Agastache rugosa, a member of the mint family (Labiatae), is a perennial herb widely distributed in East Asian countries. It is used in traditional medicine for the treatment of cholera, vomiting, and miasma. This study assessed the genetic diversity and population structures on 65 accessions of Korean mint A. rugosa germplasm based on inter simple sequence repeat (ISSR) markers. The selected nine ISSR primers produced reproducible polymorphic banding patterns. In total, 126 bands were scored; 119 (94.4%) were polymorphic. The number of bands generated per primer varied from 7 to 18. A minimum of seven bands was generated by primer 874, while a maximum of 18 bands was generated by the primer 844. Six primers (815, 826, 835, 844, 868, and 874) generated 100% polymorphic bands. This was supported by other parameters such as total gene diversity ($H_T$) values, which ranged from 0.112 to 0.330 with a mean of 0.218. The effective number of alleles ($N_E$) ranged from 1.174 to 1.486 with a mean value of 1.351. Nei's genetic diversity (H) mean value was 0.218, and Shannon's information index (I) mean value was 0.343. The high values for total gene diversity, effective number of alleles, Nei's genetic diversity, and Shannon's information index indicated substantial variations within the population. Cluster analysis showed characteristic grouping, which is not in accordance with their geographical affiliation. The implications of the results of this study in developing a strategy for the conservation and breeding of A. rugosa and other medicinal plant germplasm are discussed.

In this study, the lateral loading test was performed to analyze structural performance of multi-span plastic greenhouse through full-scale experiment and numerical analysis. In order to analyze the lateral stiffness and stress, we installed 9 displacement sensors and 19 strain gauge sensors on the specimen, respectively, and load of l mm per minute was applied until the specimen failure. In the comparison between the full-scale experiment and the structural analysis results of a multi-span greenhouse with venlo-type roof according to bracing installation, there was a large difference in the lateral stiffness of the structure. By installing a brace system, the lateral stiffness measured near the side elevation of the specimen increased by up 44%. As the bracing joint used in the field did not secure sufficient rigidity, the external force could not be transmitted to the entire structure properly. Therefore, it is necessary to establish a bracing construction method and design standards in order for a greenhouse to which bracing applied to have sufficient performance.
Owing to the lack of consistent research on endangered plant species in Korea, there are insufficient data to preservespecies and expand habitats. This study analyzed the preferred habitat and threats to the survival of Aconitum austrokoreense, found on Baekwun Mountain in Gurye-gun, Gwangyang-si, Jeollanam-do Province, and classified as a level two endangered wild plant by the Ministry of Environment, by investigating major environmental factors such as climate, location, soil, and stand structure. By examining five selected sites inhabited by Aconitum austrokoreense on BaekwunMountain, this study found that the habitat had an altitude of 420 to 675 m above sea level and showed a northeast tendency, spreading over a range of inclination angles between 15° and 37°. The average number of plants across the five sites was 156. Site 4 (550 m) had the highest density of 372 plants, with an average height of 0.6 m. The average soil moisture and relative light intensity were 20.48% and 7.34%, respectively. Layer soil was presumed to be sandy loam, characterized by high sand content and good drainage. The habitat had average soil pH of 5.2, average organic matter of 16.46%, average nitrogen of 0.86%, average available phosphate of 11.86 mg/kg, average electrical conductivity of 0.44 dS/m, and average cation exchange capacity of 37.04 cmolc/kg. The total carbon in soil averaged 10.68%. From the analysis of the vegetation structure of sites inhabited by Aconitum austrokoreense, the dominant populations were Pinus koraiensis and Lindera erythrocarpa in Site 1, Magnolia obovata and Carpinus laxiflora in Site 2, Zelkova serrate and Quercus variabilis in Site 3, Staphylea bumalda and Lindera erythrocarpa in Site 4, and Morus bombycis,Styrax japonicus, and Carpinus laxiflora in Site 5. With most habitats located near trails and sap collection sites of Acer pictum, the species were exposed to artificial damage and interference threats.
A lactic acid bacteria with ${\beta}$-gal activity was isolated from Kimchi, a traditional fermented vegetable food in Korea. The isolate was identified as a Lactococcus lactis strain and named L. lactis A2. The gene encoding ${\beta}$-gal of L. lactis A2 was cloned as a 5.8 kb PstI fragment. DNA sequencing identified the complete lacA (galactoside acetyltransferase)-lacZ (${\beta}$-galactosidase) genes together with the 3' part of upstream galT (galactose-1-phosphate uridyltransferase), and the 5'region of downstream galE (UDP-galactose-4-epimerase) genes. L. lactis A2 had the same gal/lac operon structure as in L. lactis subsp. lactis 7962. Other genes of the Leloir pathway are most likely to be located in the 5'upstream of the 5.8 kb fragment on the A2 chromosome. Sequences downstream of galE were different from those of L. lactis subsp. lactis 7962.

