Biochemical Composition of Marine Microalgae and Their Potential Antimicrobial Activity

  • This study is to investigate biochemical compositions of two species of marine microalgae, Chlorella ellipsoidea of Chlorophyta and Tetraselmis suecica of Prasinophyta, and to assess their potential antimicrobial activities. Crude protein, lipid and carbohydrate for C. ellipsoidea were $43.15\%$, $12.63\%$ and $13.09\%$, respectively, and those for T. suecica were $44.95\%$, $4.80\%$ and $24.05\%$, respectively. The major amino acids of the two micro algae were aspartic acid, glutamic acid, glycine, alanine, valine, leucine, lysine and proline, and no significant difference between the amino acid compositions of both micro algae was observed. The major sugars for both microalgae were glucose, galactose and mannose, and glucose contents showed the highest level, $58.70\%$ for C. ellipsoidea and$57.86\%$ for T. suecica. The major mineral contents of both micro algae for 100g were Ca (3,114mg in C. ellipoidea and 3,389mg in T. suecica) and followed by Na (2,881mg), K (548mg) and Mg (545mg) for C. ellipsoidea and Na (1,832 mg), Mg (1,510mg) and K (548mg) for T. suecica. In the content of ATP-related compound, hypoxanthine in C. ellipsoidea and IMP in T. suecica were absolutely dominant compounds. The highest content of fatty acid in C. ellipsoidea was 20:4, $27.15\%$ and that in T. suecica was 18:3 (w-6), $18.10\%$. In case of physiologically important polyunsaturated fatty acids like eicosapentaenoic acid (20: 5) and docosahexaenoic acid (22: 6), both microalgae possessed just trace amounts but was rich in arachidonic acid (20: 4). Vitamin content in both microalgae was significantly high in choline and inositol. In antimicrobial activity by water- and fat-soluble fraction of the micro algae, hexane extract in the fat-soluble fraction of C. elliposidea inhibited the growth of Bacillus subtilis by $96\%$ bactericidal activity and tetrachlorocarbon extract of T. suecica indicated relatively excellent antimicrobial activity $(81\%\;bactericidal\;activity)$ against Escherichia coli. Hot water extract among water-soluble fraction of both micro algae almost suppressed the growth of Staphylococcus aureus by $96\%$ bactericidal activity.

Differentially expressed serum proteins associated with calcium regulation and hypocalcemia in dairy cows

  • Objective: Hypocalcemia is an important metabolic disease of dairy cows during the transition period, although the effect of hypocalcemia on biological function in dairy cows remains unknown. Methods: In this study, proteomic, mass spectrum, bioinformatics and western blotting were employed to identify differentially expressed proteins related to serum Ca concentration. Serum samples from dairy cows were collected at three time points: 3rd days before calving (day -3), the day of calving (day 0), and 3rd days after calving (day +3). According to the Ca concentration on day 0, a total of 27 dairy cows were assigned to one of three groups (clinical, subclinical, and healthy). Samples collected on day -3 were used for discovery of differentially expressed proteins, which were separated and identified via proteomic analysis and mass spectrometry. Bioinformatics analysis was performed to determine the function of the identified proteins (gene ontology and pathway analysis). The differentially expressed proteins were verified by western blot analysis. Results: There were 57 differential spots separated and eight different proteins were identified. Vitamin D-binding protein precursor (group-specific component, GC), alpha-2-macroglobulin (A2M) protein, and apolipoprotein A-IV were related to hypocalcemia by bioinformatics analysis. Due to its specific expression (up-regulated in clinical hypocalcemia and down-regulated in subclinical hypocalcemia), A2M was selected for validation. The results were consistent with those of proteomic analysis. Conclusion: A2M was as an early detection index for distinguishing clinical and subclinical hypocalcemia. The possible pathogenesis of clinical hypocalcemia caused by GC and apolipoprotein A-IV was speculated. The down-regulated expression of GC was a probable cause of the decrease in calcium concentration.

