Relationship between Transferable Embryos and Major Metabolite Concentrations in Holstein Donor Cows

  • This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.

Antitumoral, Antioxidant and Antimicrobial Activities of Solvent Ftactions from Grifola umbllatus (저령추출물의 항암, 항산화 및 항균효과)

  • Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.

  • Rhtological properties of surimi gel from white fishes by acid (acid surimi) and alkali (alkali surmi) process and effect of chemicals on gelation were investigated by punch and dynamic tests. The breaking force and deformation values of heat-induced gel of acid surimi were less than their values of alkali and conventional surimi gel, and whiteness was greatly decreased, Gel point of acid surimi was decreased but it of alkali surimi was increased with increasing moisture content in the range of 80 to $85\%$. Storage modulus of acid surimi was the highest vaule in pH 6.8, but that of alkali surimi showed high value at neutral and slightly alkali pH. Propylene glycol increased storage modulus in $20\~50^{\circ}C$, hut urea and 2-mercaptoethanol suppressed it. Potassium bromide improved storage modulus in $20~80^{\circ}C$, The results suggest that alkai process is used for making surimi instead of conventional method.

  • Fabricatio of the potassium-titanate fiber with K2O${\cdot}nTiO_2$ composition and coating of electrically conductive Sb-doped $SnO_2$ (ATO: Antimony Tin Oxide) layer on the fiber on the fiber were the fiber were the aims of this work. The fiber fabricated by slow-cooling technique showed the mean length of $15{\mu}m$ and mean diameter of $0.5{\mu}m$. Three different coating methods i.e, sol-gel, co-precipitation and urea technique, were attempted to coat the conductive ATO layer on the potassium-titanate fiber. The influences of coating method, concentrations of ATO(5∼70wt%) and Sb (0∼20wt%), temperature in the range of $450\;to\;800^{\circ}C$, number of washing (3∼4 times) on the resistivity of the ATO coated fiber were examined in details. The fiber coated ATO by coprecipitation exhibited lower resistivity of 103${\Omega}{\cdot}$cm at the 30 wt% of ATO, and showed nearly constant low value of $60{\Omega}{\cdot}cm\;to\;90{\Omega}{\cdot}$cm at the higher concentration of ATO.

  • The analytical measurement range (AMR) is the range of analyte values that a method can directly measure on a specimen without any dilution, concentration, or other pretreatment not part of the usual assay process. The linearity of the AMR is its ability to obtain test results which are directly proportional to the concentration of analyte in the sample from the upper and lower limit of the AMR. The AMR validation is the process of confirming that the assay system will correctly recover the concentration or activity of the analyte over the AMR. The test specimen must have analyte values which, at a minimum, are near the low, midpoint, and high values of the AMR. The AMR must be revalidated at least every six months, at changes in major system components, and when a complete change in reagents for a procesure is introduced; unless the laboratory can demonstrate that changing the reagent lot number does not affect the range used to report patient test results. The AMR linearity was total protein (0-16.6), albumin (0-8.1), total bilirubin (0-18.1), alkaline phosphatase (0-1244.3), aspartate aminotransferase (0-1527.9), alanine aminotransferase (0-1107.9), gamma glutamyl transpeptidase (0-1527.7), creatine kinase (0-1666.6), lactate dehydrogenase (0-1342), high density lipoprotein cholesterol (0.3-154.3), sodium (35.4-309), creatinine (0-19.2), blood urea nitrogen (0.5-206.2), uric acid (0-23.9), total cholesterol (-0.3-510), triglycerides (0.7-539.6), glucose (0-672.7), amylase (0-1595.3), calcium (0-23.9), inorganic phosphorus (0.03-17.0), potassium (0.1-116.5), chloride (3.3-278.7). We are sure that materials for the AMR affect the evaluation of the upper limit of the AMR in the process system.

Criteria of Nitrate Concentration in Soil Solution and Leaf Petiole Juice for Fertigation of Cucumber under Greenhouse Cultivation in Gyeonggi region

