대추나무 미친병에 관한 연구 (I) -병식물의 내외형태학적 특징 및 그 명명에 대해서-

  • Since the peculiar virus disease of chinese date tree (Zizyphus jujuba Mill. var. inermis Rehd.) has been noted in South Korea around 1950, 70% to 80% of the economically important trees have been either completely destroyed or infected with the virus, severe damage has been noted, particularly, across the area ranged from middle east to the middle part of Korea, including Seoul area. Yoon-Koock-Byung in 1958 first reported the disease and descirbed it might be caused by a kinds of yellows. But he did not conform in his paper that the disease is pecisely caused by yellows virus. The authors, hereby intend to identify the true cause of the desease of the chinese data tree by studying the external symptoms of the disease and the internal morphological characteristics of the diseaset plant which shows various abnormalities in contrast to the healthy checks. In view of fact that leaves of the infected plants become yellowish in color similar to the peach yellows, aster yellows, it is likely to be identifiable as the common yellows. Furthermore, the abnormal characteristics observed by the authors are as follow: The floral organs such as petals, sepals, stamens, and pistil turn into vegetative leaves, the leaves on heavily infected plant appear as small sized one and also showing as a common witch's broom like symptom. There are also an occuring of numerous advantitious shoots developed from both of stems and roots. The amount of photosynthetic starch grains increases in parenchymatous cells, necrosis takes place in mesophyll, Particularly, Palisade Parenchyma in the leaves of infected plants are distinguished in contrast to the healthy checks. From the symptoms and the present experimetns described above, the authors are believed that the disease of chinese data tree is not caused by the yellows. It appears the disease is rather similar to the symptoms of sandal spike virus which was noted in India early in this centry. But the host plant of standal disease, Santalum albun L. and the insect vector, Jassus indicus Wal., have never been reported in Korean flora and the founa. The termperature and the otehr environmental factors is quite different Korea and India. Thus the authors believe that the peculiar disease must be an endemic new virus origin in Korea and must be called as "shoot cluster disease of chinese date tree."

STRAINS OF FOOT-AND-MOUTH DISEASE VIRUS IN DIFFERENT DISTRICTS OF BANGLADESH

  • An investigation was carried out to find out the strains of foot-and-mouth disease (FMD) virus in 24 districts of Bangladesh. A total of 505 FMD virus samples were collected from June, 1989 to June, 1991 and tested by complement fixation test (CFT). Of these, 276 (54.7%) were found positive for different strains of FMD virus and the rest 45.3% were either negative or anticomplementary. Strains identified were O, C, Asia-1 and sub-strains $A_5$ and $A_{22}$. Strain O was found to be most prevalent(39.8%) followed by Asia-1 (5.7%), C (5.3%), $A_5$ (3.4%) and $A_{22}$ (0.4%). Prevalence of sub-strain $A_5$ was reported for the first time in Bangladesh. District-wise typing of FMD virus has been done which would be helpful for appropriate vaccination programme in different districts of Bangladesh for control of the malady.

Incidence and Distribution of Virus Diseases on Cucumber in Jeonnam Province During 1999-2002

  • Disease incidences of cucumber virus diseases in Jeonnam province were 52.5%, 16.1%, 35.2%, and 50.9% in 1999, 2000, 2001, and 2002, respectively. Rod- and flexuous rod-shaped virus particles were observed with the frequencies of 63.2% and 10.5%, respectively from the samples collected in 1999 under EM observation. Rod-shaped virus particles are considered as tobamovirus while flexuous rod shaped particles are considered as potyviruses. To further confirm their nature, total of 312 diseased virus samples were collected from 2000 to 2002, and tested by RT-PCR. Disease incidences of tobamoviruses including Cucumber green mottle mosaic virus and Kyuri green mottle mosaic virus were 48.7% and 3.8%, respectively while those of potyviruses including Zucchini yellow mosaic virus, Papaya ringspot virus, and Watermelon mosaic virus were 15.7%, 9.3%, and 5.1%, respectively. Interestingly, Cucumber mosaic virus was hardly detected. About 5.8% of tested samples were infected with more than one virus. Tobamovirus infection was consistently observed from September to December regardless of planting time, whereas infection of potyviruses was observed in many cucumber cultivating areas where it was planted in September and October.

