Diagnosis of Freemartinism in Korean Native Cattle by Amplification of Two Different Male-Specific DNA Sequences

  • The freemartinism is the most frequent form of intersexuality found in cattle, and females of heterosexual twins become sterile. With increase of twinning rates due to transfer of multiple embryos derived from in vitro fertilization, it is of great economic value to establish early diagnosis of freemartins to remove infertile individuals from breeding stock. In the present study polymerase chain reaction (PCR) of two different Y-chromosome specific segments (BRY.l and AMX/Y) was performed to identify freemartins from twins and less common single born freemartins in Korean Native Cattle (KNC). Two male-specific sequences were amplified in all heterosexual twins tested (n=5). In addition, Y-specific PCR products were detectable in one of the single born females (n=4) with visible genital abnormalities. These results suggest that the sensitivity of PCR-based assay may be sufficient to detect freemartinism in single born females as well as female partners of heterosexual twins in KNC.

Design of discriminant function for thick and thin coating from the white coating (백태 중 후태 및 박태 분류 판별함수 설계)

  • Introduction: In Oriental medicine, the status of tongue is the important indicator to diagnose one's health, because it represents physiological and clinicopathological changes of inner parts of the body. The method of tongue diagnosis is not only convenient but also non-invasive, so tongue diagnosis is most widely used in Oriental medicine. By the way, since tongue diagnosis is affected by examination circumstances a lot, its performance depends on a light source, degrees of an angle, a medical doctor's condition etc. Therefore, it is not easy to make an objective and standardized tongue diagnosis. In order to solve this problem, in this study, we tried to design a discriminant function for thick and thin coating with color vectors of preprocessed image. Method: 52 subjects, who were diagnosed as white-coated tongue, were involved. Among them, 45 subjects diagnosed as thin coating and 7 subjects diagnosed as thick coating by oriental medical doctors, and then their tongue images were obtained from a digital tongue diagnosis system. Using those acquired tongue images, we implemented two steps: Preprocessing and image analyzing. The preprocessing part of this method includes histogram equalization and histogram stretching at each color component, especially, intensity and saturation. It makes the difference between tongue substance and tongue coating was more visible, so that we can separate tongue coating easily. Next part, we analyzed the characteristic of color values and found the threshold to divide tongue area into coating area. Then, from tongue coating image, it is possible to extract the variables that were important to classify thick and thin coating. Result : By statistical analysis, two significant vectors, associated with G, were found, which were able to describe the difference between thick and thin coating very well. Using these two variables, we designed the discriminant function for coating classification and examined its performance. As a result, the overall accuracy of thick and thin coating classification was 92.3%. Discussion : From the result, we can expect that the discriminant function is applicable to other coatings in a similar way. Also, it can be used to make an objective and standardized diagnosis.

An Experimental Study on the Effects of Low Level Laser Irradiation on the Cell Viability of Cultured Fibroblast (저출력레이저조사가 배양섬유아세포의 생존력에 끼치는 영향에 관한 실험적 연구)

  • This study was performed to investigate the effects of infrared and visible light laser irradiation on cell viability of human gingival fibroblast. For the present study, the author used cultured fibroblast originated from sound gingiva which were fifth of sixth passage. Laser machine utilized here were stomalaser which irradiate infrared (GaAs diode) and red (HeNe) laser in turn with pulse wave pattern or continuous wave pattern, and the machine had several frequency mode presented by regeneration, relaxation and analgesic modalities. Cultured fibroblast samples were divided by this modalities of cell counts and laser exposure time which were 7-seconds of 150 seconds, respectively. 1 day after laser irradiation, each cell-well was treated with MTT and measured optical density with ELISA. The obtained results were as follows : 1. There was a tendency of increasing optical density in proportion to irradiation time in groups of $1\times10^4$ cell per well but in groups of $5\times10^3$ cell per well, reverse phenomena were observed. 2. The difference of optical density according to frequency modalities were not showed significantly except several cases in groups of $5\times10^3$ cell per well. 3. In general, cell viability of cultured human gingival fibroblast wer not showed consistent feature by low level laser irradiation.

  • Purpose: Autogenous bones are frequently used because of their lack of antigenicity, but good osteoconductive and osteoinductive properties. This study evaluated the biological behavior of perforated and nonperforated cortical block bone grafts. Methods: Ten nonsmoking patients who required treatment due to severe resorption of the alveolar process and subsequent implant installation were included in the study. The inclusion criteria was loss of one or more teeth; the presence of atrophy of the alveolar process with the indication of reconstruction procedures to allow rehabilitation with dental implants; and the absence of systemic disease, local infection, or inflammation. The patients were randomly divided into two groups based on whether they received a perforated (inner surface) or nonperforated graft. After a 6-month healing period, a biopsy was performed and osseointegrated implants were installed in the same procedure. Results: Fibrous connective tissue was evident at the interface in patients who received nonperforated grafts. However, full union between the graft and host bed was visible in those who had received a perforated graft. Conclusions: We found that cortical inner side perforations at donor sites increased the surface area and opened the medullary cavity. Our results indicate an increased rate of graft incorporation in patients who received such perforated grafts.

