Isolation of $\beta$-1,4-D-arabinogalactanase Producing Strain and Enzyme Purification ($\beta$-1,4-D-arabinogalactanase 생산균주의 분리 및 효소정제)

  • Alkalophilic Bacillus sp. HJ-12 producing $\beta $-1, 4-D-arabinogalactanase was isolated from soil in the alkalic condition, pH 10.0. $\beta $-1, 4-D-arabinogalactanase was maximaly produced in the medium consisting of 2% soybean arabinogalactan (SAG), 0.5% yeast extract, 0.5% polypeptone, 0.5% NaCl, 0.1% K$_{2}$HPO$_{4}$, 0.02% MgSO$_{4}$$\cdot $7H$_{2}$O, 0.1% Na$_{2}$CO$_{3}$ under the aerobic condition (pH 8.2). $\beta $-1, 4-D-arabinogalactanase is inducible enzyme so that its activity has been increased 10 fold in the SAG medium than in the glucose medium. Through the ammonium sulfate precipitation, DEAE- Sephadex A-50 ion chromatography, and Sephadex G-75 gel chromatography procedures, this enzyme was purified with a single protein of 11% vield and 110 fold's purity. $\beta $-1, 4-D-arabinogalactanase is endo type enzyme producing ollgosaccharide from SAG.

  • This study was performed to investigate the effect of two step austenitized treatment on the mechanical properties and fracture characteristic of the ductile cast iron and austempered ductile cast iron(ADI). The obtained results of this study were as follows. The matrix structures of specimens were changed differently by austenitizing heat treatment. Microstructure of austempered ductile cast iron obtained by two step austenitized treatment was bainitic ferrite and retained austenite. With two step austenitized treatment, vield strength, tensile strength and hardness decreased, while the elongation increased.

  • Recently, ozone is utilized in various fields and its needs are expanding. In this paper, glass and ceramic plate tyre ozonizer have been fabricated to investigate discharge parameter(input power, flow rate of supplied gas, electrode form, etc.) effect to discharge. And the conditions of discharge parameter have been investigated for optimum ozone generation. Ozone concentration is continuously increased with increasing input power for same discharge space, and ozone yield is also increased until maximum point after that it is saturated. Ozone concentration is inversely proportional to flow rate of supplied gas but ozone generation and ozone vield characteristics are improved.

  • Isothermal pyrolysis of high density polyethylene(HDPE), polypropylene(PP) and polystyrene(PS) was performed at $450^{\circ}C$, respectively. The effect of pyrolysis time on yield and product composition was investigated. Conversion and liquid yield obtained during HDPE pyrolysis continuously increased with time up to 80minutes, but those of PP and PS did not largely change after 35minutes. Each liquid product formed during the pyrolysis was classified into gasoline, kerosene, light oil and wax according to the distillation temperature based on the petroleum product quality standard of Korea Petroleum Quality Inspection Institute. The major liquid product of HDPE pyrolysis was light oiH34 wt.% based on the amount of HDPE treated) and the amounts of the other liquid ingredients(gasoline, kerosene and wax) were almost the same. On the other hand, the pyrolysis of PP produced 27 wt.% gasoline, 22 wt.% kerosene, 24 wt.% light oil and 13wt.% wax, and the pyrolysis of PS produced 56 wt.% gasoline, 12 wt.% kerosene, 9 wt.% light oil and 13 wt.% wax.

  • This document illustrates the way to control of coiling temperature(CTC) of stip tail part in minimill process. The coiling temperature (CT) is very important fact in hot rolling process because the mechnical properties of strip depend on it. In mini-mill pocess, the speed pattern of rolling is different from that of conventional hot rolling. We have a lot of difficulties in controlling the coiling temperature at strip tail part, because after the strip tail is passed out the final stand, it's impossible to control the coiling temperature by using coiling speed. So we have CT deveiation in this gauge, about 160$^{\circ}C$ below in comparison with target CT. It's clear that deviation of mechanical properties(tensile strength, vield ratio etc) of strip to direction of length is so large, so we could not produce middle carbon steel, like this temperature condition. New coiling temperature control of strip especially in strip tail was developed.   • Existing cooling technologies no longer provide adequate heat dissipation due to excessive heat generation caused by the growing component density on electronic devices. An ionic gas pump can be used for the thermal management of micro-electronic devices, since the size of pump can be reduced to a micrometer scale. In addition, the gas pump allows for gas flow control and generation without moving parts. This ideal property of gas pump gives rise to a variety of applications. However, all these applications require maximizing the wind velocity of gas pump. In this study a barrier discharge type gas pump, with a needle-shaped corona electrode instead of a plate-shaped corona electrode, has been investigated by focusing on the corona electrode shape on the wind velocity and wind generation yield. As a result, the enhanced wind velocity and wind generation yield of 1.76 and 3.37 times were obtained with the needle-shaped corona electrode as compared with the plate-shaped corona electrode of the proposed barrier discharge type gas pump.