  • Current overseas research reveals that among the pathogens causing flacherie of silkworm, damage by infectious flacherie virus is the most serious, but little research in this fold has been reported in Korea. This experiment was undertaken to observe the occurrence of infectious flacherie virus by means of biological environmental conditions associated with occurrence of virus discase and interaction of the virus of flacherie and Bacillus spp. isolated from flacherie silkworm, and to determine ways to check infection by the virus during the rearing of silkworms. The results obtained are as follows: 1. The pathogen, infectious flacherie virus observed in Korea, is proved to be round in shape and 26-30m$\mu$ in diameter under observation with electron microscope, 2. The infectious flacherie virus-disease occurred apparently in conditions of nutritional disturbance such as shortage of diet or rearing in high temperature and humidity during the 3-4th instar. 3. The percentage of disease-occurrence was increased remarkably, and the latent period was shortened in the case of simple inoculation of virus suspension as compared with the suspension added with bacteria. 4. The application of calcium hydroxide in the silkworm-rearing bed is able to check infection of virus disease.

Development and Characterization of Monoclonal Antibodies against Nucleoprotein for Diagnosis of Influenza A Virus

  • Influenza, which is a highly contagious disease caused by the influenza A virus, continues to be a major health concern worldwide. Although the accurate and early diagnosis of influenza virus infection is important for controlling the spread of this disease and rapidly initiating antiviral therapy, the current influenza diagnostic kits are limited by their low sensitivity. In this study, we developed several new influenza nucleoprotein (NP)-specific monoclonal antibodies (mAbs) and compared their sensitivity and specificity of those with commercially available anti-NP mAbs. Three mAbs, designated M24.11, M34.3, and M34.33, exhibited higher reactivities to recombinant NPs and A/Puerto Rico/8/1934 (H1N1) viral lysates compared with the commercial mAbs, as assessed using enzyme-linked immunosorbent assays. M34.3 and M34.33 showed higher reactivities with A/California/04/09 (pandemic H1N1) and A/Philippines/2/82 (H3N2) viral lysates than the commercial mAbs. In contrast, M24.11 had marked reactivity with H3N2 but not with pandemic H1N1. Immunofluorescent confocal microscopy showed that the three mAbs effectively detected the presence of influenza virus in lung tissues of mice infected with A/Puerto Rico/8/1934. These results indicate that the newly developed M34.3 and M34.33 mAbs could be useful for the development of influenza diagnostics.

Hepatitis C Virus Nonstructural Protein 5A Interacts with Immunomodulatory Kinase IKKε to Negatively Regulate Innate Antiviral Immunity

  • Hepatitis C virus (HCV) infection can lead to chronic hepatitis, liver cirrhosis, and hepatocellular carcinoma. HCV employs diverse strategies to evade host antiviral innate immune responses to mediate a persistent infection. In the present study, we show that nonstructural protein 5A (NS5A) interacts with an NF-κB inhibitor immunomodulatory kinase, IKKε, and subsequently downregulates beta interferon (IFN-β) promoter activity. We further demonstrate that NS5A inhibits DDX3-mediated IKKε and interferon regulatory factor 3 (IRF3) phosphorylation. We also note that hyperphosphorylation of NS5A mediates protein interplay between NS5A and IKKε, thereby contributing to NS5A mediated modulation of IFN-β signaling. Lastly, NS5A inhibits IKKε-dependent p65 phosphorylation and NF-κB activation. Based on these findings, we propose NS5A as a novel regulator of IFN signaling events, specifically by inhibiting IKKε downstream signaling cascades through its interaction with IKKε. Taken together, these data suggest an additional mechanistic means by which HCV modulates host antiviral innate immune responses to promote persistent viral infection.

Comparison of Virus Elimination Methods for Disease-free Seedlings of the Apple Dwarfing Rootstock (사과 왜성대목 무독묘 생산을 위한 바이러스 제거 방법 비교)

