대서와 보라밸리 감자 두 품종의 이화학적 특성 (Physico-chemical Characteristics of Atlantic and Bora Valley Potato)

  • Comparisons of the physico-chemical characteristics between white Atlantic potatoes and violet Bora Valley potatoes were performed. According to the results, Bora Valley had 7% higher moisture content than Atlantic, but there were no differences in crude fat and crude ash contents. Bora Valley, however, had about 3% less crude protein content than Atlantic. In a vitamin C analysis by HPLC, the Atlantic and Bora Valley potatoes contained 14% and 39% vitamin C, respectively, indicating that Bora Valley contained about twice as much of the vitamin as Atlantic. In a vitamin C analysis by HPLC, the Atlantic and Bora Valley potatoes contained 14% and 39% vitamin C, respectively, The dietary fiber content of Bora Valley was 3.12 g per 100 g higher than that of the Atlantic variety. While Atlantic contained neither glucose nor fructose, both sugars were measured in the Bora Valley potatoes, and there was no difference in sucrose content between the cultivar.

  • Potato plants carrying the Ry gene are extremely resistance to a number of potyviruses, but it is not known which variety expressed the resistance. In this investigation, combined classical and molecular techniques were used to identify virus resistance potatoes. Mechanical inoculation of 32 varieties of Korean potato cultivars, with potato virus Y (PVY), induced various symptoms, such as mosaic, yellowing, necrosis, mottle, vein clearing and vein bending. Different virus spreading patterns were observed, such as highly sensitive, moderate and resistant to $PVY^o$ inoculated leaves in different cultivars. From the results of double antibody sandwich-enzyme links immunosorbant assays (DAS-ELISA), coupled with reverse transcription polymerase chain reaction (RT-PCR), Winter valley and Golden valley were found to be highly susceptible and resistant cultivars to $PVY^o$ respectively. TEM was used as a complementary method to conform the localization of the virus in leaf tissues. TEM detect virus particles in Golden valley, where, ELISA and RT-PCR were unable to detect the CP gene. However, the interior part of the tissues was severely deformed in $PVY^o$ infected Winter valley, than Golden valley The Ry gene is involved in an induced response in $PVY^o$ infected Golden valley plants. The methods described in this study could be applied for the screening and development of antiviral potatoes.

국내산 신품종 감자의 영양성분 및 이화학적 특성 (Nutrient Components and Physicochemical Properties of New Domestic Potato Cultivars)

  • (주)포테이토밸리에서 개발한 감자 신품종인 구이밸리, 보라밸리, 고구밸리와 기존 품종인 수미의 영양성분과 이화학적 특성을 조사하였다. 일반성분의 경우 조단백은 수미가, 회분은 고구밸리가, 탄수화물은 보라밸리가 가장 높게 나타났으며, 지방산은 linoleic acid, linolenic acid와 같은 필수지방산의 비율이 높으면서 품종간 함량 차이는 거의 없었다. 무기질 함량은 고구밸리가 가장 높았으며, K와 P가 주요 무기질인 것으로 나타났다. 구이밸리는 전분 함량이 높고 환원당 함량은 낮아 가공용 감자로 이용하기에 적합한 품종이었고, 식이섬유와 vitamin C는 고구밸리가 기타 품종에 비해 월등히 높은 함량을 나타내었다. 페놀산의 경우 chlorogenic acid, caffeic acid 그리고 ${\rho}-coumoric$ acid 등이 정량되었고 chlorogenic acid와 caffeic acid가 주요 페놀산이었으며, 보라밸리가 가장 높은 페놀산 함량을 나타내었다. DSC 분석 결과 호화개시 온도는 수미에 비하여 밸리 품종이 비교적 높은 호화개시 온도를 나타내었고, 피크온도도 유사한 경향이었으며, 호화 엔탈피는 구이밸리가 가장 높은 값을 나타내었다. RVA 상의 페이스트 온도는 DSC 결과에서의 호화개시온도 경향과 유사하였으며, 구이밸리가 가장 높은 최고점도와 최종점도를 나타내었다.

