Isolation and Characterization of Exopolysaccharide-Producing Bacteria from Korean Fermented Vegetables (전통 침채류 유래 다당 생산균의 분리 특성)

  • Four bacteria producing viscous exopolysaccharides (EPSs) were isolated from Korean fermented vegetables (Cucumber kimchi, Young radish kimchi, Green onion kimchi) using a selection medium intended for isolating bacteria with tannin-degrading activity. They were identified phylogenetically by 16S rDNA sequence analysis and found to be very close to Enterobacter cowan ii, Escherichia senegalensis, Enterobacter asburiae, and Enterobacter ludwigii. Strain CK31, the most efficient EPS-producer, produced a heteropolysaccharide with an approximate molecular weight of 420 kDa. The neutral sugar fraction of the EPS was composed of rhamnose, fucose, arabinose, mannose, galactose, and glucose.

Effects of Web-Based Nutrition Counseling on Dietary Behavior and Food Intake in Hyperlipidemic Patients (고지혈증 환자의 식행동 및 식품섭취에 웹기반 영양상담의 영향)

  • The purpose of this study was to investigate the effect of the web-based nutrition counseling on the dietary behavior and food intake of hyperlipidemic patients and to evaluate the web-based nutrition counseling program. Forty hyperlipidemic patients, twenty of them were hypercholesterolemia and the other twenty were hypertriglyceridemia, participated in the web-based nutrition counseling program. At the first nutrition counseling, the patients were interviewed and then follow-up nutrition counselings were accomplished four times during eight weeks through a web-based internet program. The web-based nutrition counseling program was consisted of diet management, nutrition education and nutritional assessment and the follow-up counseling program. The dietary behavior score increased significantly in both groups after the nutrition counseling program(p<0.01). The diet therapy knowledge score also increased significantly (p<0.01). For food frequency, vegetable (p<0.01), fruit (p<0.05) and seaweed (p<0.01) consumption increased significantly, while meat (p<0.01), egg (p<0.01), snack (p<0.01) and instant food (p<0.01) intake decreased significantly in both groups after the nutrition counseling program. The evaluation of the web-based nutrition counseling program by the patients showed above average in all domains, specially the evaluation scores of younger aged patients habituated to practice computer were better than those of older aged patients. Therefore, this study shows that the web-based nutrition counseling program is effective for improving dietary behavior and food consumption the patients. In addition, these results indicate that internet presents us with potential as a new medium for nutrition counseling in informationized society.

Homologous Expression and Quantitative Analysis of T3SS-Dependent Secretion of TAP-Tagged XoAvrBs2 in Xanthomonas oryzae pv. oryzae Induced by Rice Leaf Extract

  • Xanthomonas oryzae pv. oryzae (Xoo) produces a putative effector, XoAvrBs2. We expressed XoAvrBs2 homologously in Xoo with a TAP-tag at the C-terminus to enable quantitative analysis of protein expression and secretion. Addition of rice leaf extracts from both Xoo-sensitive and Xoo-resistant rice cultivars to the Xoo cells induced expression of the XoAvrBs2 gene at the transcriptional and translational levels, and also stimulated a remarkable amount of XoAvrBs2 secretion into the medium. In a T3SS-defective Xoo mutant strain, secretion of the TAPtagged XoAvrBs2 was blocked. Thus, we elucidated the transcriptional and translational expressions of the XoAvrBs2 gene in Xoo was induced in vitro by the interaction with rice and the induced secretion of XoAvrBs2 was T3SSdependent. It is the first report to measure the homologous expression and secretion of XoAvrBs2 in vitro by rice leaf extract. Our system for the quantitative analysis of effector protein expression and secretion could be generally used for the study of host-pathogen interactions.

