Effect of Vitamin $B_{12}$ Addition on the Hatching Rate and Production of Resting Eggs of Freshwater Rotifer, Brachionus calyciflorus Pallas (Vitamin $B_{12}$ 첨가가 담수산 rotifer, Brachionus calyciflorus Pallas의 내구란의 생산과 부화율에 미치는 영향)

  • This study was performed to investigate the effect of vitamin $B_{12}$ addition on the production and hatching rate of resting egg of freshwater rotifer, Brachionus calyciflorus. Different concentrations of vitamin $B_{12}$ was added into concentrated freshwater Chlorella before fed to rotifer and rotifer was cultured in 250 mL flask. The maximum density of rotifer was 138 inds./mL at 6 ppm and it increased with an increase of vitamin $B_{12}$ addition. Mixis rate and fertilization rate of rotifer were $63.0\%$ and $89.6\%$, respectively, at 6.0 ppm. The production of resting eggs per mL was 231 eggs/mL at 1.5 ppm, but there was no significant difference among vitamin $B_{12}$ addition levels. The highest production of resting eggs per 10,000 rotifers and Chlorella dry weight (g) were 11,289 eggs and 121,252 eggs, respectively, at the 6 ppm. The hatching rate of resting egg was not significantly different among vitamin $B_{12}$ addition levels, but significantly higher than the control (without addition of vitamin $B_{12}$ ). These results showed that vitamin $B_{12}$ addition could affect production and hatching rate of resting egg of freshwater rotifer, B. calyciflorus.

  • The concentrations of dissolved vitamin $B_{12}$, thiamine and biotin in the water of Gyokpo coast, were determined by microbiological assay methods. Also the relations between the distribution of B group vitamin and other environmental factors were studied. Vitamin $B_{12}$ was assayed with Euglena gracilis strain Z, thiamine with Cryptococous albidus and biotin with Achromo bacter sp. yH-51. It was found that the concentration of B group vitamin in the water of Gyokpo coast were normal level : vitamin $B_{12}$; 1.36-3.95 ng/l, thiamine ; u-0.4 ng/l and biotin; 1.40-14.60 ng/l. The concentration of B group vitamin was high in summer than in winter. In the water of Gyokpo coast during summer, B group vitamin occurred slightly lower level than normal, the concentration suficiently neccessary for phytoplankton development. The concentration of biotin was positively correlated with abundance of phytoplankton, but not aerobic heterotrophic bacteria. It was suggested that the concentration of biotin in water might be much more influenced with the growth of phytoplankton and any environmental factors than bacteria and the other vitamin, especially.

The Effect of Vitamin B6 Deficiency on the Utilization of Fuel and Blood Cholesterol Profile with Regular Exercise-Training in Rats (비타민 B6 부족이 정기적인 운동 훈련시 연료의 이용과 혈액 콜레스테롤 성상에 미치는 영향)

  • The purpose of this study was to determine whether vitamin B6(B6) deficiency affects fuel utilization and blood cholesterol profile with exercise-training. Twenty-four rats were fed a B6 deficient(-B6) diet or a control (+B6) diet for 5 weeks and either exercised(EX) or nonexercised (NE). EX rats were exercised on treadmill(10$^{\circ}$, 0.5-0.8km/h) for 20 minutes everyday. Glucose(GLU), glycogen (GLY), protein(PRO), trglyceride(TG), free fatty acid(FFA), total cholesterl(TC), HDL-cholesterol(HDL-C) and LDL-choleterol(LDL-C) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. There was a vitamin effect on the level of P-GLU, P-TG, M-TG, L-GLY, L-PRO and an exercise effect on the level of P-PRO, P-FFA, M-PRO, L-GLY, L-TG, P-TC, P-HDL-C, P-LDL-C. Compared to +B6 rats were lower and there were no differences in P-GLU, P-FFA, P-TG. M-GLY, L-TG, P-TC and P-HDL-C. In EX group, the level of P-TG was higher and M-PRO was lower in -B6 rats. There were no differences in M-GLY, L-TG, P-TC and P-HDL-C. These results suggest that a lowered intake of vitamin B6 may impair the adaptation of animals to fuel metabolism related to a decrease of fatty acid oxidation and attenuates the exercise-traning effect on blood lipid profile.

