Simultaneous Determination of B Group Vitamins in Supplemented Food Products by High Performance Liquid Chromatography-Diode Array Detection

  • A simple HPLC-DAD method was developed and validated to determine B group vitamin content (thiamine, riboflavin, nicotinamide, pantothenic acid, pyridoxine and folic acid) in supplemented food samples, i.e., infant formula, cereal, low-calorie food, a multi-vitamin pill and a vitamin drink. In this study, the most significant advantages were simultaneous determination of the six B group vitamins in various food matrices and a small number of sample treatment steps that required only an organic solvent, acetonitrile. Moreover, this method prevents reduction of column durability, because the mobile phase does not contain ion-pairing reagents. Analytes were separated on a Develosil RPAQUEOUS $C_{30}$ (4.6 mm ${\times}$ 250 mm, 5 ${\mu}M$ particle size) column with a gradient elution of acetonitrile and 20 mM phosphate buffer (pH 3.0) at a flow rate between 0.8 and 1.0 mL/min. Detection was performed at 275 nm, except for that of pantothenic acid (205 nm). The calibration curves for all six vitamins showed good linearity with correlation coefficients ($r^2$) higher than 0.995. The developed method was validated with respect to linearity, intra- and inter-day accuracy and precision, limit of quantification (LOQ), recovery and stability. The method showed good precision and accuracy, with intra- and inter-assay coefficients of variation less than 15% at all concentrations. The recovery was carried out according to the standard addition procedure, with yields ranging from 89.8 to 104.4%. This method was successfully applied to the determination of vitamin B groups in supplemented food products.

  • Purpose: The aim of this study is to examine the effects of egg consumption and suggest proper guidelines for consumption of eggs by determining the relationship between eggs and cholesterol. Methods: Literature review was conducted on the relationship between nutritional, functional properties of eggs and serum cholesterol, as well as cardiovascular disease. Results: Eggs, which are a good protein food with complete amino acid composition, contain vitamin A, riboflavin, vitamin $B1_2$, folic acid, vitamin D, vitamin E, vitamin K, calcium, iron, choline, selenium, ${\beta}$-carotene, lutein, zeaxanthin, etc. However the egg yolk has a high cholesterol content, which is associated with chronic diseases, including heart disease and hypertension. As a result, its intake is subject to regulation. Outbreak of heart disease by yolk intake can show different results depending on the characteristics of the subjects, amount of egg intake, and the implications of other foods eaten. It is difficult to determine whether eggs are beneficial, as they are the main supplying source for other major nutritive elements as well. Several research studies insist that when cholesterol intake increases by 100 mg, the level of serum cholesterol increases by 2.2~4.5 mg/dL and when serum cholesterol increases by 1%, the risk of heart disease increases by 2%. This indicates that a large intake of eggs can increase the risk of heart disease. Although the cholesterol of egg yolk and serum cholesterol are correlated, it is insufficient to conclude that only cholesterol and not other components are related to heart disease. In fact, other components in egg such as various unsaturated fatty acids and phospholipids could be related as well. Rather than concluding egg as a 'good' or 'bad' food according to its cholesterol content, it is important to define egg as a part of dietary patterns. Conclusion: Generalizing an indiscriminate and uniform amount of egg intake for all seems inadequate. However, patients with diabetes or heart disease should pay particular attention to the amount of egg intake. As for the norm, eating egg with vegetables as a substitute for other animal products seems beneficial.

Analysis of Amino Acids, Vitamins and Minerals of Fruiting Body of Fomitopsis pinicola (소나무 잔나비버섯의 아미노산, 비타민 및 무기물 분석)

  • The nutritional composition of Fomitopsis pinicola (F. pinicola) fruiting body has been analyzed for medicinal and edible uses. The contents of crude fibers, carbohydrates, crude protein, moisture, crude fats and ashes were 43.3%, 26.3%, 12.8%, 12.6%, 3.3% and 1.7%, respectively. Eighteen amino acids were found in F. pinicola. Among total amino acids, glutamate content was the highest (457 mg/100 g dry mushroom) and arginine, glycine, valine, aspartate and isoleucine were followed. Concerning free amino acids, glutamine, arginine, trytophan, and glutamate were dominant. The vitamin E content was the highest (276 mg/100 g dry mushroom), then vitamin H and vitamin B, were followed. The mineral contents were as follows: K 165.06 mg, P77.57 mg, Mg 46.11 mg, Fe 21.56 mg, and Ca 16.90 mg based on 100 g dry mushroom.

