Macro Mineral Responses to Caffeine in Serum and Urine of Healthy Young Females(I) -Na, K, Ca, P, Mg- (Caffeine 섭취에 따른 성인 여자의 혈액과 소변중 다량 무기질 함량 변화에 관한 연구(I) -Na, K, Ca, P, Mg-)

  • This study was undertaken to investigate the acute effect of caffeine consumption on the change of mineral concentration in serum and urinary mineral excretion in healthy young females. On two separate mornings at one week intervals, each subject drank a coffee which contained no caffeine and 3mg/kg body weight caffeine. To obviate dietary effects on mineral concentration in serum and urine, each subject fasted at least ten hours before consuming the test beverage. At one, two, three and four hours, serum and urine production collected seperately for measurement of sodium, potassium, calcium, phosphorus and magnesium concentration. The results were as following : 1) Mean age of subjects was 20.6$\pm$0.32, Mean body mass index of subjects was 21.64$\pm$0.89, which was within $\pm$10% of ideal body weight. 2) Total urine volume of caffein groups for 4 hour after caffeine consumption was higher than that of decaffeine one, but urine pH was unchanged after caffeine consumption. Total urinary four hour excretion of creatinine was not affected by caffeine consumption and creatinine clearance also was not different from the control value. 3) In serum, mean three hour content of sodium(p<0.01) and phosphorus was higher in the subject given the caffeine. Mean serum magnesium and calcium contents were lower in caffeine group than that of decaffeine one. Mean serum magnesium content for three hour after caffeine ingestion was affected by caffeine consumption(p<0.001). Mean serum content of potassium was unaffected by caffeine consumption. 4) Total urinary four hour excretion of sodium, increased significantly after caffeine consumption(p<0.05), while total urinary four hour excretion of potassium, calcium, phosphorus and magnesium was unchanged after caffeine intake. Urinary excretion of Na, Ca, P and Mg was greatest at one hour after caffeine consumption, especially urinary sodium and potassium excretion was significantly high(p<0.05, p<0.01). The above results show that only 3mg caffeine per kg body weight increase the urinary macro mineral excretion in healthy young females.

Quantity and Characteristics of Manure Produced Holstein Dry Cow (홀스타인 건유우의 분뇨배설량과 이화학적 특성)

  • This study was carried out to investigate the quantity of Holstein dry cow manure excreted and their characteristics. The results obtained in this study were summarized as follow; The average body weight of the Holstein dry cow during experiment was 619.9kg. The feed intake(DM basis) and water consumption was 10.7, 38.6kg/day/head, repectively. The manure production of Holstein dry cow was 44.8kg/day/head (feces 26.9, urine 17.9kg). The moisture content of feces and urine was 84.5%, 95.3%, respectively. Wastewater pollutant concentration of $BOD_5$(Biochemical Oxygen demand), $COD_{Mn}$ (Chemical Oxygen demand), SS(Suspended Solids), T-N(Total Nitrogen) and T-P(Total Phosphorus), excreted from Holstein dry cow was 16,874,55,763, 87,333, 2,353, $368mg/{\ell}$ in feces and 5,621, 8,673, 518, 2,423, $3mg/{\ell}$ in urine, repectively. The fertilizer content of manure, N(Nitrogen), $P_2O_5$(Phosphoric acid) and $K_2O$(Potassium oxide) was 0.24, 0.08, 0.15% in feces and 0.24, 0.001, 0.30% in urine, respectively.

Correlation between Heart Rate Variability and Urine Analysis of Women with Urinary Disturbance (배뇨장애를 호소하는 여성의 자율신경계 특성과 요검사와의 상관관계 분석)

  • Objectives: The purpose of this study was to examine correlation between Heart Rate Variability and urine analysis of women with urinary disturbance. Methods: We studied 34 patients visiting ${\bigcirc}{\bigcirc}$hospital from January 2010 to September 2010. The subjects were categorized in two groups, symptom group (n=11) and no symptom group(n=23). We studied the difference of Heart Rate Variability and urine analysis between two groups by Student T-test and correlation between Heart Rate Variability and urine analysis by Pearson's correlation coefficient test using SPSS for windows (version 17.0). Results: Occult blood of symptom group was significantly higher than no symptom group. SDNN, TP and HF of symptom group was significantly lower than no symptom group. Occult blood and SDNN, occult blood and RMS-SD, occult blood and HF significantly showed negative correlation coefficient. pH and TP, pH and LF significantly showed positive correlation coefficient. Conclusion: The results suggest that urinary disturbance can be related to decreased activity of autonomic nervous system. Also urine from women with urinary disturbance tend to show higher occult blood.