  • This study was conducted to simultaneous analysis methods for water soluble vitamins B group (vitamin $B_1$, vitamin $B_2$, nicotinic acid, nicotinamide, vitamin $B_6$) which is used as health functional foods etc. Analytical methods of water-soluble vitamins B group by HPLC were established through instrumental analytical conditions, and the examination of data such as domestic and foreign reliable methods, and papers of journal. HPLC method analyzing water soluble vitamins B group was established using Capcell Pak C18 UG 120 column in 270 nm through test of columns. The validation has been performed on the method to determine linearity, accuracy, limits of quantification (LOQ) and repeatability for water soluble vitamins B group. An excellent linearity ($r^2=0.999$) was observed for vitamin $B_1$, vitamin $B_2$, nicotinic acid, nicotinamide, vitamin $B_6$ in the concentration range ($0.1{\sim}2{\mu}g/mL$). Observed recovery of vitamin $B_1$ was found to be between 100 and 103%, vitamin $B^2$ was found to be between 104 and 112%, nicotinic acid was found to be between 82 and 85%, nicotinamide was found to be between 121 and 124% and vitamin $B_6$ was found to be between 95 and 104%. LOQ of vitamin $B_1$ was found to be $0.04{\mu}g/mL$, vitamin $B_2$ was found to be $0.05{\mu}g/mL$, nicotinic acid was found to be $0.15{\mu}g/mL$, nicotinamide was found to be $0.08{\mu}g/mL$ and vitamin $B_6$ was found to be $0.63{\mu}g/mL$. Repeatability precision for vitamin $B_1$ was found to be 0.4%, vitamin $B_2$ was found to be 0.4%, nicotinic acid was found to be 0.5%, nicotinamide was found to be 0.7% and vitamin $B_6$ was found to be 0.4% relative standard deviation (RSD). Also, verify the accuracy of the simultaneous analysis methods, we monitored the labeled contents of the health functional foods and children's preferred foods.

Change of ROS Generation and Antioxidant Enzyme Activity of Flavonol Quercetin in the Presence of Vitamin E, L-Ascorbit acid, Reduced Glutathione on the B16F10 Murine Melanoma Cells (B16F10 세포에서 Quercetin과 Vitamin E, L-Ascorbic acid, 환원형 글루타치온과의 병용 투여에 의한 활성산소종 발생과 항산화 효소의 활성 변화)

  • It has been known that quercetin, a bioflavonoid widely distributed in fruits and vegetables as dietary-derived flavonoid and exert significant multiple biological effects such as antioxidant and anti-inflammatory, anti-tumor effects. In addition, it has been shown to have a chemoprotective role in cancer, though complex effects on signal transduction involved in cell proliferation and angiogenesis. The present study investigated whether quercetin can enhance antioxidant enzyme activity (glutathione peroxidase: GPx, superoxide dismutase: SOD, catalase: CAT) and regulate the reactive oxygen species (ROS) generation in the presence of vitamin E, L-ascorbic acid, reduced glutathione (GSH) on B16F10 murine melanoma cells. After 48h treatment of cells with quercetin in the presence of vitamin E, L-ascorbic acid, GSH, we measured the cytotoxicities by MTT assay. The cells exhibited a dose-dependent inhibition in their proliferation in the presence of vitamin E, L-ascorbic acid, GSH respectively. We also investigated the effects of antioxidant enzyme activity and ROS generation. The antioxidant enzyme activity of quercetin in the presence of vitamin E was stronger than GSH, L-ascorbic acid, the same treatments decreased ROS generation in B16F10 murine melanoma cells. Taken together, these result demonstrate that the antioxidant effect of quercetin can enhanced in the presence of vitamin E and it might plays an important role in anti-oxidative effects.