Lactobacillus zymae GU240 was previously isolated from Kimchi, a Korean fermented vegetable, as a strong GABA producer. The strain showed β-galactosidase (β-Gal) activity on MRS agar plates with X-gal. When growth and β-Gal activities of GU240 were measured using MRS (glucose, 2%, w/v) and MRSL (lactose, 2%, w/v) broths, cells were found to grow slowly in MRSL, and the β-Gal activity (36 units at 4 h) was lower than that of cells grown in MRS (94 units at 16 h). The highest OD600 value of the culture in MRS was 1.6 at 24 h at 37℃, whereas that of the culture in MRSL was 0.6 at 16 h. β-Gal activity of the culture in MRS reached the maximum (95.6 u/ml) at 16 h, decreased thereafter, and was not detected at 48 h. β-Gal activity for culture in MRSL reached its highest (36 u/ml) at 4 h and decreased gradually, but some activity (11.05 u/ml) still remained at 72 h. The structural gene encoding β-Gal in L. zymae GU240 was cloned as a 3.1 kb fragment, and DNA sequencing confirmed the presence of complete lacLM genes. lacLM genes from L. zymae GU240 showed 98-99% homologies in nucleotide sequences with other lacLM genes from L. brevis. Reverse transcription (RT)-PCR confirmed the operon structure of lacLM. The results indicated that L. zymae GU240 might be in the process of losing the ability to grow rapidly on lactose-containing medium, such as milk, due to adaptations to plant environments, including kimchi.

This research was conducted to obtain the basic information for establishment of standard guidelines in the design and installation of roof ventilation system in single-span plastic greenhouse. To achieve this, the greenhouse structure & characteristics, cultivation status, and ventilation system were investigated for single-span greenhouse with roof ventilation system cultivating the Cucurbitaceae vegetables, watermelon, cucumber, and oriental melon. Most of single-span watermelon greenhouse in Haman and Buyeo area were a hoop-style and the ventilation system in those greenhouses mostly consisted of two different types of 'roof vent (circular or chimney type) + side vent (hole) + fan' and 'roof vent (circular type) + side vent (hole or roll-up type)'. The diameter of circular and chimney-type vent was mostly 60cm and the average number of vents was 10.5 per a bay with vent spacing of average 6.75m. The ratio of roof vent area to floor area and side vent area in the single-span watermelon greenhouse with ventilation fan were 0.46% and 7.6%, respectively. The single-span cucumber greenhouse in Haman and Changnyeong area were a gable roof type, such as even span, half span, three quarter and the 70.6% of total investigated single-span greenhouses was equipped with a roof ventilation fan while 58.8% had a circulation fan inside the greenhouse. The ratios of roof vent area to floor area in the single-span cucumber greenhouse ranged from 0.61 to 0.96% and in the case of the square roof vent, were higher than that of the circular type vent. On average, the roof ventilation fan in single-span cucumber greenhouse was equipped with the power input of 210W and maximum air volume of $85.0m^3/min$, and the number of fans was 9.75 per a bay. The number of roof vent of single-span oriental melon greenhouse with only roll-up type side vent ranged from 8 to 21 (average 14.8), which was higher than that of other Cucurbitaceae vegetables while the vent number of the greenhouse with a roof ventilation fan was average 7 per a bay.
A new Asiatic hybrid lily 'Redsky' cultivar was selected from a cross between 'Vivaldi' cultivar with pale pink color flower as mother plant and 'Prato' with orange colored flower in 2000 year. The selected bulb line was propagated and cultivated for line nomenclature as 'Gonong-F-na-10'. The survey of the growth and flower characteristics of 'Gonong-F-na-10' were conducted from 2005 to 2007 on summer cropping system under structure in Highland Agriculture Research Center, NICS, RDA. After all 'Gonong-F-na-10' was named as 'Redsky'. The flowers of the 'Redsky' cultivar had not soft hair and fragrance, and had orange red (O-R N30D) colored petals with dark brown spots in center of flower. The stem color was dark pink and brown caused by anthocyanin pigment, the stigma and pollen colors were brown. Flowering direction was upward. The flowering date was July 7 so early flowering. The plant height was 97.2 cm so long, number of flowers was 4.9 ea, and length of flower bud was 9.3 cm. The bulb weight was about 34.8 g and its bulb size was about 14.8 cm in circumference. 'Redsky' cultivar was registered as a new variety with No. 3119 in Korea Seed and Variety Service on June of 2010.
