Dental caries is a bacterial disease of the dental hard tisssus, characterized by a localized, progressive, molecular disintegration of tooth structure. The action of Leuconostoc lactis 51 about plaque formation and replication by Streptococcus mutans was studied as follows. 1. Lower amount of plaque was produced at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone on the wires in the beaker. 2. Fewer cells of S. mutans were replicated at the mixed culture of S. mutans and L. lactis 51 than S. mutans alone. 3. In M17Y broth, viable cells of S. mutans and L. lactis 51 increased for 12 hours, and decreased for 24 hours. In M17YS broth, viable cells of S. mutans showed time-dependent decrease at mixed culture of S. mutans and L. lactis 51. 4. The culture supernatant of L. lactis 51 didn't inhibit the replication of S. mutans and the formation of artificial plaque. 5. Sucrose and frutose were extracted from the culture supernatant of L. lactis 51 in M17YS broth. These results suggest that L. lactis 51 isolated from the oral cavity inhibits the replication of S. mutans and the formation of artificial plaque.
Total viable cells and lactose non-fermenting cells were counted from animal feedstuffs (n=65). And isolation of Gram negative lactose nonfermenting enterobacteria and antibiotics susceptibility of isolates were performed. 1. The ranges of total viable cells / lactose non-fermenters in animal feedstuffs from Korean cattle were counted as 9$\times$$10^4$-1$\times$$10^7$ / 1$\times$$10^2$-6$\times$$10^3$, milking cow as 1$\times$$10^4$-2$\times$$10^8$ / 2$\times$$10^2$-8$\times$$10^3$, pig as 1$\times$$10^4$-1$\times$$10^6$ / 2$\times$$10^2$-6$\times$$10^3$, and chicken as 7$\times$$10^4$-1$\times$$10^9$ / 4$\times$$10^2$-1$\times$$10^5$ cfu/g, respectively. 2. Among the 214 isolates from feedstufs, 87 from Chinan(n=23), 66 from Changsu (n=23) and 61 from Mooju(n=19) were isolated. Of these isolates, 60 from pigs (n: 19), 51 from milking cows(n=15), 45 from chikens(n=11) and 58 from Korean cattle(20) were isolated. 3. Among the 6 genuses of Gram negative lactose nonfermenting enterobacili, Salmonella sp, Y pseudotuberculosis, Ent agglomerans and Sal choleraesuis were frequently encountered. 4. A majority of isolates were sensitive to 19 antibiotics, singly or in combination. These isolates were completely susceptible to Cp, Gm, Imp and Pi, 93% to Ak and To, 73% to Cax and Ts, 66% to Cft and Tim, 46-53% to Caz, Cf and Cz, 33-40% to Am, Azt, Cfz and Ti, and 6% to Cfx, in order, but not susceptible to Crm. 5. Among the antibiotic resistant strains, a total of 23 resistant patterns was noted, and of these Crm 40(18.7%), Am Cf Cfx Cfz Crm Ti 27(12.6%), each of Azt Ctx Crm and Azt Cax Caz Cft Cfx Crm 22(10.3% ) were frequently encountered.
Purpose: The purpose of this study was to investigate the far-infrared emissivity of patented ocher quilt cotton fabrics and to investigate the microorganisms that survived the washing of cotton fabrics up to 20 times. Methods: A 16S rRNA assay was performed using a far-infrared radiometer and a single colony in which microorganisms grew in nutrient media. Results: The far-infrared emissivity of ocher quilt was 0.902 (90.2%) at 5~20 ㎛ at 40℃, and the radiation energy was 3.63 × 102 w/m2. The number of viable cells was 2.0 × 102 cells/ml in ocher duvet cotton fabric, and no viable bacteria found in regular cotton fabric. The base sequence of 16S rRNA of B-2 strain isolated into single colonies was 1,419 bases, and the base sequence of strain A-4 was 1,284 bases. The base sequence of 16S rRNA of these two strains showed high homology with Bacillus spp. The B-2 bacteria showed high homology with 99.0% of the 16S rRNA sequence of B. aryabhattai EF114313 and 99.0% of the A-4 bacteria of B. bingmayongensis AKCS01000011. Consequently the colony strain B-2 finally identified as B. aryabhattai BJ-2 and A-4 as B. bingmayongensis BJ-4 strain. Concusions: Soil Bacillus strains survived in ocher quilt cotton fabric after 20 washing. The material can be useful because quilt cotton fabric emits a large amount of far-infrared and far-infrared radiation energy.