Some Factors Affecting Bone Mineral Density of Korean Rural Women (농촌 성인 여성들의 골밀도에 영향을 미치는 요인 분석 연구)

  • The factors affecting bone mineral density (BMD) of 103 rural women aged 30 to 76 years were investigated. Data for food and nutrient intake was obtained by 24-hour recall method. BMD of lumbar spine(L1-L4), femoral neck, ward's triangle and trochanter was measured by XR-series x-ray bone densitometer. Serum was collected and analyzed for total Ca, P and ionized Ca (Ca++)content. Relationship between the factors and BMDs was analyzed by Pearson's correlation coefficient(r) and multiple regression analysis. The results are summarized as follows. 50% of the subjects under 50 years of age($\leq$49 yr group) and 86.4% of the subjects from 50 up($\geq$50yr group) were classified as osteopenia or osteoporosis. Mean body weight, height and BMI were 153.1cm, 59.1kg and 25.0kg/$m^2$ in $\leq$49 yr group and 151.9cm, 55.9kg, and 24.2kg/$m^2$ in $\geq$50 yr group. BMDs of lumbar spines and femurs ranged from 0.84 to 1.05g/$m^2$ and from 0.67 to 1.16g/$m^2$ in $\leq$49 yr group, and ranged from 0.67 to 0.85kg/$m^2$ and from 0.68 to 0.44g/$m^2$ in $\geq$50 yr group, respectively. On the whole, the BMDs were reduced to 83.8 to 94.2% of peck bone mass in $\leq$49 yr group and 55.2 to 78.9% of those in $\geq$50 yr group. Mean daily intake of Ca was much less than the Korean RDA, \67.2% in $\leq$49 yr group and 62.3% in $\geq$50 yr group. The average concentration of total Ca, P and Ca++ in serum were within normal range in both age group. Both age and height were significantly related with BMD in both age group but the relationship tended to be stronger in $\geq$50 yr group than in $\leq$49 yr group. Body weight was also a potent determinant of BMD only in 50 yr group. In $\leq$49 yr group, total food intake was positively related with BMDs of ward's triangle, L1 and L2 and intake of cereals and grain products, sugars and sweets, milk and dairy products was positively related with BMDs measured in this study. On the contrary, intake of eggs, oil and fats were positively related with a few BMDs in 50 yr group. The BMDs were positively affected by intake of energy, protein, carbohydrate, Ca, P and Fe in $\leq$49 yr group and those of protein, fat Ca, P, vitamin B1, vitamin B2 and vitamin C in $\geq$50 yr group. It was noteworthy that serum Ca++ concentration was positively related with BMDs of lumbar spine in boty age groups. According to multiple regression analysis, the four factors, age, body weight, height and BMI additionally accounted for 21% of the variance in BMD of trochanter in $\leq$49 yr group and only two factors, age and C a intake accounted for 38% of that of femoral neck in $\geq$50 yr group. Further investigation is necessary to make sure of the relations between BMD and serum Ca++ level.

Stable Transformation via Callus Formation and Rhizogenesis from the Cultures of Hypocotyl Explant of Chinese Cabbage (배추의 배축절편으로부터 캘러스와 뿌리 발생을 통한 안정적 형질전환)

