The antioxidant activity of steamed ginger and its protective effects on obesity induced by high-fat diet in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Ginger, a root vegetable, is known to have antioxidant and antiobesity effects. Preparation, such as by steaming, can affect the chemical composition of prepared root vegetables or herbs and can change their functional activities. In the present study, we investigated the protective effects of steamed ginger against oxidative stress and steatosis in C57BL/6J mice fed a high-fat diet. MATERIAL/METHODS: The levels of polyphenols and flavonoids in two different extracts of steamed ginger, i.e., water extract (SGW) and ethanolic extract (SGE); as well, their antioxidant activities were examined. Forty male C57BL/6J mice were fed a normal diet (ND, n = 10), high-fat diet (HFD, 60% fat, w/w, n = 10), HFD supplemented with 200 mg/kg of SGE or garcinia (GAR) by weight (SGED or GARD, respectively, n = 10) for 12 weeks. Serum chemistry was examined, and the expressions of genes involved in lipid metabolism were determined in the liver. Histological analysis was performed to identify lipid accumulations in epididymal fat pads and liver. RESULTS: The SGE had higher contents of polyphenols and flavonoids and higher DPPH and $ABTS^+$ free radical scavenging activities compared to those of SGW. Treatment with SGE or GAR significantly decreased the HFD-induced weight gain. Both SGE and GAR significantly reduced the high serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein levels induced by HFD. Compared to ND, HFD significantly increased hepatic TC and TG levels. SGE or GAR supplementation significantly decreased the increase of hepatic lipids by HFD. Interestingly, SGE had a more significant effect in reducing hepatic TC and TG levels than GAR. Furthermore, hepatic genes involved in lipogenesis and lipolysis were altered in both the SGED and GARD groups. CONCLUSIONS: The present study indicates that steamed ginger supplementation can decrease plasma TC and TG and can inhibit liver steatosis by regulating the expressions of hepatic genes.

국내 다소비 십자화과 새싹채소 추출물의 항산화 활성 (Antioxidant activities of commonly used Brassica spp. sprout vegetables in Korea)

  • 본 연구에서는 국내에서 다소비 되는 십자화과 새싹채소 7종(콜라비, 적무, 브로콜리, 배추, 유채, 무순 및 다채)의 항산화 및 지방세포 내 ROS 생성 억제 활성을 측정하였다. 총 페놀 및 총 플라보노이드 함량을 측정한 결과, 총 페놀 함량은 콜라비 새싹과 무순 추출물이 $23.97{\pm}0.46mg\;TAE/g$$24.40{\pm}1.24mg\;TAE/g$으로 유의적으로 가장 높은 값을 나타내었고, 총 플라보노이드 함량은 무순 추출물이 $15.30{\pm}1.35mg\;CE/g$으로 다른 새싹채소 추출물에 비해 높은 값을 나타내었다. DPPH radical 소거능과 ORAC value를 측정한 결과, 콜라비가 DPPH radical 소거능($IC_{50}=1.95mg/mL$)과 ORAC value($79,032.5{\mu}M\;TE/g$)에서 모두 항산화능이 뛰어난 것으로 나타났다. 한편, 다채를 제외한 새싹채소 추출물 6종 모두에서 세포 내 ROS 생성량을 감소시킴을 확인하였다. 이상의 결과로부터, 십자화과 새싹채소 중 콜라비 새싹과 무순 추출물의 경우 항산화활성 및 지방세포 내 ROS 생성 억제 활성을 가지며, 천연물 유래 항산화 소재로써 활용 가능성이 높은 것으로 기대된다.

