Anti-Inflammatory Effects of the Chemical Compounds Obtained from Celastrus hindsii in RAW264.7 Cells

  • Celastrus hindsii (family Celastraceae) is located abundantly in the United States, China, and Vietnam, where it is utilized as a traditional herbal and traditional drug for the care of cancer. However, the antioxidant and anti-inflammatory effects of Celastrus hindsii extract are unknown. In our research, the antioxidant activity of Celastrus hindsii leaf extract was investigated, and then anti-inflammatory efficacy of C. hindsii extract in lipopolysaccharide (LPS)-induced RAW264.7 cells. First, our results revealed that C. hindsii extract showed powerful antioxidant capability. Moreover, the application of C. hindsii extract significantly reduced nitric oxide (NO) production without cytotoxic effects. Furthermore, C. hindsii extract reduced the generation of pro-inflammatory cytokines, like as interleukin (IL)-6, IL-1β, and TNF-α. Our results are the first to confirm the anti-inflammatory capability of C. hindsii extract in RAW264.7 cells.

Changes in volatile compounds in rice-based distilled soju aged in different types of containers (숙성기간과 저장용기를 달리한 쌀 증류식 소주의 휘발성 향기성분 변화)

  • In this study, volatile compounds in 13 aged and 3 commercial rice-distilled soju samples were isolated by headspace solid phase microextraction (HS-SPME) and analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 85 volatile components including 35 esters, 15 alcohols, 5 ketones, 3 aldehydes, 15 miscellaneous, and 14 unknowns were identified. Esters and alcohols were the largest groups among the quantified volatiles. Differences in volatile compounds among the distilled soju samples and possible sample groupings were examined by principal component analysis of the GC-MS datasets. The first and second principal components (PC1 and PC2, respectively) explained 51.94% of the total variation across the 16 samples. The samples aged in oak containers had higher concentrations of ketones, aldehydes, and miscellaneous compounds. In the positive direction of PC1, oak-aged samples were observed, while, pot-aged samples were observed on the far negative side. Furthermore, samples aged for longer periods, such as 18 months, were observed in the positive direction of PC2.

Effect of roasting degree of barley on aroma characteristics of boricha (보리의 로스팅 정도가 보리차의 향 특성에 미치는 영향)

  • The objective of this study was to evaluate the effect of roasting degree of barley on aroma characteristics of boricha (barley tea) using solvent-assisted flavor evaporation-gas chromatography-mass spectrometry and gas chromatography-olfactometry. Fifteen volatile compounds including pyrazine, ethylpyrazine, butyrolactone, and guaiacol were considered important volatile compounds, which are generated by roasting barley, because concentrations of those volatiles were significantly increased (p<0.05) as roasting degree of barley was darker. Guaiacol (smoky), furfuryl alcohol (burnt sugar), and furfural (caramel) were detected as aroma-active compounds of boricha with high intensity. Aroma intensity of aroma-active compounds in boricha increased with increase in roasting degree of barley. However, one unknown compound with burnt smell was detected as off-flavor in dark roasted barley. Therefore, it is implied that medium roasting of barley is desirable during boricha manufacturing. Furthermore, boricha manufactured with steamed barley contained more abundant volatile flavor compounds, which may lead to better aroma quality of boricha.

Antioxidant and Anticancer Activities of Artemisia annua L. and Determination of Functional Compounds (개똥쑥의 항산화 및 항암활성과 기능성 물질의 탐색)

  • The antioxidant and anticancer activities of leaves and stems from Gaeddongssuk (Artemisia annua L.) were investigated. The leaves and stems were extracted with water and 80% ethanol, respectively. The antioxidant and growth inhibition activities toward cancer cells by the 4 kinds of extracts were tested. In addition, phenolic compounds from A. annua were identified through quantitative analysis using HPLC. Antioxidant activities significantly increased in a does-dependent manner, and those of water and ethanol extracts of the leaves were stronger than those of the stems. Growth inhibition activities of the leaf extracts on HeLa and AGS cells were higher than those of the stem extracts. In particular, the ethanol extract of the leaves had growth inhibition activities of 61.07% and 57.24% against HeLa and AGS cells, at the concentration of $500\;{\mu}g$/mL, respectively, which were the highest among all the extracts. Phenolic acid and catechin contents of the A. annua extracts as determined by HPLC were higher in the leaves than in the stems. Flavonols were identified only in the leaves. The data suggest that the antioxidant and anticancer activities of A. annua extracts were due to phenolic compounds as well as unknown biological compounds in A. annua.