Background: Somatic cell nuclear transfer (SCNT) is used widely in cloning, stem cell research, and regenerative medicine. The type of donor cells is a key factor affecting the SCNT efficiency. Objectives: This study examined whether urine-derived somatic cells could be used as donors for SCNT in pigs. Methods: The viability of cells isolated from urine was assessed using trypan blue and propidium iodide staining. The H3K9me3/H3K27me3 level of the cells was analyzed by immunofluorescence. The in vitro developmental ability of SCNT embryos was evaluated by the blastocyst rate and the expression levels of the core pluripotency factor. Blastocyst cell apoptosis was examined using a terminal deoxynucleotidyl transferase dUTP nick end-labeling assay. The in vivo developmental ability of SCNT embryos was evaluated after embryo transfer. Results: Most sow urine-derived cells were viable and could be cultured and propagated easily. On the other hand, most of the somatic cells isolated from the boar urine exhibited poor cellular activity. The in vitro development efficiency between the embryos produced by SCNT using porcine embryonic fibroblasts (PEFs) and urine-derived cells were similar. Moreover, The H3K9me3 in SCNT embryos produced from sow urine-derived cells and PEFs at the four-cell stage showed similar intensity. The levels of Oct4, Nanog, and Sox2 expression in blastocysts were similar in the two groups. Furthermore, there is a similar apoptotic level of cloned embryos produced by the two types of cells. Finally, the full-term development ability of the cloned embryos was evaluated, and the cloned fetuses from the urine-derived cells showed absorption. Conclusions: Sow urine-derived cells could be used to produce SCNT embryos.

A number of experimental and epidemiological studies have implicated that antiestrogenic effects of estrogen-like compounds in legumes and plant seeds are responsible for lowering breast cancer risk in human. However, few studies have been conducted to illustrate the possible chemopreventive effects of Korean traditional food materials. This study was performed to determine the cytotoxic and apoptotic effects of yellow soybeans, black soybeans and brown rice extracts on hormone-dependent and hormone-independent human breast cancer cells. Methanol-or acetone-soluble fractions of soybeans or brown rice were incubated with hormone-dependent cells (MCF-7) or hormone-independent cells (MDA-MB-231). Cell cytotoxicity was measured by MTT assay at 24, 48 and 72 hrs of incubation. Apoptotic effects of these extracts toward breast cancer cells were also determined at 48 hrs of incubation by measuring DNA fragmentation. Results indicated that the acetone-soluble fraction of brown rice exerted strongest cytotoxic effect on MCF-7 ceIls, although other fractions also reduced the number of viable MCF-7 cells after 48 hrs of incubation. Both acetone and methanol soluble fractions of all samples exerted a significant cytotoxicity towards MDA-MB-231 cells after 24 hrs of incubation, and acetone and methanol soluble fractions of brown rice were especially effective in these cells. At 48 hrs of incubation, methanol fractions of all three samples induced apopotosis of MDA-MB-231 cells. These results indicate methaol or acetone soluble fractions of yellow soybeans, black soybeans and brown rice induce cytotoxicity in both hormone-dependent and hormone-independent breast cancer cells. Therefore, possible mechanisms of cell cytotoxicity do not necessarily include antiestrogenic effects of soybean or brown rice extract. A possible anticarcinogenic effect of brown rice methanol-soluble fraction may mediated through their apoptotic effect. Further studies are requried to elucidate responsible compounds and mechanisms involved in observed anticarcinogenesis.