천연 조미향상물질의 첨가에 의한 Maillard 반응에서 Meat-like Flavor의 개발 (Development of Meat-like Flavor by Maillard Reaction with Addition of Natural Flavoring Materials)

  • Meat-like flavor를 생성하고자 당과 아미노산 혼합액인 0.2 M cystine-0.1 M lactose-0.1 M maltose를 대조구로 하여 천연 조미향상물질(HVP, HAP, YE)을 첨가하였다. 천연 조미향상물질의 첨가는 대조구에 농도별로 단독 첨가하였으며 단독 첨가시 선정된 농도의 HVP, HAP나 YE를 혼합하여 그 영향을 비교하였다. Meat-like flavor 생성에 유사한 조건인 $100^{\circ}C$에서 8시간 동안 마이야르 반응시킨 반응액은 흡광도, 색, 관능적 성질 및 휘발성 향기성분을 분석하였다. 그 결과 HVP, HAP, YE를 첨가 농도별로 첨가하였을 때 반응 시간과 첨가 농도가 증가할수록 420 nm와 278 nm에서의 흡광도가 증가하였다. 또한 첨가 농도별 색의 변화는 HVP, HAP와 YE 모두 b값은 증가 경향을 나타낸 반면 L값은 감소 경향을 나타내었다. 관능 검사에서 meat flavor에 근접한 각 조미향상물질의 첨가 농도는 1.16% HVP, 0.94% HAP, 1.48% YE이었고 가장 meat flavor에 근접한 반응액은 대조구에 HVP 1.16%와 HAP 0.94%를 혼합 첨가한 반응액이었다. 대조구와 HVP 단독 첨가구 및 HVP와 HAP 혼합 첨가구에 대한 향기 성분을 비교하고자 GC/MS로 분석한 결과 hydrocarbon류 1종, aldehyde류 9종, ketone류 5종, ester류 1종, alcohol류 5종, aromatics(benzene)류 2종, furan류 2종, sulfur compound류 1종을 확인할 수 있었다.

Analysis of Residual Triflumizole, an Imidazole Fungicide, in Apples, Pears and Cucumbers Using High Performance Liquid Chromatography

  • The present study was conducted to monitor the level of triflumizole residues in fruits (apple and pear) and vegetable (cucumber) samples in order to assess risk posed by the presence of such residues to the consumer. Triflumizole was applied at a recommended dose rate to apple and pear pulps and to a cucumber sample. The samples were collected at harvesting time following several treatments (three and/or four treatments). Triflumizole was extracted with methanol and re-extracted into dichloromethane. The presence of triflumizole was determined by HPLC with UV detection at 238 nm following the cleanup of the extract by open preparative chromatographic column with Florisil. The versatility of this method was evidenced by its excellent linearity (> 0.999) in the concentration range between 0.2 and 4.0 mg/kg. The mean recoveries evaluated from the untreated samples spiked at two different fortification levels. 0.1 and 0.4 mg/kg, and ranged from 87.5${\pm}$0.0 to 93.3${\pm}$2.6 for the tested fruits and vegetable, respectively, and the repeatability (as relative standard deviation) from three repetitive determinations of recoveries were no larger than 6%. The calculated limit of detection was 0.02 mg/kg and the minimum detectable level of 4 ng for triflumizole was easily detected. When triflumizole was sprayed onto the apple trees three times at 50-40-30 and 40-30-21 days prior to harvesting and four times onto the pear trees at 40-30-21-14 days prior to harvesting, the mean residual amounts of 0.05 and 0.06 mg/kg for apples and pears, respectively, were not detected in all of the treatments. When the cucumber sample was fumigated four times at 7, 5, 3 and 1 day prior to harvesting, the mean residual amount was not detectable. Triflumizole can be used safely when sprayed (wettable powder, 30% active ingredient) and fumigated (10%) 4 times at 14 and 1 day prior to harvesting to protect the fruits and vegetable, respectively.

Fruit and vegetable discards preserved with sodium metabisulfite as a high-moisture ingredient in total mixed ration for ruminants: effect on in vitro ruminal fermentation and in vivo metabolism