Characterization of Diethyl Phthalate(DEP) Removal using Ozone, UV, and Ozone/UV Combined Processes (오존, UV, 오존/UV 혼합 공정을 이용한 Diethyl Phthalate(DEP)의 제거특성 연구)

  • Three candidate processes(ozone alone, UV alone and ozone/UV combined processes) were evaluated for the removal of diethyl phthalate(DEP). Of the candidates, the ozone/UV process showed the highest removal efficiency of DEP. To elucidate a major oxidant for DEP oxidation in the ozone/UV process, the effects of pH and hydroxyl radical($OH^{\circ}$) scavenger were investigated. As a result, it was found that $OH^{\circ}$ plays a important role for DEP elimination. Meanwhile, the direct reaction between ozone and DEP was negligible. Observing the pseudo first-order rate of DEP removal in ozone alone and ozone/UV processes, the different pattern was obtained from two processes. The ozone/UV process was well plotted following the pseudo first-order. but in the ozone alone process the rate was divided into fast and slow phases. DEP degradation characteristics in ozone alone and ozone/UV was also investigated by observing the HPLC spectrum. We detected unknown compounds that were guessed to DEP byproducts and observed the formation and disappearance of the unknown compounds according to reaction time. Observing of high removal of TOC in ozone/UV combined process, it was found that DEP and DEP byproducts are completely oxidized by ozone/UV combined process.

Isolation of formaldehyde-responsive proteins in Arabidopsis (Formaldehyde에 반응하는 애기장대 단백질의 분리)

  • Plant can detoxify the effect of the volatile organic compounds (VOC) such as formaldehyde and toluene, however, mechanisms of VOC detoxification are largely unknown in plant system. This study was performed to investigate phenotypic changes of Arabidopsis seedlings upon treatment of either formalin or toluene. Formalin treatment up to twenty four hours didn't cause any significant phenotypic damages on the leaf surface of 27 DAG Arabidopsis seedlings. However, the protein profile of formalin-treated seedlings was significantly different from that of mock control. Using automated electrophoresis system, the molecular weight of each formaldehyde-responsive protein (FRP) was predicted and its formaldehyde-dependent expression was confirmed at transcription level by quantitative real-time RT-PCR analysis. Four FRPs isolated in this study are the novel proteins with unknown functions but highly homologous to the stress-related proteins.

Evaluation of In vitro and In vivo Screening Methods for Estrogenic Activity of Endocrine Disruptors (내분비계 장애물질의 에스트로겐 활성에 대한 In vitro 및 In vivo 검색시험법)

  • The purposes of our study were to optimize the conditions of the screening and testing methods for endocrine disruptors, to characterize these assays using several compounds with well-defined endocrine activity, and to compare the sensitivity between these assays currently undergoing validation. Two in vitro test systems, MCF-7 cells proliferation (E-screen assay) and competitive binding to estrogen receptors (ER) were selected to evaluate the estrogenic effects. 17$\beta$-Estradiol (E2) and diethylstilbestrol (DES) were used as a positive control in vitro test. Also, E2 and ethinyl estradiol (EE) were used as a positive control in vivo uterotrophic assay. In in vitro test, E2 and DES showed a strong estrogenic response at concentration of 1.0 nM. In uterotrophic assay, E2 (0.3 $\mu\textrm{g}$/kg) and EE (0.3 $\mu\textrm{g}$/kg) produced a significant increase in uterus and vagina weight in both immature and ovariectomized rats. Although we did not com-pared the specificity between in vivo and in vitro assays, these assay systems may serve as a good tool for endocrine disruptors screening methods. Our data indicate that these assay systems exhibit some difference in their sensitivity to the same estrogenic compounds. Therefore, as a first rapid screening assay for estrogenic activity qf unknown chemicals, at least two assay systems should probably be carried out with a view of high sensitivity and standardization conditions. Also, a careful validation tests are necessary to obtain a reasonable degree of reproducibility.

