육계 사료 내 박테리오파지 CP의 첨가가 생산성, 영양소 소화율, 혈액특성, 도체특성 및 분내 미생물 조성에 미치는 영향 (Effects of Dietary Supplementation of Bacteriophage CP on Growth Performance, Nutrient Digestibility, Blood Profiles, Carcass Characteristics and Fecal Microflora in Broilers)

  • 본 연구는 박테리오파지의 급여가 육계의 생산성, 영양소 소화율, 혈액 특성, 장기무게 및 분내 미생물 조성에 미치는 영향을 알아보기 위해 시험을 실시하였다. 사양 시험은 1일령 ROSS 308(♂, ♀) 340수 공시하였고, 시험 개시 체중은 $41.14{\pm}0.17g$으로 31일간 실시하였다. 시험 설계는 1) CON(basal diet), 2) CP05(CON + bacteriophage CP 0.05%), 3)CP10(CON + bacteriophage CP 0.10%), 4) CP15(CON + bacteriophage CP 0.15%)로 4개 처리를 하여 처리당 5반복, 반복당 17수씩 완전 임의 배치하였다. 1~14일령의 생산성에 있어서는 처리구 간 유의적 차이가 없었다. 그러나 15~31일령에 있어서는 증체량 및 사료 섭취량에서 CP15 처리구가 CON 처리구보다 높게 나타났고, 전체 시험 기간 동안의 증체량에 있어서도 CP10 및 CP15 처리구가 CON 처리구보다 유의적으로 높게 나타났다. 영양소 소화율 및 혈액 특성에 있어서 처리구 간 차이를 나타내지 않았다. 도체 특성 중 가슴육의 보수력 있어서 박테리오파지를 첨가한 처리구가 CON 처리구보다 유의적으로 높게 나타났고, 장기 무게 중 F낭의 무게에 있어 CP05 처리구가 CON 처리구에 비해 유의적으로 높게 나타났다. 분내 미생물 조성에 있어서는 처리구간 유의적 차이를 나타내지 않았다. 결론적으로, 육계 사료 내박테리오파지를 첨가 시 증체량을 향상시키고, F낭의 무게를 증가시키며, 가슴육의 보수력이 증가하는 효과를 나타내었다.

Utilization of Processed Pine Needle Powder in Broiler Chicken Diets

  • This study examined the effects of dietary processed pine needle tree powder (PPNP) on growth performance, visceral organ parameters, ileal digestibility, blood metabolites and ileal morphology of broiler chickens for 5-week after hatch. In total, 220 1-day-old male Ross 308 broiler chicks were randomly allocated to 30 pens (seven birds per pen), and each pen was assigned to one of five dietary treatments, a diet without PPNP (control) and diets with 0.5%, 1.0%, 1.5% and 2.0% PPNP. The body weight and feed intake were measured weekly. One bird from each pen was selected randomly and gently euthanized to collect the blood, visceral organs and digesta samples on 2, 3 and 5 week of age. Birds fed 2.0% PPNP showed decreased (P<0.05) overall body weight, average daily gain and average daily feed intake compared to those fed control, 0.5 and 1.0% PPNP. The small intestine length increased (P<0.05) as the level of PPNP in the diet increased. A deeper crypt depth was observed (P<0.05) when birds fed 2.0% PPNP compared to those fed the control diet at week 2. A higher V:C ratio was observed (P<0.05) in birds fed 1.0% PPNP than in those fed other treatments at week 3. Ileal protein digestibility improved (P<0.05) when birds fed 0.5% and 1.0% PPNP compared to that in those fed the control diet at week 3. The feed conversion ratio, ileal dry matter digestibility and blood urea nitrogen and cholesterol, and gizzard and ceca weights were not affected (P>0.05) by supplementation of any levels of PPNP. These results demonstrated that inclusion of PPNP in broiler diets could be a possible factor in improving overall growth performance suggesting early gut development when 1% PPNP is supplemented.