Genetic, management, and nutritional factors affecting intramuscular fat deposition in beef cattle - A review

  • Intramuscular fat (IMF) content in skeletal muscle including the longissimus dorsi muscle (LM), also known as marbling fat, is one of the most important factors determining beef quality in several countries including Korea, Japan, Australia, and the United States. Genetics and breed, management, and nutrition affect IMF deposition. Japanese Black cattle breed has the highest IMF content in the world, and Korean cattle (also called Hanwoo) the second highest. Here, we review results of research on genetic factors (breed and sex differences and heritability) that affect IMF deposition. Cattle management factors are also important for IMF deposition. Castration of bulls increases IMF deposition in most cattle breeds. The effects of several management factors, including weaning age, castration, slaughter weight and age, and environmental conditions on IMF deposition are also reviewed. Nutritional factors, including fat metabolism, digestion and absorption of feed, glucose/starch availability, and vitamin A, D, and C levels are important for IMF deposition. Manipulating IMF deposition through developmental programming via metabolic imprinting is a recently proposed nutritional method to change potential IMF deposition during the fetal and neonatal periods in rodents and domestic animals. Application of fetal nutritional programming to increase IMF deposition of progeny in later life is reviewed. The coordination of several factors affects IMF deposition. Thus, a combination of several strategies may be needed to manipulate IMF deposition, depending on the consumer's beef preference. In particular, stage-specific feeding programs with concentrate-based diets developed by Japan and Korea are described in this article.

스타티스 ‘오션 블루’의 자가영양배양시 광도, 환기횟수 및 $\textrm{CO}_2$ 농도가 소식물체의 기내 생육에 미치는 영향 (Autotrophic Growth of Limonium spp. 'Ocean Blue' Plantlets In Vitro as Affected by PPF, NAEH and $\textrm{CO}_2$ Concentration)

  • 자가영양배양시 광도,환기횟수 및 $CO_2$농도가 스타티스 ‘Ocean Blue’ 소식물체의 생장에 미치는 영향을 조사하였다 MS 배지에서 광흔합영양해양을 통하여 증식된 스타티스 식물체에서 2~3장의 잎을 가진 줄기 절편체를 채취하여 배양용기당 4개씩 치상하였다. 절편체는 온도 25$\pm$1$^{\circ}C$. 상대습도 70~ 80%인 배양실에서 cool-white 형광램프로 명기동안 70, 150, 또는 220$\mu$mol. $m^{-2}$ . $s^{-1}$의 광을 공급하면서 41일 동안 배양하였다. MS 기본배지를 사용하였으며, sucrose와 vitamin은 첨가하지 않았다. 광도가 높아질수록 생체 중,건물중 및 엽면적이 증가하였다. $CO_2$농도를 높여준 경우 초장, 생체중, 건물중 및 엽면적이 증가하였다. 뿌리길이와 엽록소 농도는 광도나 $CO_2$의 영향을 받지 않았다. 배양용기 내의 환기횟수를 높이고, 고광도의 $CO_2$를 공급할수록,그리고 PPF를 증가시킬수록 스타티스 소식물체의 자가영양배양에서의 생육이 촉진되었다.

미생물에 의한 살어성물질의 생성 및 그 작용 (Production and Action of Microbial Piscicidal Substance)

