Gene Transfer Optimization via E. coli-driven Conjugation in Nocardiopsis Strain Isolated via Genome Screening (유전체 스크리닝으로 선별된 Nocardiopsis 균주의 대장균 접합을 통한 유전자 도입전략 최적화)

  • Actinomycetes, Gram positive soil bacteria, are valuable microorganisms which produce useful secondary metabolites including antibiotics, antiparasitic substances, anti-cancer drugs, and immunosuppressants. Although a major family of actinomycetes, known as streptomycetes, has been intensively investigated at the molecular level for several decades, a potentially valuable and only recently isolated non-streptomycetes rare actinomycetes (NSRA) family has been poorly characterized due to lack of proper genetic manipulation systems. Here we report that a PCR-based genome screening strategy was performed with approximately 180 independently isolated actinomycetes strains to isolate potentially valuable NSRA strains. Thanks to this simple PCR-based genome screening strategy we were able to identify only seven NSRA strains, followed by 16S rRNA sequencing for confirmation. Through further bioassays, one potentially valuable NSRA strain (tentatively named Nocardiopsis species MMBL010) was identified which possessed both antifungal and antibacterial activities, along with the presence of polyketide synthase and non-ribosomal peptide synthase genes. Moreover, Nocardiopsis species MMBL010, which was intrinsically recalcitrant to genetic manipulation, was successfully transformed via E. coli-driven conjugation. These results suggest that PCR-based genome screening, followed by the establishment of an E. coli-driven conjugation system, is an efficient strategy to maximize potentially valuable compounds and their biosynthetic genes from NSRA strains isolated from various environments.

Antifungal Activity of Methylene Chloride Fraction of Pimpinella brachycarpa Against Aspergillus niger (참나물 Methylene Chloride 분획의 Aspergillus niger에 대한 항진균 활성)

  • In order to develop safe and economic novel antifungal agents, we prepared 73 methanol extracts from medicinal and edible herbs and examined their 365 solvent fractions using n-hexane, methylene chloride, ethylacetate, butanol and water residue based on the sequential organic solvent fraction method. When using the various fractions in the screening step for antifungal activity, we discovered ethylacetate fraction of Morus alba L., methylene chloride fraction of Pimpinella brachycarpa (MCPB), and n-hexane fraction of Salvia miltiorrhiza Bunge, which all have activities in methanol extracts, as potential sources of antifungal agents. Amongst these, the antifungal activity of P. brachycarpa has not to date been reported on. In addition, the mycelial growth inhibition and spore germination inhibition activities of MCPB against A. niger were confirmed by disc-diffusion assay in a 10 day culture. The MIC and MFC of MCPB were determined as 0.25 and 0.5 mg/ml, respectively. The MCPB has no hemolytic activity against human RBC at 0.5 mg/ml and glycoside-flavonoids are theorized to be active constituents. These results suggest that MCPB has a prominent antifungal activity and that the application of sequential organic solvent fractions, instead of simple natural product extracts, is useful in the screening process of novel bioactive substances.

Determination of p-Anisic Acid and Methylparaben by Using High Performance Liquid Chromatography (HPLC를 이용한 p-아니식애씨드와 메칠파라벤의 분리 분석법 개발)

  • In this study, we developed a HPLC method for the separation and analysis of methylparaben and p-anisic acid, which are commonly used as a synthetic preservative and natural preservative, respectively. Methylparaben and p-anisic acid have same molecular weight (152.15 g/mol), similar structure and same maximum absorption wavelength (250 nm), thus they showed same retention time (RT) value (13.3 min) in HPLC experiment. We observed that two substances are separated on C18 column after methylparaben was derivatized selectively through the acetylation reaction. Instead, RT of the acetylated methylparaben was moved to 23.9 min from 13.3 min. The average retention time was $23.9{\pm}0.1min$ and peak area values was $5042882{\pm}4778$. In addition it showed a high linearity in the calibration curve with a correlation coefficient (R2) of 0.9999658. Detection and quantitation limits were $1.47{\mu}g/mL$ and $4.44{\mu}g/mL$, respectively. In conclusion, the developed method can be useful for separation and analysis of preservatives with similar structure in cosmetic fields.