  • Objectives : This study was undertaken to verify the effects of Gyeongshingangjeehwan18 (GGEx18) on obesity using ob/ob male mice. Methods : Eight-week old mice (wild-type C57BL/6J and ob/ob) were used for all experiments. Wild-type C57BL/6J mice were used as lean control and obese ob/ob mice were randomly divided into 5 groups: obese control, GGEx15, GGEx16, GGEx17, and GGEx18. After mice were treated with several kinds of GGEx for 11 weeks, body weight gain, feeding efficiency ratio, plasma lipid and glucose metabolism. Results : 1. Compared with obese controls, GGEx-treated mice had lower body weight gain and feeding efficiency ratio, the magnitudes of which were prominent in GGEx16 and GGEx18. 2. Consistent with their effects on body weight gain, GGEx16 and GGEx18 not only decreased plasma triglycerides levels, but also increased HDL-cholesterol concentration. 3. CT analysis revealed that visceral fat areas were decreased in all treatment groups compared with obese control mice. The decrease in visceral fat area was prominent in GGEx16 and GGEx18, although they were not statistically significant. 4. The size of adipocytes were significantly decreased by GGEx18, whereas the adipocyte number per unit area was significantly increased, suggesting that GGEx18 decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGEx16 and GGEx18, and the inhibitory effect was most effective in GGEx18. 5. Plasma GOT and GPT concentrations were significantly lower following GGEx16 and GGEx18 treatment compared with obese controls. Organ weights were not changed by GGEx treatment, indicating GGEx do not show any toxic effects. Conclusions : These results suggest that GGEx may regulate obesity. Of the 4 compositions, GGEx18 seems to be most effective in improving obesity and lipid disorders.

고지방식이 유도 비알콜성지방간 마우스 모델에서 육신국(六神麯)의 지방간(脂肪肝) 개선효과 (Massa Medicata Fermentata improves fatty liver in high fat diet-fed nonalcoholic fatty liver disease's mouse model)

  • Objectives : This study was undertaken to verify the effects of Massa Medicata Fermentata (MMF) on nonalcoholic fatty liver disease (NAFLD) using high fat diet-fed male mice. Methods : Fifty four male C57BL/6N mice (age matched) were used for all experiments. Nine standard chow diet-fed mice were used as normal group and forty five high fat diet-fed obese mice were randomly divided into 5 groups: control, atorvastatin-10mg/kg, MMF(1)-62.5mg/kg, MMF(2)-125mg/kg and MMF(3)-250mg/kg. After all groups were treated with several kinds of diets for 8 weeks, we measured body weight gain, adipose tissue weights, plasma lipid and glucose metabolism, visceral organ weights, histological analysis for liver on the mice. Results : MMF-treated mice had lower body weight gain compared with controls. Among MMF-treated mice, the effect was magnified in MMF(2). MMF(3)-treated mice had lower blood plasma total cholesterol (TC) and glucose level compared with controls. MMF decreased hepatic lipid accumulation, liver fibrosis and liver inflammation of mice compared with controls. The effects was maximized in MMF(2) and atorvastatin. Blood plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyltransferase (${\gamma}$-GT) concentrations tends to be decreased by MMF compared with controls. Blood plasma AST, ALT, ${\gamma}$-GT concentrations and organ weights were not changed by MMF, indicating that all three kinds of MMF do not show any hepatotoxicity. Conclusions : These results suggest that MMF improves NAFLD by reducing body weight gain, hepatic lipid accumulation, liver fibrosis, liver inflammation.

  • This treatise is written in order to solve the important contradiction between the two theories; in oriental medicine psychological function is responsible for heart, but in western one it is responsible for brain. So we take the methods of studying in the aspects of morphological characteristics(MC) and visceral manifestation theory(VMT, 藏象論) and others about two organs-heart and brain. Brain(頭腦) is preferred to understand as a structure which is manifesting mental activity of heart. So the brain can be named with external heart(外心) corresponding to the relation of kidney(外 and external kidney. Saying conversely, the nutritional foundation of the mental function is the blood of heart, but the enlightening and insightful features of mentality make it's own residence move to the organ in the uppermost and positive site, that is head. And the close relationships on mental functions between heart and brain were discussed in various aspects, like investigation on east and west etymological literature, or Jiu gong and Taoist theory as well as Me and VMT, These understandings can make us know about the pathology of brain by itself. It has deep relations with heart fire and heart blood and kidney essence, and gastrointestinal function and liver with lung additionally. In another point, it makes the highly complicated psychological functions to be explained free from body relatively, and so can do a role in the complement of the strict 5 viscera theory.

