Heavy Metal Tolerance of Novel Papiliotrema Yeast Isolated from Vietnamese Mangosteen

  • Three yeast strains (Hue-1, Hue-8, and Hue-19) with strong heavy metal tolerance were isolated from mangosteen from Hue city, Vietnam. They exhibited identical phenotype and phylogeny. Sequence analysis of the D1/D2 region of the LSU rRNA gene and the internal transcribed spacer (ITS) region demonstrated that the closest relative of these strains is Papiliotrema sp. with 2.12% and 3.55-3.7% divergence in the D1/D2 domain, and ITS domain, respectively. Based on the physiological, biochemical, and molecular data, the three strains belong to a novel species of Papiliotrema genus, for which the name Papiliotrema huenov sp. nov. is proposed. These strains are highly tolerant of heavy metals compared to other yeasts, being able to grow in the presence of 2 mM Pb (II), 2 mM Cd (II), and up to 5 mM Ni (II), but no growth was observed in the presence of 1 mM As (III).

  • Acanthochitonidae Pilsbry, 1893 is one of the chiton families characterized by 9 pairs of sutural tufts on a well-expanded girdle that overgrows plates. It is found in intertidal and subtidal zones worldwide. Of its eight genera, only two have been previously reported in South Korea: Acanthochitona Gray, 1821 and Notoplax H. Adams, 1862. Here we report Leptoplax doederleini (Thiele, 1909) as the first record of the genus Leptoplax in South Korea and describe and compare its morphological features with N. kaasi Hong, Dell'Angelo and Van Belle, 1990. In addition, we provide the partial sequence of the mitochondrial DNA cytochrome c oxidase subunit I gene as a DNA barcode sequence record.

  • The purpose of this study was to measure and observe Memes of people in Korean modern architectural discourse. After extracting Memes from the text of 'SPACE' magazines, network analysis was used with the Memes which were obtained. The results of this study were as follows; 1. Korean modern architectural discourse field was influenced by people in order of American, French and Korean. 2. Korean modern architectural discourse field was influenced by people in order of Le Corbuiser, Mies Van der Rohe, Louis Kahn, Kim Soogeun and Gilles Deleuze.

Anti-inflammatory and PPAR Subtypes Transactivational Activities of Phenolics and Lignans from the Stem Bark of Kalopanax pictus

  • A new compound, kalopanaxin F (3), and 11 known compounds (1, 2, 4-12), were isolated from the stem bark of Kalopanax pictus. Their structures were elucidated on the basis of chemical and spectroscopic methods. Five of the compounds (2, 3, 5, 6, and 12) significantly inhibited $TNF{\alpha}$-induced NF-${\kappa}B$ transcriptional activity in HepG2 cells in a dose-dependent manner, with $IC_{50}$ values ranging from 6.2 to 9.1 ${\mu}M$. Furthermore, the transcriptional inhibitory function of these compounds was confirmed based on decreases in COX-2 and iNOS gene expression in HepG2 cells. Compounds 3-7, 9, and 12 significantly activated the transcriptional activity of PPARs dose-dependently, with $EC_{50}$ values ranging from 4.1-$12.7{\mu}M$. Compounds 4 and 5 exhibited $PPAR{\alpha}$, $PPAR{\gamma}$, and $PPAR{\beta}({\delta})$ transactivational activities in a dose-dependent manner, with $EC_{50}$ values of 16.0 and 17.0, 8.7 and 16.5, 26.2 and 26.3 ${\mu}M$, respectively.

  • The title compound (C13H17NO2S) is monoclinic, space group P21/a, with a = 13.756 (3), b = 9.310(4), c = 21.305(3) $\AA$, $\beta$ = 95.0。, Z = 8, V = 2718.11 $\AA$3, Dc = 1.23$g·{\cdot}cm-3$, (Mo k$\alpha$) = 0.71069$\AA$,$\mu$ = 2.18 cm-1, F(000) = 1071.86, T = 298, R = 0.085 for 2935 unique observed reflections with I >2.0$\sigma$(I). The structure was solved by direct methods. The C-H bond lengths and the methyl groups are fixed and refined as their ideal geometry by allowing to ride on the parent atoms. Both molecules A and B have almost same structures except for two terminal ethyl groups. The ethylene-like skeleton including the nitro group in one molecule is nearly perpendicular to the plane of the methylbenzene group and two ethyl groups form a cis-type structure which has the dfferent orientations between two molecules; in the molecule A, two terminal methyl groups being the opposite directional arrangement against the plane of its skeleton, while in the B, with the same directional structure from its plane. The molecules in the crystal are packed together by non-bonded van der Waals forces.

