한국에서의 감자 바이러스병 발생과 그 연구에 대한 고찰 (Review on the Occurrence and Studies of Potato Viral Diseases in Korea)

  • The occurrence of potato(Sotanum tuberosum) viral diseases caused by Potato virus X(PVX), Potato virus Y (PVY), Potato leafroll virus(PLRV), Potato vims S(PVS), Potato virus M(PVM), Potato virus A(PVA), Potato virus T(PVT), Alfalfa mosic virus(AIMV), Tobacco mosic virus(TMV), Potato mop top virus(PMTV) Tobacco rattle virus(TRV) and Potato spindle tuber viroid(PSTVd), potato witches' broom phytoplasma, have been identified so far in Korea. Major viral diseases such as PVX, PVY and PLRV had been studied more deeply, however, the others are just identified and only partially characterized since the first study on the relation between PVX nucleic acid and virus protein by Kim in 1961. The most studies on potato viral diseases are mainly focused on the problems of seed potato production. The National Alpine Agricultural Experiment Station(NAAES), since it began its activities in 1961, has given special attention to this problem by doing studies to identify, characterize and control potato virus diseases. This effort resulted in the development of new potato virus detection methods as a basis for elaborating new method of control, such as the production of seed potato free of virus and the selection of new virus-resistant transgenic potatoes. The further studies of potato viral diseases required would be fallowings: the continuous monitoring for the occurrence of identified or not identified potato viruses in Korea, the isolation of resistant viral genes, the development of control method for the non-persistently transmitted viruses like PVY, special vectors such as nematode and fungus transmitted viruses, TRV and PMTV and the development of control methods against potato viral diseases by viral cross protection, therapy, transgenic plant, and the use of the agents or molecules, such as virus inhibitors and antiviral proteins, etc., blocking viral replication.

무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구 (Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

Porcine epidemic diarrhea virus: an update overview of virus epidemiology, vaccines, and control strategies in South Korea

  • Porcine epidemic diarrhea virus (PEDV) has posed significant financial threats to the domestic pig industry over the last three decades in South Korea. PEDV infection will mostly result in endemic persistence in the affected farrow-to-finish (FTF) herds, leading to endemic porcine epidemic diarrhea (PED) followed by year-round recurrent outbreaks. This review aims to encourage collaboration among swine producers, veterinarians, and researchers to offer answers that strengthen our understanding of PEDV in efforts to prevent and control endemic PED and to prepare for the next epidemics or pandemics. We found that collaboratively implementing a PED risk assessment and customized four-pillar-based control measures is vital to interrupt the chain of endemic PED in affected herds: the former can identify on-farm risk factors while the latter aims to compensate for or improve weaknesses via herd immunity stabilization and virus elimination. Under endemic PED, long-term virus survival in slurry and asymptomatically infected gilts ("Trojan Pigs") that can transmit the virus to farrowing houses are key challenges for PEDV eradication in FTF farms and highlight the necessity for active monitoring and surveillance of the virus in herds and their environments. This paper underlines the current knowledge of molecular epidemiology and commercially available vaccines, as well as the risk assessment and customized strategies to control PEDV. The intervention measures for stabilizing herd immunity and eliminating virus circulation may be the cornerstone of establishing regional or national PED eradication programs.

Development of a Multiplex Polymerase Chain Reaction Assay for Detecting Five Previously Unreported Papaya Viruses for Quarantine Purposes in Korea

  • There are concerns about the introduction and spread of plant pests and pathogens with globalization and climate change. As commercial control agents have not been developed for plant viruses, it is important to prevent virus spread. In this study, we developed a multiplex polymerase chain reaction (PCR) detection method to rapidly diagnose and control three DNA (papaya golden mosaic virus, Lindernia anagallis yellow vein virus, and melon chlorotic leaf curl virus) and two RNA (papaya leaf distortion mosaic virus and lettuce chlorosis virus) viruses that infect papaya. Specific primer sets were designed for the virus coat protein. Performing PCR, clear bands were observed with no non-specific reaction. Our multiplex PCR method can simultaneously detect small amounts of DNA/RNA to diagnose five viruses infecting papaya and prevent the spread of the virus.

