than preterm milk. PUFA and P/S ratio in preterm milk were higher than those in fullterm milk. On the other hand, SFA and MUFA contents were higher in the fullterm milk. Vitamin E which is related to PUFA concentration in colostrum was also higher in the preterm milk than fullterm milk.

  • This experiment was conducted to study the effect of dietary vitamin E on the lipid composi-tion of rats treated with carbon tetrachloride. Rats were divided into five groups I. e. C(soybean oil with vitamin E(40mg/kg diet)) EF(CCl4 with vitamin E deficient diet) 40E(CCl4 with vitamin E(40mg/kg diet) 400E(CCl4 with vitamin E(400mg/kg diet) 800E(CCl4 with vitamin E(800mg/kg diet) Body weight gain and food efficient ratio were not observed significant differences. The liver weight was significantly increased in the CCl4 treated groups but the liver weight of 800E group was significantly lower than that of EF group. In EF and 40E groups kidney weights were significantly higher compared to that of C group. The content of total lipid in liver of 40E and 400E groups were significantly higher than that of control group but in 400E and 800E groups the contents were significantly lower compared with that of EF group. The content of triglyceride in liver was significantly higher in the CCl4 treated group and the content of cholesterol was significantly higher in the EF and 40E groups but those of 800E group were significantly lower compared with those of EF group. In CCl4 administration groups were significantly higher than those of control group and 400E and 800E groups were significantly lower than those of EF group.

Effect of Dietary Vitamin A and E Peroxidative Deterioration of Heart in Adriamycin-Treated Rats (식이중의 Vitamin A와 E가 Adriamycin을 투여한 흰쥐의 심근손상의 원인인 과산화작용에 미치는 영향)

  • The present study was designed to evaluate the protective effects of dietary vitamin A or E. and of combination of vitamins A and E on peroxidative deterioration of heart in adriamycin-treated rats. Male Sprague-Dawley rats were assigned to 5 groups according to the dietary supplementation of vitamin A or E Except control rats a dose of 2mg ADR/kg of B. W was injected to these animals intraperitoneally on the same day every week. Adriamycin treatment significantly decreased the weight gain of experimental rats compared with that of control rats, But this decrement was not modified by dietary supplementation of vitamin A or E. Lipid peroxide values of plasma were elevated by ADR treatment. The combined use of ADR and dietary vitamin A or E significantly reduced these values, The interaction between vitamins A and E seemed to be present in the lipid peroxide value of plasma. Catalase and superoxide dismutase(SOD) activities in rat heart were decrased by ADR treatment but glutathione peroxidase(GSH-Px) activity was elevated. Dietary supplmen-tation of vitamin A or E enhanced the heart catalase and SOD activities. except only vitamin A-supplemented group. GSH-Px activity of rat heart tended to be decreased by dietary supple-mentation of vitamin A or E. With ADR treatment polyunsaturated fatty acids such as archido-nic acid(20:4) and docosahexaenoic acid(22:6) were decreased in rat heart. However dietary supplementation of vitamins A and E reduced this decrease. The retinol and tocopherol contents of rat plasma were decreased by ADR treatment. Dietary vitamin A or E influence vitamin A or E content of plasma. The interaction between dietary vitamins A and E was observed in vitamin A or E level of rat plasma.

Nutrient Analyses of Sustainable Ready-to-Use-Supplemental Food (RUSF) developed with East African Ethnic Plant Resources (동아프리카 자원식물을 활용하여 개발된 지속가능한 영양보충식품의 영양성분 분석)

  • The objectives of the study were to analyze the various nutrient contents of ready-to-use-supplemental-food (RUSF) developed with Eastern African ethnic plants and compare them with commercially available supplemental food (CASF) for infants. RUSF was made with three African plants, including Ficus sur, Amaranthus caudatus, and Prosopis juliflorafa and locally available food ingredients in DR Congo and Ethiopia and was cooked at $180^{\circ}C$ for 20 min in a conventional oven. Macronutrients were analyzed by the AOAC method. Contents of 17 amino acids were analysed with amino acid analyzer, and vitamin $B_1%$ niacin, vitamin $B_6$, and vitamin C were analyzed with HPLC. Isomers of vitamin E were analyzed by GC-FID. The price of RUSF was 0.133 USD per 100g. Contents of macronutrients were not significantly different between developed RUSF and CASF. Results of amino acid analysis showed that most amino acid contents were similar or higher in RUSF compared with CASF. However, glutamic acid and lysine contents were less in RUSF than in CASF. Vitamin E and vitamin $B_1$ contents were more than five times higher in RUSF (22.40, 6.26 mg/100 g, respectively) than in CASF (4.00, 0.80 mg/100 g, respectively). Niacin was about three times higher while vitamin $B_6$ was almost 20 times higher in RUSF than in CASF. However, ascorbic acid was significantly lower in RUSF (3.43 mg/100 g) than in CASF (42.08 mg/100 g). Our study found that the developed localized RUSF showed higher nutrient contents in most micronutrients analyzed except for ascorbic acid. Out of 17 amino acids, glutamic acid and lysine contents in RUSF were less than in CASF. Therefore, further study is needed in order to improve these nutrients contents.