  • In this paper, mechanisms of gene editing technologies including ZFN, TALENS and CRISPR were briefly discussed with mutual advantages and disadvantages. Classification criteria of gene edited, site-directed mutagenesis (SDN) crops for regulatory purpose were also discussed. The number of studies using CRISPR technology was high and studies conducted on Arabidopsis thaliana and rice were highest, followed by tobacco, tomato, wheat, and corn. It has been applied to a variety of plants such as other grain crops, flower crops, vegetable crops, and fruit trees. The number of studies focused on practical application or commercialization in the future were also increasing yearly, and the scope of studies also expanded to include research on metabolic engineering for mass production of useful proteins or substances, development of disease resistant crops against viruses, bacteria, and fungi, abiotic environmental stressresistant crops, and increased yields. In addition to this, it was revealed that application range is becoming more diversified, including the development of parthenocarpic tomatoes, hybrid rice lines using male sterility and increased shattering resistance Brassica napus. It was also revealed that the number of CRISPR gene edited crops permitted by the USDA(APHIS) increases yearly, to be released in the international seed market soon.

Effect of Gamma Irradiation on Meat Quality in Chicken Breast during Cold Storage (감마선 처리에 따른 닭고기 가슴육의 냉장 저장 중 품질 특성)

  • The influence of gamma irradiation with low doses (0.5, 1.0, and 1.5 kGy) on the meat quality of chicken breast was determined for 6 days of storage at $4^{\circ}C$. The pH of irradiated chicken was lower than that of the control at day 1 (p<0.05), while no difference was found among irradiation at different doses. Drip loss was higher in irradiated chicken with doses of 0.5 and 1.0 kGy compared to the control at day 1 and day 3 (p<0.05). Lightness (L) at day 1 and day 6 was increased due to irradiation (p<0.05). Redness (a) increased with irradiation doses of 1.0 and 1.5 kGy at day 1 and day 3 (p<0.05). However, no significant differences were found in yellowness (b). The number of aerobic plate count of irradiated chicken during storage was significantly lower than the control (p<0.05). E. coli and coliforms were not detected after irradiation at any dose. Also, irradiation doses of 1.0 and 1.5 kGy significantly reduced volatile basic nitrogen (VBN) and 2-thiobarbituric acid reactive substances (TBARS) values compared to the control during storage (p<0.05). These results suggest that low dose gamma irradiation can be useful to extend the shelf life of chicken breast during cold storage.

Response of Photobacterium phosphoreum to Heavy Metal (Photobacterium phosphoreum의 중금속에 대한 반응성)

  • Photobacterium phosphoreum was used in order to study response to heavy metal including $HgCl_2$, $CdCl_2$, $MnSO_4$ and $ZnSO_4$ in view of developing monitoring system for toxic substances. The concentrations of heavy metal causing 50% reduction($EC_{50}$) in bioluminescence intensity were determined with both free and immobilized P. phosphoreum. The bioluminescence responses were examined at various concentrations of heavy metal after 10, 20 and 30 min of exposure. The linear correlation between Gamma values and concentrations of heavy metal was obtained and $EC_{50}$ was calculated from the linear correlation. The significant inhibitory concentrations for bioluminescence emission were found to be 0.05mg/L for $HgCl_2$, 25mg/L for $CdCl_2$, 50mg/L for $MnSO_4$ and 12.5mg/L for $ZnSO_4$, respectively. The free cell and disc type were shown to be more sensitive to heavy metal than cells mixed with Na-alginate or immobilized on Sr-alginate. However, the linear regression curves were derived from the Sr-alginate immobilized cells indicating the immobilization method is a useful tool for monitoring of heavy metal under more stable condition of bioluminescence.

Effects of Feeding Purple Rice (Oryza sativa L. Var. Glutinosa) on the Quality of Pork and Pork Products

  • Purple rice is a strain of glutaneous rice rich in anthocyanins and ${\gamma}$-oryzanol. Both types of compounds are involved in antioxidant and lipid metabolism of mammals. Three experimental diet types were used which consisted approximately by half either of purple rice, white rice or corn. Diets were fed to $3{\times}10$ pigs growing from about 30 to 100 kg. Meat samples were investigated either as raw or cured loin chops or as smoked bacon produced from the belly. Various physicochemical traits were assessed and data were evaluated by analysis of variance. Traits describing water-holding capacity (drip, thaw, and cooking losses) and tenderness (sensory grading, shear force) of the meat were mostly not significantly affected by the diet type. However, purple rice feeding of pigs resulted in lower fat and cholesterol contents of loin and smoked bacon compared to white rice, but not compared to corn feeding except of the fat content of the loin. The shelf life of the raw loin chops was improved by purple rice as well. In detail, the occurrence of thiobarbituric acid reactive substances after 9 days of chilled storage was three to four times higher in the white rice and corn diets than with purple rice. The n-6:n-3 ratio in the raw loin chops was 9:1 with purple rice and clearly higher with 12:1 with the other diets, meat lipids. Level and kind of effect of purple rice found in raw meat was not always recovered in the cured loin chops and the smoked bacon. Still the impression of flavor and color, as well as overall acceptability were best in the smoked bacon from the purple-rice fed pigs, whereas this effect did not occur in the cured loin chops. These findings suggest that purple rice has a certain, useful, bioactivity in pigs concerning meat quality, but some of these effects are of low practical relevance. Further studies have to show ways how transiency and low recovery in meat products of some of the effects can be counteracted.