  • Apple (Malus domestica) is one of the most economically important fruits in Korea. But virus infection has decreased sustainable production of apple and caused the serious problems such as yield loss and poor fruit quality. Virus or viroid infection including Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV) and Apple scar skin viroid (ASSVd) has been also reported in Korea. In many cases, apple is infected with virus and viroid with no specific symptoms, the damage caused by the virus are unaware significantly. In our research, we tried to eliminate viruses in the rootstock for the disease-free seedlings of the apple dwarfing rootstock M.9 and M.26. The method of virus elimination was meristem culture, heat($37^{\circ}C$, 6weeks) treatment and chemistry($Ribavirin^{(R)}$) treatment. The analytical methods commonly used for the detection of virus is Enzyme-linked Immuno-Sorbent Assay(ELlSA) and Reverse Transcription-polymerase Chain Reaction(RT-PCR). RT-PCR method was more 30% sensitive than ELISA method. Efficiency of method eliminate virus appeared meristem method > heat treatment > chemistry treatment. The higher acquisition rate of disease-free seedlings is 30~40% on meristem treatment. In meristem treatment, the apple dwarfing rootstock M.9 gained infection ratio of ACLSV, ASPV and ASGV were 45%, 60% and 50% respectively. In the apple dwarfing rootstock M.26, infection ratio of ACLSV, ASPV and ASGV were 40%, 55%, 55%, respectively. Based on our results, it was found that most effective method of disease-free seedlings apple dwarfing rootstocks was by meristem treatment than heat method and chemistry treatment.

Development of a Reliable Technique to Eliminate Sweet potato leaf curl virus through Meristem Tip Culture Combined with Therapy of Infected Ipomoea Species

  • In vitro elimination of Sweet potato leaf curl virus (SPLCV) from infected sweet potato is difficult due to low number of virus-free plants obtained from meristem tip culture and long growth period required for the virus detection. In this study, efficient production of the SPLCV-free sweet potato by in vitro therapy coupled with a PCR assay for virus detection was investigated. Infected shoots cultured on Murashige and Skoog medium were treated at three different temperatures for 7 weeks followed by meristem tip culture on the medium with or without ribavirin at 50 mg/L. The regenerated plantlets were tested for virus infection by a PCR assay. The results showed that the both heat- and cold-treatments, and addition of the ribavirin did not have significant effect on efficiency of the virus elimination. The meristem size, however, greatly affected the survival rate. Meristems sized over 0.4 mm survived better than smaller ones (0.2-0.3 mm). The PCR assay was approved to be a rapid, sensitive and reliable for the SPLCV detection in regenerated plantlets. Therefore, combination of cultivating meristem tips sized 0.4-0.5 mm on the medium at $22^{\circ}C$ without ribavirin and detection of SPLCV in the regenerated plantlets by the PCR assay was an efficient system for the SPLCV elimination from infected sweet potato.

  • An acute fatal infectious disease in rabbits has been outbroken in Korea since 1985. This disease has been characterized as an acute hepatitis caused by viruses. However, viral pathogenesis in rabbit viral hepatitis leading to sudden death remain unclear. This report dealt with the electron microscopic findings on the spleen of experimentally infected rabbits, because spleen is one of the affected organs which have high titer of virus by a haemagglutination test. A typical crystalline array of virus was not found in the splenic cells of infected rabbits with acute hepatitis. Virus-like particles were seen within the phagosome of macrophages of the spleen. Ultrastructural changes in the spleen were severe with the lapse of time after inoculation. From these results, virus-like particles in the spleen were supposed to be phagocytosed by macrophage during viremia, while active replication of virus occurred in the liver. It was concluded that sudden death in this viral disease was caused by hepatic coma and/or circulatory disturbance.

Differential diagnosis among Marek's disease, reticuloendotheliosis and avian leukosis by polymeras chain reaction (중합효소연쇄반응을 이용한 닭 종양성 질병의 감별진단에 관한 연구)

  • The present study attempted to apply polymerase chain reaction (PCR) to develop a rapid differential diagnosis among Marek's disease, reticuloendotheliosis and avian leukosis. The primers chosen to detect Marek's disease virus (MDV) flank the 132bp tandem direct repeat of the MDV genome. The primers selected for reticuloendotheliosis virus (REV) and avian leukosis virus (ALV) are based on proviral long terminal repeats of spleen necrosis virus and Rous-associated virus-2 genomes, respectively. The specific PCR products of MDV, REV and ALV were observed with each primer and the reaction was not cross-reacted among the viruses. MDV-specific DNA was also amplified from the MDV-induced lymphoma (MDCC-MSB1) but not from the REV-induced tumor and ALV-induced lymphoma (LSCC-1104B1). In addition, proviral DNA of REV from REV-induced tumor and proviral DNA of ALV from ALV-induced lymphoma were also amplified by REV-specific and ALV-specific PCRs, respectively. Therefore these three PCR methods may be used to rapidly differentiate among MDV, REV and ALV-associated tumors in diagnosis.
