Effects of Nutritional Supplementation of Cereal-Vegetable Diet on the Growth of Rats (I) - Especially on Protein Metabolism - (곡류.야채식이의 영양소 보완이 흰쥐의 성장에 미치는 영향 (I) - 단백질 대사를 중심으로 -)

  • This study was designed to find out the nutritional defect of general Korean diet and the effective way of nutritional supplementation. Seventy weanling Sprague-Dawley male rats weighing 51.8$\pm$0.9g were blocked into ten groups and fed ten different diets ad libitum for eight weeks: Standard groups(st gp) was given 72% sugar-20% casein diet: Cereal-vegetable group(c-v gp) was fed cereal-vegetable diet(c-v diet) composed of rice, barley, soybean, spinach and cabbage: the other eight groups were fed c-v diets supplemented with casein, vitamin $B_{2}$, calcium, vitamin A, vitamin B2 and A, vitamin A and calcium, vitamin $B_{2}$ and calcium, vitamin A and $B_{2}$and calcium, respectively, on the basis of each nutrients content of standard diet. The results were as follows: 1. Food intakes and body weight gains in all the experimental groups were significantly lower than st gp. Among experimental groups, casein gp and vit B2 gp tended to gain more body weights than c-v gp. 2. Through all the experimental period, F.E.R., pp.E.R., and NDPcal% did nod show significant differences among all the experimental groups and st gp. 3. The weights of liver, kidney, and gastrocnemius were significantly lower in all the experimental groups as compared with st gp. But brain and sex organ weights did not show differences among all the groups. 4. Nitrogen contents of total carcass, liver, and gastrocnemius in all the experimental groups tended to be increased as compared with st gp, and among experimental groups, they tended to be increased by casein supplementation and decreased by ca supplementation. 5. Apparent nitrogen digestibility, urinary and fecal nitrogen excretion, the amount of nitrogen retained, and N.P.U. did not show any significant differences among all the groups. 6. Serum total protein concentrations did not show any significant differences among all the groups.

Nutrients Contents in Different Parts of Pickly Pear(Opuntia humifusa) and Possible Anti-Breast Cancer Effect (천년초선인장의 부위별 영양성분 분석과 정상 및 암세포에 미치는 효과)

  • In order to examine the nutritional value of prickly pear(Opuntia humifusa), contents of ash, protein, fat, minerals and vitamins were determined on freeze-dried stem, fruit, seed and root from plants harvested in autumn. The average moisture contents for stem, fruit, seed and root were 67~87%. Crude ash content determined on dry weight basis was 2~3%. Crude protein existed mostly in seed(2.95%) and root(2.37%). Crude fat was detected mainly in seed(4.49%). Contents of major minerals(mg/100 mg dry weight) was generally higher in stem. Ca in stem(4,142.30) and fruit(2,790.86) were much higher than in seed(43.37). P in stem, seed and fruit were 448.19, 263.20 and 161.59, respectively. Stem also displayed more abundant Mg(1,110.86), Zn(35.62) and Mn(37.07). However, fruit contained higher amounts of Fe(13.38) and Se(0.15). Vitamin A was negligible in all plant parts. Vitamin E contents in fruit and stem were 1.78 mg and 1.22 mg/mg dry weight, respectively. Vitamin C was detected mostly in fruit(445.40) and stem(260.94). Use of a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide-based microtiter assay of cell viability demonstrated an anti-proliferative effect of O. humifusa extract on the MCF-7 estrogen-dependent human breast cancer cell line.

Physicochemical Properties of Dongchimi Added with Gatt (Brassica juncea)

  • To improve Dongchimi (watery radish kimchi) quality and preservation, 0, 3, 5, 10, and 15% of gatt (Brassica juncea; leaf mustard) per radish was added. Chemical characteristics were determined during fermentation at $10^{\circ}C$ for 45 days. Total acidity increased slowly by addition of gatt during initial fermentation period, and 15% treatment showed lowest total acidity. Total vitamin C content increased initially in all treatments depending on gatt content, and decreased thereafter. Period for reaching maximum value was delayed by addition of gatt. Highest total vitamin C was found in 15% treatments. In the case of reducing sugar, 5% treatment showed highest contents. Lactic, succinic, and tartaric acid contents consistently increased during fermentation, while those of malic and citric decreased. Turbidity and total solid contents of Dongchimi liquid increased in all treatments as fermentation proceeded, although the extent was rather suppressed by addition of gatt. Colorimetric lightness values decreased, while the initial increased and then decreased in redness and yellowness. Addition of gatt at above 15% weight level per radish accelerated fermentation at the later fermentation stage thus it should be avoided. Most changes in typical characteristics of fermentation were similar depending on treatments. More acceptable Dongchimi could be prepared by fermenting at 5% gatt concentration under given conditions.