새로운 관절염 치료제 CONP01의 생식독성연구: 랫드 배 · 태자 발생시험 (Reproductive Toxicity Study of CONP01, a New Antiarthritic Agent: Developmental Study in Rats)

  • A developmental study of CONP01, a new antiarthritic agent, was conducted in Sprague-Dawley rats. Dosage of CONP01 0, 111, 333, and 1000 mg/kg/day were administered to dams orally from day 6 to day 16 of gestation. Two-third of dams per group were subjected to caesarean section on day day 20 of pregnancy for examination of their fetuses, and the remaining one-third of dams per group were allowed to deliver naturally for postnatal examination of their offspring. There was no change in the dams body weights, food consumptions, specific clinical sings and gross findings. There was significant decrease only in the absolute and relative weights of right ovary in 111 mg/kg treatment group, when compared with the vehicle control, whereas other organ weights were not changed. Moreover, no increase in the frequencies of external, visceral and skeletal malformation of fetuses were observed in the treated groups. These results suggest that the oral NOAEL (no observed adverse effect level) of CONP01 may be over 1,000 mg/kg in dams and fetuses of rats.

야관청혈탕이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 영향 (Effect of Yagwan-cheunghyeoltang on Obesity in Rats Induced by High Fat Diet)

  • Background : Obesity, the syndrome caused by a high fat diet, is a disease. At the same time, obesity causes diabetes mellitus, hyperlipidemia, and cardiovascular disease. Recently, its prevalence rate is increasing. Yagwan-cheunghyeoltang (YCT) used in this experiment is the prescription of Yagwanmoon added to Cheunghyeol-tang which is reported to be very effective in weight loss controlling and serum cholesterol. It is also reported that Yagwanmoon has significant antioxidant effects and YCT has a significant effect on blood glucose control. Objectives : This study was conducted to experimentally evaluate the effects of YCT on obesity in rats induced by high fat diet. Methods : The experiment was conducted with 4-week-old male rat s divided into 5 groups. They were a normal diet group, a high fat diet group, a positive drug control group, a 1% YCT group, and a YCT 3% group, and were tested for eight weeks. After four weeks of inducing obesity by a high fat diet, rats were allowed to lose weight by following the normal diet group, approximately 30% compared with 10 rats in each group were determined as still obese. Changes in body weight and organ weight and serum cholesterol, triglyceride, glucose-density, low density lipoprotein cholesterol, antioxidant activity were checked. Results : In the experimental groups, we observed weight loss and visceral fat reduction, improvement of liver function, reduction of serum glucose, activation of HMG-CoA reductase inhibitor, reduction of concentrations of leptin and it showed a significant effect on antioxidants and lipid peroxidation. Conclusions : YCT has significant effects on the regulation of hyperlipidemia and lipid peroxidation associated with obesity and has significant effects on, antioxidants and lipid peroxidation, too. Additional clinical studies are needed.

Interaction of physical trainings and coffee intakes in fuel utilization during exercise in rats

  • This study investigates the impact of exercises, coffee intakes, and physical trainings on fuel utilization in rats. Ninety-six rats were fed a control diet with either water (C) or coffee (CF; 0.12 g freeze-dried instant coffee/100 g body weight/d). Additionally, the animals go through physical training (TC and TCF) or no training (NTC and NTCF) for 4 weeks. For physical training, animals have to exercise on treadmills for 30 minutes (5 d per week, $15^{\circ}$ incline, 0.5-0.8 km/h). At the end of week 4, the animals in each group were subdivided into three exercise groups: before exercise (BE), during exercise (DE), and after exercise (AE). The DE rats exercised on treadmills for 1 hour immediately before being sacrificed. Hemoglobin, hematocrit, glucose, glycogen, protein, triglyceride (TG), and free fatty acid (FFA) levels in the plasma, liver, and skeletal muscle of the rats were compared accordingly. Organ weights were also measured. Coffee-training interaction had a significant impact on heart weight, visceral fat, hemoglobin, hematocrit, liver glycogen in DE and AE, and liver triglyceride in DE and AE. Exercise (meaning exercised on a treadmill for 1 hour immediately before being sacrificed) training interaction was significant in liver glycogen, muscle glycogen in control diet and control diet with coffee, FFA and muscle TG levels at control diet with coffee group. Exercise-coffee interactions significantly influenced the FFA with no training groups. Exercise-coffee-training interaction significantly effects on FFA, Liver TG and Muscle TG. Coffee intakes can increase lipolysis during exercising but coffee consumptions delay the recovery of liver glycogen levels in trained rats after exercising. Coffee can be an effective ergogenic aid during exercise for physically trained rats.