Determination of Catechin Compounds in Korean Green Tea Infusions under Various Extraction Conditions by High Performance Liquid Chromatography

  • Liquid chromatographic methods with UV and fluorescence detection have been used to determine the levels of (+)-catechin, (-)-epicatechin, (-)-epicatechin gallate, and (-)-epigallocatechin gallate in Korean green tea infusions. The extracts of Korean green tea leaves or powders in water at various temperatures (100 ${^{\circ}C}$, 80 ${^{\circ}C}$, 60 ${^{\circ}C}$) and time, were washed with chloroform and re-extracted to ethyl acetate. The ethyl acetate phase was dried and re-dissolved in methanol and analyzed. Five catechin compounds were separated by gradient elution. The flavonoids were found decomposed on prolonged extraction, thus exhaustive extraction by a Soxhlet apparatus was found useless for green tea. Some unknown components were found in the extracts at 100 ${^{\circ}C}$. When the green tea was filtered and re-extracted with new fresh water, still some flavonoids were extracted. However, the contents of flavonoids in the third extract were found negligible. The flavonoid extraction rate of green tea powders was higher than that of green tea leaves, but flavonoid decomposition of green tea powders was also faster than that of green tea leaves. The traditional way of drinking green tea was found appropriate in view of flavonoids intake.

Evaluation of Sampling Methodology for the Measurement of Polycyclic Aromatic Hydrocarbons in the Atmosphere (대기 중 다환방향족 탄화수소의 측정을 위한 시료포집방법의 비교평가)

  • This study was carried out to investigate the influence of different sampling methods on the measured concentrations of polycyclic aromatic hydrocarbons (PAH) both in the vapor and particulate phases, and to evaluate the effects of ambient temperature and sampling duration on the losses of PAH associated with particle samples due to volatilization. The experimental protocol of this study is consisted of two parts. The first part is related to the comparison of PAH concentrations measured by 4 different sampling systems, each of which involves different sampling principles for comparison purposes, including a medium-volume sampler with XAD-2 adsorbent, a high-volume sampler with polyurethane foam (PUF), two identical low-volume samplers: one with XAD-2 and the other with PUF, respectively. The second part of this study is to quantitatively estimate the losses of particulate PAH samples by volatilization during sampling, using two identical low-volume samplers: one was used for changing the filters every 3 hrs, 6 hrs, 12 hrs, and 24 hrs sampling, while the other was maintained for continuous 48 hours sampling without changing the filter. The concentrations of volatile PAH including 2-3 rings appeared to be significantly affected by the type of adsorbent. Measured levels of these lower-molecular weight PAH by XAD-2 adsorbent were much higher than those by PUF for both high-volume and low-volume sampling. PUF was found to give rise to unknown components that interfered with the PAH analysis, even after extensive clean-up. In addition, the retention efficiency of PUF for lower molecular weight PAH was subject to a large variation, being significantly influenced by sampling conditions such as ambient temperature. However, the effect of sampling methods with different adsorbents on the measured levels of semi-volatile compounds including 4 rings PAH such as fluoranthene, pyrene, BaA and chrysene, was not so much significant as more volatile PAH compounds. It was also clear from this study that volatilization losses of the semi-volatile PAH collected on the filters were inevitably occurred during prolonged sampling, and hence the results obtained from conventional sampling methods may not be expected to yield an accurate distribution of PAH between the vapor and particulate phases.

Cytotoxic Effects of Phytophenolics from Caesalpinia mimosoides Lamk on Cervical Carcinoma Cell Lines through an Apoptotic Pathway

  • Background: Extracts of Caesalpinia mimosoides Lamk has been reported to possess anticancer effects, but the active ingredients and the anti-cancer mechanisms are still unknown. Materials and Methods: The effects of a C mimosoides Lamk extract on cell proliferation and apoptosis induction in human cervical carcinoma cell lines, namely HeLa, SiHa, and C33A, as well as in normal Vero cells, were investigated. Results: Treatment with 5 active fractions (F17-F21) of C mimosoides Lamk methanol extracts inhibited cell viability in a dose- and time-dependent manner. Neutral red assays indicated that treatment with F21 significantly decreased the viability of all cervical cancer cell lines compared to F21-treated normal cells. In addition, HPLC analysis revealed that F21 contained multiple phenolic compounds, namely gallic acid, caffeine, vanillic acid, ferulic acid and resveratrol. F21 had the lowest IC50 and, therefore, a much higher cytotoxicity than F20, F17, F19, and F18 by 20-, 25-, 46- and 47- fold, respectively. Analysis of activation of the apoptosis pathway using a caspase 3/7 activity assay revealed that F21 treatment resulted in a considerable increase in caspase activation in all cancer cell lines tested. At the same concentration of F21, HeLa cells had the highest caspase activity (6.5-fold) compared to the control. Conclusion: C mimosoides Lamk may be of value as an alternative therapeutic agent, especially in combination with other compounds offering possible of synergy of action. Moreover, HPV- and non-HPV-related cervical cancer cells may differ in their responses to treatment regimens.