The Changes of Vitamin C Content in Yulmoo Mulkimchi According to the Shift of Fermentation Temperature (발효 온도의 변화에 따른 열무 물김치 중 비타민 C의 함량 변화)

  • The vitamin C contents in Mulkimchi (watery kimchi prepared with Yulmoo, leaf radish) were analyzed during its fermentation and storage at 30$^{\circ}C$, 15$^{\circ}C$ and 4$^{\circ}C$, respectively. The total amount of vit. C in Mulkimchi fermented for 1 day at 30$^{\circ}C$ was rapidly increased to the highest level of 20 mg%. At this time, Mulkimchi showed pH 4.5 and 0.2% of total acidity expressed in lactic acid. The total microbes of the kimchi amounted to 10$\^$8/ cfu/$m\ell$ which are mostly found to be lactic acid bacteria. However, the amount of vitamin C in Mulkimchi was decreased gradually during the storage at 30$^{\circ}C$, 15$^{\circ}C$ and 4$^{\circ}C$ for 9 days. In the Mulkimchi fermentated at 15$^{\circ}C$, the maximum amount of vitamin C was 15 mg% at 4 to 5 day-fermentation. The pH of this kimchi was 4.0 and its acidity was 0.15% as lactic acid. When the storage temperature was dropped to 4$^{\circ}C$ after 1 day-fermentation at 15$^{\circ}C$, its vitamin C didn't reach the above level after 9 day storage. As the fermentation temperature was kept at 4$^{\circ}C$, the content of vitamin C in Mulkimchi remained at 10 mg% of its initial storage and didn't change through the 9 days storage.

Evaluation of Internal Quality of Kale Leaf by Non-Destructive Color Measurement (비파괴적 엽색분석을 통한 케일 잎의 내적품질 평가)

  • Kale leaf had similar contents of vitamin C, $NO_3$, p, Ca, Mg, and Fe with reported values. Among these internal quality factors, $NO_3$ content which has been a concern recently, ranged from 139 to 429 mg in 100 g fresh kale leaf, Kale showed high vitamin C content ranging from 106 to 203 mg in 100 g fresh leaf. The relative concentration of chlorophyll and b value color had a high correlation coefficient (r) with vitamin C, $NO_3$, Mg and Fe content. The relationship between $NO_3$ content and the relative concentration of chlorophyll was given by the following linear equation: $NO_3$ content : 21.55 + (5.907 ${\times}$ the relative concentration of chlorophyll) with an r of $0.910^{{\ast}{\ast}}$. Correlation between $NO_3$ content and b value was also very dependable (r = $-0.901^{{\ast}{\ast}}$). Vitamin C content and the relative concentration of chlorophyll showed a high correlation, r = $-0.858^{{\ast}{\ast}}$. Among internal quality factors, vitamin C content increased with decreasing $NO_3$ content and their correlation coefficient was high (r = $-0.795^{{\ast}{\ast}}$). Consequently, $NO_3$ content of kale leaf could be inferred from an external nondestructive method, such as the relative concentration of chlorophyll. We may be able to produce high quality kale leaves containing high amount of vitamin C and low content of $NO_3$ using this method.

  • Seakso 5, a maize hybrid, was applied for variety in 2021 by Gangwon Agricultural Research and Extension Services in Korea. This variety was bred to produce a purple extract of com husk. It shows purple color in the husk and cob and contains abundant anthocyanins. In this study, to obtain basic data on purple com 'Seakso 5' husk extract (PCHE), a husk extract was prepared and the contents of vitamins, amino acids and fatty acids were analyzed. The Seakso 5 husks were extracted with 30% alcohol and concentrated, after adding dextrin to the concentrate, it was spray-dried to prepare an extract. The PCHE were analyzed for vitamins, amino acids, and fatty acids by referring to the Food Code. As a result, three types of vitamins were detected, and their contents were vitamin B1 0.14 mg/100g, vitamin B2 2.30 mg/100g, and vitamin C 11.32 mg/100g. A total of 15 amino acids were detected in the PCHE, including aspartic acid and glutamic acid, and the content of aspartic acid was the highest at 3.02 mg/g. Simultaneous analysis of fatty acids Among 37 types, saturated fatty acids were palmitic acid (C16:0), stearic acid (C18:0), monounsaturated fatty acids were oleic acid (C18:1(n-9)), polyunsaturated fatty acids were linoleic acid(C18:2(n-6)c) and a-Linolenic Acid (C18:3(n-3)) were detected. Among the detected fatty acids, the content of palmitic acid was the highest at 40.75 mg/100g. In the future, These analysis results will be used as reference data for temporary food ingredient applications by the Ministry of Food and Drug Safety.

Effects of Temperature and Relative Humidity on Water Soluble Vitamin Contents in Commercial Vitamin Tablet (저장 온도 및 상대습도가 비타민 정제 중 수용성 비타민 함량의 변화에 미치는 영향)

  • Effects of temperature and relative humidity on contents of water-soluble vitamins (vitamins $B_1,\;B_2,\;B_3,\;B_6$, and C) of two commercial tablets ("Multivitamin Dandelion" and "Chewable vitamin C") were investigated. When stored at various temperatures (25, 35, and $45^{\circ}C$) with cap, all measured vitamins were stable and degraded very slowly during 24 weeks of storage; low relative humidity (11% RH) without cap also provided stability during storage period. At higher relative humidities (75 and 100% RH), contents of all water-soluble vitamins, except vitamins $B_2\;and\;B_3$, decreased significantly at early storage period. These results showed that stability of water-soluble vitamins is highly dependent on relative humidity rather than storage temperature.