  • Recently, the miniaturization and digitalization for the inspection devices of point-of-care testing (POCT) are rapidly evolving. In the urine test, a lot of researches on index paper technology are being conducted because people can be self-diagnosed through visual color comparison using a urine test paper, Dipsick. The purpose of this study is to analyze the RGB values from the color changes on Dipstick Pad, which isused for urine test, using a smartphone camera. To this end, the primary, analytes in urine wasdiabetes-related parameters such as glucose, ketone body and pH, which is the most frequently tested elements, and we pursuited to quantify the changes in dipstick color caused from artificial urine containing different ranges of sugar, ketone body, and pH. In this experiment, changes in RGB values under bright and dark illuminances were compared, and changes in RGB value were monitored as a function of concentration of analytes under the ambient illumination of laboratory. As a result, color separation at the bright luminance region was good, but it did not appearat the low luminance region, and the changed profiles in RGB value under different illuminances was suggested to correct the problem of the color separation algorithm.

  • The extraction of trace cobalt, copper, nickel, cadmium, lead and zinc in urine samples of organic and alkali metal matrix into chloroform by the complex with a dithizone was studied for graphite furnace AAS determination. Various experimental conditions such as the pretreatment of urine, the pH of sample solution, and dithizone concentration in a solvent were optimized for the effective extraction, and some essential conditions were also studied for the back-extraction and digestion as well. All organic materials in 100 mL urine were destructed by the digestion with conc. $HNO_3$ 30 mL and 30% $H_2O_2$ 50 mL. Here, $H_2O_2$ was added dropwise with each 5.0 mL, serially. Analytes were extracted into 15.0 mL chloroform of 0.1% dithizone from the digested urine at pH 8.0 by shaking for 90 minutes. The pH was adjusted with a commercial buffer solution. Among analytes, cadmium, lead and zinc were back-extracted to 10.00 mL of 0.2 M $HNO_3$ from the solvent for the determination, and after the organic solvent was evaporated, others were dissolved with $HNO_3-H_2O_2$ and diluted to 10.00 mL with a deionized water. Synthetic digested urines were used to obtain optimum conditions and to plot calibration-eurves. Average recoveries of 77 to 109% for each element were obtained in sample solutions in which given amounts of analytes were added, and detection limits were Cd 0.09, Pb 0.59, Zn 0.18, Co 0.24, Cu 1.3 and Ni 1.7 ng/mL, respectively. It was concluded that this method could be applied for the determination of heavy elements in urine samples without any interferences of organic materials and major alkaline elements.

Monitoring the Reproductive Status of Dairy Cows by Urinary Pregnanediol Glucuronide

  • This study was undertaken with the aim to establish a reliable radioimmunoassay (RIA) system for urinary pregnanediol glucuronide (PdG) and to employ it for monitoring the reproductive status of dairy cows. Urine and blood samples were collected from the Holstein cows both pregnant and non-pregnant. The samples were then investigated for evaluating the relationship between progesterone ($P_{4}$) in blood and PdG in urine adjusted with or without urinary creatinine basis. Biweekly urine collection was employed for three cows in estrous and those artificially inseminated, while urine from pregnant cows was collected on a monthly basis. P_{4}$ and PdG levels were measured by enzymeimmunoassay (EIA) and RIA techniques, respectively. Our results indicated the sensitivity of PdG for RIA being 35 pg/tube and the recovery rate of 100%. Urinary creatinine concentrations also fluctuated within a day, but change at midday was not noteworthy. Regardless of the time of urination the change in concentrations of PdG was relatively smaller and did not vary significantly. The urinary PdG concentration showed periodic changes as that with serum P_{4}$ levels during the cow's estrus cycle. The correlation coefficient rose when creatinine level in urine was adjusted but the change was also not significant. The concentrations of PdG during the luteal phase were detected between 8.2 and 17.4 ng/ml, three to five times higher than that in the follicular phase. The concentration of PdG from pregnant cows (21 days after conception) was three to four times higher than in the nonpregnant cows. Our finding suggests that the determination of urinary PdG could be reliably employed for early pregnancy detection. The urinary PdG level continued to raise until 30 days pre-partum while the concentration reached its peak at 30 ng/ml, after which it started to fall 18 to 30 days before parturition and finally fell to its nadir value one week after parturition. As the correlation coefficient between the urinary PdG and serum P_{4}$ was higher than that corrected by urinary creatinine it can be suggested that the adjustment is not needed. The concentrations of urinary PdG could be maintained stably for 2 days in urine samples stored at room temperature and extended to 8 days when the samples were pretreated by boiling for 30 minutes. In conclusion urinary PdG concentration even without the need for creatinine basis adjustment can be used directly for monitoring the reproductive status of dairy cows.