  • 진균류인 Aspergilus flavus의 대사생성물인 aflatoxin이 발암성물질이카는 것이 알려진 이래 비병원성 미생물이라고 ale어왔던 여러가지 미생물의 독성물질 생성여부에 관한 연구가 많이 이루어져 우리 식생활에 미생물독 즉 mycotoxin에 대한 문제는 더욱 중요하게 되였다. 본 실험에서는 토양에서 분리한 방선균속 균주가 생성하는 살어성물질의 기본적인 성질은 pH 3~7사이는 안정하였으나 PH 8이상에서는 매우 불안정하였으며 열에 대해서도 비교적 불안정한 물질이었으며 어종과 어체중에 따른 독성의 비교는 체중이 증가함에 따라 생존시간이 길었지만 버들붕어와 미꾸라지가 타어종에 비해서 본 독성물질에 대한 저항성이 매우 켰다. 일반적으로 미생물독의 생물에 대한 작용$^{(24~26)}$은 hemolytic necrotizing, proteolytic, neurotoxic, cardiotoxic, hepatotoxic, nephrotoxic 등을 들 수 있으나 대부분의 독성물질은 상기 독작용 가운데 한 두 가지로서 그 주인이 된다. 본 물질의 잉어에 대한 각 조직의 succinic dehydrogenase 활성에 대한 조해작용을 조사한 결과 근육, 심장, 뇌 조직에 강한 조해현상을 나타내었다. 본 독성물질생성에 대한 금속 ion의 영향을 조사한 결과 $Cu^{2+}$, $Pb^{2+}$의 첨가가 크게 관계한다는 것을 알았으며 기타 본물질성장에 carbohydrate, amino acid, vitamin이 미치는 영향을 조사해 보고자 하며 어류이외의 타 온혈동물의 조직에 본독성물질을 작용시켜 그 결과를 알고자 한다. 한편 본 물질이 잉어의 각 조직에 대한 succinic dehydrogenase 활성에 미치는 정도 및 본 물질생성에 미치는 금속염의 영향을 볼 때 송$^{19~22}$ 등이 분이한 물질과는 차이가 있음을 알았다.소되었다. Alkaline protease는 배양 24시간에서 48시간까지는 0.1%구와 0.5%구가 대조구보다 효소력이 높았고 그 이상의 인삼엑기스가 첨가된 처리구보다 효소력이 낮았다. 그러나 48시간 이후부터는 효소력은 인삼엑기스 첨가량 순으로 감소를 나타내었다. period of May 13th-June 4th 1976. The study data was collected by the direct interview method based on the questionnaire which the investigator made out. The study result was analyzed by percentage, t - test. The findings can be summarized as follows: 1. General characteristics of nurses doing duty on puerperal ward: a. Nurses′average age was 24.8 years old. b. 84.6% had educational background of 4 years of college. c. 69.2% had a religion. d. 53.8% were married. e. 53.8% had clinical experience of 1 year -3 years. f, 61.5% did duty on puerperal ward during 1 year -3 years. g. 46.2% desired to do duty on obstetric ? gynecologic ward. 2. General characteristics of the women who were studied during their puerperal period: a. Women′s average age was 26.4 years old. b. 79.5% had educational background above high school.

Different Sources and Levels of Copper Supplementation on Performance and Nutrient Utilization of Castrated Black Bengal (Capra hircus) Kids Diet

  • Twenty eight 3-4 month old castrated Black Bengal kids (Capra hircus) were used to determine the effects of source and level of dietary copper (Cu) concentration on their performance and nutrient utilization. Cu was supplemented (0, 10, 20 and 30 mg/kg diet DM) as copper sulfate ($CuSO_4$, $5H_2O$) or copper proteinate (Cu-P). Kids were fed a basal diet containing maize (19.5%), soybean (17.0%), deoiled rice bran (56.5%), molasses (4.0%), di-calcium phosphate and salt (1.0% each) and mineral and vitamin mixture (0.5% each) supplements at 3.5% of body weight to meet NRC (1981) requirements for protein, energy, macro minerals and micro minerals, excluding Cu. The basal diet contained 5.7 mg Cu/kg, 122.5 mg Fe/kg, 110 mg Zn/kg, 0.26 mg Mo/kg and 0.32% S. $CuSO_4$ or Cu-P was added to the basal diet at the rate of 10, 20 and 30 mg/kg. Kids were housed in a well ventilated shed with facilities for individual feeding in aluminum plated metabolic cages. Blood samples were collected from the jugular vein on d 0, 30, 60 and 90 to determine hemoglobin (Hb), packed cell volume (PCV), total erythrocyte count (TEC), total leukocyte count (TLC) and serum enzymes (alkaline phosphatase, alanine transferase and aspertate transferase). A metabolism trial of 6 days duration was conducted after 90 days of experimental feeding. Statistical analysis revealed that source and level of Cu supplementation improved live weight gain (p<0.04) and average daily gain (p<0.01). No significant contribution of source and level of Cu to alter serum serum enzymes was evident. Goats fed Cu-P tended to have higher Hb, PCV and TEC than with $CuSO_4$ supplementation. Cu-P increased digestibility of ether extract (EE, p<0.02) and crude fiber (p<0.05) and showed an increasing trend (p<0.09) for digested crude protein (CP) and crude fiber (CF). Supplemental dose of Cu linearly improved (p<0.02) digestibilities of dry matter (DM), organic matter (OM), EE and nitrogen free extract (NFE). Though the absorption of nitrogen (N) was not affected (p>0.10) by both source and dose of Cu, N retention was affected (p<0.04) and there was a significant $Source{\times}Dose$ interaction (p<0.05). Final body weight (BW) was not influenced (p>0.10) by the source of Cu but increasing dose of Cu increased (p<0.04) the BW of kids. TDN intake (g/kg $W^{0.75}$) was higher (p<0.05) with the increased dose of Cu and there was a significant $Source{\times}Dose$ interaction. It was concluded that supplementation of Cu from different sources and varying dose level in a concentrate based diet may improve performance, nutrient utilization and plane of nutrition in castrated Black Bengal kids. The effects on performance and nutrient utilization are more pronounced with Cu-P than $CuSO_4$ supplementation. Higher dose of Cu showed better result than lower dose.