Efficacy of Lens Shielding Device to Prevent Cataract with Radiotherapy for Orbit or Ocular Adnexal Tumor (안와 및 안부속기 종양의 방사선치료에서 백내장의 예방을 위한 렌즈보호 장치의 효용성)

  • Radiotherapy which is the most effective for orbit lymphoma has been used increasingly due to the increase of orbit or ocular adnexal tumor patients. Curative effects and convalescence have been being more satisfied thanks to remarkable development of cancer chemotherapy and medical treatments, but side effects such as cataract, dry eye and retinopathy still break out. Thus, in this study, a Lens Shielding Device (LSD hereafter) was designed to prevent occurring of cataract due to radiation therapy for orbit lymphoma and its efficacy through dosimetry were evaluated. And in this paper, its manufacturing process was also explained. LSD is composed of a cover body covering the lens and a side fixing part supporting the cover body. To measure radiation, the patient therapy conditions were simulated and the measurement of the radiation was conducted with Thermo Luminescence Detector (TLD) and Markus chamber. The average TLD value was 5.7% and the TLD value and Markus chamber value were acquired as 4.2% and 5.1% respectively at 6 mm depth where zero lens center was located. Only 1.5Gy ($300Gy{\times}\;5%$) or 5% of total 30Gy with 9 MeV electron beam is estimated to affect on patient's lens. That is smaller dose than the threshold value of cataract (2GY) or the value (5Gy) that was reported to cause cataract in clinical conditions. Thus, these findings suggest that LSD be very useful for prevention of cataract during radiotherapy for malignant lymphoma of orbit and ocular adnexa. Furthermore, it might be possible to reduce patient's discomfort caused by alien substances and to make it easier to fix the device with customized manufacturing manners.

A Bioassay Method with Radish Seedlings for Bioactive Substances (무우 유묘(幼苗)를 이용(利用)한 식물생리활성물질(植物生理活性物質)의 생물검정법(生物檢定法))

  • Some experiments were conducted to utilize the raphanus test as a bioassay method for plant growth regulaters and hormonal herbicides. Among seven radish cultivars, the response of Jangbaegminong was the most sensitive and quantitative to ${\alpha}$-NAA and its coefficient of variation value was the lowest. The optimum application time of test compounds was estimated seven to nine days after seeding. After ${\alpha}$-NAA treatment the curvature angle of radish cotyledonary petiols was sharply increased to the maximum value at 24 hours after and was changed by light and temperature conditions. The curvature response was observed by application of ${\alpha}$-NAA, phenoxy and indole compounds but often plant growth regulators, $GA_3$ benzyladenine, ABA and homobrassinolide was not detected by this method. So this bioassay was useful to evaluate the activity of a compound as an auxin or a hormonal herbicide.

Optimization of Germinated Brown Rice Cookie with Added Spinach Powder (시금치 가루를 첨가한 발아현미쿠키의 최적화)

  • Germinated brown rice is a useful material that contains natural nutrients and biologically active substances. This study was conducted to develop a optimal composite recipe for functional germinated brown rice cookie with added spinach (spinacia oleracea L) powder and with high preference in all age groups. The experimental design utilized herein was based on the Central composite design methodology of response surface, which included 16 experimental points, including 2 replicates for spinach, sugar, and butter. The physical, mechanical, and sensory properties of the test were measured, and these values were applied to the mathematical models. A canonical form and perturbation plot showed the influence of each ingredient on the mixed final product. The results of spread ratio decreased significantly with increases in spinach powder, sugar, and butter(p<0.01). L value shows that lightness decreased significantly with an increase in sugar(p<0.05), but the a and b values were not significant. Sensory evaluation found significant values for color(p<0.05), flavor(p<0.05), texture(p<0.05) and overall quality(p<0.05) in the predicted model. The optimum formulation by the numerical and graphical methods was calculated as follows: spinach powder 3 g, sugar 35.5 g butter 54.4 g.
