Vaccination is one of the most important and cost-effective methods of preventing infectious diseases. Over the past decade, scientific in molecular biology and immunology have improved understanding of many diseases and led to the development of novel strategies for vaccination. An ideal vaccine would induce effective immunity specific for the type of infection, have long duration, require minimal or no boosters, have safety, would not induce adverse reaction, and be easy to administer. The desire to meet these criteria has resulted in the development of vaccines that do not depend on the use of the viable disease agent. It is not the intent of this review to give an extensive review of the field of vaccinology, but rather to address characteristics of conventional and genetically engineered vaccines.
One of the practical limitations with the use of liposomes for delivery of the pharmaceutical substances such antigens is that liposomes are relatively unstable in storage. In order to extend the stability of liposome in storage without affecting their functional activity, solution-type liposomes were dehydrated to form a structurally intact dry liposomes. Comparative immunological evaluation was carried out for both dry and solution-type liposomes containing gag-V3 chimera, consequently it was found that dry liposomes elicited both humoral and cellular response as efficiently as solution-type liposemes did against the same gag-V3 antigen. Especially, long-term stability of the liposomes was remarkably enhanced by the dehydration made to loposomes without a significant change in its ability to elicit immune response in vivo. These results indicate that dry pH-sensitive liposome may become an effective delivery and adjuvant system for general vaccine development.
The vaccines has been developed over the first two hundred years since Jenner's smallpox vaccination. In modern days, vaccination has had the largest impact on the incidence and persistence of infections. Although natural infection induces lifelong immunity, the assumption that the vaccine also confers permanent protection has been reconsidered following outbreaks of measles in students who had been vaccinated 15-20 years prior to infection in the US in the 1980s. Clinical studies have proposed several mechanisms such as vaccine failure in some individuals and the subsequent loss of immunity after vaccination. An ideal vaccine is relatively easy to define, but few real vaccines approach the ideal. Many difficulties account for the failure in producing these ideal vaccines. However, recent advances in methods for studying immune response to pathogens have provided a better understanding of immune mechanisms. Based on these findings, the development of good vaccine formulations allowing stimulation of optimal and prolonged protective immunity and immunization policies or schedules should lead to the introduction of vaccines for previously resistant organisms.
The Ministry of Food and Drug Safety of Korea made an official announcement in March 2018 that the total number of inoculations of Hantaan virus vaccine ($Hantavax^{(R)}$) would change from 3 to 4. Some aspects of this decision remain controversial. Based on the characteristics of Hantaan virus (HTNV) and its role in the pathogenesis of hemorrhagic fever with renal syndrome, it might be difficult to develop an effective and safe HTNV vaccine through the isolate-inactivate-inject paradigm. With the development of high-through-put 'omics' technologies in the 21st century, vaccinomics has been introduced. While the goal of vaccinomics is to develop equations to describe and predict the immune response, it could also serve as a tool for developing new vaccine candidates and individualized approaches to vaccinology. Thus, the possibility of applying the innovative field of vaccinomics to develop a more effective and safer HTNV vaccine should be considered.
본 연구의 목적은 해산어류와 갑각류에 심각한 위해를 끼치고 있는 V. harveyi에 대한 백신개발을 위하여 V. harveyi 백신생산에 적합한 배양배지를 탐색하고 적정 투여량을 조사하며 또한 S. parauberis 백신과 혼합백신의 형태로 투여시 백신효능에 대해 조사하는 것이다. 이를 위하여 2-2'-dipyridyl이 첨가되거나 첨가되지 않은 TSB와 BHIB에 배양 후 FKC 백신을 제작한 후 넙치에 투여하여 응집항체가의 생산변화와 공격실험에서의 상대생존율을 비교분석하였다. 또한 V. harveyi 백신의 적정 투여량을 정하기 위하여 어체중 kg 당 10mg 또는 20mg을 투여하여 면역반응을 비교하였으며 S. parauberis 백신을 혼합한 백신을 투여한 후 면역반응을 비교하였다. 그 결과 2-2'-dipyridyl이 들어간 TSB와 BHIB에 배양된 V. harveyi 백신은 응집항체형성과 방어력에서 차이를 보이지 않았다. 또한 백신 투여량에 따른 응집항체가에 있어서 큰 차이는 없었으나 어체중 kg 당 10mg을 투여한 실험구가 조금 높은 방어력을 나타내었다. S. parauberis 백신과 혼합한 dual 백신을 투여시 V. harveyi 백신만을 단독으로 투여했을 때와 비교해 방어력이 현저히 증가하였으며 특히 어체중 kg당 두가지 백신을 각각 10mg씩 혼합하여 투여한 실험구에서는 28일동안 폐사가 전혀 일어나지 않아 백신효능이 매우 뛰어난 것으로 나타났다. 결론적으로 넙치를 위한 V. harveyi 백신을 개발할 때는 2-2'-dipyridyl가 첨가된 TSB에 배양 후 제작된 V. harveyi 백신을 어체중 kg당 10mg의 투여량으로서 S. parauberis 백신과 혼합투여하는 것이 효능과 경제적인 면에서 바람직하다고 생각된다.

