  • Objective : Lonicerae Flos has detoxifying properties and been used as antipyretic, antibacterial and antitumor. Fermentation of herbal medicine is known to increase the absorption, enhance effectiveness, decrease herbal toxicity and reduce side-effects. This study was performed to measure the effects of fermented Lonicerae Flos on influenza A/WSN (H1N1) virus replication. Material and Methods : Lonicerae Flos was fermented by Lactobacillus casei PM1. Fermented Lonicerae Flos was treated for 12 hours to MDCK (Mardin Darby canine kidney) cells, then cell-virulence was observed by MTT assay for 12 hours, 24 hours, and 36 hours after treatment. Following cases were conducted for 0, 10, 100, and $1000{\mu}g/ml$ concentrations of fermented Lonicerae Flos under the same time-frame; the fermented Lonicerae Flos was treated to MDCK cells before and after contamination by A-type influenza virus. The fermented Lonicerae Flos and the virus were mixed directly. The influence was observed by MTT assay and plaque assay. Results : These findings suggest that the fermented Lonicerae Flos inhibited the virulence of influenza A virus in MDCK cells and suppressed the plaque forming colonies induced by influenza A virus. Furthermore, pretreatment with fermented Lonicerae Flos was more effective than post-treatment. The titer of influenza virus was reduced for all before and after influenza A virus inoculation.

  • The hepatitis B virus(HBV) is a small, enveloped virus with a circular, double-stranded DNA genome. HBV causes active and chronic hepatitis worldwide, including Korea, and is considered to be a major factor for liver cirrhosis and hepatocellular carcinoma. In contrast to the wealth of knowledge on the gene structure and expressional regulation, immunological and pathological mechanisms for HBV-induced hepatocellular injury are not well known. In the present study, vaccinia virus which has been demonstrated to be a useful eukaryotic expression vector was used to clone the gene for HBV surface antigen, L(S+preS2+preS1). The recombinant vaccinia virus vector, pMJ-L, which contains L surface antigen gene of adr-type HBV was constructed, and subseouently used for making recombinant vaccinia virus VV-$\textrm{HBV}_{L}$. Expression of the HBV antigen was examined by immunofluorescent antibody (IFA) test using mouse monoclonal anti-hepatitis B surface antigen. HBsAg was detected in the recombinant virus indicating that the VV-$\textrm{HBV}_{L}$ expressed S antigen successfully. The HBV-Vaccinia Virus recombinant obtained in this study is currently being used for studying the immunological aspects of HBV infection.

  • The primary culture of human nasal epithelial cells was performed using the inferior nasal turbinate tissues, and infected with Hantaan virus to examine the hypothesis of airborne transmission of Hantaan virus in humans. The primary culture cells were identified as epithelial cells by morphologic and immunologic analyses. The viral antigens were detected in the primary human nasal epithelial cells infected with Hantaan virus by immunofluorescence staining. The ICAM-1 induction by Hantaan virus or $IFN-{\gamma}$ was examined in the primary human nasal epithelial cells and human lung fibroblasts (WI-38). Hantaan virus induced the surface ICAM-1 in WI-38 cells in a time-dependent manner, and $IFN-{\gamma}$ induced the surface ICAM-1 in a dose-dependent manner in HNEC and WI-38 cells. These results revealed that the human nasal epithelial cells are susceptible to Hantaan viral infection supporting the hypothesis of airborne transmission of Hantaan virus in humans. The human lung fibroblasts also might have an important role in the pathogenesis of Hantaan virus through the induction of ICAM-1.

  • The biological wastewater treatment system is known to have an important role in reducing the quantify of enteric virus in water environments. To clarify the roles of activated sludge microbes in decreasing the virus infectivity, the behavior of the virus in bacteria, protozoa, and metazoa was examined by pure or mixed culture system using poliovirus type 1(Lsc, 2ab strain). In the bacterial culture systems, the virus infectivity in the liquid phase decreased by a reversible adsorption of the virus to the bacteria or bacterial flocs. On the other hand, in the protozoa and the metazoa culture systems using T. pyriformis and P. erythrophthalma, respectively, with a variety of bacterial strains as prey, the main virus decrease mechanism of reversible adsorption in early stage was changed to irreversible predation, which was not eluted in this study. The virus decrease was more effective in the P. erythrophthalma culture system, which had high predation and floc forming abilities. However, in the mixed culture system of Z. ramigera and P. erythrophthalma, the more rapid reversible adsorption of virus to Z. ramigera flocs preceded the irreversible predation of P. erythrophthalma.

