Role of certain nutritional supplements and biological regulators in the epilepsy

  • Certain dietary contents, biological supplements might influence the occurrence or treatment of epilepsy. Some studies have found that the supplementation with individual nutrients reduced seizure frequency or improved other aspects of health in patients with epilepsy. Potentially beneficial dietary interventions include treating blood glucose dysregulations. Identifying and avoiding allergenic foods, and avoiding suspected triggering agents such as alcohol, aspartame, and monosodium glutamate. The Atkins diet (very low in carbohydrates) is a less restrictive type diet that may be effective in some cases. Nutrients that may reduce seizure frequency include vitamin B6, magnesium, vitamin E, manganese, taurine, dimethylglycine, and omega-3 fatty acids. Administration of thiamine may improve cognitive function in patients with epilepsy. Supplementation with folic acid, vitamin B6, biotin, vitamin D, and L-carnitine may be needed to prevent or treat deficiencies resulting from the use of anticonvulsant drugs. Vitamin K1 has been recommended near the end of pregnancy for women taking anticonvulsants. Melatonin may reduce seizure frequency in some cases, and progesterone may be useful for women with cyclic exacerbations of seizures. In most cases, nutritional therapy is not a substitute for anticonvulsant medications. However, in selected cases, depending on the effectiveness of the interventions, dosage reductions or discontinuation of medications may be possible. However, nutrient supplementation may be necessary to prevent or reverse the effects of certain deficiencies that frequently result from the use of antiepileptic drugs.

Whitening Effect of 3-O-Cetyl-L-Ascorbic Acid (3-O-Cetyl-L-Ascorbic Acid의 미백 개선 효과)

  • Deficiency of collagen regeneration, denaturalization of fibers, increased melanin synthesis and reactive oxygen species are important factors inducing deterioration of healthy skin function. They induce freckles and decrease in skin elasticity. Vitamin C, vitamin A and their derivatives have been used as cosmetic ingredients for improvement of these problems but they have various problems. We have been developing the various derivatives of these ingredients. In this study, we investigated whitening effect of 3-o-cetyl-L-ascorbic acid (VCCE), a new vitamin C derivative. The VCCE inhibited melanogenesis in a dose dependent manner(44 % at $20\;{\mu}g/mL $) and tyrosinase expression. For 8 weeks, we also investigated skin brightening effect of VCCE on pigmented spots in UV-irradiated human skin. In results, VCCE showed a statistically significant skin whitening effect by mechanical and visual evaluation. Taken together, our findings suggest that the VCCE has potential benefits as an active ingredient for whitening cosmetics.

Synthesis and Biological Evaluation of Phosphonate Analogues of 1 $\alpha$, 25-Dihydroxyvitamin $D_3$

  • A new series of phosphonate side chain analogues of 1$\alpha$,25-dihydroxyvitamin $D_3$ (1) have been synthesized. Antiproliferative activities of theses analogues (8a,b and 9a,b) using human keratinocyte cell shows that analogues which have natural A-ring show higher activity than unnatural A-ring series and almost equally active to 1 $\alpha$,25-Dihydroxyvitamin $D_3$(1) at 1 $\mu$M level.

Influence of UV Irradiation Procedures on the Concentration of Vitamin $D_{3}$ and 25-Hydroxyvitamin $D_{3}$ in the Liver and Skeleton of Broiler Chicks (자외선 조사방법이 육계 병아리의 간장과 골격중 Vitamin $D_{3}$ 및 25-Hydroxyvitamin $D_{3}$ 농도에 미치는 영향)

