Biochemical Study on the Environmental Preservation of Lake -Distribution of Chemical Compounds and $Vitamin\;B_{12}$ of Okjeong Lake- (호수의 환경보전에 있어서의 생물화학적 연구 -옥정호의 영양염과 $Vitamin\;B_{12}$의 함량-)

  • Concentration of dissolved $vitamin\;B_{12}$ in the water of Okjeong lake have been determined by microbiological assay methods on the 20th of August, 1983. Also the relations between the $vitamin\;B_{12}$ and other physical, chemical and biological environmental factors were investigated. $Vitamin\;B_{12}$ was assayed with Euglena gracilis Z strain. The results obtained in this investigation were as follows : 1. Generally, the values of COD and $Cl^-$ were higher than values reported in 1981. 2. The concentration of nutrient salts in the lake water was found to be at a higher level: above the general criterion in a eutrophic lake viz., phosphorous 0.175-0.225 mg/l. 3. Chlorophyll a contents in this lake were $1.1-8.7{\mu}g/l$, 4. The concentration of $vitamin\;B_{12}$ in the water fairly high: 1.25-6.0 ng. It is apparent that concentrations of the $vitamin\;B_{12}$ as well as nutrient salts were high in the water of Okjeong lake. It was found that Okjeong lake was in eutrophic.

The risk of MTHFR variants, folate and vitamin B$_{12}$ deficiencies and hyperhomocysteinaemia during pregnancy associated with short gestational age and reduced birth weight (임산부에서의 Methylenetetrahydrofolate reductase (MTHFR) 유전자 변이, 엽산 및 비타민 B$_{12}$ 결핍과 고호모시스틴 혈증이 재태기간과 출산아의 체중에 미치는 영향)

  • The purpose of this study was to evaluate whether the MTHFR variants, folate and vitamin $B_{12}$ deficiencies increase the risk of hyperhomocysteinaemia and adverse pregnancy outcome such as short gestational age or reduced birth weight. Healthy pregnant women (n=136; 24-28 gestational weeks; 20-40 years old), who visited Ewha Womans University Hospital for prenatal care, participated in this study. At the time of delivery, trained nurses recorded the pregnancy outcome from medical chart. We determined maternal MTHFR polymorphisms (C to T subsitution at nucleotide 677) and measured serum homocyteine, vitamin $B_{12}$, and folate concentrations. We compared serum homocysteine level by MTHFR genotype, serum folate and serum vitamin B12 levels using ANOVA. To evaluate the association between serum homocysteine level and pregnancy outcome, we compared the gestational age and birth weight by serum homocysteine levels using multiple regression analysis, adjusting for other potential predictors. Mean level of serum homocysteine was highest among pregnant women of the MTHFR variants with low levels of serum folate and vitamin $B_{12}$. Regarding association with birth outcome, we found the relationship between homocysteine levels and increased gestational age (p=0.03) and reduced birth outcome (p>0.05). Our data demonstrates that serum level of folate and vitamin $B_{12}$ among pregnant women affects significantly serum homocysteine levels, and the genetic polymorphism of MTHFR modulates the relationship between them. However, we did not have conclusive evidence of association between high homocysteine level and adverse pregnancy outcome such as preterm or low birth weight.

Clinical Significances of Serum Vitamin $B_{12}$, Folate and Eerritin Levels in Patients with Malignant Tumors (악성종양에서 혈청 비타민 $B_{12}$, 엽산 및 Ferritin의 진단적 의의)

  • In order to evaluate the clinical significances of the serum vitamin $B_{12}$, folate and ferritin levels in patients with malignant tumors, the levels were measured in 10 normal contol subjects, 70 patients with malignant tumors, 7 patients with liver cirrhosis and 25 patients with other benign diseases. The results are as follows: 1) In normal control subjects, mean serum values for vitamin $B_{12}$, folate and ferritin level were $588.80{\pm}131.58pg/ml,\;5.59{\pm}1.52ng/ml\;and\;89.22{\pm}42.78ng/ml$ retrospectively 2) There was no significant difference in serum levels between patients with benign diseases and normal control subjects. 3) The serum vitamin $B_{12}$ and ferritin levels in patients with liver cirrhosis were significantly higher than in normal control, and the serum folate levels in these patients were lower than in normal control subjects. 4) The serum vitamin $B_{12}$ and ferritin levels in patients with malignant tumors were significantly higher than in normal control subjects, and the serum folate levels in these patients were significantly lower than in normal control subjects. The above results suggest that the serum vitamin $B_{12}$ and ferritin may be useful as tumor markers in patients with malignant tumors.