Salmonella typhi Ty21a is an attenuated strain of S. typhimurium and used for oral typhoid vaccine. In an attempt to increase the stability of Ty21a manufacturing typhoid vaccine, the stability of freeze-dried Ty21a including additives at various temperature conditions was studied. In order to investigate the freeze-drying rate of Ty21a according to various absorbance, Ty21a was lyophilized by using 8% sucrose as a stabilizer. (omitted)
In this study we investigated the effect of vaccine efficacy and immune activities by feeding additives several natural herbal remedies extracts of Sweet Wormwood (Artemisia annua), Galla Rhois, Oriental raisin tree (Havenia Dulicis) and 6-mixed herb to Oilve flounder, Paralichthys olivaceus. Each group of fish fed with the herbal remedies added feed and basic formula during the seven weeks and vaccinated with Edwardsiella tarda Formalin killed vaccine after 4 weeks feeding. Relative Percentage Survival values (RPS) in the group was assessed by the challenge with E.tarda. All groups with the herbal remedies added feed enhanced growth rate, but there were not significantly different in lysozyme activity and agglutination titer. In a challenge experiment with E.tarda, RPS in the all groups feeding additives natural herbal remedies was higher than that of the control group. These results suggest that the natural herbal remedies extracts of Sweet Wormwood (Artemisia annua), Galla Rhois, Oriental raisin tree (Havenia Dulicis) and 6-mixed herb would be effective to enhance efficacy of vaccination to Olive flounder.
Objective: This study aimed to develop information materials on vaccine's safety and adverse events which can be utilized by healthcare professionals when prescribing, dispensing and administering vaccines and also by non-healthcare professionals such as pharmaceutical distributors of vaccine. Methods: Information materials regarding vaccines from domestic and foreign governmental organizations, academies, medical organizations and pharmaceutical companies were reviewed. Advisory Committee which consisted of experts in the areas of the vaccine's safety verified the contents and the final information material. Results: Based on the collected data, we developed general guidelines including vaccine constituents, safety information and adverse events of each vaccines, storage and handling, and labeling information. The information materials were developed for both healthcare professionals and non-healthcare professionals such as vaccine distributors. Conclusion: Information materials on vaccine's safety and adverse events developed from this study could be utilized to provide useful information on the vaccine to the medical institutions and distributors.
본 연구는 코로나19 백신에 대한 대학생의 인식을 조사하기 위해 20세 이상 대학생 415명을 대상으로 구글설문을 이용한 설문조사를 수행하였다. 그 결과 집단면역을 위해 '코로나19 백신접종이 필요하다고 생각한다'는 결과가 평균 3.90으로, '백신접종의 필요성'은 65.8%가 인식하고 있었으나, '백신의 안전성'에 대해서는 35.4%가 부정적 평가를 보였다. 코로나19 백신접종 의향은 '순서가 오면 바로 접종할 것이다'가 34.7%로, 현재 코로나19 백신의 예방접종에 대해서는 긍정적이지 않았다. 백신접종을 원하지 않는 이유로는 '백신의 이상반응'이 65.3%, '백신 자체에 대한 불신'이 25.8%이었다. 또한 선진국과 저개발국가와의 백신공급을 불평등으로 인식하고 있었고, 코로나19 백신의 이상반응에 두려움을 느낀다는 평균 3.94로 조사되었다. 따라서 보다 효과적으로 코로나19 백신에 대한 정보를 습득할 수 있도록 대학생이 많이 사용하는 매체를 통한 홍보 방법의 연구, 플랫폼 개발 및 교육이 요구 된다.

The potency of influenza vaccine is determined based on its hemagglutinin (HA) content. In general, single radial immunodiffusion (SRID) assay has been utilized as the standard method to measure HA content. However, preparation of reagents for SRID such as antigen and antibody takes approximately 2~3 months, which causes delays in the development of influenza vaccine. Therefore, quantification of HA content by other alternative methods is required. In this study, we measured HA contents of H1N1 antigen and H1N1 influenza vaccine by reverse phase-high performance liquid chromatography (RP-HPLC) methods. The presence of HA1 and HA2 was investigated by silver staining and Western blot assay. In addition, accuracy and repeatability of HA measurement by RP-HPLC were evaluated. Comparison of HA concentration by SRID and RP-HPLC revealed a precise correlation between the two methods. Our results suggest that RP-HPLC assay can replace SRID in the event of a pandemic flu outbreak for rapid vaccine development.
