Norinyle 복용이 체내대사에 미치는 영향에 대한 연구 (A Study on Metabolic Effects of Norinyle Administration on Female Guinea Pig)

  • In order to observe metabolic effects of an oral conceptive, Norinyle, on female Guinea pig, the changes of ascorbic acid amount and alkaline phosphatase activity in the liver and serum were determined, and histochemical changes of the uterus were observed by microscopic and electronmicroscopic methods by administration of Norinyle with or without ascorbic acid. The results obtained are summerized as follows: 1) The metabolic changes were clearly influenced by the administration of Norinyle alone, but the changes were diminshed by administration of Norinyle with ascorbic acid. 2) The adimnistration of Norinyle influenced to increases the requirment of ascorbic acid in the liver. 3) The uterus weight of the Norinyle administered group was much increased, while the weight was less increased in the group of administered Norinyle with ascorbic acid than the control. 4) The Norinyle administration was brought about an atrophy of endometrium, of uterus especially, functional layer, that the formation of glands were inadequately and the fromation of basal layer and stroma were diminished. 5) An acute infarction on the all layers of the uterus was developed at 9th and 25th days of Norinyle administration and 20th day of Norinyle with ascorbic acid administration. 6) A hypertrophy of stromal and endovascular cells were observed on the groups administered of Norinyle alone(group II ) or Norinyle with ascorbic acid(group IV). 7) It was observed that amount of collagen fiber in the basal and muscular layeres of uterus were diminished under a microscopical observation by the special stained specimen on the Norinyle administered group, but the amount and distribution of reticulin fiber were not changed significantly. 8) The fille structure of outer functional layer of the uterus were significantly changed by administration of Norinyle which were shown irregurarity of nuclear membrane, poor development ana significant expansion of enaoplasmic reticulum, decreases of the amount of ribosome due to slip off, increases of the number of dense bodies, obvious formation of vaccule, an4 decreases the amount of collagen in inner and outer layer of the stroma. 9) The amount of ascorbic acid in the serum did not much changed but the amount in the liver was much decreased by the administration of Norinyle, And the administration of Norinyle with ascorbic acid induced for a significant diminishing on the changes of uterus which might be able to developed by the administration of Norinyle alone.

Interferon Tau in the Ovine Uterus

  • The peri-implantation period in mammals is critical with respect to survival of the conceptus (embryo/fetus and associated extraembryonic membranes) and establishment of pregnancy. During this period of pregnancy, reciprocal communication between ovary, conceptus, and endometrium is required for successful implantation and placentation. At this time, interferon tau (IFNT) is synthesized and secreted by the mononuclear trophectodermal cells of the conceptus between days 10 and 21~25. The actions of IFNT to signal pregnancy recognition and induce or increase expression of IFNT-stimulated genes (ISGs), such as ISG15 and OAS, are mediated by the Type I IFN signal transduction pathway. This article reviews the history, signaling pathways of IFNT and the uterine expression of several IFNT-stimulated genes during the peri-implantation period. Collectively, these newly identified genes are believed to be critical to unraveling the mechanism(s) of reciprocal fetal-maternal interactions required for successful implantation and pregnancy.

Spatio-temporal Expression and Regulation of Dermatopontin in the Early Pregnant Mouse Uterus

  • During endometrial differentiation the extracellular matrix (ECM) changes dramatically to prepare for implantation of the embryo. However, the genes regulating the ECM build-up in the uterine endometrium during early pregnancy are not well known. Using the PCR-select cDNA subtraction method, dermatopontin was identified in the uterus of a pregnant mouse on day 4 of gestation. Dermatopontin mRNA increased dramatically on day 3, and was at its highest level at the time of implantation. Administration of RU 486 significantly inhibited mRNA expression by day 4 of gestation, but ICI 182,780 did not. Progesterone markedly induced dermatopontin expression in ovariectomized uteri within 4 h of administration, whereas estrogen had little effect. In silico analysis revealed progesterone receptor binding sites in the dermatopontin promoter region. Decidualization did not induce expression of dermatopontin; instead dermatopontin mRNA became strongly localized at the interimplantation site. In situ hybridization revealed that expression gradually decreased in the luminal epithelial cells as pregnancy progressed, whereas it increased in the stromal cells. The pattern of localization and the changes of intensity of dermatopontin mRNA coincided with those of collagen. Collectively, these results strongly suggest that dermatopontin expression is steroid-dependent. They also suggest that, at the time of implantation, dermatopontin expression is primarily regulated spatio-temporally by progesterone via progesterone receptors, and is modulated by the decidual response during implantation. Dermatopontin may be one of the regulators used to remodel the uterine ECM for pregnancy.

