Genetic Variation in Geographically Peripheral Populations of Bupleurum euphorbioides (Apiaceae) with Comparison to a Widespread Congener, B. longiradiatum

  • Bupleurum euphorbioides is isolated and restricted to high mountains in Korea northeastern China. Its conservation depends on whether it is threatened by inbreeding or a loss of genetic diversity. We compared the genetic variability in B. euphorbioides with B. longiradiatum, a widespread congener, to understand how they differ in their population genetic structure. Although B. euphorbioides showed a little lower genetic variability than B. longiradiatum, $F_{IS}$ statistics for most loci were strongly positive in both B. euphorbioides (0.445) and B. longiradiatum (0.553). In addition, B. euphorbioides showed higher mean $F_{ST}$ value than B. longiradiatum (0.297 vs 0.194). It might be due to the polycarpic nature of B. longiradiatum, which holds higher genetic potentials effectively in homogeneous environment than the monocarpic B. euphorbioides. The results suggested that B. euphorbioides is a genetically viable species, and that they are threatened primarily by environmental factor.

Serotype and pathogenicity of Erysipelothrix rhusiopathiae isolated from the cases of acute septicemic swine erysipelas (급성폐혈증형 돼지단독예에서 분리한 Erysipelothrix rhusiopathiae의 혈청형 및 병원성)

  • The serotypes and pathogenicity of 47 isolates of Erysipelothrix rhusiopathiae from pigs with acute septicemic swine erysipelas were investigated. The serotypes were determiend by agar gel precipitation test using autoclave extracted antigens. All of the 47 isolates(42 strains from Youngnam and fi fotrains from Kyunggi province) were serotype lao Five field isolates of Erhusiopathiae serovar la were tested for pathogenicity in mice. All of 5 selected isolates were pathogenic for mice and $LD_{50}$ was $3.7{\times}10^3$ viable cells. Mice immunized subcutaneously with live vaccine did not die after challenge with virulent isolates of E rhusiopathiae.

  • Bacillus thuringiensis serovar israelensis H 14 strain was cultured in 4 different fermentation M-media and then measured the rates of their growths and the productions of endotoxin crystals front the media. Out of the four M-media the production of endotoxin crystals and spores was maximal in M-4 medium (pH 9). The wet weight of the cells grown in the 150$m{\ell}$ culture was approximately 3.901g and the number of viable spores was 1.53$\times$10$^{12}$ per nil and the ratio of the endotoxin over the total cell weight was 18.54%. The generation time was about 89.3 minutes in the M-1 medium, 124.1 minutes in the M-2, 97 minutes in the M-3, 130.8 minutes in the M-4. The proper pHs for the production of the endotoxin appeared to be 6.5 to 7.5.

  • The viability of Bifidobacterium longum KCTC 3128, entrapped in calcium alginate beds in simulated gastroenteric juices (gastric and bile salt solution), was tested to evaluate the influences of several parameters (gel concentration, bead size, and initial cell number). The death rate of B. longum in beads after being sequentially exposed to simulated gastric juices and bile salt solution decreased propertionally with increasing both the alginate gel concentration and bead size. The number of initial cell loading in beads affected the numbers of survivors after being exposed to these solutions, while the death rate of the viable cells were not affected. From the results obtained, the influence of entrapment parameters on the survival of bifidobacteria was quantitatively and systematically evaluated by using a mathematical method.

Changes in Fermentation Characteristics of Kimchi Added with Leek (부추첨가 김치의 발효특성 변화)

  • The effect of addition in different amounts of leek(4, 8, 12% respectively) during fermentation of kimchi was investigated. Fermentation characteristics such as pH, acidity and total reducing sugars as well as microbiological properties were determined. During fermentation, pH was more slowly lowered in kimchi added with leek than in control and titrable acidity of these kimchi was lower than that of control. Viable cells of total bacteria and lactic acid bacteria in these kimchi were higher than that of control during fermentation. Content of total reducing sugars was higher than that of control. Three kinds of reducing sugars such as fructose, glucose and galactose were detected and the dominant one appeared to be fructose. These results suggested that addition of leek seems to retard fermentation of kimchi due to their anti microbial actvity.