Comparison of Virulence Factors of Enterococci from Intestinal Drugs, Infant Feces and Clinical Isolates (정장제, 신생아 분변 및 병원에서 분리한 장구균의 병독성인자 비교)

  • Three isolates, E. faecium P1, P2 and P3, from intestinal drugs of three phamaceutical companies, four clinical vancomycin resistant isolates, E. faecium V1, V2, V3 and E. faecalis V4, and three isolates, E. faecalis DW01, DW07 and DW14, from infant feces were tested for the presence of virulence genes, ace, agg, esp, efaA, gelE, sprE, vanA and vanB as well as fsrABC, regulatory genes of gelE and sprE, cylMBA, cytolysin activation genes and cpd, cob and ccf, pheromone genes by PCR and for their phenotype activities such as protease, biofilm formation, cell clumping and hemolysis. The genes encoding cell surface adherence proteins, ace, agg, esp and efaA, were predominantly amplified from the vancomycin resistant strain V4 and the fecal isolates DW01, DW07 and DW14. Both protease and biofilm formation activity were detected only from E. faecalis V4 from which the PCR products of gelE and spreE as well as fsrABC were amplified. The pheromone genes were amplified from the V4, DW01, DW07 and DW14 strains and these strains showed clumping activity. Biofilm formation was observed from the strains DW01, DW07 and DW14, all of which produced PCR products of pheromone, and V4 as well. Whole cytolysin regulator genes were amplified from DW01, DW07 and DW14 and ${\beta}$-hemolysis activity was detected from these strains. Any virulence genes or activities except the pheomone gene ccf were not detected from the pharmaceutical isolates, E. faecium P1, P2 and P3.

Detection of Vancomycin Resistance Genes and Antibiotic Resistance Characteristics of Enterococcus spp. Isolated from Inland Pollution Sources Near Shellfish Farms on the West Coast of South Korea (서해안 패류양식장 인근 육상오염원에서 분리한 장구균의 Vancomycin 내성 유전자 검출 및 항생제 내성 특성)

  • In this study, 143 strains of Enterococcus spp. were isolated from inland pollution sources near shellfish farms on the west coast of South Korea. Not all isolated Enterococcus spp. strains possessed vancomycin resistance genes (VanA and VanB). However, since vancomycin-resistance Enterococcus (VRE) have been detected not only in the clinical field but also out in the world, it is possible that the VRE gene may be transferred to other bacterial strains commonly found in coastal waters where seafood is produced. It is important to monitor trends in the appearance of VRE. In addition, antimicrobial resistance patterns of isolates were examined in this study. Overall antimicrobial resistance rates were high: ciprofloxacin (32.2% of isolates resistant), chloramphenicol (30.8%), quinupristin/dalfopristin (19.6%), and tylosin (15.4%). Eight E. faecium strains (6.2%), out of the 129 strains assessed, showed multidrug resistance. All multidrug-resistant E. faecium showed resistance to erythromycin, quinupristin/dalfopristin, tetracycline, and tylosin, in all 14 strains. All multidrug-resistant E. faecalis showed resistance to erythromycin, quinupristin/dalfopristin, tetracycline, and tylosin. Both multidrug-resistant E. faecium and multidrug-resistant E. faecalis showed common resistance to erythromycin, quinupristin/dalfopristin, tetracycline, and tylosin.

Promoter Analysis of the Cell Surface-abundant and Hypoviral-regulated Cryparin Gene from Cryphonectria parasitica

  • Cryparin, encoded as a single copy gene (Crp) of the chestnut blight fungus Cryphonectria parasitica, is the most abundant protein produced by this fungus. However, its accumulation is decreased remarkably in C. parastica strains containing the double-stranded (ds) RNA virus Cryphonectria hypovirus 1. To characterize the transcriptional regulatory element(s) for strong expression and viral regulation, promoter analysis was conducted. Serial deletion of the Crp promoter region resulted in a step-wise decrease in promoter activity, indicating a localized distribution of genetic elements in the cryparin promoter. Promoter analysis indicated two positive and a repressive cis-acting elements. Among them, the promoter region between nt -1,282 and -907 appeared to be necessary for hypoviral-mediated down-regulation. An electrophoretic mobility shift assay (EMSA) on the corresponding promoter region (-1,282/-907) indicated two regions at (-1,257/-1,158) and (-1,107/-1,008) with the characteristic AGGAGGA-N42-GAGAGGA and its inverted repeat TCCTCTC-N54-TCCTCCT, respectively, appeared to be specific binding sites for cellular factors.

Development of Molecular Markers for Xanthomonas axonopodis Resistance in Soybean

  • A single recessive gene, rxp, controls the bacterial leaf pustule (BLP) resistance in soybean and in our previous article, it has been mapped on linkage group (LG) D2 of molecular genetic map of soybean. A total of 130 recombinant inbred lines (RILs) from a cross between BLP-resistant SS2-2 and BLP-susceptible Jangyeobkong were used to identify molecular markers linked to rxp. Fifteen simple sequence repeat (SSR) markers on LG D2 were screened to construct a genetic map of rxp locus. Only four SSR markers, Satt135, Satt372, Satt448, and Satt486, showed parental polymorphisms. Using these markers, genetic scaffold map was constructed covering 26.2cM. Based on the single analysis of variance, Satt372 among these four SSR markers was the most significantly associated with the resistance to BLP. To develop new amplified fragment length polymorphism (AFLP) marker linked to the resistance gene, bulked segregant analysis (BSA) was employed. Resistance and susceptible bulks were made by pooling equal amount of genomic DNAs from ten of each in the segregating population. A total of 192 primer combinations were used to identify specific bands to the resistance, selecting three putative AFLP markers. These AFLP markers produced the fragment present in SS2-2 and the resistant bulk, and not in Jangyeobkong and the susceptible bulk. Linkage analysis revealed that McctEact97 $(P=0.0004,\;R^2=14.67\%)$ was more significant than Satt372, previously reported as the most closely linked marker.