Peri-implanting 단계의 돼지배아 EST 연구 (The EST Study of the Peri-implanting Porcine Embryos)

  • 임신초기의 배아는 peri-implantation 단계에서 매우 극적인 형태학적변화가 일어나는데, 이는 임신 인식 인자로 작용하는 배아에서 제공된 signal(s)에 의해서 시작되어지며, 나아가서 모성자궁과 배아의 상호 신호전달이 임신의 시작과 유지에 필수적인 인자로 작용한다. 배아형태의 급격한 리모델링에 관련된 세포학적, 생화학적, 유전학적 연구를 위하여, 또한 자궁과 배아의 상호 신호전달에 관여하는 잠재적 유전자 군을 발굴하기 위하여, peri-implantation 시기의 돼지배아를 이용하여 expresses sequences tag (EST) 분석을 실행하였다. 돼지배아 EST 분석으로 임신초기 특히 전 착상 단계에서 발현되는 유전자들의 카탈로그(Transcriptome)를 작성하였다. 그중에서 6개의 clone을 선택하여 그 발현 양식을 배아 및 자궁 등에서 관찰한 결과, 각각의 유전자들은 조직, 세포 유형 및 임신 시기에 따른 특이적인 발현 현상을 나타내었다. 본 연구결과는, 배아와 자궁 내막에서의 유전자 발현이 임신 시기에 따라서 다이내믹한 상호 조절 작용을 하고 있음을 나타낸다. 이는 전 착상단계의 모성자궁에서 배아와 자궁 내막의 상호 신호전달이 전 착상 단계의 배아의 급격한 형태학적 변화를 가능하게하고 또한 착상에 필요한 적절한 자궁 내부 환경을 제공하고 있음을 보여준다.

합곡(合谷), 삼음교(三陰交) 자침(刺鍼)이 백서(白鼠) 자궁(子宮) 운동(運動) 및 Cyclooxygenase-2 발현(發現)에 미치는 영향(影響) (Effects of HapKok (LI-4) , SamUmGyo (SP-6) Acupuncture on Uterine Motility and Cyclooxygenase-2 Manifestation in Rats)

  • By the activation of ovary hormone, many morphological changes occur in the epithelial cell lines and muscle cells in rat uterus. These two cells in uterus are important to the implantation of embryo, maintaining pregnancy and starting parturition. One important change associated with the morphological change of these two cells in uterus is the change on prostaglandin(PG) metabolism. Its presence and synthesis in endometriurn and myometrium in uterus affects estrous cycle and the start of embryo implantation in uterus. It also performs as an important modulator in parturition. So the abnormally weak expression of PG causes difficulty during labor and over-expression causes pre-term labor. PG biosynthesis starts from either free or liberated arachidonic acids from membrane phospholipid by phospholipase. Such arachidonic acids are converted into PG catalyzed by Cyclooxygenase. Under normal physiological condition, Cyclooxygenase-1(COX-1) having 602 units of amino acids controls the synthesis of PG. It acts as a local hormone regulating vasomodulation of blood flow, flexible muscle movement, increasing the blood permeability and contributing the protective role in preserving integrity of the stomach lining and Cyclooxygenase-2 (COX-2) is induced by the inflammation, pregnancy and increased its expression until parturition. Lipid metabolite like PG is located in uterine and expression of COX-2 increased with pregnancy. Increased expression of COX proteins in epithelial cells and myometrial cells are told to increase the muscle contractility in uterus but decreased right after the labor in rat. It is a good sign indicating that COX proteins are deeply related to the start of labor. Currently, Several studies report the use of PG and COX-2 inhibitor as medication for controlled abortion or to prevent pre-term labor but they entail various side-effects. Our study proposed to suggest use of acupuncture as an another mediator to control abortion or pre-term labor without causing unnecessary side-effects by those medicines. Two acupuncture sites, LI-4 & SP-6 were selected due to their known efficacy. From the immunohistochemical staining of COX-2, normal expression of COX-2 protein in nonpregnant SD rat's uterus revealed that COX-2 protein was primarily detected in the lumina epithelial lining and in the epithelial cell lining contacting the stromal cells. High resolution optical microscopic scanning revealed distinguishable staining in the myometrial mucosa. LI-4 acupuncture administered nonpregnant rat's uterus showed strong expression for COX-2 in endometrium contacted with lumina epithelial lining of rat uterus and in myometrial mucosa. Stromal cells showed more staining than untreated nonpregnant rat's uterus and stronger staining in stromal cells contacting myometrial layer compared to untreated nonpregnant rat's uterus. SP-6 acupuncture administered nonpregnant rat's uterus showed weak expression for COX-2 in myometrial layers and stromal cells but no staining was visible in lumina epitheliai and glandular epithelial cells. Few stromal cells and myometrial mucosa were positively stained for COX-2. Pregnant SD rat's uterus was also immunostained for COX-2 expression after 18 days of pregnancy. Unlike to untreated nonpregnant rat's uterus, luminal epithelial cells were not positively stained for COX-2 but stronger staining for COX-2 was revealed in stromal cells. LI-4 acupunctured SD rat's uterus had very strong expression of COX-2 in luminal epithelial lining. Few stromal cells showed stronger positive COX-2 staining and myometrial layers also showed more expression than untreated pregnant rat. SP-6 acupuncture administered pregnant SD rat's uterus showed positive expression of COX-2 in epithelial cells of luminal mucosa layer but weaker than that of LI-4 acupuncture treatment's case. However, strong positive staining was revealed in stromal mucosa and myometrial layers. Virgin SD rat's uterus motility index during LI-4 acupuncture was 66.52 % (Prob〉T = 0.0197) compared to its motility before the acupuncture treatment but the motility index was slighdy elevated up to 79.58 % (Prob〉T = 0.1175) after the acupuncture. During the SP-6 acupuncture treatment for 30 minutes, uterus motility index was 90.52 % (Prob〉T = 0.1832) showing lesser decrement but consequently reached similar motility index decreasal to 79.95 % (Prob〉T = 0.0215) after the acupuncture treatment as LI-4 showed. LI-4 acupuncture tend to be a quick treatment to reducing the uterus motility in a virgin rat but eventually both two acupuncture administration created very similar reduction of uterus motility seeing the index after the both acupunctures. The uterus movement monitored during the LI-4 acupuncture administered for 30 minutes, Pregnant SD rat showed decreased motility down to 77.90 % (Prob〉 T = 0.0076) compared to uterus motility before the acupuncture and it continuously decreased down to 71.81 %(Prob〉T = 0.0214) after the removal of needle. The statistical analysis using paired t-test showed significance difference for both two motility indexs at =0.05. SP-6 acupuncture administered to pregnant SD rat also had similar pattern of decreasing uterus motility index down to 74.70 % (Prob〉T = 0.1730) during the initial 30 minutes acupuncture administration and it was continuously lowered to 71.52 % (Prob〉T = 0.0155) after the acupuncture. The paired t-test resuit for SP-6 suggest prompt response of uterus motility index to the SP-6 acupuncture treatment but consequently reached same level of inducing the motility reduction as LI-4 at =0.05 level.