The identification of tumor-specific antigens has represented a critical milestone in cancer diagnosis and therapy. Clinical research in this area for leukemia has also been driven over the past few decades by the hope that surface antigens with restricted tissue expression would be identified. Disappointingly, only a small number of the leukemic antigens identified to date, meet sufficient criteria to be considered viable immunophenotypic markers. In this paper, we nominate anti-JL1 monoclonal antibody as an immunodiagnostic and immunotherapeutic candidate for leukemia. The JL1 molecule appears to be a novel cell surface antigen, which is strictly confined to a subpopulation of limited stages during the hematopoietic differentiation process. Despite the restricted distribution of the JL1 antigen in normal tissues and cells, anti-JL1 monoclonal antibody specifically recognizes various types of leukemia, irrespective of immunophenotypes. On the basis of these findings, we propose JL1 antigen as a tumor-specific marker, which shows promise as a candidate molecule for diagnosis and immunotherapy in leukemia, and one that spares normal bone marrow stem cells.
This study was designed to evaluate the viability of canine trachea after cryopreservation for two months. Eight cervical tracheal rings were resected in three dogs and both ends were anastomosed. The resected tracheal segments were cryopreserved and stored in liquid nitrogen at -196oC for two months. Two months later, the cryopreserved segments were thawed. Half of each segment was implanted into the abdomen of its donor animal and the other half was cultured in tissue media. Two weeks later, the animal was sacrificed. The native cervical trachea was removed to serve as a control and the abdominally implanted trachea was removed for study. At that time, both specimens were also cultured in tissue media. Tracheal epithelial viability was assessed histologically by using an inverted microscope. The epithelial cells were confirmed immunohistochemically using monoclonal antibodies against cytokeratin and epithelial membrane antigen. Control and cryopreserved segments showed good, viable epithelial cells, but the implanted segments showed slightly depressed viability. We conclude that canine tracheal epithelium can survive after cryopreservation for two months, but the implanted trachea will be slightly damaged by ischemia before revascularization, even if omental wrapping is used.

The application of steroids has steadily increased thanks to their therapeutic effects. However, alternatives are required due their severe side effects; thus, studies on the activities of steroid derivatives are underway. Sugar derivatives of nandrolone, which is used to treat breast cancer, as well as cortisone and prednisone, which reduce inflammation, pain, and edema, are unknown. We linked O-glucose to nandrolone and testosterone using UDP-glucosyltransferase (UGT-1) and, then, tested their bioactivities in vitro. Analysis by NMR showed that the derivatives were 17β-nandrolone β-ᴅ-glucose and 17β-testosterone β-ᴅ-glucose, respectively. The viability was higher and cytotoxicity was evident in PC12 cells incubated with rotenone and, testosterone derivatives, compared to the controls. SH-SY5Y cells incubated with H2O2 and nandrolone derivatives remained viable and cytotoxicity was attenuated. Both derivatives enhanced neuronal protective effects and increased the amounts of cellular ATP.
Insect cells were grown and infected with baculovirus for the production of recombinant protein. Later infection gave the lower expression of recombinant protein. This indicates that the expression rate is lower at higher cell concentration. This phenomena provides a well-posed optimization problem with respect to the infection time. The optimal infection time was experimentally shown to exist for the maximum productivity of recombinant protein. Also, the expression increased with the addition of 5% silkworm hemolymph. This is considered to be due to the increase of intracellular viruses and the longevity of viable cells after the infection. The production of ${\beta}$-galaclosidase increased about ten-fold with the addition of yeastolate and silkworm hemolymph for high cell density and high expression, respectively.

We investigated the thin films of poly(3-hexylthiophene) (P3HT) and C61-butyric acid methylester (PCBM) prepared by ultrahigh vacuum (UHV) electrospray depositioin (ESD) by using in-situ XPS, UPS and ambient-pressure AFM. The morphology, chemical structures, and interface properties of these materials, most widely used for bulk heterojunction organic solar cells, were studied depending on the ESD solution compositions and concentrations. We found that the solution conductivity and flow rate as well as applied voltage are the important parameters for stable electrospray and film formation. These results suggest that UHV ESD is a viable method for the deposition of multilayers of polymers under UHV condition. We also discuss the energy level alignment for the various deposition conditions at the interface, which is one of the most important operating parameters of the bulk heterojunction organic solar cells.