  • The effects of environmental conditions, initial dissolved oxygen concentrations, pH, and the presence of electron carrier vitamin B$_{12}$ , on the reduction rate of TNT by Fe$^{0}$ was Quantitatively analyzed using a batch reactor. In all experiments, TNT reduction was best described with a first order reaction and the reduction rate decreased with the increase in the initial DO concentration. However, the specific reaction rate did not decrease linearly with the increase in the initial DO concentration. In the presence of HEPES buffer 0.2 and 2.0 mM(pH 5.7$\pm$0.2), the specific reaction rate increased more than 5.8 times, which showed reduction rate is rather significantly influenced by the pH of the solution. To test the possibility of reaction rate enhancement, well-known electron carrier(or mediator), vitamin B$_{12}$ has augmented besides Fe$^{0}$ . In the presence of 8.0 $\mu\textrm{g}$/L of vitamin B$_{12}$ , the specific reaction rate increased as much as 14.6 times. The results indicate that the addition of trace amount of vitamin B$_{12}$ can be a promising rate controlling option for the removal of organics using a Fe$^{0}$ filled permeable reactive barrier.

A Study on the Composition of Seasoning Using Lentinus edodes. (표고버섯을 이용한 조미료의 주요성분에 관한 연구)

  • The major compositions of Lentinus edodes seasoning(LES) compose of 30.3% of Lentinus edodes fruit body, 18.1 % of anchovy (Engraulis japonicus) powder and 51.6% of sea tangle (Laminaria japonica), prawn and green tea powder and so on were investigated in order to evaluate the nutritional value. Sodium concentration in LES was high up to 5,042.99 mg/IOO g and K, Ca, P, Mg, Fe, Zn, Mn and Cu were followed. There were nineteen total amino acids in LES. The glutamic acid content was high up to 4.62 mg/100 mg and glycine, leucine, alanine, aspartic acid were followed. Among twenty free amino acids, glycine content was high up to 2.37 mg/100 mg and glutamic acid, asparanine, proline, taurin and aspartic acid were followed. The contents of vitamin C in LES was high up to 224.78 mg/100 g and pantothenic acid, vitamin B$_{6}$, niacin, vitamin E, folic acid, vitamin B$_{l}$ vitamin B$_{2}$, vitamin D$_{3}$, vitamin A, vitamin B$_{12}$ and vitamin $K_{l}$ were followed.wed.

Effects of Food and Vitamin B12 on the Growth of a Freshwater Rotifer (Brachionus calyciflorus) in the High Density Culture (고밀도 배양에 있어 먹이종류와 공급량 및 vitamin B12 첨가에 따른 담수산 rotifer (Brachionus calyciflorus)의 성장)

  • This study investigated the effects of food type (condensed freshwater Chlorella, dried Chlorella, dried Spirulina, dried Schizochytrium, baker's yeast and $\omega-yeast$) and amount, and supplementation of vitamin $B_{12}$ on the growth of freshwater rotifer (Brachionus calyciflorus) in high density culture. Growth of rotifers fed condensed freshwater Chlorella was the highest and its density ranged $7.65-8.14{\times}10^3\;inds./mL.$ The primary lipid acids of rotifers fed condensed freshwater Chloyella were linoleic and linolenic, and their amount ($\%$ of total fatty acids) were $48.8\%\;and\;26.8\%,$ respectively. This suggests that condensed freshwater Chlorella would be an effective diet for high quality and quantity rotifers, which in turn serve as live food for freshwater fish larvae. Growth rate of rotifers with Chlorella supplementation increased as amount of supplementation increased up to 1.5 and 2.5 mg at 28 and $32^{\circ}C$, respectively. However, undissolved ammonia toxicity and packing volume of Chlorella in culture medium, reached the optimal conditions for the stable and effective cultivation of rotifers when amount of condensed freshwater Chlorella was 1.5 mg in dry weight per 1,000 rotifers at $28^{\circ}C\;and\;32^{\circ}C$ Growth of rotifers in condensed freshwater Chlorella with vitamin $B_{12}$ supplementation was significantly higher than that of rotifers without supplementation. However, no significant difference was found among the different concentrations of vitamin $B_{12}.$ Therefore, vitamin $B_{12}$ could improve the growth of rotifers (B. calyciflorus).