A Review of Mechanisms of Implantation

  • Implantation is a highly organized process that involves an interaction between a receptive uterus and a competent blastocyst. In humans, natural fecundity suggests that the chance of conception per cycle is relatively low (~30%) and two-third of lost pregnancies occur because of implantation failure. Defective implantation leads to adverse pregnancy outcomes including infertility, spontaneous miscarriage, intrauterine fetal growth restriction and preeclampsia. With use of advanced scientific technologies, gene expression analysis and genetically-engineered animal models have revealed critical cellular networks and molecular pathways. But, because of ethical restrictions and the lack of a mechanistic experiment, comprehensive steps in human implantation have still not been completely understood. This review primarily focuses on the recent advances in mechanisms of implantation. Because infertility is an emerging issue these days, gaining an understanding the molecular and hormonal signaling pathway will improve the outcome of natural pregnancy and assisted reproductive technology.

Successful laparoscopic surgery of accessory cavitated uterine mass in young women with severe dysmenorrhea

  • Accessory cavitated uterine mass (ACUM) is a rare and unique condition seen in young women. We report cases of ACUMs in two patients, a 14-year-old girl and a 25-year-old woman, both with complaints of severe dysmenorrhea that had started at menarche and had progressively worsened since. A large cystic lesion was localized in the anterolateral wall of the myometrium separate from the endometrium, which was difficult to distinguish from congenital uterine anomalies. Laparoscopic excision of the ACUMs was successful and completely resolved the dysmenorrhea. Early investigation of severe dysmenorrhea in young women can provide appropriate management and relieve symptoms.

발정주기 동안 생쥐 자궁에서의 Aquaporin-4와 -8 유전자의 발현 (Expression of Aquaporin-4 and -8 Genes in Mouse Uterus during the Estrous Cycle)