  • Objective: Our recent series of laboratory- and large-scale experiments confirmed that under aerobic and anaerobic conditions, sodium metabisulfite (SMB) was effective in preserving nutrients and antioxidant capacity of highly perishable fruit and vegetable discards (FVD). Hence, the purpose of this study was to examine how partial inclusion of SMB-treated FVD in total mixed ration (TMR) influences in vitro ruminal fermentation, whole-tract digestibility, nitrogen metabolism, blood metabolites, and voluntary feed intake of sheep. Methods: The FVD were mixed thoroughly with 6 g SMB/kg wet biomass and kept outdoors under aerobic conditions for 7 days. Four TMRs including four levels of SMB-treated FVD (as-fed basis) at 0%, 10%, 20%, and 30% (equaling to 0%, 1.9%, 3.8%, and 5.7% on dry matter basis, respectively), were prepared as replacement for corn grain. The ruminal fermentation metabolites were studied using an in vitro gas production test. Four mature male Corriedale sheep were assigned at random to the 4 diets for two separate sub-experiments; i) digestibility trial with four 21-d periods, and ii) voluntary feed intake trial with four 28-d periods. Results: Inclusion of SMB-treated FVD in the TMR tended to quadratically increase partitioning factor. No effect was seen on total-tract digestibility of organic matter, ether extract, crude protein, and acid detergent fiber, except for neutral detergent fiber digestibility that tended to linearly increase with increasing SMB-treated FVD in the TMR. The progressive increase of FVD preserved with SMB in the diet had no effect on nitrogen metabolism. Treatment had no effect on serum antioxidant capacity and blood metabolites assayed. Voluntary feed intake was not impaired by inclusion of SMB-treated FVD in the TMR. Conclusion: It appears that FVD preserved with SMB can be safely incorporated into TMR as replacement of corn grain without impairment of nutrient metabolism and feed intake.

Development of Cabbage Juice Medium for Industrial Production of Leuconostoc mesenteroides Starter

  • Leuconostoc mesenteroides is used as a starter to produce high-quality kimchi products. In this study, an efficient and economical cabbage juice medium (CJM) was developed by process optimization of cabbage extraction and pasteurization and by compositional supplementation of various lacking nutrients. The pasteurized cabbage juice was determined to be a good medium candidate to cultivate L. mesenteroides, showing maximal cell numbers ($9.85{\times}10^8CFU/ml$) after 24 h. Addition of sucrose and yeast extract with soy peptone resulted in increment of bacterial cell counts in CJM, showing the supplementing effect of the lacking nutrients. Furthermore, addition of shell powder gave a protective effect on bacterial cells by preventing pH decline and organic acid accumulation in CJM, resulting in a 2-fold increase of bacterial counts. The optimized composition of CJM was 70% cabbage juice diluted with water, 0.5% (w/v) sucrose, 1% (w/v) yeast extract, 1% (w/v) soy peptone, and 1.5% (w/v) ark shell powder. The CJM developed in this study was able to yield a comparable level of bacterial counts with MRS medium and reduced the cost by almost 10-fold.

Inhibitory activities of Perilla frutescens britton leaf extract against the growth, migration, and adhesion of human cancer cells

  • BACKGROUND/OBJECTIVES: Perilla frutescens Britton leaves are a commonly consumed vegetable in different Asian countries including Korea. Cancer is a major cause of human death worldwide. The aim of the current study was to investigate the inhibitory effects of ethanol extract of perilla leaf (PLE) against important characteristics of cancer cells, including unrestricted growth, resisted apoptosis, and activated metastasis, using human cancer cells. MATERIALS/METHODS: Two human cancer cell lines were used in this study, HCT116 colorectal carcinoma cells and H1299 non-small cell lung carcinoma cells. Assays using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide were performed for measurement of cell growth. Soft agar and wound healing assays were performed to determine colony formation and cell migration, respectively. Nuclear staining and cell cycle analysis were performed for assessment of apoptosis. Fibronectin-coated plates were used to determine cell adhesion. RESULTS: Treatment of HCT116 and H1299 cells with PLE resulted in dose-dependent inhibition of growth by 52-92% (at the concentrations of 87.5, 175, and $350{\mu}g/ml$) and completely abolished the colony formation in soft agar (at the concentration of $350{\mu}g/ml$). Treatment with PLE at the $350{\mu}g/ml$ concentration resulted in change of the nucleus morphology and significantly increased sub-G1 cell population in both cells, indicating its apoptosis-inducing activity. PLE at the concentration range of 87.5 to $350{\mu}g/ml$ was also effective in inhibiting the migration of H1299 cells (by 52-58%) and adhesion of both HCT116 and H1299 cells (by 25-46%). CONCLUSIONS: These results indicate that PLE exerts anti-cancer activities against colon and lung cancers in vitro. Further studies are needed in order to determine whether similar effects are reproduced in vivo.