Development of New Potato Cultivars for the Utilization of Healthy Food with High Biological Function

  • Potatoes have been recognized for a long time as one of the major food crops as well as horticultural crops. Potato production as a table food has been decreased in developed countries, while it has been steadily increased in the third world countries for it importance as food source. It is a new trend to look for the food, not only as a feeding crop but also healthy food. It is also time for the potato producers to look for the potato having high economic value as found in medicinal plants. There are great diversities in potato species, indicating that valuable compounds can be found in different amounts, depending on potato species. We screened the cultivars, breeding clones, and germplasms based on the vitamin C, Vitamin E, antioxidant compounds, diverse sugar types, important amino acids, and other valuable compounds. We could select the breeding clones KC003, 98Wl17, 99j717, and Vally 8 (A group) due to their high levels of antioxidant compounds, and it can be said that most of the red and purple colored potato clones belong to the A group. In the contents of essential amino acids, ‘Taebook Valley’,‘Summer Valley’ and other breeding clones were found to be high in amount. We also made crosses between breeding clones with high biological function and low agronomic traits and low biological function with high quality in agronomic characteristics . The patterns of genetic trends of these offsprings in comparison with their parents will be reported as well. And the potential of using potato as antibody production of anti-cancer will be discussed.

Clinical Effects of Intake of Juice Valley and Gogu Valley toward Fecal Microflora of Healthy Human Volunteers

  • Juice Valley and Gogu Valley were administered to twelve healthy young volunteers for 4 weeks to study their clinical effects on human intestinal microflora. Changes in fecal microflora, fecal moisture, and fecal pH were observed for Juice Valley intake. Administration of Juice Valley significantly increased numbers of Bifidobacterium and Lactobacillus from 8.69 and 7.02 to 10.89 and 9.02 (Log CFU/g wet feces), respectively, whereas those of Clostridium perfringens and Escherichia coli decreased. Moisture content of feces increased, and fecal pH decreased after 4 weeks of Juice Valley intake, intake of Gogu Valley slightly increased growth responses of Bifidobacterium and Lactobacillus and decreased growth responses of C. perfringens and E. coli. Su-mi potato, as a reference, had no effect on Bifidobacterium and Lactobacillus numbers. This study confirmed Juice Valley has better effects than Gogu Valley and Su-mi, and has important role on growth promotion and inhibition of human intestinal bacteria.

Physicochemical Properties of Starches Isolated from New Potato Cultivars in Korea

  • Physicochemical properties of starches isolated from 3 new potato cultivars developed by Potato Valley Ltd. were investigated and compared to those of starch isolated from 'Superior' being distributed prominently in Korea. Significant differences were observed in physicochemical properties such as granule size, amylose content, phosphorus content, water binding capacity, swelling power, solubility, and in vitro digestibility of starches from different potato cultivars. Thermal properties were evaluated using differential scanning calorimetry (DSC), and onset gelatinization temperatire ($T_o$), peak temperature ($T_p$), and enthalpies of gelatinization (${\triangle}H$) of different potato cultivars ranged as $58.0{\pm}0.3-61.7{\pm}0.4^{\circ}C,\;63.7{\pm}0.2-$ $66.5{\pm}0.0^{\circ}C$, and $15.6{\pm}0.5-17.0{\pm}0.3$ J/g, respectively. Pasting properties were evaluated using a rapid visco analyzer (RVA), and pasting temperature, peak viscosity, and final viscosity of different potato cultivars ranged as $65.0{\pm}0.1-68.9{\pm}0.1^{\circ}C$, $8,163.7{\pm}196.3-9,035.7{\pm}6.4$ cp, and $4,397.7{\pm}166.7-7,025.0{\pm}271.3$ cp, respectively. Especially, 'Gogu valley' starch showed the highest values in the amylose and phosphorus content among tested potato cultivars and higher water binding capacity, swelling power, and solubility than those of other tested starches. And it also showed high pasting temperature, peak viscosity, trough viscosity, and final viscosity as compared to other tested starches.

폴리페놀, 안토시아닌과 비타민 C 함량이 우수한 감자 계통 선발 (Selection of the Excellent Potato Clones Based on Total Polyphenol, Anthocyanin and Vitamin C Contents)

  • 본 연구는 기능성 식품 소재용 감자 계통을 선발하고자, 35개의 계통 및 대조구 품종인 '수미', '다솜밸리' '고구밸리'의 총 폴리페놀, 총 안토시아닌, 비타민C의 함량을 분석하였다. 분석결과, 계통별 함량차이가 큰 것으로 나타났으며, 그 가운데서 총 폴리페놀 함량은 KPG16 계통이 $105.08mg{\cdot}100g^{-1}\;FW$, 총 안토시아닌 함량은 KPG13 계통이 $4.78mg{\cdot}100g^{-1}\;FW$, 비타민 C 함량은 KPG20 계통이 $22.16mg{\cdot}100g^{-1}\;FW$으로 각각 높게 나타났다. 종합적으로 분석한 결과, KPG5계통이 총 폴리페놀, 총 안토시아닌, 비타민 C 함량이 각각 103.95, 3.15, $12.12mg{\cdot}100g^{-1}\;FW$ 으로 3성분을 고루 함유하고 있음을 확인할 수 있었으며, 추후 기능성 품종 선발의 소재로 활용될 수 있을 것으로 사료된다.