Evaluation of vitamin $B_6$ intake and status of 20- to 64-year-old Koreans

  • BACKGROUND/OBJECTIVES: Recent research regarding vitamin $B_6$ status including biochemical index is limited. Thus, this study estimated intakes and major food sources of vitamin $B_6$; determined plasma pyridoxal 5'-phosphate (PLP); and assessed vitamin $B_6$ status of Korean adults. MATERIALS/METHODS: Three consecutive 24-h diet recalls and fasting blood samples were collected from healthy 20- to 64-year-old adults (n = 254) living in the Seoul metropolitan area, cities of Kwangju and Gumi, Korea. Vitamin $B_6$ intake and plasma PLP were analyzed by gender and by vitamin $B_6$ supplementation. Pearson's correlation coefficient was used to determine associations of vitamin $B_6$ intake and plasma PLP. RESULTS: The mean dietary and total (dietary plus supplemental) vitamin $B_6$ intake was $1.94{\pm}0.64$ and $2.41{\pm}1.45mg/day$, respectively. Median (50th percentile) dietary intake of men and women was 2.062 and 1.706 mg/day. Foods from plant sources provided 70.61% of dietary vitamin $B_6$ intake. Only 6.3% of subjects consumed total vitamin $B_6$ less than Estimated Average Requirements. Plasma PLP concentration of all subjects was $40.03{\pm}23.71nmol/L$. The concentration of users of vitamin $B_6$ supplements was significantly higher than that of nonusers (P < 0.001). Approximately 16% of Korean adults had PLP levels < 20 nmol/L, indicating a biochemical deficiency of vitamin $B_6$, while 19.7% had marginal vitamin $B_6$ status. Plasma PLP concentration showed positive correlation with total vitamin $B_6$ intake (r = 0.40984, P < 0.0001). CONCLUSIONS: In this study, vitamin $B_6$ intake of Korean adults was generally adequate. However, one-third of subjects had vitamin $B_6$ deficiency or marginal status. Therefore, in some adults in Korea, consumption of vitamin $B_6$-rich food sources should be encouraged.

The effect of Vitamin $B_2$ Deficiency on the Utilization and Recuperation of Fuel in Exercising Rats (비타민 $B_2$ 부족이 운동중 연료의 이용과 회복에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin B2 deficiency on the utilization and recuperation of fuel in exercising rats. Thirthy six rats were fed either a vitamin B2 deficient diet (-B2) or a control diet (+B2) for 3 weeks and then subdivided into 3 groups : non-exercise group(NE), exercise group (ES), exercise and recuperation group (ER). ES group were exercised on treadmill (10$^{\circ}$, 0.5-0.8km/h) for 2 hours and ER group were recuperated three days with the respective diet after exercise. Glucose (GLU), glycogen(GLY), protein(PRO), triglyceride(TG) and free fatty acid(FFA) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. Compared to + B2 rats, in NE group, the level of P-GLU and L-GLU of -B2 rats was lower, L-PRO and M-GLY was higher and there were no differences in P-PRO, P-FFA, L-TG and M-TG. In ES group, the level of P-GLU was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLU, L-TG, M-GLY, M-TG and M-PRO. In ER groups, the level of P-GLU and L-TG was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLY, L-PRO, M-GLY, M-TG and M-PRO. These results suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise and may lead a sluggish recuperation related to fuel stores after exercise.