식방풍의 혈관성 치매에 대한 예방과 치료효과 검증 (The Protective and Recovery Effects of Peucedanum Japonicum Thunberg for Vascular Dementia)

  • Objectives : During several thousand years, Peucedanum Japonicum Thunberg has been considered as a vegetable side dish in Korea. There is folk knowledge that Peucedanum Japonicum Thunberg prevents vascular disease such as stroke. To identify the effects Peucedanum Japonicum Thunberg, we made up its extract and named it as KH020. Then, we employed common carotid artery ligation (CCAl) surgery for vascular dementia model (VDM), and two types doses of per os (per oral: p.o) treatment. Methods : To confirm prevention and recovery effects for vascular dementia, we treated two doses (100, 400 mg/kg) KH020 in male C57BL/6 mouse during 7 days. After treatment, animals were CCAl operated, and given time to recover. Then, animal were tested in a Y-maze and passive avoidance test. Results : Y-maze results demonstrated that cognition and memory performance were decreased in the VDM group, compared to the sham group. KH020 treatment abolished these effects significantly. The results from the passive avoidance test showed the same phenomenon, but it was not statically significant. Conclusions : Therefore, KH020 prevents the onset of vascular dementia. In future studies, we will evaluate KH020 in regard to alzheimer dementia.

-Aspergillus Parasiticus R-716의 생육 및 aflatoxin 생성에 미치는 채소추출물의 영향- (Effects of Vegetable Extracts on the Growth and the Aflatoxin Production by Aspergillus Parasiticus R-716)

  • 15종의 채소를 Chloroform으로 추출물을 조제하여 공시균(Aspergillus parasiticus R-716)의 생육 및 aflatoxin 생성에 미치는 영향을 조사하였다. 그 결과 마늘, 생강, 무우 및 양배추의 순으로 공시균의생육을 저해하였으나, 가지, 상치 등은 오히려 생육을 촉진하였다. 또한 균체생성시 공시균의 aftlatoxin기 생성은 무우, 생강, 쑥갓, 양배추의 순으로 저해하였으나, 우엉, 고추는 오히려 증가시켰다. 특히 무우는 균체 g당 aflatoxin 함량을 감소시켜 가장 효과적인 채소로 나타났으며, 고체배지에서도 무우 30g에 해당되는 추출물의 첨가로 약 $80\%$의 aflatoxin생성이 저해되어 쌀, 보리배지에서 각각 484 및 $191{\mu}g$의 aflatoxin만이 생성되었다.

A Study on the Dyeing of Ramie Fabric Treated with Medicinal Plant IV. The Natural Dyeing on Ramie Fabric Using Mentha arvensis Herba

  • This study was performed to investigate the effect of Mentha arvensis Herba extract on the treatment of chromaticity and colorfastness. Mentha arvensis Herba has been used as a Korean medicine. It is effective in headache and stress. It is also good as a aphtha and in treating cold. In the long history of Korea, dyeing has been applied for a means representing the grace of natural and inner esthetic consciousness of man. Vegetable dyes give us such great benefits, diversified color, but no pollution. And ramie fabric has distinctive features such as beautiful brilliance, elegance, and strong durability. So, it is regarded as a special product of Korea traditionally. These studies were carried out to treat with acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was died with Mentha arvensis Herba extract. The results of experiment showed as follows: First, the chromophoric degree was the highest in acetate iron but not distinction in another mordants. Second, the light colorfastness was the highest in non treated and dichloride copper, but alum was the lowest. Third, the discoloration was alum and dichloride copper showed first grade in washing colorfastness. Abrasion colorfastness was not significant in this test. According the previous results, Mentha arvensis Herba has an efficiency in removing aphtha and in treating cold. So it is considered that Mentha arvensis Herba can be applied effectively to headache and stress.