Quantity and Characteristics of Manure Produced by Holstein Heifer at Different Seasons (홀스타인 육성우의 계절별 분뇨 배설량 및 특성에 관한 연구)

  • This study was conducted to determine the volume of Holstein heifers manure excreted and its characteristics. The average dry matter intake of heifers was 6.7 kg/head/day. The intake rate was lowest in spring among four seasons. The average dry matter intake rate during spring, summer, fall, and winter was 4.6, 8.3, 7.1, and 6.8 kg/head/day, respectively. The average water intake of heifers was $19.3{\ell}/head/day$. The wale. consumption was highest value ($21.8{\ell}/head/day$) in summer and lowest values ($18.3{\ell}/head/day$) in spring and winter. Values were found not to be statiscally different for the four seasons. The average manure production of heifers (average live weight was 363.1 kg) was 20.3 kg/head/day and it was 5.6% of live animal weight. The manure production during spring, summer, fall, and winter was 13.7, 23.5, 25.0, and 20.2 kg/head/day, respectively. Production during spring was lower than the other seasons (p<0.05). A higher correlation between live weight and manure production ($R^2=0.7816$) and between live weight and feed intake ($R^2=0.7296$) was observed for heifers. Correlations between manure production and water intake and between manure production and feed intake were found to be relatively low for heifers. The moisture content of feces was 83.5% and that of urine 94.6%. The pH of feces and urine were in the ranges of 7.4 and 7.5, respectively. The $BOD_5$, COD, SS, T-N, T-P concentrations of the heifer feces were 18,048, 50,114, 119,833, 2,519, and $427mg/{\ell}$, respectively. Heifer urine showed lower levels of $BOD_5(5,434mg/{\ell})$, COD$(6,550mg/{\ell})$, SS$(825mg/{\ell})$, T-N$(3,616mg/{\ell})$, and $T-P(28mg/{\ell})$ than feces. The fertilizer nutrient concentrations of heifer feces was 0.25% N, 0.1% $P_2O_5$ and 0.14% $K_2O$. Urine was found to contain 0.36% N, 0.006% of $P_2O_5$ and 0.31% $K_2O$.

Enantioselective Determination of Cetirizine in Human Urine by HPLC

  • In order to study the simultaneous determination of (+)- and (-)-cetirizine in human urine we have developed a chiral separation method by HPLC. A chiral stationary phase of $\alpha$$_1$-acidglycoprotein, the AGP-CSP was used to separate the enantiomers. The pH of the phosphate buffer, as well as the content of the organic modifier in the mobile phase, markedly affected the chromatographic separation of (+)- and (-)-cetirizine. A mobile phase of 10 m㏖/1 phosphate buffer (pH 7.0)-acetonitrile (95 : 5, v/v) was used for the urine assays. Ultraviolet absorption was monitored at 230nm and roxatidine was employed as the internal standard for quantification. (+)-Cetirizine, (-)-cetirizine and the internal standard were eluted at retention times of 12, 16, and 32 mins, respectively. The detection limit for cetirizine enantiomers was 400 ng/$m\ell$ of urine. A pharmacokinetic study was conducted with the help of 5 healthy female volunteers who were administered with a single oral dose of racemic cetirizine (20 mg). The peak area ratios provided by the cetirizine enantiomers were linear(r>0.997) over a concentration range of 2.5-200 ${\mu}g/ml$. The peak of the excreted cetirizine enantiomers appeared in the urine sample during the period of 1-2 hrs following the administration of the oral dose. The excreted level of (+)-cetirizine was slightly higher than (-)-cetirizine but the difference was not statistically significant. However, this method appears to have applications for enantioselective pharmacokinetic studies of racemic drugs.