  • Hypocotyl explants of Chinese cabbage (cvs. "Jeong Sang") produced transgenic calli on callus induction medium (MS salt, B5 vitamin, 5 mg/L acetosyringone, 1 mg/L 2,4-D, 3% sucrose, 400 mg/L cefotaxime, 100 mg/L paromomycin, pH 5.8) after cocultivation with strains of Agrobacterium tumefaciens (EHA101, LBA4404, GV3101) harboring the pPTN290 containing paromomycin-resistance gene as a selectable marker, and then they transferred to root induction medium (1/2MS salt, MS vitamins, 2% sucrose, 100 mg/L paromomycin, 100 mg/L cefotaxime, pH 5.8) and shoot induction medium (MS salt, B5 vitamin, 4 mg/L $AgNO_3$, 4 mg/L 6-benzyladenine, 3 mg/L alpha-naphthaleneacetic acid, 100 mg/L paromomycin, 100 mg/L cefotaxime, 3% sucrose, pH 5.8) in order. There was a significant difference in the frequency of transgenic calli depending on Agrobacterium strains. In particular, the highest frequency (6.1%) of transgenic calli was obtained from the hypocotyls cocultivated with EHA101 strains. Also, the frequency (%) of transgenic root and plants from each transgenic callus clone were obtained with 60.7% and 38.2% in EHA101, with 8.3% and 0% in LBA4404, with 20.5% and 85.7% in GV3101 strains, respectively. They were grown to maturity in a greenhouse and normally produced $T_2$ seeds. GUS histochemical assay for progeny ($T_2$) revealed that the transgenes was expressed in the plant genome, and progeny analysis from 7 independent transgenic events demonstrated that the transformants transmitted the transgene as a single or multiple functional locus.

Chemical Composition and Bioactivity of Korean Green Tea (Camellia sinensis) Pollen collected by Honey Bee (한국 녹차 화분의 화학적 조성과 기능성 연구)

  • In this study, we investigated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of green tea (Camellia sinensis) pollen grains collected by Apis mellifera bees, for use as a health food. The crude protein and fat content was estimated at 26.14% and 3.49%, respectively. Eighteen amino acids were identified in green tea pollen, including 8 essential amino acids and 10 non-essential amino acids. The predominant amino acids were glutamic acid, proline and aspartic acid accounting for about 33.3% of total free amino acids. The concentration of vitamin C was the highest value of 35.7%, followed by $B_3$ and $B_2$ among the detected vitamins. The predominant minerals were potassium (790.32 mg/100g), followed by phosphorus (707.52 mg/100g) and sulfur (302.67 mg/100g), whereas copper, zinc and sodium were detected as minor elements. The antioxidant activity and phenolic content accounted for 33.8% at $500{\mu}g/mL$ extract and $2.55 {\mu}g/mg$, respectively.

Effects of Dry Roasting on the Vitamin E Content and Microstructure of Peanut (Arachis hypogaea)

  • Effects of roasting on vitamin E content, color, microstructure and moisture of peanuts, and vitamin E content in peanut oils prepared from the roasted peanuts were investigated. Runner-type peanuts were roasted at 140, 150, and $160^{\circ}C$ for 10-20 min. As roasting temperature and time increased, the CIELAB $L^*$ value of peanuts decreased while $a^*$ and $b^*$ values increased, resulting in formation of the golden brown color of roasted peanuts. Moisture ratio (M/Mo) and color $b^*$ value of peanuts roasted at 140 to $160^{\circ}C$ showed a correlation of $b^*=21.61\;(M/Mo)^2-40.62\;(M/Mo)+34.12$ ($R^2=0.9123$). Overall changes in the tocopherol contents of peanuts and peanut oils were significantly affected by roasting temperature and time (p<0.05). Roasting at $140^{\circ}C$ caused a slight increase in the levels of tocopherols of peanuts over roasting time up to 20 min (p<0.05). There was no significant change in the tocopherol levels of peanuts during roasting at $150^{\circ}C$ for 20 min (p>0.05). At $160^{\circ}C$, the levels of tocopherols significantly decreased during the initial 10 min of roasting (p<0.05) while there was no extended loss after 10 min, resulting in about 5, 12, 20, and 10% losses of ${\alpha}$-, ${\beta}$-, ${\gamma}$- and ${\delta}$-T, respectively. After 20 min, total tocopherols decreased by 18%. However, tocopherol contents of pressed peanut oils significantly decreased at all roasting temperatures (p<0.05). After roasting peanuts at $160^{\circ}C$ for 20 min, about 84% of initial ${\alpha}$-T in peanut oils was retained. ${\alpha}$-T was the most stable to roasting while ${\gamma}$-T was the least. Swollen epidermal cells on the inner surface and broken cell walls of parenchyma tissue of peanut cotyledon were observed in peanuts after roasting at $160^{\circ}C$ for 15 min. Severe changes in microstructure of peanut by roasting would contribute to vitamin E stability because of exposure of oil droplets in peanuts to oxygen.