충남(忠南) 일부지역(一部地域)의 영양실태(營養實態) 및 임상조사(臨床調査) (청양군 운곡면을 중심으로) (A Nutritional and Clinical Survey on people in Chung Nam Area)

  • This study is based on data from the nutritional and clinical survey in Chung Nam area, namely Myl-Yang-Ri, Yoon-Gok-Myon, Chyung Yang Koon. Under the direction of two Nutrition professors and two clinical pathology professors it was carried by 35 Dae Jeun medical Junior College students majoring in nutrition and clinical pathology from 18 July to 25 July 1978. The nutrition surveys were carried out with subjects in village from a total 67 households, 36 of them were randomly selected. The clinical surveys were carried out with subjects in village from a total 382 inhabitant, 154 inhabitant were randomly selected and 109 peoples were subjected to stool examination. The results obtained in this study are summarized as follows. 1) Nutritional survey a) Food Intake The average food intake per person per day in survey area was 929.9 grams (95.5% in vegetables and 4.5% in animal foods). The average consumption of the basicfood groups per person per day was 965g for meats and legumes (10.4% of the total food in take). 268.9g for fruits and vegetables (29% of the total food intake), 559.1g for cereals and potatoes(60.1% of the total food intake), 5.2g for milk and small fishes and 0.2g for fats and oils. b) Nutrient Intake The average daily consumption of calories and nutrients was 2054.1 kcal and 61.2g for total proteins, 223.2mg for calcium, 9.5mg for iron, 4914.4 IU for vitamin A, 1.5mg for thiamin, 1.2mg for riboflavin, 19.9mg for niacin and 54.7mg for ascorbic acid. When these figures are compared with the recommended allowances for Korean, the calories and nutrients intakes were insufficient. Especially the intakes of the calcium were lower than the recommended allowance which are 500 milligrams per day. c) Kinds of food stuffs consumed The kinds of food stuffs consumed by the subjects were 47 figures total. Generally these kinds of food were vegetables food. 2) Clinical Survey a) The mean value of hemoglobin from 72 males was 13.2g/dl, that of 82 females was 12.3g/dl. b) The proportion of low hemoglobin (<12.2g/dl for male and <11.3g/dl fo female) for male was 20.8%, females was 11.0% and from 24 male & female (both sexes) were 15.6% c) The mean value of Hematocrit of males was 39.6%, that of females was 37.4%. d) The mean value of MCHC of males was 33.9%, that of females was 33.6%. e) The ABO blood group was distributed as following; 30.5%, for group O, 29.2% for group A, 24.0% for group B, and 16.3% for AB group. Biochemical race index was 1.13. f) The over all prevalence rate of heminthic infectious by rate of each helminth was as following; Ascaris lumbricoides 33.9%, Thrichocephalus trichiurus 11.9%, Hookworm 0.9%, Hymenolepis diminuta 0.9%, Trichostromgylus orientalis 0.9% and the prevalence rate of two more helminthic infection was 7.2%.

Is the Agricultural Work a Risk Factor for Koreans Elderly Spinal Sagittal Imbalance?