  • This study was carried out to elucidate the time course variation of vitamin $D_{3}$ (V$D_{3}$) and 25-hydroxyvitamin $D_{3}$ [25(OH)$D_{3}$] contents in the liver and skeleton of 3-wk old broiler chicks when treated with different UV irradiation procedure. Day-old Hubbard chicks were fed vitamin D deficient diet for 3 wk and exposed to medium wave ultraviolet(UVB) light with different irradiation procedures. Procedure I was 30 min continuous irradiation(O.204 mJ /$cm^{2}$) and Procedure II was three seperate 10 min irradiation with 12 h intervals, and Procedure III was three seperate 10 min irradiation with 24 h intervals. The liver and skeleton samples were collected at 10 different times between 0000~2400 h after the last irradiation. The V$D_{3}$ and 25(OH)$D_{3}$ fractions wereseparated by Sep-Pak silica cartridge and the concentrations were determined by normal phase HPLC. The mean content of V$D_{3}$ in the liver of the birds treated by Procedure II was 6.68 ng /g, which was higher than 5.60 and 5.30 ng /g from Procedure I and Ill, respectively(P<.O5). With regard to the effect of elapsed time after UVB irradiation on the V$D_{3}$ concentration of the liver, 96 h treatment showed the highest value(13.08 ng/g)(P<.05). There was a significant(P$D_{3}$ were significantly(P$D_{3}$ in the skeleton of tibia and femur, there were no significant differences among Procedure I, II and III, but significant differences were found among those from various elapsed time after irradiation, The highest value was shown at 96 h(O.99 ng /g) treatment, and interaction between irradiation procedure and elapsed time was not significant. With regard to the mean content of 25(OH)$D_{3}$ in bone, the Procedure II(18.79 ng /g) and III(17.73 ng /g) showed higher values than Procedure I did (P<.05), and the 12 h elapsed time showed the highest value(31.17 ng /g) among 10 treatments (P<.05), however, there was no significant interaction between exposing procedure and elapsed time. In conclusion, the Procedure II would he more desirable than Procedure I or III to produce more V$D_{3}$ and 25(OH)$D_{3}$ in the liver and skeleton of birds by exposing to the UVB light. Also, it was verified that 25(OH)$D_{3}$ increases more quickly than V$D_{3}$ in both tissues tested and is utilized more quickly to recover from the rickets of chicks.

Effect of Rudbeckia laciniata Extract on Physiological Activity of HaCaT Cells (삼잎국화 추출물의 피부세포 생리활성 효과)

  • The objective of the present investigation was to obtain vitamin, mineral, flavonoid, and polyphenol profiles of Rudbeckia laciniata (RL), and to examine the effects of extract of RL (RLE) on various physiological activities of HaCaT keratinocyte for the utilization of RL as natural raw materials to develop functional food. To accomplish this purpose, we checked the contents of the general nutrients of RL. The contents of vitamin A, vitamin $B_1$ and vitamin $B_2$ were $7.49{\mu}g/g$, $51.96{\mu}g/g$, and $132{\mu}g/g$ respectively, while vitamin C and vitamin $D_3$ were not detected. The contents of mineral such as Ca, K and Fe were 2.01 mg/g, 6.06 mg/g and 0.03 mg/g respectively. Total flavonoid contents of RLE were 0.25 mg/g, and total polyphenol were estimated as 1.43 mg/g. Because RL contains high levels of vitamin A which is associated with skin aging, we investigated the effect of RLE on physiological function of keratinocytes with respect to skin aging. We found that RLE significantly increased the growth rate of HaCaT cells and reduced ultraviolet radiation B (UVB)-induced cellular toxicity. Also, the extract of Rudbeckia laciniata attenuated the UVB-induced reactive oxygen species (ROS) generation in a dose-dependent manner in HaCaT cells. In addition, treatment with the extract dose-dependently increased migration activity of HaCaT cells. Thus, these findings indicated that RLE could regulate the physiological activity of keratinocytes, and may be used to develop functional foods.

Effect of UV-B Irradiation on the content of vitamin D$_2$, color and flavor pattern in Lentinus edodes (자외선 B파 조사가 표고버섯의 비타민 D$_2$ 함량, 색도 및 향기패턴에 미치는 영향)

  • Lentinus ededes were irradiated with Utraviolet-B(UV-B) at 0, 2.5, 5.0 J/$\textrm{cm}^2$ levels while culturing in sawdust medium, and the changes in vitamin D$_2$ content, color and flavor of the mushroom were analysed by high-performance liquid chromatography (HPLC), Hunter-Lab Chromatometer and Gas Chromatography-Surface Acoustic Wave (GC-SAW) electric nose, respectively. Irradiation of 0, 2.5, 5.0 J/$\textrm{cm}^2$ doses of UV-B increased the content of vitamin D$_2$ in the mushroom significantly, which was 157∼206% higher than the control group. There was a significant difference in L value between the control group and UV-B irradiated (2.5 and 5.0 J/$\textrm{cm}^2$) groups. The changes of flavor pattern were detected by GC-SAW electric nose. But the color and flavor changes were not detrimental to the quality of the mushroom. The results suggested that UV-B irradiation is a good way to increase the vitamin D$_2$ content of Lentinus edodes during cultivation.