Composition of Vitamin A, E, $B_l$ and $B_2$ Contents in Korean Cow's Raw Milk in Korea (국내산 원유 중 비타민 A, E, $B_l$$B_2$ 함량에 관한 연구)

  • This study was performed to investigate the changes of vitamin A, vitamin E, vitamin $B_l$ and vitamin $B_2$ contents in cow's raw milk collected from dairy farms in Chungcheong-do and Jeolla-do for a year. The contents of fat soluble vitamin A and E were changed as seasonal effect, but water soluble vitamin $B_l$ and $B_2$ contents were not changed as seasonal effect. Vitamin A content in cow's raw milk was as follows [minimum ${\sim}$maximum (mean), ug/100 mL]; $35.1{\sim}59.0$ (44.4) in spring, $36.7{\sim}65.6$ (50.0) in summer, $28.7{\sim}61.2$ (46.8) in autumn and $29.9{\sim}57.8$ (43.1) in winter. In case of vitamin E was as follows [minimum${\sim}$maximum (mean), ug/100 mL]; $28.3{\sim}59.2$ (45.8) in spring, $39.6{\sim}69.9$ (58.8) in summer, $35.0{\sim}62.8$ (46.2) in autumn and $26.0{\sim}55.4$ (41.5) in winter. In case of vitamin $B_l$ was as follow [minimum${\sim}$maximum (mean), ug/100 mL]; $27.7{\sim}57.9$ (42.84) in spring, $32.4{\sim}66.1$ (49.39) in summer, $34.1{\sim}63.7$ (46.69) in autumn and $20.6{\sim}61.4$ (43.20 in winter. The amounts of vitamin $B_2$ in cow's raw milk was as follows [minimum${\sim}$maximum (mean), ug/100 mL]; $150{\sim}182$ (160 in spring, $145{\sim}185$ (163) in summer, $149{\sim}180$ (166) in autumn and $148{\sim}190$ (167) in winter.

Relationships between Vitamin $B_6$ Status of Maternal-Umbilical Cord (임신 말 모체 및 제대혈의 비타민 $B_6$ 농도와 임신결과와의 상관성)

  • The purpose of this study was to evaluate the concentration of vitamin B6 in 16 pregnant-infant pairs and 15 nonpregnant women and to investigate the relationships between vitamin B6 status of maternal-umbilical cord plasma and pregnancy outcomes. dietary intake was obtained from semiquantitative frequency questionnaire. The daily mean energy and protein intakes were higher than the recommended dietary allowance, while daily vitamin B6 was only 74% of RDA in pregnant and 73% of RDA in nonpregnant women. The main sources of vitamin B6 were vegetables and fruits in pregnant women, while cereal and starch in nonpregnant women. The plasma PLP and PL levels of pregnant women were 14.85nmol/l and 20.56nmol/l, significantly lower than those of nonpregnant women. the PLP/PL ratios of pregnant and nonpregnant women were 1.65 and 0.33, indicating that the levels of vitamin B6 was altered during pregnancy. The PLP and PL levels of umbilical cord plasma were 63.55nmol/l and 32.25nmol/l, respectively. The vitamin B6 levels of umbilical cord plasma were significantly higher than that of maternal plasm. This finding indicates that the uptake of vitamin B6 in the fetus may be due to an active placental transport mechanism. The PLP level of maternal plasma correlated positively with that of umbilical cord plasma, showing the PLP concentration of umbilical cord plasma is affected by maternal vitamin B6 status. The maternal plasma PL level showed a positive correlation to infant birth weight. The positive association has bee also found between plasma PL level of umbilical cord and Apgar 1 min score.

In EX group, the levels of P-FFA, L-PRO of -{TEX}$B_{6}${/TEX} rats were higher. P-TG, L-TG of -{TEX}$B_{6}${/TEX} rats were lower and there was no difference in L-GLY. In EG group, the levels of P-GLU, P-PRO, P-RG, L-PRO of -{TEX}$B_{6}${/TEX} rats were lower and there were no differences in L-GLY, L-TG, M-TG and M-GLY. these results suggest that a lowered intake of vitamin {TEX}$B_{6}${/TEX} may impair the recuperation of aminals after exercise related to exercise fuel stores although there is a compensation among stored fuel utilization during exercise.