A Study on the Influence of Sulphur-compound to Vitamin C in the different Vegetable during Cooking (유황화합물질이 조리시 야채류중의 Vitamin C 함량에 미치는 영향에 대하여)

  • In order to study the influence of sulphur-compound on the vitamin C contents of different vegetables-i.e. Korean green pepper, leaf, Bilum, leaf, Soybean sprouts, Mung bean sprouts, Cucumber-the garlic powder and minced garlic were used while the vegetables were cooked. The results garlic juice during scaling the vegetables the vitamin C contents of them were higher as much as 9.2~12.7% 2. When the scaled vegetables were seasoned the vitamin C contents were higher as much as 8.9~`17.5% by adding garlic powder and 7.3~14.9% by adding minced garlic. 3. Vitamin C loss of the cooked vegetables with room condition for 30 min, was higher as much as 15.2~34.5% compared to just cooking. 4. Vitamin C contents of vegetables after 30 min from cooking, compared to just cooking were higher of vegetables after 30 min from cooking, compared to just cooking were higher as much as shown below. 1) ~13.3% by adding garlic powder. 2) 4.0~9.3% by adding minced garlic.

Effects of Heated Oil on Lipid Peroxidation in Rat Liver (가열유가 흰쥐 간장폐의 지질과산화에 미치는 영향)

  • To study effects of heated oil on lipid peroxidation in rat liver, rats were fed 3 and 6 weeks by intubating oils heated for l1(HA group) and 24 hours (HB group) at 18$0^{\circ}C$. The contents of lipid peroxides and vitamin E, and the activities of super oxide dismutase and glutathione peroxidase in liver were measured. Histological changes of the liver tissue were observed. In both HA and HE groups, the contents of lipid peroxides and the activities of superoxide dismutase were increased, but the activities of glutathione peroxidase and the contents of vitamin E in liver were decreased when compared to the control group which was fed fresh cora oils. During the oil feeding period, the activities of superoxide dismutase and the contents of vitamin E were not significantly changed, but the activities of glutathione peroxidase were decreased, and lipid peroxides were increased in the 3 weeks than in 6 weeks. In HB liver, the heterochromatin of nucleus increased, mitochondria swellen, cristae in mitochondria disappeared, fat droplet and secondary lysosome increased and lumen of rough endoplasmic reticulum enlarged, compared with that of the control group. These phenomena in HA group were less pronounced.

EFFECTS OF VITAMIN E AND SELENIUM SUPPLEMENTATION TO DIETS CONTAINING AFLATOXIN B1 ON THE CONTENTS OF LIVER LIPIDS AND VARIOUS BLOOD PARAMETERS IN RATS

  • Ninety Wistar male rats were used to study the effects of vitamin E and Se supplementation to diets containing aflatoxin $B_1$ on the contents of liver lipids and various blood parameters. Two levels of dietary aflatoxin (0 and 1 ppm), 3 levels of vitamin E (30, 60 and 120 IU/kg), and 3 levels of Se (0.1, 1 and 2 ppm) were used to design a $2{\times}3{\times}3$ factorial experiment. Rats, weighing about 200 g, were randomly allotted to 18 cages, 5 rats per cage. The aflatoxin significantly (p < .05) decreased growth rate, feed intake and feed efficiency. Aflatoxin increased the glucose level and decreased the cholesterol level in blood significantly. Levels of blood triglyceride, total protein, and albumin were not affected by aflatoxin, vitamin E or Se. Activities of blood alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were significantly increased by aflatoxin; however, the glutathione peroxidase (GSH-Px) activity in the blood was decreased by aflatoxin even in the presence of Se. The vitamin E supplementation decreased the AST activity significantly, while GSH-Px activity increased significantly as the levels of dietary Se increased. The levels of total cholesterol and free cholesterol in the liver were significantly lower in rats receiving aflatoxin, while the extra vitamin E supplementation increased these hepatic cholesterol levels. It was concluded that the extra dietary vitamin E or Se supplementation might partially alleviate some of the harmful effects of aflatoxin in rats.