Quality Changes in Pulp-containing Apple Juice upon Addition of Vitamin C (비타민 C 첨가에 따른 미세과육 함유 사과주스의 저장 중 품질변화)

  • We investigated changes in the quality of pulp-containing apple juice, during storage, after addition of various amounts of vitamin C, which was stable over time. Neither sugar content nor acidity level varied when vitamin C was added. The pH was slightly lower (pH 4.29-4.30) in juice with added vitamin C than in unsupplemented juice (pH 4.40). The L and b color values fell as vitamin C content rose and the storage period was extended. In sensory evaluation tests, taste and overall acceptability were higher for juice to which vitamin C had been added to 0.02% (w/v) than for unsupplemented juice. Vitamin C levels fell less during storage at $4^{\circ}C$ than at higher temperatures. The alcohol-soluble color (ASC) value fell as the amount of added vitamin C rose, and tended to be lower when juice was stored at $4^{\circ}C$ compared to $37^{\circ}C$. In summary, apple juice containing pulp was optimally stored at $4^{\circ}C$ after addition of 0.02% (w/v) vitamin C

Quality Characteristics of Semi-Dried Red Pepper (Capsicum Annuum L.) Using Hot-Air drying (반건조 고추(Capsicum Annuum L.)의 건조조건에 따른 품질 특성)

  • The quality characteristics of semi-dry red pepper (SDRP) (Capsicum annuum L.) were compared with raw and dry red pepper (DRP). Raw red pepper was divided into pieces and the semi-drying treatment involved air-drying at $65^{\circ}C$. The study focused on describing the characteristics of semi-dry red pepper in comparison with dry red pepper. Factors considered were reduced drying period, ASTA color, capsanthin, capsaicinoids, free sugars and vitamin C content. ASTA color, capsanthin, capsaicinoids, free sugars and vitamin C content of SDRP were higher in SDRP than in DRP for 15 g or less water per 100 g. Red pepper powders made using the semi-drying method showed the highest amount of glucose and vitamin C. Capsanthin content in SDRP (151.6155.9 mg/100 g) was significantly higher than for DRP (133.4 mg/100 g). The capsaicinoid content of SDRP was about 13-25% higher than in DRP. The ASTA values (148.7159.3) for SDRP were much higher than for DRP (139.5). The vitamin C and free sugar content of SDRP was 40-76% and 20-40% higher, respectively, than for DRP. and $20{\sim}40%$, respectively, by SDRP compared to DRP.

The Correlation and Comparison of Quality Characters of Several Tomato Cultivars Grown by Hydroponics (수경재배한 토마토의 품종별 품질특성 비교 및 품질특성간 상관관계)

  • This study was conducted to compare the quality characters of 15 different tomato cultivars ('Dotearang Dia', 'Super Dotearang', 'Super sunroad', 'Hoyong', 'Mirokku', 'Baccuhs', 'Mascara', 'Poseidon', 'Radido', 'Madison', 'Rapsodie', 'Solomon', 'Amaral', 'Picasso', 'Tymaxx') and find out the correlation among the quality characters in order to get basic informations for exporting tomato. The $a^*/b^*$ value of fruit surface showed from 2.8 to 4.8, but there was no significant difference in fruit surface color value, such as $a^*$, $b^*$ and $a^*/b^*$ among the cultivars. The respiration and ethylene production rate also were not shown any significantly difference among cultivars. The firmness of fruit was higher in 'Picasso', 'Tymaxx' and 'Madison' cultivars. The contents of soluble solids was higher in 'Hoyong' and 'Dotearang Dia' cultivars. The vitamin C content was higher in 'Super Sunroad' and 'Dotearang Dia' cultivars. The titratable acidity was higher in 'Hoyong' cultivars. The sugar/acid ratio that depended on soluble contents was lower in 'Amaral', 'Picasso', 'Tymaxx' cultivars that showed lower soluble contents. Conclusionally, The firmness that was one of the most important quality character in exporting tomato was higher in European cultivars, but soluble solide and vitamin C content were higher in Japanese cultivars. The significant correlation coefficient values were determined (more than p = 0.05) between $a^*/b^*$ and other quality characters. The results suggested that surface color was the most suitable character represented tomato qualities. The high coefficient value (r = 0801) between $a^*/b^*$ and the contents of soluble solids. The firmness and the contents of soluble solids that is the most important factor for exporting showed significant negative correlation (r = -0.611).