  • To develop a technique for efficient management of fertility for cucumber in greenhouse, a quick test method to quantify nitrate ($NO_3{^-}$) content in soil solution and leaf petiole juice using a simple instrument that are easy to use for farmers was investigated. N fertilizer (urea) was applied at 0, 50, 100 and 200% levels of the recommended application rate from 30 days after transplanting to harvest by soil fertigation treatments. Stable results were obtained from analysis of nitrate ($NO_3{^-}$) using top $10^{th}$ or $11^{th}$ leaf petioles collected between 10 to 11 am in the morning. Under the semiforcing culture, $NO_3{^-}$ content of leaf petiole juice was highest at 60 days after transplanting (DAT) at all fertigation treatments. Appropriate $NO_3{^-}$content of leaf petiole juice was $2,418{\pm}78{\sim}2,668{\pm}118$ at 45 DAT, $3,032{\pm}90{\sim}3,332{\pm}63$ at 60 DAT, $2,709{\pm}50{\sim}3,158{\pm}155$ at 75 DAT, $2,535{\pm}49{\sim}2,907{\pm}83$ at 90 DAT, and $2,242{\pm}48mg\;L^{-1}$ at 105 DAT. In addition, appropriate $NO_3{^-}$ content of soil solution was $167{\pm}9{\sim}212{\pm}15$ at 45 DAT, $83{\pm}10{\sim}112{\pm}12$ at 60 DAT, $49{\pm}3{\sim}92{\pm}6$ at 75 DAT, $71{\pm}9{\sim}103{\pm}9$ at 90 DAT, and $73{\pm}9mg\;L^{-1}$ at 105 DAT. The cucumber yield at 100% N level of fertigation was $7,770kg\;10a^{-1}$ and no difference in yield was found at 200% N level of fertigation. However, there was 12% decrease in yield at 50% N fertigation and, 17% decrease at 0% N fertigation. Under retarding culture, $NO_3{^-}$ concentration of leaf petiole juice was highest at 55 days after transplanting (DAT) at all fertigation treatments. Appropriate $NO_3{^-}$ content of leaf petiole juice was $2,464{\pm}102{\sim}2,651{\pm}33$ at 45 DAT, $3,025{\pm}71{\sim}3,314{\pm}84$ at 55 DAT and $2,488{\pm}92mg\;L^{-1}$ at 65 DAT, respectively. Appropriate $NO_3{^-}$ content of soil solution was $111{\pm}10{\sim}155{\pm}14$ at 45 DAT, $93{\pm}7{\sim}147{\pm}14$ at 55 DAT, $67{\pm}4mg\;L^{-1}$at 65 DAT, respectively. The cucumber yield at 50% N fertigation was not different from $1,697kg\;10a^{-1}$ of 100% N fertigation level and even with that of the 200% N fertigation. However, there was 21% decrease in yield at 0% N fertigation.

Protective effects of Sasa quelpaertensis Leaf Residue Extract against Potassium Oxonate-induced Hyperuricemia in Mice (생쥐에서 제주조릿대 잎 잔사 추출물의 고요산 혈증 저감 효과)

  • Leaves of Sasa quelpaertensis Nakai are used in folk medicine for their anti-inflammatory, antipyretic, and diuretic properties. To ensure efficient utilization of S. quelpaertensis leaf, we previously reported a preparation method for phytochemical-rich extract (PRE) using the leaf residue, which was produced after hot water extraction. This study was undertaken to evaluate the hypouricemic potential of S. quelpaertensis leaf PRE in potassium oxonate (PO)-induced hyperuricemic mice. The administration of PRE significantly reduced serum uric acid (UA), blood urea nitrogen (BUN), and serum creatinine levels and increased urine UA and creatinine levels in the PO-induced hyperuricemic mice. It also reduced liver UA levels and xanthine oxidase (XA) activity. A histological analysis revealed that PRE administration protected against PO-induced liver damage, pointing to anti-inflammatory and cytoprotective effects in PO-induced hyperuricemic mice. We analyzed the transcriptome response to PRE administration in PO-induced hyperuricemic mice using RNA sequencing (RNA-Seq) in kidney tissues. The administration of PRE mainly enriched genes involved in mediating immune and inflammatory responses and the metabolic pathway. A Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the metabolic pathway, purine metabolism, and antibody biosynthesis were the major pathways altered in the PRE and PO groups. These results suggest a potential role for PRE in the prevention and treatment of hyperuricemia with inflammation.

Effects of Herbal Medicine-added Cheonggugjang extract on Improvement of the Symptoms in Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유발된 흰쥐의 당뇨 증상 개선에 미치는 한약재 첨가 청국장 추출물의 영향)

