Development of an Effective Extraction Method for the Quality Control of Eriobotrae Folium : Determination of Triterpenic Acids (비파엽 품질 비교 분석을 위한 Triterpenic Acid의 추출 방법)

  • In result of analysis for the content determination of ursolic acid(UA) and oleanolic acid(OA) in extract of Eriobotryae folium using HPLC with UV detector, UA in chloroform extract of Moo-mok variety was showed highest content(2.7843 mg/g). And OA in ethyl acetate extract of Dae-bang variety was showed highest content(0.5898 mg/g). These result suggest that direct extraction using organic solvent(chloroform or ethyl acetate) was useful method for rapid quantitative analysis of UA and OA without preprocessing such as drying or fractionation.

  • 배풍등, 등혹 및 ginsenoside Rh$_1$의 간암세포에 대한 세포독성작용, 인삼 Rh$_1$의 세포보호작용, 비파의 ursolic acid 생체방어기전 활성화 및 산화억제작용, ononin의 radical 제거능을 검토하였다. 그 결과 배풍등 CHCl$_3$분획 및 등혹 CHCl$_3$분획의 간암세포에 대한 강한 세포독성작용을 나타내었으며 배풍등 CHCl$_3$은 sarcoma 180 이식 종양조직의 성장을 유의성있게 억제하였다. Ursolic acid는 지질과산화, 단백질 산화억제와 catalase, GSH S-transferase를 활성화시켰다. 인삼 saponin은 SOD 및 nonprotein-SH를 증가시키고, 지질과산화를 억제시켰다. Ginsenoside Rh$_1$ 및 Rh$_2$는 각각 radical에 대한 세포보호작용과 간암세포 세포독작용을 나타내었다.

  • Four ursane type triterpenes have been isolated from the methanolic extract of the aerial parts of Agrimonia pilosa Ledeb. (Rosaceae) through repeated silica gel and reverse-phase C-18 column chromatography. Their chemical structures were elucidated as ursolic acid (1), pomolic acid (2), tormentic acid (3), and corosolic acid (4) on the basis of their MS, $^1H-$, and $^{13}C-NMR$ spectral data. Compounds 2 and 4 were isolated from the genes of Agrimonia for the first time.

  • From the fruits of Crataegus pinnatifide BUNGE ursolic acid (1), $quercetin-3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (2), (-)-epicatechin (3), $quercetin-3-O-{\beta}-D-galactopyranoside$ (4) and quercetin (5) have been isolated and identified on the basis of their physicochemical properties and spectroscopic evidences in comparison with authentic samples. Among isolated compounds, quercetin showed significant inhibitory effect against MAO.

  • Prunasin and pomolic acid together with a mixture of ursolic and oleanolic acids, (-)-epicatechin and ${\beta}-sitosterol$ glucoside were isolated from the roots of Chaenomeles japonica. Among these compounds, the isolation of pomolic acid and prunasin is the first report from the genus Chaenomeles.

  • From the roots of Rubus parvifolius L., four triterpenoidal sapogenins, ursolic acid 1. $2\alpha$-hydroxyursolic acid 2. causapic acid 3. $2\alpha$, $3\beta$, $19\alpha$-trihydroxyurs-12-en-23.28-dioic acid 4 and one triterpenoidal glycoside. suavissimoside $R_1$ 5. were isolated. The structures were elucidated by spectroscopic methods and chemical transformations. Compound 4 was first isolated as free form.

  • The ether soluble fraction of the methanol extract from the fruits of Crataegus pinnatifida var. psilosa was applied to silica gel column chromatography, eluted step-gradiently with a mixture of dichloromethane and methanol, repeatedly, and five compounds were isolated. These compounds are identified as ursolic acid(I), corosolic acid(II) and euscapic acid (III), ${\beta}-sitosteryl-3-O-{\beta}-_D-glucopyranoside(IV)$ and hyperin(V).

  • Hyaluronidase is one of the mucopolysaccharide-splitting enzyme and is related to the permeability of the vascular system and inflammation. An anti-hyaluronidase assay guided fractionation of the methanolic extract of Uncariae Ramulus et Uncus has furnished a pentacyclic triterpene, ursolic acid (compound I). Compound I exhibited hyaluronidase inhibitory activity with $IC_{50}$ value of 0.15 mM, and disodium cromoglycate showed the inhibitory activity with $IC_{50}$ value of 1.78 mM as a positive control.

  • Two lupane-type triterpenes, lupenone (1) and lupeol (2), a phytosterol, ${\beta}-sitosterol$ (3), two ursane-type triterpenes, $3{\beta}-acetoxy$ ursolic acid (4) and ursolic acid (5), a lignan, (-)-lyoniresinol $3a-O-{\beta}-D-xylopyranoside$ (6), and two flavanol glycosides, $catechin-7-O-{\beta}-D-xylopyranoside$ (7) and $catechin-7-O-{\beta}-D-apiofuranoside$ (8) were isolated from the stem bark of Sorbus commixta (Rosaceae). Their chemical structures were identified by physicochemical and spectroscopic methods.

  • The photochemical study of Viscum album (Loranthaceae) led to the isolation of twelve compounds, lupeol (1), betulonic acid (2), betulinic acid (3), terminic acid (4), ursolic acid (5), $\beta$-sitosterol (6), $\alpha$- spinasterol (7), oleanolic acid (8) , 5-hydroxy-1- (4′-hydoxyphenyl) -7- (4"-hydroxyphenyl) -hepta-1- en-3-on (9), 2′-hydroxy -4′, 6′- dimethoxychalcone -4-O-glycoside (10) ,2′-hydroxy-4′, 6′-dimethoxychalcone -4-O- [apiosal(1longrightarrow12)]glucoside (11) and syringin (12). Their structures were established by chemical and spectroscopic methods. The cytotoxicity of the isolated compounds was evaluated by sulforhodamine B assay against five cultured human tumor cell lines.