The Effects of Environment-Friendly Diets on the Growth Performance, Nutrient Digestibility, Fecal Excretion, Nitrogen Excretion and Emission Gases in Manure for Growing Pigs (환경친화적인 사료의 급여가 육성돈의 성장 능력, 영양소 소화율, 분 배설량, 분뇨내 질소배설량 및 악취 가스에 미치는 영향)

  • Two experiments were used to determine the effects of environment-friendly diets on growth performance, fecal excretion, nitrogen excretion and emission gases in manure for growing pigs. In experiment 1, ninety six crossed pigs(Landrace×Yorkshire×Duroc) were allocated into four treatments. Treatments were AME(adequate ME diet, 3,265 kcal/kg), LME(lower ME diet, 3,100 kcal/kg), LME 0.05(lower ME diet+α- galactosidase & β-mannanase 0.05%) and LME 0.10(lower ME diet+α-galactosidase & β-mannanase 0.10%). Pigs fed AME diet had lower ADFI(Average Daily Feed Intake) than pigs fed other diets(p<0.05). DM(Dry Matter) digestibility in pigs fed AME and LME 0.10 diets had greater than pigs fed LME diet(p<0.05). Energy digestibility is higher in pigs fed AME and LME 0.10 diets than other treatments(p<0.05). In experiment 2, twenty four crossbred pigs(33.71 kg average BW) were used in a 14-d metabolism experiment. The pigs were housed in individual cages equipped with plastic bed flooring. Treatments were CP(Crude protein) 18% without Bacillus sp., CP 18% diet+Bacillus sp. 0.05%, CP 14% without Bacillus sp. and CP 14% diet+Bacillus sp. 0.05%. Nitrogen intake was higher for CP 18% diets than CP 14% diets(p<0.05). DM, N(Nitrogen) and energy digestibility were affected by probiotics(p<0.05). With the high CP in diets, Energy and N digestibility, urine N percent, urine N excretion and total N excretion were increased significantly compared to low CP in diets(p<0.05). Among the treatments, DM and N digestibilities, feces N excretion, N absorption were decreased significantly(p<0.05), however, feces excretion, feces N, urine N percent, urine N excretion and total N excretion were increased significantly(p<0.05) when pigs fed without probiotics diets compare to pigs fed with probiotics diets. DM and N digestibility, feces excretion, feces N excretion, urine N percent, urine N excretion, total N excretion, N absorption and N adsorption ratio were CP×probiotic interactions in p<0.05. Ammonia(p<0.01) and H2S(p<0.05) in manure were lower in CP 14% diets than CP 18% diets. Also, ammonia and H2S in manure were CP×probiotic interactions in p<0.05. In conclusion, low energy and reduction of CP dietary added enzyme and probiotics improved nutrient digestibility and reduced odors emission in manure for growing pigs.

Urine Collection from Disposable Diapers in Infants and Young Children (유소아에서 일회용 기저귀를 이용한 소변 채취 방법)

  • Purpose : Urine collection using a sterile adhesive bag for urinalysis has been used commonly in infants and young children. However, this method has had some drawbacks. So, this study was performed to evaluate the usage of disposable diapers as a substitute for the routine urine collection method. Methods : 60 mL of self voided fresh urine was collected from 99 patients. Half of the urine was poured on the disposable diapers which did not contain absorbent gel beads. After 1 hr, we obtained the urine samples(extracted urine) by compressing the wet diapers using a 50 ml syringe. Routine, microscopic and biochemical analyses were performed on the other half of fresh urine and extracted urine. Then we compared each result by correlative analysis. Results : 198 samples from 99 patients were evaluated. The results of routine urinalysis except WBC and biochemical urinalysis showed a significant correlation between the two groups(P<0.05). The relative coefficients of urine SG, pH, glucose, protein, blood and leukocytes between the two groups were 0.964, 0.938, 0.977, 0.956, 0.931 and 0.738, respectively. Those of urinary sodium, potassium, chloride and creatinine were 0.997, 0.998, 0.995 and 0.998, respectively Microscopic examinations showed lower relative coefficients than the other results, 0.740 for RBC and 0.602 for WBC, but these were still significant(P<0.05). Conclusion : The results of the urine analysis with extracted urine from diapers correlates well with that of fresh urine. This new method is very helpful and can be used as an alternative of urine collection, especially for infants and young children. (J Korean Soc Pediatr Nephrol 2005;9:128-136)