Breeding of Vivid Pink Standard Carnation 'Pink Beryl' with Round Petals for Cut Flower (둥근 꽃잎에 화색이 선명한 분홍색 스탠다드 카네이션 'Pink Beryl' 육성)

  • A new standard carnation Dianthus caryophyllus 'Pink Beryl' was bred from a cross of red standard cultivar 'Francesco' and purple standard cultivar 'Saturn' in 1996 at the National Institute of Horticultural & Herbal Science. After investigation of the characteristics for 4 years from 1997 to 2000 and registered to the official breeding variety of Rural Development Administration in 2000. 'Pink Beryl' is pink standard cultivar with round edge of petal for cut flower. It has medium resistance to Fusarium wilt. To avoid calyx splitting caused by high temperature, this cultivar is recommended to grow over $8^{\circ}C$ at night and under $25^{\circ}C$ at days during flower bud differentiation period.

Biological Characterization of Paenibacillus polymyxa JE201 with Antifungal Activity Against Fungal Leaf Spot Disease of Aster scaber (취나물 점무늬병을 억제하는 Paenibacillus polymyxa JE201의 생물학적 특성)

  • Koreans consume cham-chwi (Aster scaber thunb.) as a common vegetable in a meal because of its bitter taste and rich flavor. In addition, it is the crop with the most residual pesticides detected in the last five years. Among the detected pesticides, the most common was azoxystrobin, which is a drug used primarily to prevent the leaf spot disease of A. scaber caused by Septoria sp.. We isolated the microorganisms that antifungal activity against Septoria sp.. The optimum incubation conditions (temperature, pH and growth medium) were examined for the growth of the isolates. Additionally, cellulase and protease activity and siderophore production ability were also examined. According to 16S rRNA sequencing of the isolate was affiliated to Paenibacillus polymyxa JE201. Largest inhibition zone measuring up to 9.2 mm was observed for P. polymyxa JE201 after 7 days of inoculation. P. polymyxa JE201 strain showed antifungal activity against various fungal phytopathogens Altanaria sp., Botrytis cinerea, Colletotrichum acutatum, Fusarium oxysporum, Phytophthora capsici, Ph. drechesleria, Rhizoctonia solani and Stemphylium sp.. Based on these observations, P. polymyxa. JE201 can be used as a promising biocontrol agent for preventing the leaf spot disease and other phytopathogens.

Development of transgenic disease-resistance root stock for growth of watermelon.(oral)

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. Influence of Isolation Temperature on Isolating Diverse Lactic Acid Bacteria from Kimchi and Cultural Characteristics of Psychrotrophs

  • Kimchi is a traditional Korean fermented vegetable that is stored and fermented at low temperatures. However, kimchi lactic acid bacteria (LAB) are typically isolated under mesophilic conditions, which may be inappropriate for isolating the diverse LAB. Therefore, this study investigated the suitable conditions for isolating various LAB from kimchi. Here, LAB were isolated from four kimchi samples using MRS, PES, and LBS media and varying isolation temperatures (30, 20, 10, and 5℃). Then, MRS was selected as the suitable medium for LAB isolation. A comparison of culture-dependent and culture-independent approaches indicated that 5℃ was not a suitable isolation temperature. Thus, the number and diversity of LAB were determined at 30, 20, and 10℃ using 12 additional kimchi samples to elucidate the effect of isolation temperature. With the exception of two samples, most samples did not substantially differ in LAB number. However, Leuconostoc gelidum, Leuconostoc gasicomitatum, Leuconostoc inhae, Dellaglioa algida, Companilactobacillus kimchiensis, Leuconostoc miyukkimchii, Leuconostoc holzapfelii, and Leuconostoc carnosum were isolated only at 10 and 20℃. The growth curves of these isolates, except Leu. holzapfelii and Leu. carnosum, showed poor growth at 30℃. This confirmed their psychrotrophic characteristics. In Weissella koreensis, which was isolated at all isolation temperatures, there was a difference in the fatty acid composition of membranes between strains that could grow well at 30℃ and those that could not. These findings can contribute to the isolation of more diverse psychrotrophic strains that were not well isolated under mesophilic temperatures.