소 수정란의 전염성질병 예방 (Infectious Disease Control of Bovine Embryos)

  • Based on the current importing and exporing regulations for disease control of embryo transfer, some important microorganisms and their control possibilities are reviewed. The results reviewed were sumrnarized as follows: 1. Regulations regarding to the import of embryos vary between importing and exporting countries, but exporting countries examine the donor and embryos for the heaith certification by the requirements of importing countries. 2. Organisms that infect the gametes are 5 kinds of viruses and the diseases caused by them could not be controlled or eradicated using embryo transfer. 3. Organisms that do not infect the gametes are 4 kinds of viruses and the causal organisms are potential candidates for control or eradication by embryo transfer. 4. Organisms that penetrate the zona pellucida and infect the embryo are 6 kinds of viruses including bovine viral diarrhea virus. 5. Organisms that cannot penetrate the zona pellucida or do not infect the embryo are 15 kinds of viruses and the removal from their contaminations are recommended by proper washing procedure and antisera treatment. Bovine and porcine parvovirus, porcine pseudorabies virus and vesicular stomatitis virus are included in these organisms. 6. Bovine embryos that artificially exposed to various pathogenic organisms such as bovine herpes virus, IBR virus, bluetongue virus, bovine viral diarrhea virus and Brucella abortus in vitro are discussed about their infection by several treatments.

Occurrence of Two Tobamovirus Diseases in Cucurbits and Control Measures in Korea

  • Two Tobamoviruses, Cucumber green mottle mosaic virus (CGMMV) and Zucchini green mottle mosaic virus (ZGMMV), occurred in Korea in 463 ha in 1998, 33.9 ha in 1999, and 44.2 ha in 2000. CGMMV was detected in watermelon, cucumber, oriental melon, and melon, whereas ZGMMV was mainly detected in zucchini squash. Thirty-six CGMMV isolates wee classified into three types by analysis of single strand cDNA conformational polymorphism (SSCP) of the coat protein gene. In a comparison of serological relationships among CGMMV, ZGMMV, and Kyuri green mottle mosaic virus (KGMMV), the three tobamoviruses specifically reacted with each homologous antibody in the double-antibody sandwich enzyme-linked immunosorbent assay and rapid imunofilter paper assay (RIPA), although ZGMMV and KGMMV were slightly biologcially similar. In a survey of the three tobamoviruses in cucurbitgrowing field in Korea by RIPA, CGMMV and ZGMMV were detected but KGMMV was not found in commercially growing cucurbit crops so far. Seed contamination ratio of CGMMV in bottle gourd seeds tested was 84%, while seed trasmission ratio from the virus-contaminated seeds was 2.0%. Soil transmission ratio was 0-3.5% in fields naturally infested with CGMMV or ZGMMV. Control measures of the virus diseases are roguing and sanitation. These suggest that it is important to rogue the first infected crops, which include the seed and soil, especially early in the season. This may be practicable to control the diseases because CGMMV and ZGMMV have a narrow host range restricted to cucurbitaceous crops.

Development of a Multiplex PCR for Simultaneous Detection of Blueberry Red Ringspot Virus and Blueberry Scorch Virus Including an Internal Control

  • Blueberry red ringspot virus (BRRSV) and blueberry scorch virus (BlScV) are included in the quarantine virus list managed by the Korean Animal and Plant Quarantine Agency. A multiplex polymerase chain reaction (PCR) assay with an internal control was developed for the simultaneous detection of both viruses. The specific primers used here were designed based on the highly conserved regions of the genomic sequences of each virus, obtained from the National Center for Biotechnology Information nucleotide databases. The primers were designed to amplify a partial sequence within coat protein (CP) for detecting BRRSV and a partial sequence within the CP-16 kDa for detecting BlScV. 18S ribosomal RNA (rRNA) was used as internal control, and the primer set used in a previous study was modified in this study for detecting 18S rRNA. Each conventional PCR using the BRRSV, BlScV, and 18S rRNA primers exhibited a sensitivity of approximately 1 fg plasmid DNA. The multiplex PCR assay using the BRRSV, BlScV, and 18S rRNA primers was effective in simultaneously detecting the two viruses and 18S rRNA with a sensitivity of 1 fg plasmid DNA, similar to that of conventional PCR assays. The multiplex PCR assay developed in this study was performed using 14 blueberry cultivars grown in South Korea. BRRSV and BlScV were not detected, but 18S rRNA was all detected in all the plants tested. Therefore, our optimized multiplex PCR assay could simultaneously detect the two viruses and 18S rRNA in field samples collected from South Korea in a time-efficient manner. This approach could be valuable in crop protection and plant quarantine management.