Sclerotinia rot caused by Sclerotinia sclerotiorum frequently causes serious and unpredicable yield losses of the leaves of perilla growing under structured cultivation as vegetable in Korea. Temperature for mycelial growth ranged from 5 to $30^{\circ}C$ with optimum temperature d $20^{\circ}C$. Sclerotia were formed fewer at low temperature, but their dry weight was heavier than that at high temperature. The apothecia were formed from the sclerotia that buried up to 3 cm soil depth at $15^{\circ}C$ in moisture condition. The incidence of perilla Sclerotinia rot caused by S. sclerotiorum was observed throughout the growing season at greenhouse. The occurrence of this disease was especially severe from January to February of low temperature period. The average incidence rates of this disease was up to 15%. The significant occurrence of this disease was showed mainly in the continuous cropping field for more than five years. The incidence of this disease increased according to the increase of continuous cropping year. The incidence rates of this disease reached up to 20% in the continuous cropping field for ten years. Also it was firstly investigated the natural infection caused by S. sclerotiorum on weed plants, Gnaphalium affine and Mazus pumilus in farmer's field. The casual fungus showed pathogenicity on 11 weed plants species tested, and more severe pathogenicity on G. affine, Latuca indica and Ixeris dentata included in the family Compositae. This result suggests that effective crop rotation and weed eradication can be the method for organic control of perilla sclerotinia rot, and sudden outbreaks of this disease on perilla growing under structure after paddy rice call be explained by the presence of weed hosts.
In this study, the structural experiment was conducted with two types of specimens to investigate the mechanical behavior of the column-rafter-purlin connection of an arch-type greenhouse under monotonic loading. Based on the experimental results, the flexural performance was analyzed for two types of connections, and connection classification was attempted. Type B showed 77% of flexural performance compared to Type A, and both types showed that the rigidity and flexural strength did not reach the level of the full rigid. The behavior of the column-rafter-purlin connection was dominated by local buckling due to deformation of the weld and fasteners. As a result of connection classification by AISC standard, both Type A and B connections showed a result that did not meet the rigid connection performance assumed during design, and were classified as simple connection. Therefore, the connection performance evaluation and classification results show that the greenhouse design should be made in consideration of connection performance and in order to design a reliable greenhouse structure, a study on establishing clear design standards for the greenhouse connection is necessary.
This study was carried out in order to optimize the manufacturing condition of fried garlic flakes as well as to investigate the physicochemical properties of the flakes. Fried garlic flake samples were prepared as follows: garlic was sliced by a thickness of 1.5 mm, 2.0 mm, 2.5 mm, which were measured by a thickness gage. The samples were fried in vegetable oil under different temperatures of $140{\sim}150^{\circ}C$, $160{\sim}170^{\circ}C$ and $180{\sim}185^{\circ}C$. The compression strength depending on the height (h) was measured in order to find the thickness effect by the rheometer (force control: 50 N, h: 3.25 mm). Moreover, the sample with 1.5 mm thickness showed crisp phenomena of the split compared with the crush shape of the 2.0 mm and 2.5 mm thick samples. The result of strength for time dependence showed a sample with a thickness of 1.5 mm, which was measured 5~9 times more than the 2.0 mm and 2.5 mm thick samples. We thought the reason that the 1.5 mm sample had less response power equivalent to compression force than the other samples. Alliin has been found to affect the immune responses in the blood, it is a derivative of the amino acid cysteine and is also quite heat stable. The LC system with a UV detection at 210 nm consists of a separation on a Zorbax TMS column and isocratic elution with water and ACN as a mobile phase. The alliin contents of raw and fried garlic flake under $140{\sim}150^{\circ}C$, $160{\sim}170^{\circ}C$ and $180{\sim}185^{\circ}C$ were 18.10 mg/mL, 14.0 mg/mL, 11.6 mg/mL and 11.1 mg/mL, respectively. The decrement of alliin content under different temperature was a small quantity hence, we confirmed that the increasing manufacturing temperature was not affected by the alliin content. Examining for the particle structure of fried garlic flakes by a polarization microscope, the color of the sample treated at $160{\sim}170^{\circ}C$ was pure yellow. Furder, the fiber shaped particle, which has an effect on the tough texture, almost did not appear compared to the different temperature conditions. Finally, the sensory test for the preference of fried garlic flake under different conditions was carried out and the scores for various sensory characteristics were surveyed. According to the physicochemical measurements and sensory evaluation, we confirmed that the optimum manufacturing condition of fried garlic flake was 1.5 mm thick at a temperature of $160{\sim}170^{\circ}C$.