  • Recently, intensive research has been conducted to develop innovative methods for diagnosing plant diseases based on hyperspectral technologies. Hyperspectral analysis is a new subject that combines optical spectroscopy and image analysis methods, which makes it possible to simultaneously evaluate both physiological and morphological parameters. Among the physiological and morphological parameters are classifying healthy and diseased plants, assessing the severity of the disease, differentiating the types of pathogens, and identifying the symptoms of biotic stresses at early stages, including during the incubation period, when the symptoms are not visible to the human eye. Plant diseases cause significant economic losses in agriculture around the world as the symptoms of diseases usually appear when the plants are infected severely. Early detection, quantification, and identification of plant diseases are crucial for the targeted application of plant protection measures in crop production. Hence, this can be done by possible applications of hyperspectral sensors and platforms on different scales for disease diagnosis. Further, the main areas of application of hyperspectral sensors in the diagnosis of plant diseases are considered, such as detection, differentiation, and identification of diseases, estimation of disease severity, and phenotyping of disease resistance of genotypes. This review provides a deeper understanding, of basic principles and implementation of hyperspectral sensors that can measure pathogen-induced changes in plant physiology. Hence, it brings together critically assessed reports and evaluations of researchers who have adopted the use of this application. This review concluded with an overview that hyperspectral sensors, as a non-invasive system of measurement can be adopted in early detection, identification, and possible solutions to farmers as it would empower prior intervention to help moderate against decrease in yield and/or total crop loss.

WLDF: Effective Statistical Shape Feature for Cracked Tongue Recognition

  • This paper proposes a new method using Wide Line Detector based statistical shape Feature (WLDF) to identify whether or not a tongue is cracked; a cracked tongue is one of the most frequently used visible features for diagnosis in traditional Chinese Medicine (TCM). We first detected a wide line in the tongue image, and then extracted WLDF, such as the maximum length of each detected region, and the ratio between maximum length and the area of the detected region. We trained a binary support vector machine (SVM) based on the WLDF to build a classifier for cracked tongues. We conducted an experiment based on our proposed scheme, using 196 samples of cracked tongues and 245 samples of non-cracked tongues. The results of the experiment indicate that the recognition accuracy of the proposed method is greater than 95%. In addition, we provide an analysis of the results of this experiment with different parameters, demonstrating the feasibility and effectiveness of the proposed scheme.

Review of Bladder cancer (방광암의 이해)

  • Bladder cancer is one of the most common cancers affecting men and women and thus has a profound impact on health care. The majority of patients (75%) with newly diagnosed urothelial tumors have non-muscle invasive disease confined to the bladder mucosa or the lamina propria. The most important risk factors for the development of bladder cancer are smoking and occupational exposure to toxic chemicals. Painless visible hematuria is the most common presenting symptom of bladder cancer. Cystoscopy and urine cytology are currently the recommended tools for diagnosis of bladder cancer. Excluding muscle invasion is an important diagnostic step, as outcomes for patients with muscle invasive bladder cancer (MIBC) are less favorable. For non-muscle invasive bladder cancer (NMIBC), the high rate and frequency of recurrence and the concern for disease progression - especially in patients with high-risk tumors - mandate careful strategies for tumor surveillance. The surveillance strategies should be based on available prognostic factors and in particular data from the EORTC risk tables.

  • 본 논문에서 적외선(IR; Infrared), 자외선(UV; Ultraviolet), 가시광선(VR; Visible ray)의 영역에서 촬영한 설진 영상으로부터 가장 효과적인 설태 인식을 위한 최적 혀 영상 분석 방법을 제안한다. 제안한 방법은 설진에서 혀 영상 촬영을 위한 최적 파장 범위와 해당 파장에서 설태 분석에 최적의 컬러 영상을 선정한다. 최적 영상 선정을 위해서는 각 파장별로 촬영한 혀 영상을 LAB, HSV, YcBcR, RGB 컬러모델로 변환하고, 변환된 영상들로부터 설태와 비설태 영역의 히스토그램(Histogram)을 분석에 의해서 영역-분별력을 측정한다. 실험 결과 설진에서 설태 인식을 위한 최적 혀 영상은 자외선 영역에서의 RGB 컬러모델로 나타났다.

  • Massive hydrothorax complicating continuous ambulatory peritoneal dialysis (CAPD) is relatively rare. A 67-year-old male and a 23-year-old female patients during CAPD presented massive pleural effusion. They have been performing peritoneal dialysis due to end-stage renal disease for 8 months and 2 weeks respectively. We injected $^{99m}Tc$-labelled radiopharmaceutiral (phytate and MAA, respectively) into peritoneal cavity with the dialysate. The anterior, posterior and right lateral images were obtained. The studies reveal visible radioactivity in the right chest indicating the communication between the peritoneal and the pleural space. After sclerotherapy with tetracycline, the same studies reveal no radioactivity in the right chest suggesting successful therapy. We think nuclear imaging is a simple and noninvasive method for the differential diagnosis of pleural effusion in patients during CAPD and the evaluation of therapy.

  • Sj${\ddot{o}}$gren's syndrome is a chronic autoimmune exocrinopathy that destroys salivary and lacrimal gland tissue. We report an unusual case of this disease in a 54-year-old woman who presented with multiple and bilateral parotid cystic masses. The multiple, small, bead-like cysts were clearly evident in the computed tomography sections in this patient, a visible reminder that this may be the initial presentation in a patient with Sj${\ddot{o}}$gren's syndrome. As the case illustrates, Sj${\ddot{o}}$gren's syndrome should be included in the differential diagnosis of multiple and bilateral cystic parotid lesions.