  • Objective : This study was conducted to investigate antidiabetic effects of chunggugjang with medicinal herbal complex (CJ) in streptozotocin (STZ)-induced animal models. Method : STZ (65 mg/kg) was injected intraperitoneally to induce diabetes. Then rats were divided into 5 groups ; NG (normal diet + 0.9% saline), COS (STZ +saline 5 mL/kg), COB (STZ + fermented soybean(100 mg/kg), CJ 100/200 (STZ+CJ(100 and 200 mg/kg), CJ 300/600 (STZ+CJ(300 and 600 mg/kg). 4 weeks later, oral glucose tolerance test (OGTT) was performed. After sacrificing rats, serum levels of blood urea nitrogen (BUN), creatinine, aspartate amino transferase (AST), alanine amino transferase (ALT), total cholesterol (TC), and triglyceride (TG) were measured and histological changes were observed. Result : Body weight change and food efficiency ratio (FER) were decreased in the CJ 300/600 group than in the COS group. But, there was no change in water intake. Serum levels of glucose, AST, ALT and BUN were lower in the CJ 300/600 group than in the COS group. Also, TG, TC, and creatinine were decreased in the CJ 300/600 group than in the COS group. According to OGTT, 120 minutes postprandial glucose levels were lower in the CJ 300/600 group than in the COS group. In addition, administration of CJ extracts restored histopathological damage. Conclusion : The results suggest that CJ can be used as a functional material for diabetes treatment as it has the effect to improve pathological symptoms in STZ-induced diabetic rats.

Effect of Fertilizer Deep Placement on Rice and Soybean Yield Using Newly Developed Device for Deep Fertilization (신개발 심층시비장치를 이용한 심층시비가 벼와 콩 수량에 미치는 영향)

  • Nitrogen fertilizer is an essential macronutrient that requires repeated input for crop cultivation. Excessive use of nitrogen fertilizers can adversely affect the environment by discharging NH3, NO, and N2O into the air and leaching into surrounding water systems through rainfall runoff. Therefore, it is necessary to develop a technology that reduces the amount of nitrogen fertilizer used without compromising crop yields. Fertilizer deep placement could be a technology employed to increase the efficiency of nitrogen fertilizer use. In this study, a deep fertilization device that can be coupled to a tractor and used to inject fertilizer into the soil was developed. The deep fertilization device consisted of a tractor attachment part, fertilizer amount control and supply part, and an underground fertilizer input part. The fertilization depth was designed to be adjustable from the soil surface down to a depth of 40 cm in the soil. This device injected fertilizer at a speed of 2,000 m2/hr to a depth of 25 to 30 cm through an underground fertilizer injection pipe while being attached to and towed by a 62-horsepower agricultural tractor. Furthermore, it had no difficulty in employing various fertilizers currently utilized in agricultural fields, and it operated well. It could also perform fertilization and plowing work, thereby further simplifying agricultural labor. In this study, a newly developed device was used to investigate the effects of deep fertilizer placement (FDP) compared to those with urea surface broadcasting, in terms of rice and soybean grain yields. FDP increased the number of rice grains, resulting in an average improvement of 9% in rice yields across three regions. It also increased the number of soybean pods, resulting in an average increase of 23% in soybean yields across the three regions. The results of this study suggest that the newly developed deep fertilization device can efficiently and rapidly inject fertilizer into the soil at depths of 25 to 30 cm. This fertilizer deep placement strategy will be an effective fertilizer application method used to increase rice and soybean yields, in addition to reducing nitrogen fertilizer use, under conventional rice and soybean cultivation conditions.

Assessment of Combined Administration of Hypoglycemic Agents and Herbal Extracts (Pyeongwi-san or HyangsaPyongwi-san) on Blood Glucose Levels in Type 2 Diabetes Mellitus: A Retrospective Study (표준 치료를 받고 있는 제2형 당뇨병 환자에서 혈당강하제와 평위산 및 향사평위산 병용 투여가 혈당에 미치는 영향 및 안정성 연구)

  • Abstract: This retrospective study delved into the effects and safety considerations associated with the concomitant usage of hypoglycemic agents and herbal extracts, specifically Pyeongwi-san (PWS) or HyangsaPyongwi-san (HSPWS) in the context of type 2 diabetes mellitus management. Methods: The investigation involved 38 inpatients with type 2 diabetes mellitus who received PWS or HSPWS treatment at Kyung Hee University Korean Medical Hospital from January 2012 to December 2022. By investigating clinical attributes and conducting laboratory assessments, this study aimed to discern the impact of these herbal extracts on blood glucose levels, encompassing fasting blood sugar (FBS) and mean 2-hour postprandial glucose (PP2) levels. Furthermore, the safety profile of the herbal extracts was assessed by comparing liver function indicators, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), and γ-glutamyl transferase (GGT), alongside kidney function markers, such as blood urea nitrogen (BUN) and creatinine (Cr). Results: Following the administration of the herbal extracts, no statistically significant alterations in FBS and mean PP2 levels emerged compared to the baseline levels. Notably, the safety evaluation revealed no significant differences in liver and kidney function parameters following herbal extract administration. Conclusion: The results of this research indicate that using PWS or HSPWS alongside hypoglycemic medications could be a beneficial additional method for addressing digestive symptoms in individuals with type 2 diabetes mellitus. Notably, this combination seems to have no negative interactions with other drugs.