  • In this work, the effect of the culture medium composition on microbial growth and ethanol production in Clostridium autoethanogenum culture was investigated to enhance the ethanol productivity. D-Ca-pantothenate, vitamin B12 (as vitamins), and sodium sulfide (as sulfur source) were selected as examined components, and the effects of components' concentration on cell growth and ethanol production was investigated. For D-Ca-pantothenate concentrations varing from 0.5, 5, 50 and 500 mg/L, a slight increase in the ethanol production was observed at the 0.5 mg/L, but negligible differences in microbial growth and ethanol production were measured for the concentration ranges examined. The effect of vitamin B12 concentrations from 0.1, 1.0, 10, and 100 mg/L on the microbial growth and ethanol production was investigated, and it was found that the ethanol production using a 0.1 mg/L of vitamin B12 concentration increased by 245% compared to that of using the basic medium concentration (10 mg/L). The effect of sodium sulfide concentrations (0.5, 5, and 10 g/L) on the microbial growth and ethanol production was also studied, and the inhibition of microbial growth was observed when the sodium sulfide usage was over 0.5 g/L. In conclusion, changes in D-Ca-pantothenate and sodium sulfide concentrations did not affect the ethanol production, whereas even a 100 times lower concentration of vitamin B12 than that of the basic medium improved the production.

The Relationship between Risk Factors for Cardiovascular Disease and Levels of Plasma Total Homocysteine, Folate and Vitamin {TEX}$B_{12}${/TEX} in Koreans

  • The elevation of total plasmahomocysteine is now an established risk factor for cardiovascular disease. Plasma folate and vitamin {TEX}$B_{12}${/TEX} influence Hcy metabolism as cofactors. In this study, we studied the relationship of major risk factors for cardovascular disease, including advanced age, male gender, obesity, hypertension, hyperglycemia, and dislipidemia and plasma homocyteine, folate and vitamin {TEX}$B_{12}${/TEX} levels in Koreans. A total of 195 adult Koreans participated. The subjects were divided into three groups according to how many major conventional risk factors of cardiovascular disease they had: no risk, low risk (1~3 risk factors) and high risk (>3 risk factors) groups. As the number of risk factors increased, the plasma homocysteine levels significantly increase, while the plasma folate levels significantly decreased. The plasma homocysteine levels re higher in males than in females. The subjects with hyperglycemia had higher plasma homocysteine levels than the subjects without the risk factor. Also the subjects with dislipidemia had higher plasma homocysteine levels than the subjects without the risk factor. The plasma folate and vitamin {TEX}$B_{12}${/TEX} levels were significantly lower in males tan females. However, there were no significant differences in plasma folate and vitamin {TEX}$B_{12}${/TEX} levels between the subjects with or without other risk factors. These results indicate that plasma homocysteine levels were positively related with risk factors for cardiovascular disease and plasma folate levels were negatively related with the risk factors for cardiovascular disease. Also, we conclude that plasmahomocysteine levels might be related to the combination of risk factors, rather than an individual risk factor.

Analysis and verification of vitamin B12 in animal foods for update of national standard food composition table (국가표준식품성분표 개정을 위한 동물성 식품 비타민 B12 분석 및 검증)

  • In order to create the national food nutrient database, a total of 41 animal foods (ham, seafood, edible insects and eggs) were analyzed for their vitamin B12 content and the applied immunoaffinity-HPLC was verified. Ham vitamin B12 contents were 0.30-0.65 ㎍/100 g. Seafood showed relatively high vitamin B12 level, where the values of fermented clam were the highest (26.80 ㎍/100 g) followed by fermented pollack roe. Vitamin B12 was not detected in silkworm pupae and beetles, while relatively high levels were found in the two-spotted cricket imago (6.70 ㎍/100 g). Chicken and quail egg yolk had roughly 100- and 30-times higher vitamin B12 levels as compared to their egg white. Vitamin B12 contents in quail and chicken eggs were significantly enhanced by boiling (p<0.05). Results based on accuracy (97-102% recovery) and precision (<5% RSD) indicate that this study provides reliable vitamin B12 information on animal foods consumed in Korea.