  • Respiratory syncytial virus (RSV) and influenza virus are the most significant pathogens causing respiratory tract diseases. Composite vaccines are useful in reducing the number of vaccination and confer protection against multiple infectious agents. In this study, we generated fusion of RSV G protein core fragment (amino acid residues 131 to 230) and influenza HA1 globular head domain (amino acid residues 62 to 284) as a dual vaccine candidate. This fusion protein, Gcf-HA1, was bacterially expressed, purified by metal resin affinity chromatography, and refolded in PBS. BALB/c mice were intranasally immunized with Gcf-HA1 in combination with a mucosal adjuvant, cholera toxin (CT). Both serum IgG and mucosal IgA responses specific to Gcf and HA1 were significantly increased in Gcf-HA1/CT-vaccinated mice. To determine the protective efficacy of Gcf-HA1/CT vaccine, immunized mice were challenged with RSV (A2 strain) or influenza virus (A/PR/8/34). Neither detectable viral replication nor pathology was observed in the lungs of the immune mice. These results demonstrate that immunity induced by intranasal Gcf-HA1/CT immunization confers complete protection against both RSV and homologous influenza virus infection, suggesting our Gcf-HA1 vaccine candidate could be further developed as a dual subunit vaccine against RSV and influenza virus.

  • The virus infection rates and disease symptoms of three major citrus viruses, citrus tristeza virus (CTV), satsuma dwarf virus (SDV), and citrus tatter leaf virus (CTLV), were investigated at 35 citrus orchards in Cheju Island from 1995. The infection rates of CTV, SDV, and CTLV were 69.8%, 8.6%, and 9.3%, respectively. However, depending on cultivars there were significant differences in the infection rates. The infection rates of CTV were highest in early satsuma mandarin (Citrusunshiu) with 80.9% and lowest in very early satsuma mandarin with 51.9%. In SDV, the highest was in very early stasuma mandarin with 23.1% and the lowest was in early satsuma mandarin with 6.3%. And the highest infection rate in very early satsuma mandarin with 17.9% and the lowest in tangors with 7.3% in CTLV. The symptoms of virus-infected citrus were very diverse; small and abnormal shape of fruits, abnormal leaves such as narrow boat and small spoon shapes of leaves, stem-pitting on the twig, bud-union crease and swelling of the graft part, reduction of the plant vigor and poor yields.

  • Hantavirus is a genus of the Bunyaviridae family causing two serious diseases, hemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS). Puumala virus is a member of hantavirus originally found in Europe, and its natural reservoir is Clethrionomys glareolus. It is also associated with the human disease nephropathia epidemica, a milder form of HFRS. To identify the hantaviruses in bats, bats were collected from Jeong-Sun, Won-Joo, Chung-Ju and Hwa-Cheon area in Korea, and nested RT-PCR was performed with serotype specific primer from M segment. Interestingly, Puumala virus was detected in bats (Rhinolophus ferrum-equinum) only from Won-Joo. The 327 bp nested RT-PCR product, was sequenced. The sequence database search indicates that the sequence is homologous to the published sequence of Puumala viruses. The sequence similarities were ranged from 71% to 97%. The highest sequence similarity was 97% with Puumala virus Vranicam strain, and the lowest was 71% with Puumala virus K27 isolate. Puumala virus Vranicam strain was isolated from a bank vole (Clethrionomys glareolus) in Bosnia-Hercegovina. Puumala virus K27 was isolated from human in Russia. This analysis confirms that bats (Rhinolophus ferrum-equinum) in Korea are natural reservoir of Puumala virus.

  • A survey of virus infection in garlic plants cultivated in Korea was conducted for three years. Most virus-infected garlic plants (Allium sativum) showed typical symptoms on the leaves such as yellow mosaic, stripes, and distortion. Through immunosorbent electron micro-scopy and RT-PCR analysis, the complex mixtures of viruses including garlic viruses of the genus Allerivirus, gaylic strain of Leek yellow stripe virus of the genus Potyvirus, and Garlic latent virus of the genus Carlavirus were identified in the virus-infected garlic plants. Among these viruses, Allexivirus was the most frequently detect-ed in the regions surveyed. Using sets of differential primers for Allexivirus genomes, two members of the genus were amplified and sequenced from the purified viruses. The deduced amino acid sequences for the coat proteins and the nucleic acid binding proteins of two viruses showed high homologies to Garlic virus A (CarV-A) and Garlic virus D (GarV-D) of Allekivirus. This is the first report of GarV-A and GarV-D in Korea. This suggests that Allexivirus in gavlic plants in Korea was mixed and varied. Phylogenetic analyses showed that the genus Allexivirus was diversi(ied by the processes of accumulation and evolution of viruses in garlic plants due to the long period of repeated vegetative propagation.