Environmental Control in the Plant Factory System Influences Year-Round Production of Allium hookeri Leaves (삼채(Allium hookeri) 잎 연중생산을 위한 식물공장 환경제어 효과)

  • The demand for the fresh leaf of hooker chive, which is mainly used as functional roots and contains dietary sulfur or saponin, is increasing, but the leaves are only harvested 3-4 times per year under conventional field conditions. A plant factory system with different light qualities or intensities was applied for year-round production of the fresh leaves. Hooker chive (Allium hookeri) roots were hydroponically cultured under the plant factory with a mixture of blue plus red LEDs (Light-Emitting Diodes) and fluorescent lights for 50 weeks. Maximum leaf growth was attained with the 1.5 dS/m EC in the culture medium under the plant factory. The average leaf and shoot numbers of hooker chive grown hydroponically under a mixture of 200 µmol/m2/s LEDs increased by 147% and 140%, respectively compared to those under 100 µmol/m2/s LEDs at the 10th harvest. The leaf length of hooker chive grown under the LEDs treatment with the lowest light intensity significantly increased by 27% compared with the natural light treatment at the 10th harvest. However, there was no significant difference in leaf pigmentation between natural and 200 µmol/m2/s LEDs treatments. Plant factory with the mixture LEDs of blue and red lights can be applied for year-round production of hooker chive fresh leaves to ensure a stable supply of leafy vegetable throughout the year.

Combination of Enrichment and PCR in Rapid Semi-Quantification of Bacillus cereus in Fresh-Cut Vegetables

  • Widespread consumption of fresh-cut vegetables without cooking results in ingestion of major foodborne pathogens including Bacillus cereus. In this study, we aimed to develop a method to rapidly detect B. cereus in fresh-cut vegetables by combining commercial PCR analysis with enrichment of the pathogenic levels. A mixture of B. cereus strains (KCTC1013, KCTC1014, KCTC1092, KCTC1094, and KCTC3624) was inoculated on the surface of fresh-cut cabbage lettuce (20 g) and baby leafy vegetables (10 g) to concentration 1, 2, 3, 4, and 5 log CFU/g. Eighty milliliters of TSB with 0.15% polymyxin B was used for cabbage lettuce, and 90 mL of medium was used for baby leafy vegetables and incubated at 42℃ for 0, 2, 3, 4, 5, 6, and 7 h. One milliliter of the enriched media was plated on mannitol-egg yolk-polymyxin agar for quantification, and another 1 mL was used for DNA extraction for PCR analysis. Additionally, the minimum number of sub-samples to be tested from a pack of fresh-cut vegetable samples was determined using 5 sub-samples. The results from this study showed that for detecting B. cereus in fresh-cut cabbage lettuce, 3, 4, 5, 6, and 7 h enrichment were required to at least detect 5, 4, 3, 2, and 1 log CFU/g of B. cereus, respectively. B. cereus in fresh-cut baby leafy vegetables could be detected after 2, 3, 4, 5, and 6 h of enrichment at 5, 4, 3, 2, and 1 log CFU/g, respectively, using a combination of enrichment and PCR analysis. To determine if a pack of fresh-cut vegetable is positive, the minimum number of sub-samples should be 3. These results can be used to develop a rapid detection method to semi-quantify B. cereus in fresh-cut vegetable samples combining enrichment and PCR.

  • This experiment was conducted to investigate the opimum media, media amount and planting density of nutri-culture for the high quality and yield of wild vegetable, Liguliaria fischeri Turez. The fried rice hull was recognized best media among perlite, sand and fried rice hull for nutri-culture of Ligularia fischeri Turcz. The yield on fried rice hull media at early growing stage was lower, but higher than in other media at late growing stage. Therefore, total yield in fried rice hull media was 15-29% higher than that of the existing perlite media. Growth and yield of Ligularia fischeri Turcz. were not significantly different between medium amounts, therefore considering economical efficiency and stability, the optimum medium amount was 105 $\iota/m^3$, Yield by planting density increased in the high density and 50 plants/$m^2$ was considered as proper planting density in nutri-culture of Liguliaria fishcheri Turcz.