Evaluation of Individual Glucosinolates, Phytochemical Contents, and Antioxidant Activities under Various Red to Far-Red Light Ratios in Three Brassica Sprouts (적색/원적색광 조사 비율에 따른 3종 배추과 채소 새싹의 Glucosinolate 함량 및 항산화 기능성 평가)

  • The aim of this study was to evaluate the individual glucosinolate (GSL), total phenol, total flavonoid, and vitamin C content, and antioxidant activity under various light quality condition, mainly focusing on red (R) to far-red (FR) light ratios in three Brassica sprouts (radish, Chinese cabbage, and broccoli). Three R/FR ratio of 0.6, 1.3, and 2.0 were exposed to 5-day old sprouts for 48 h in a controlled environment, and the targeted phytochemical contents and antioxidant activities were compared with three separate control plot of dark, fluorescent, and red:blue 8:2 conditions. Total GSL content was highest in broccoli among the cultivars throughout the respective treatments, and increased with the increasing of R/FR ratio in the broccoli sprouts, while the content showed non-significant results in the Chinese cabbage sprouts. The progoitrin, a major GSL in Chinese Cabbage and broccoli, content decreased by upto 38% and 69%, respectively, with decreasing the R/FR ratio compared to the control plots (fluorescent, red:blue 8:2, and dark condition). The contents of phenol, flavonoid, and vitamin C were lowest in dark condition in all the three Brassica sprouts. The total phenol content and antioxidant activities increased with decreasing the R/FR ratio in all the Brassica sprouts, while total flavonoid and vitamin C content showed different patterns depending upon the Brassica sprouts. These results suggest that additional use of FR is expected to improve the functional quality of Brassica sprouts in different ways.

A Study on the develpoment of new recipes of 5-Korean Wild Vegetables (한국산 야생식용식물의 조리과학적 연구)

  • The Contents of proximate compositions, vitamin C and minerals(Ca, P, Fe and Zn) of five Korean wild vegetables were analyzed. Twenty four cooking recipes with three of the vegetables were developed and their taste characteristics were evaluated by 12-trained panels. The five Korean wild vegetables are Cassia tora(C.t.), Lycium chinese Mill(L.c.), Trichosanthes quadricirra MIQ(T.q.), Polygonatum japonicum MORR et DECAIS(P.j.) and Articum lappa L.(A.l.). They have been known as the efficacious plants agaginst diabetes mellitus. In our previous work, their hypoglycemic effects have been studied through streptozotocin-induced diabetic rats and fed on the wild vegetables. Contents of moisture were 14~86% in the samples and L.c. showed relatively higher content of crude protein(16.4%). High level of crude fat was seen in C.t.(9.4%). Contents of vitamin C were also relatively high in the wild vegetables(14.2~42.0 mg%). Analysis of minerals revealed the high level of Ca in L.c. leaves, P in A.l. and Fe in C.t. and L.c. leaves. All of the samples contained Zn in 5.0~14.6 mg%. Twelvetrained taste panels indicated that the 24-preparations of L.c. leaves, fruites and P.j. were well accepted in taste, color and odor. Although L.c. fruit gruel, L.c. cream soup and P.j. saute were evalulated as less acceptable, their sensory scores were in 3.6~5.4 which were not undesirable. The newly developed cooking recipes of L.c. fruits and leaves showed better scores in color and texture, P.j. in texture and odor.

Classification of Korean Green Tea Products Based on Chemical Components

  • The prices of domestic green tea products are relatively expensive and price differences within products of the same levels of quality are various. Also, there is no basic criteria on evaluation of green tea quality. To group 43 commercial green tea products into several parts by the principal component and cluster analyses, this work was done by use of 8 chemical constituents which were analyzed by NIR system. The principal component and cluster analyses revealed 8 groups. The first group included 16 products that had lower free amino acid and theanine contents. The second group included 5 products having higher free amino acid and theanine contents, but lower ash contents. The third group included 13 products showing medium values of 8 constituents. The IV group included 4 products having higher contents of moisture, free amino acids, and theanine. The V group included 1 product showing higher moisture but lower catechins contents. The VI group included 2 products that had higher moisture and catechins contents, but lower free amino acid and theanine contents. The VII group had higher moisture and catechins contents. The VIII group had higher ash and vitamin C contents. The free amino acid contents which were the most important in flavor evaluation of green tea quality did highly positively correlate with the contents of total nitrogen $(0.956^{**}),\;theanine\;(0.981^{**}),\;and\;caffeine\;(0.793^{**})$, but negatively with the contents of ash $(-0.884^{**})$. The catechins used as for functional ingredients did correlate with contents of caffeine(+) and vitamin C(-), respectively.