  • Objective : A primary degenerative sagittal imbalance has been considered because of unique lifestyles such as the prolonged crouched posture during agricultural work and performing activities of daily living on the floor. Previous papers have reported that sagittal imbalance disease is often seen distinctly in the farming districts of "oriental" countries such as Korea and Japan. However, this finding was only evaluated with the use of X-ray, and other factors such as magnetic resonance imaging (MRI), muscle volume, compression fracture, and laboratory results were not considered. Thus, using these, we evaluate the agricultural work-associated factors for Korean elderly spinal sagittal imbalance. Methods : We recruited 103 Korean participants who had a sagittal vertical axis (SVA) of >5 cm in this Korean Elderly Sagittal Imbalance Cohort Study. The following were evaluated : radiological parameters, MRI, compression fracture, vitamin D, parathyroid hormone, C-terminal telopeptide, osteocalcin, bone mineral density and muscle fatty change, muscle volume, and health-related quality of life from patients' survey. Moreover, in this survey, the farmers' annual working hours were investigated. Subsequently, we analyzed the associated factors for spinal sagittal imbalance depending on occupation. Results : A total of 46 participants were farmers, and the others were housewives, sellers, and office workers. The farmer group had more SVA (141 vs. 99 mm, p=0.001) and pelvic tilt (31° vs. 24°, p=0.004) and lesser lumbar lordosis (20° vs. 30°, p=0.009) and thoracic kyphosis (24° vs. 33°, p=0.03) than non-farmer group. A significantly positive correlation was noted between the working hour and SVA in the farmer group (p=0.014). The visual analogue scale score for back pain (8.26 vs. 6.96, p=0.008) and Oswestry Disability Index (23.5 vs. 19.1, p=0.003) in the farmer group were higher than that in the non-farmer group, but the Short Form-36 score was not significantly different between the two groups. The Mini-Mental State Exam score was significantly lower in the farmer group than in the non-farmer group (24.85 vs. 26.98, p=0.002). Conclusion : The farmer group had more sagittal imbalance and back pain in proportion to the working hours even though the muscle and bone factors and general laboratory condition were not significantly different between the two groups. These results supported that the long hours spent in the crouched posture while performing agricultural work were a risk factor for severe sagittal imbalance.

무기질 불균형: 골 탈석회화와 연조직 석회화 (Mineral Imbalance: Bone Decalcification and Soft Tissue Calcification)

  • 여러 단백질과 다양한 무기질(칼슘 등)을 함유한 단단한 기관인 골은 정상적으로 다른 연조직 세포로 무기질을 공급하는 역할을 수행하지만, 비타민 결핍, 노화, 폐경기 및 대사성 질환 등으로 인해서 골다공증이 유발되고 동시에 다양한 연조직(심장, 대동맥, 신장, 허파, 췌장 등)의 석회화가 빈번히 유도된다. 같은 중간엽 줄기세포로부터 유래되는 연조직을 구성하는 세포와 조골세포 사이에서는 상호 횡간의 분화가 될 수 있는 여지가 있어, 연조직 세포는 칼슘 축적으로 골을 형성하는 조골세포와 유사한 세포로 분화될 수 있다. 연조직을 이루는 다양한 연조직 장기의 칼슘 축적으로 인한 석회화는 치명적인 장기손상으로 생명을 위협할 수 있으므로 적극적인 예방과 치료가 중요할 것이다. 골다공증과 연조직 석회화는 상호 밀접한 연계성이 있어 한 가지의 질병이 진행되면 다른 질병이 연속적으로 발병할 우려가 있으므로 초기 질병에 대한 적극적인 치료가 필요하다. 향후 초고속고령화 사회 진입으로 이의 두 질환의 급격한 증가가 예상되므로, vitamin K와 D를 비롯한 다양한 무기질을 균형적으로 조절할 수 있는 식습관과 다량의 항산화제를 함유한 음식물 섭취로 이의 질환을 예방할 수 있을 것이다.

콩 품종별 감식초 절임 중 성분의 변화와 항산화 효과 (The Change of Compositions and Antioxidant Effect in Soybean Cultivars Pickled in Persimmon Vinegar)

  • 여러 가지 종 품종에 감식초를 처리하여 성분의 변화와 항산화 효과에 대하여 살펴본 결과는 다음과 같다. 아미노산 함량은 글루타민산이 가장 많았고, 이어서 아스파르트산, 라이신, 로이신, 알기닌 순이었다. 주요 유리당은 glucose, fructose 및 sucrose이었으며, 초절임에 의해 sucrose의 함량이 크게 감소하였고, glucose와 fructose의 함량은 증가하였다. Maltose는 초절임한 대원과 일품검정에서만 발견되었다. 품종별 원료 콩의 리놀레인산의 함량은 $49.3{\sim}57.1%$의 범위였으며 리놀레닌산 함량은 각각 $7.8{\sim}8.9%$의 범위였으며 초절임에 따른 큰 변화는 보이지 않았다. 품종별 원료 콩과 초절임한 콩의 품종별 무기질의 조성은 칼륨, 인, 마그네슘이 많았고 이어서 칼슘과 나트륨, 철분이었고, 초절임에 따른 큰 변화는 보이지 않았다. 비타민 $B_1$과 C의 함량 그리고 총 페놀 함량은 초절임 후에 증가하였다. 초절임에 의해 전자 공여능, 항산화 효과, 아질산염 소거능은 모두 증가하였다.