EFFECT OF CALCIUM AND VITAMIN D SUPPLEMENTATION ON BONE FORMATION AROUND TITANIUM IMPLANT IN OSTEOPOROSIS-INDUCED RATS (골다공증 유도 흰쥐에서 칼슘과 비타민 D 섭취가 티타늄 임플랜트 주위의 골 형성에 미치는 영향)

  • Purpose: The purpose of this study was to observe the effect of calcium and vitamin D to the titanium implant osseointegration in the osteoporosisinduced animal model. Material and method: Thirty-two rats, 10 weeks of age, were divided into two groups: experimental group was ingested additional calcium and vitamin D, and a control group was not. Titanium screw implant(diameter, 2.0 mm; length, 3.5 mm; pitch-height 0.4 mm) were placed into tibia of 32 rats, 16 in the control group and 16 in the experimental group. The rats were sacrificed at 1, 2, 4 and 8 weeks after implantation for histopathologic examination, histomorphometric analysis and immunohistochemistry with fibronectin and collagen type I antibody. Result: In histopathological findings, newly formed bone was seen at 2 weeks and became lamellar bone at 4 weeks, and mature trabecullar bone was seen at 8 weeks in experimental group. In control group, thickness of regenerated bone increased till 4 weeks gradually and trabecullar bone was seen at 8 weeks. In histomorphometric analysis, marrow bone density increased significantly in experimental group compared to control group. Fibronectin immunoreactivity was strong at 2 weeks in experimental group and reduced after 4 weeks gradually. But it was maintained continuously from 2 to 8 weeks in control group. Collagen type I immunoreactivity was very strong from 2 to 4 week in experimental group. And the amount of Collagen type I expression was more abundant in experimental group. Conclusion: The results of this study suggest that calcium and vitamin D supplementation promote bone healing around titanium implants in osteoporosis induced animals.

Effects of Feeding Methods (Water vs. Feed) of Vitamin Con Growth Performance and Carcass Characteristics in Broiler Chickens

  • This experiment was conducted to compare the effects of vitamin C supplemented in either feed or water on the performance and carcass characteristics of broilers during the hot season. For a 6 week feeding trial, a total of 330 broiler chicks (Ross, 4 d old, average 57 g BW) were alloted to five treatments. The treatments of vitamin C (VC) supplementation were 1) 0 ppm VC, 2) 10 ppm VC in feed, 3) 20 ppm VC in feed, 4) 5 ppm VC in water and 5) 10 ppm VC in water. During the starter phase (0-3 week), chicks on non-supplemented group grew slower (p<0.01) than the supplemented ones, and a similar trend was also noted during finisher stage and the overall stage. Feed intake was significantly (p<0.05) higher in supplemented groups and higher when fed in feed as compared with water during all stages. But feed conversion efficiency was significantly improved in non-supplemented groups compared to supplemented ones in finisher and overall stage. The digestibility of gross energy and ether extract was significantly (p<0.05) higher during starter phase in supplemented, given in feed, and at higher levels as compared with non-supplemented, given in water, and at lower levels, respectively. The bone resistance was significantly (p<0.05) higher in supplemented, supplied in feed groups as compared with their counterparts. Except breast meat, the dressing percentage and abdominal fat were also higher in supplemented group and the dressing percentage was significantly (p<0.05) higher in VC supplemented in feed as to water, but no effect of supplementation was noticed on meat color when compared between the methods of feeding (feed vs. water). The levels of VC in plasma and liver increased linearly, as the level of supplementation both in feed and water increased and it was significantly (p<0.05) higher in feed group as compared with water group. It can be concluded that, retention and availability of vitamin C in feed was higher than those in water, and supplementation of VC during summer was beneficial for poultry.

Study of 25-Hydroxyvitamin D3 concentration according to the Hepatitis B virus (Hepatitis B virus에 따른 25-Hydroxyvitamin D3 농도 영향 연구)

  • The vitamin gets through food and the sun. But there is case that density alters by certain factor in vitamin. So this research analyzed effect of the liver function factor and change of $25OH-VIT.D_3$ density. Research target is a person who undergo health medical examination in the certain university hospital medical examination center from August, 2012 to December, 2012. Comparison classified to experiment group and control group. As a result, variable that influence in $25OH-VIT.D_3$ density showed by age and B type inflammation of the liver titer. Specially, B type inflammation of the liver titer displayed reverse corelation ($R^2$=0.40). Therefore, inflammation of the liver carrier must manage liver fuction test and density of $25OH-VIT.D_3$.