  • The studies were carried out to investigate antimicrobial activity of mixed extracts of Lithospermum erythrorhizon and Scutellaria baicalensis G. and crab shell against lactic acid bacteria isolated from kimchi. The effects of Lithospermum erythrorhizon and Scutellaria baicalensis G. extracts and crab shell on shelf life of kimchi were also investigated. The growth of heterofermentative and homofer mentative lactic acid bacteria was inhibited by 95% ethanol extracts of Lithospermum erythrorhizon and Scutellaria baicalensis G. The pH of kimchi containing 1% of mixed extracts Lithospermum erythrorhizon. and Scutellaria baicalensis G., and crab shell was lower than control during fermentation for 25 days of 10oC. The viable cells of the kimchi samples were lower than that of control during fermentation. The sensory qualities of the kimchi samples were a little inferior to the control during shelf life of kimchi.

Sanitizing Effect of $\gamma$-Irradiation on Fresh Vegetable-extract Juices (감마선을 이용한 녹즙의 위생화)

  • The sanitizing effect of irradiation on the fresh vegetable extract juices was investigated. Total bacteria, coliform bacteria and total ascorbic acid were determined during the storage periods at 4oC. Chlorophyll, carotenoid, tannin, electron donating ability and peroxidase activity were determined immediately after irradiation. Results showed that the viable cells were detected below the level of 105 CFU/ml during 12 days with doses of 3 and 5 kGy. Total ascorbic acid and tannin contents increased immediately after irradiation. However, irradiation didn't affect chlorophyll and car otenoid contents, electron donating ability, and peroxidase activity. It was considered that irradiation was effective in sanitizing fresh vegetable extract juices.

Biodegradation of Formaldehyde by Peudomonas Putida H-5 (Peudomonas putida H-5에 의한 포름알데히드의 생분해)

  • This study was designed to reveal the characteristics of the strains degrading formaldehyde isolated from mud of waste water. A strain H-5 showed the highest ability of formaldehyde biodegradation among isolated strains. According to identification, the strain H-5 was ascribed to be Peudomonas putida H-5. The optimal conditions of Peudomonas putida H-5 were $30^{\circ}C$ and pH 7.0. The highest level of formaldehyde degradation was demonstrated at the concentration of 0.02~0.04% in a glucose containing medium. Formaldehyde biodegradation by Peudomonas putida H-5 indicated that this reaction was converted to the methanol and formic acid. However, methanol and formic acid did not show any effect on the growth of viable cells.

  • Chemical, physical md microbial analysis of salted baechu(Chinese cabbage) and its exudate were conducted during storage in large plastic bags(LDPE, HDPE, PVC-box) at 0$^{\circ}C$. Salinity was slightly lowered for 2 weeks storage and maintained that level thereafter in salted haechu, but in its exudate increased rapidly for 2 weeks storage and decreased thereafter. In all treatment pH decreased to 5.0 in salted baechu and to 4.5 in its exudate for 6 weeks storage. Reducing sugar content of salted baechu was 1.5∼2.0 fold higher than that of its exudate, and decresed slowly in both. Total viable cells increased rapidly for 2 weeks but maintained that level thereafter and lactic acid bacteria increased continuously until 6 weeks. Lightness of exudate decreased rapidly in PVC-box, but decreased slowly in LDPE. Compression force of salted baechu increased continuously untill 8 weeks except for PVC-box.

  • Effects of Ac pulse at low voltage on Saccharomyces cerevisiae were studied. The treatment of yeast suspensions contained 0.2m NaCl with 500mA for 350 sec at $40^{\circ}C$ was shown about 50% of lethality, whereas in the treatment of the same suspensions with 250mA for 250 sec at the temperature ($10^{\circ}C$) corresponding to about 10% of lethality, growth was completely inhibited instead of lethality. The effect of growth inhibition was due to occurrence of auxotrophic strains under experimental conditions. Detection of auxotrophic yeasts was done tentatively with the difference of the number of viable yeast cells between direct counting by methylene blue staining and plate-count on yeast morphology agar.