  • Aquaporins(AQPs)유전자는 다양한 조직의 상피세포와 내피세포에 존재하며 다량의 물 이동을 조절하는 막성 단백질로서, 세포간 또는 세포막 사이의 물 이동에 중요한 역할을 하는 것으로 알려져 있다. 발정기의 생쥐 자궁에서는 자궁내막세포의 증식과 함께 수화되는 특징을 보이며, 자궁내강으로 물이 이동되어 자궁내액의 점성이 낮아지는 현상이 나타난다. 따라서, 본 연구에서는 생쥐의 발정주기 동안 자궁에서의 형태학적인 변화와 관련하여 AQP유전자가 물 이동의 매개체로서 중요한 역할을 할 것으로 추측하여, 생쥐 발정주기 동안에 AQP유전자의 발현 양상을 역전사 중합효소 연쇄반응을 통하여 관찰하였다. 또한, laser capture microdissection(LCf을 이용하여 자궁내 세포의 종류에 따른 AQP유전자의 발현 양상을 조사하였다. AQP-4 전령체는 발정주기의 proestrus와 estrus 시기에 발현량이 유의하게 증가하는 반면, AQP-8 전령체는 동일한 시기에 유의하게 감소하는 것으로 관찰되었다. 또한, LCM 기법을 통해서 AQP-4와 -8 전령체가 자궁기질세포보다 자궁내막세포에서 강하게 발현이 유도됨을 관찰하였다. 결과적으로, 생쥐 자궁에서 AQP-4와 -8유전자가 발정주기에 따라 발현량이 변화하는 것으로 보아 난소내 호르몬인 estrogen과 progesterone에 의해 조절될 가능성이 있으며, proestrus와 estrus 시기에 자궁내강으로 자궁내액을 수송하는데 중요한 역할을 할 것으로 사료된다.

Effects of IFN-γ on IL-18 Expression in Pregnant Rats and Pregnancy Outcomes

  • The present study focused on establishing the effects of interferon-gamma (IFN-${\gamma}$) on interleukin-18 (IL-18) expression patterns and pregnancy outcomes in pregnant rats. Pregnant rats at the post-implantation stage were randomized into control, low IFN-${\gamma}$ (L-IFN-${\gamma}$) and high IFN-${\gamma}$ groups (H-IFN-${\gamma}$) that received normal saline, 100 IU/g of IFN-${\gamma}$ and 500 IU/g of IFN-${\gamma}$ vaginal muscular injection, respectively. The effects of IFN-${\gamma}$ on IL-18 expression and pregnancy outcomes were assessed systematically using several methods, including immunohistochemistry streptavidin-perosidase (SP), image pattern analysis, enzyme-linked immune-sorbent assay (ELISA), whole blood count (WBC) count, microscopy and visual observation. IL-18 was detected in the uteri of all pregnant rats, and mainly distributed in the endometrium, decidual cells, vascular endothelium and myometrium. Immunohistochemistry and image pattern analyses revealed significantly lower IL-18 expression in the H-IFN-${\gamma}$ group compared to the L-IFN-${\gamma}$ and control groups (p<0.01), indicating that high doses of IFN-${\gamma}$ induce downregulation of IL-18 in the uterus of pregnant rats. ELISA results disclosed that IL-18 expression in peripheral blood of the H-IFN-${\gamma}$ group was lower than that of the L-IFN-${\gamma}$ group (p<0.05), and significantly reduced compared to the control group (p<0.01). Moreover, the number of peripheral leukocytes in the H-IFN-${\gamma}$ group was significantly higher than those in the control and L-IFN-${\gamma}$ groups (p<0.01). Morphology analysis showed no evident differences between the L-IFN-${\gamma}$ and control groups. However, for the H-IFN-${\gamma}$ group, uterine mucosa bleeding, necrosis and excoriation were observed using microscopy. Visual observation revealed marroon, swelling, crassitude and no embryo in the uterus, which are obvious indicators of abortion. These results indicate that IFN-${\gamma}$ plays a regulatory role in IL-18 expression in the uterus and peripheral blood of pregnant rats at the post-implantation stage. Moreover, high levels (500 IU/g) of IFN-${\gamma}$ influence normal pregnancy at the early stages in rats by downregulating IL-18 expression in the uterus and peripheral blood and increasing the number of peripheral leukocytes, consequently triggering termination of pregnancy.

  • The purpose of this study is to see the radiation protection effect of the oral injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts on the small intestine and uterus of female SD Rat as a natural radiation protection agent. The experimental group was divided into four groups: Normal Control group (NC group), Injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts group (DZ group), irradiated group after injecting Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts (DZ+IR group). The whole body of SD Rat was irradiated with gamma-ray 10Gy, and the administration of oral Dendranthema zawadskii var. latilobum (Maxim.) Kitam. Extract was 2 cc (71.56 mg/day/kg) once a day for 2 weeks. For this study, chages in blood cell levels, SOD assay, small intestine and uterus were observed. In the 21st white blood cell level, the DZ+IR group recovered to a normal level, and the IR group didn't. The IR group villus length was lower than other groups on Day 1. IR group was partially recovered, and DZ+IR group was recovered like the NC group on Day 21. In the case of the first-day endometrium, the IR group was thin and the boundary was cloudy, and the DZ+IR group was thicker and the boundary was clearer than the IR group. Day 21 IR group still did not recover, and DZ+IR group recovered like NC group. This is believed to have radiation protection effects in the blood cells and small intestine and uterus of the irradiated female SD Rat, and is expected to be useful for the study of natural radiation protection materials.