Evaluation of Potato Varieties (Solanum tuberosum L.) on Fecal Microflora of Human Volunteers

  • Effects of Dasom Valley and Bora Valley on fecal microflora, fecal moisture, and fecal pH of twelve healthy human volunteers were investigated. Numbers of Bifidobacterium, Clostridium perfringens, Escherichia coli, and Lactobacillus of control group were $9.24{\pm}0.63$, $4.44{\pm}1.21$, $7.75{\pm}0.38$, and $6.98{\pm}0.81$ (Log CFU/g wet feces), respectively. During administration of Dasom Valley, numbers of Bifidobacterium and Lactobacillus were $10.70{\pm}0.44$ and $8.84{\pm}0.77$, whereas those of C. perfringens and E. coli were $2.96{\pm}1.50$ and $6.69{\pm}0.29$, respectively. Administration of Dasom Valley significantly increased growth responses of beneficial bacteria, Bifidobacterium and Lactobacillus, whereas those of harmful bacteria, C. perfringens and E. coli, significantly decreased. Moisture content of feces increased and fecal pH decreased with intake of Dasom Valley. Intake of Bora Valley slightly increased numbers of Bifidbacterium and Lactobacillus and slightly decreased those of C. perfringens and E. coli. Results indicate Dasom Valley has greater intestinal-modulating effect than Bora Valley and Atlantic. Daily intake of Dasom Valley may normalize disturbed physiological functions, resulting in improvement of growth and composition of microbial community within intestinal tract.

안지오텐신 전환효소활성 저해에 의해 항고혈압 효과를 갖는 신품종 감자 구이벨리 추출물 (Anti-hypertensive Activity of New Potato(Solanum tuberosum) Variety of Gui Valley Via Inhibition of Angiotensin Converting Enzyme)

  • This study was attempted to investigate the anti-hypertensive activity of Solanum tuberosum, cv. Gui valley. Ethanol extract of Gui vally (EGV) increased free radical scarvenge activity up to 91.6% of control at $200{\mu}g/m{\ell}$. It's anti-oxidant activity is similar to 10 uM of ascorbic acid, well known as antioxidant. EGV inhibited Angiotensin-I-converting enzyme (ACE) activity in vitro. ACE plays a important role in regulation of blood pressure and ACE inhibitors are important for the treatment of hypertension. Anti-hypertensive activity of EGV in vivo was assessed in lead acetate (LAT)-induced hypertensive rats for 8 weeks. Elevated blood pressure in control group was significantly decreased by EGV at 200 mg/kg. Also, ACE activity in blood was also suppressed by EGV treatment. Taken together, these results suggest that EGV has an anti-hypersive activity via inhibition of ACE and can be used for the treatment or prevention of hypertension.

Expression of Arabidopsis thaliana SIK (Stress Inducible Kinase) Gene in a Potato Cultivar (Solanum tuberosum L. 'Taedong Valley')

  • Osmotic stress is one of major limiting factors in crop production. In particular, seasonal drought often causes the secondary disease in the field, resulting in severe reduction in both quality and productivity. Recent efforts have revealed that many genes encoding protein kinases play important roles in osmotic stress signal transduction pathways. Previously, the AtSIK (Arabidopsis thaliana Stress Inducible Kinase) mutants have shown to enhance tolerance to abiotic stresses, accompanying with higher expression of abiotic stress-related genes than did the wild-type plants. In this study, we have transformed potato (cv. Taedong Valley) with the AtSIK expression cassette. Both PCR and RT-PCR using AtSIK-specific primers showed stable integration and expression of the AtSIK gene in individual transgenic lines, respectively. Foliar application of herbicide ($Basta^{(R)}$) at commercial application rate (0.3% (v/v)) revealed another evidence of stable gene introduction of T-DNA which includes the bar gene for herbicide resistance. Overexpression of the AtSIK gene under dual CaMV35S promoter increased sensitivity to salt stress (300 mM NaCl), which was demonstrated by the reduction rate of chlorophyll contents in leaves of transgenic potato lines. These results suggest that possible increase of osmotic tolerance in potato plants may be achieved by antisense expression of AtSIK gene.