고지방식이로 유도된 비만 생쥐모델에서 저령차전자탕의 항비만 효과 (Anti-obesity Effect of Jeoreongchajeonja-tang in a High-fat Diet-induced Obesity Mouse model)

  • Objective This study investigated the effects of Jeoreongchajeonja-tang in a high-fat diet-induced obesity mouse model. Methods The study examined 9-week-old male mice (C57bl/6J) divided into four groups: the normal(C57bl/6J-Nr), control (high-fat diet only; HFD-CTL), positive-control (high-fat diet with Garcinia cambogia), and experimental (high-fat diet with Jeoreongchajeonja-tang; HFD-JCT) groups. After 7 weeks, the body weight, food efficiency ratio, organ weight, and visceral fat weight of the mice were measured. Blood serum tests, mRNA, liver histopathology, and epididymis adipocytes were also examined. Results Compared with the Control(HFD-CTL) group, the Experimental(HFD-JCT) group given Jeoreongchajeonja-tang showed significant reductions in absolute body weight and food efficiency ratio. The serum alanine aminotransferase, total-cholesterol, triglyceride, low-density lipoprotein (LDL)-cholesterol, high-density lipoprotein (HDL)-cholesterol, insulin-like growth factor-1, and leptin levels were significantly lower in the experimental group than in the control group. The serum adiponectin levels were significantly higher in the experimental group than in the control group. Compared with the control group, the experimental group showed significant reductions in absolute abdominal subcutaneous fat, epididymal adipose tissue, kidney adipose tissue, intestine adipose tissue, and liver, kidney and spleen adipose tissue weights. The C/EBP-${\beta}$, leptin, and SREBP1c/ADD1 mRNA expression were significantly lower in the experimental group than in the control group, while the UCP-2 and adiponectin mRNA expression were significantly higher. Compared with the control group, the experimental group showed a significant reduction in the absolute adipocyte area in the liver and epididymal adipose tissue. Conclusion Jeoreongchajeonja-tang has an anti-obesity effect. Additional clinical studies are expected.

위(胃)의 생리특성 및 위기(胃氣)와 진장맥(眞藏脈)과의 상관관계에 대한 연구 (Study on Physiological Character of Stomach, and Correlation of Stomach Qi(胃氣) and True Visceral Pulse(眞藏脈))

  • By examining the relationship between physiological character of stomach and true viscera pulse(眞臟脈) with "Huangdinejing(黃帝內經)", the study shows these fallowing results. Stomach is not only one of the six bowels(六腑), it represents them. It is called as yang brightness of foot(足陽明) because it has relation with function of earth among five phases(五行), great storage(倉廩) and root of five visceras and six bowels(五臟六腑), has close relationship with triple energizers(三焦). Stomach is an important organ that feeds acquired qi(後天之氣) based on food and stomach qi (胃氣) which is a transformed form by going through digestion, is significant for life conservation. In human body, the activation of stomach qi can be known by pulse. The true viscera pulse is a pulse which predicts the period of death. If stomach qi exsits, viscera qi can be led to greater yin of hand(手太陰), however, when it does not exist, five viscera qi can not be led to greater yin of hand and this kind of situation appears the true viscera pulse. Hence, by knowing the relation between stomach qi and five viscera qi, the condition of normality, disease, and death can be known. In "Hwangdineijing" it says, stomach qi, shortly string like pulse(微弦), shortly surging pulse(微鉤), shortly floating pulse(微毛), and shortly sunken pulse(微毛) shown in four seasons are normal pulse. And also it emphasizes, normal pulse is made when four seasons, five viscera pulse, and stomach qi are in harmony. In conclusion, stomach qi is based on food for well being, and a standard of judging people whether they are living in a healthy life or not.