유자의 항 Influenza 바이러스 A형 활성에 관한 연구 (Study on the Anti-influenza Virus A type Activity of Citrus junos)

  • To evaluate anti-influenza virus activity of 113 specimens of Korean traditional medicine both water and methanol extracts were examined using haemagglutination inhibition test. The water extract from Citrus junos was found to inhibit influenza virus A/Taiwan/l/86(H1N1). The survival rates of virus were determined by in situ cellular enzyme-linked immunosorbent assay. The water extract of Citrus junos was fractionated by chromatographic separating using Amberlite XAD-4, 40% MeOH and 60% MeOH layer had antiviral activity. The half inhibition concentration $(IC_{50})$ of 40% MeOH layer on survival of influenza virus was $MIC>361.5{\mu}g/ml$ and $IC_{50}$ value of fr. 40-4 fractionated from 40% MeOH layer was $677.19{\mu}g/ml$. These results suggested that the fractions of Citus junos have potent anti-influenza A virus activity.

Chronic Hepatitis B Virus Infection and Risk of Pancreatic Cancer: A Meta-analysis

  • Objectives: A number of studies have shown that chronic hepatitis B virus infection is implicated in susceptibility to pancreatic cancer. However, the results are still controversial. This meta-analysis aimed to quantitatively assess the relationship between chronic hepatitis B virus infection and incidence of pancreatic cancer of cohort and case-control studies. Methods: A literature search was performed for entries from 1990 to 2012 using PUBMED and EMBASE. Studies were included if they reported odds ratios (ORs) and corresponding 95% CIs of pancreatic cancer with respect to the infection of hepatitis B virus. Results: Eight studies met the inclusion criteria, which included five case-control studies and three cohort studies. Compared with individuals who have not infection of hepatitis B virus, the pooled OR of pancreatic cancer was 1.403 (95%CI: 1.139-1.729, P=0.001) for patients with hepatitis B virus infection. Sub-group analysis by study design showed that the summary OR was 1.43 (95%CI: 1.06-1.94, P=0.021) when pooling case-control studies and 1.31 (95%CI: 1.00-1.72, P=0.05) when pooling cohort studies. Conclusion: Findings from this meta-analysis suggest that chronic hepatitis B virus infection may increase the risk of pancreatic cancer. This relationship needs to be confirmed by further follow-up studies.

2018년 국내 연근해 수산생물의 전염병 모니터링 (Disease monitoring of wild marine fish and crustacea caught from inshore and offshore Korea in 2018)

  • 자연수계 수산생물의 전염병 모니터링은 자연수계 및 양식 수산생물의 질병 관련성 및 상관관계 구명은 질병 발생 예방 및 확산 방지에 매우 중요한 역할을 한다. 2018년 해구 64개소에서 어류 39종 977마리 및 갑각류 14종 287마리를 선정하여 총 1,264마리에 대하여 병원체 감염 여부를 조사하였다. 어류는 법정전염병 2종(VHS, RSIVD) 및 비법정전염병 3종(MABVD, HRVD, LCD)을 분석하고 갑각류는 법정전염병 6종(WSD, IHHN, TS, IMN, YHD, WTD)을 분석하였다. 조사한 모든 시료에서 전염병이 검출되지 않았지만, 우리나라에서 조사한 자연수계 수산생물이 무병하여 청정국 또는 청정지위 선언에 중요한 정보를 제공할 것이다.
