호르텐스극구흡충을 흰쥐에게 경구 감염시키고 4주 후에 성충을 획득하여 고정, 포매 및 절편하여 조직항원 분포 부위를 관찰하였다. 호르텐스극구흡충의 항체는 호르텐스극구흡충 피낭유충을 흰쥐에게 경구 감염시키고 4주 후에 채혈한 혈청과 호르텐스극구흡충 조항원을 면역증강제와 함께 흰쥐에게 근육 주사하여 채혈한 혈청을 이용하였으며, 이 혈청을 반응시켜서 호르텐스극구흡충 조직내의 항원성 분포를 규명하였다. 한편, 면역황금표식법에 의한 미세조직의 항원성 반응 여부는 조직에 표지된 황금입자로 판정하였다. 그 결과 충체 조직에서 황금입자와 강하게 반응하는 미세조직은 표피층의 표지합포체, 난황세포의 구성물질, 유연조직의 분비과립 그리고 배설낭의 구성물질 등에서 반응이 강하게 나타났다. 이상의 결과를 볼 때 호르텐스극구흡충의 항원은 생식세포와 소화분비 및 소화관계통에서 분비 유래되는 것으로 분석되었다.
The structure of the ovary, ultrastructure of oocytes and morphological characteristics of vitellogenesis during oogenesis in female Gomphina veneriformis were investigated in clams collected from coastal waters of Samchok, Gangwon-do, Kore. In the previtellogenic oocytes, the Golgi complex was involved in the formation of a number of vacuoles. In the early vitellogenic oocytes, lipid droplets appeared among the Golgi complex, endoplasmic reticulum, and mitochondria in the cytoplasm of the oocyte were involved in the formation of lipid droplets. Coated vesicles, resulting from endocytosis appeared at the basal region of the early vitellogenic oocyte. The uptake of nutritive materials in the coated vesicles formed by receptor-mediated endocytosis appeared through the formation of coated endocytotic pits on the oolemma. In the late vitellogenic oocytes, large yolk granules were formed by a combination of small yolk granules. In the mature oocyte, a mature yolk granule in composed of three components: crystaline core, electron lucent cortex, and a limiting membrane. According to cytological and histological observations, vitellogenesis occurred by way of endogenous autosynthesis and exogenous heterosynthesis. Autosynthesis involved the conbined activities of the Golgi complex, mitochondria, rough endoplasmic reticulum, whereas heterosynthesis involved endocytotic incorporation of extraovarian precursors at the basal region of the early vitellogenic oocyte. The follicle cells which was attached to oocytes, were involved in the development of the previtellogenic and early vitellogenic oocytes as a kind of nutritive cells containing a number of glycogen particles and lipid droplets in the cytoplasm.
정상성인 여성의 머리카락에 미용실에서 일반적으로 시술하는 탈색제를 사용한 다음 탈색직후, 탈색 후 10일, 20일이 경과한 머리카락을 채취하여 머리카락의 손상정도를 고배율의 투과 및 주사전자현미경으로 관찰하였다. 탈색직후 머리카락의 표면은 정상 머리카락과 비슷한 상태로 관찰되었으며 비늘이 분리되거나 손상됨 없이 나타났다. 탈색 후 10일 경과된 머리카락은 비늘이 분리되어 있고 일부 큐티클세포의 세포질은 조각이 나 있거나 떨어져 나갔다. 이 시기에 머리카락은 비늘이 떨어져 나가면서 표면에 세포부스러기들이 그대로 부착되어 있고, 비늘의 모양은 끝 쪽이 날카로운 모양을 하고 있었다. 탈색 후 20일이 경과된 머리카락은 표면 전체가 비늘의 분리에 의해서 거칠게 나타났다.
Brain ischemia leads to overstimulation of N-methyl-D-aspartate (NMDA) receptors, referred as excitotoxicity, which mediates neuronal cell death. However, less attention has been paid to changes in synaptic activity and morphology that could have an important impact on cell function and survival following ischemic insult. In this study, we investigated the effects of reperfusion after oxygen/glucose deprivation (OGD) not only upon neuronal cell death, but also on ultrastructural and biochemical characteristics of postsynaptic density (PSD) protein, in the stratum lucidum of the CA3 area in organotypic hippocampal slice cultures. After OGD/reperfusion, neurons were found to be damaged; the organelles such as mitochondria, endoplasmic reticulum, dendrites, and synaptic terminals were swollen; and the PSD became thicker and irregular. Ethanolic phosphotungstic acid staining showed that the density of PSD was significantly decreased, and the thickness and length of the PSD were significantly increased in the OGD/reperfusion group compared to the control. The levels of PSD proteins, including PSD-95, NMDA receptor 1, NMDA receptor 2B, and calcium/calmodulin-dependent protein kinase II, were significantly decreased following OGD/reperfusion. These results suggest that OGD/reperfusion induces significant modifications to PSDs in the CA3 area of organotypic hippocampal slice cultures, both morphologically and biochemically, and this may contribute to neuronal cell death and synaptic dysfunction after OGD/reperfusion.
고압맥동전류가 탈신경근의 형태에 미치는 영향을 알아보기 위하여 조직화학적 방법과 투과전자현미경적 관찰을 하였다. 웅성 흰쥐를 정상군, 탈신경군으로 각각 8마리씩 나누어 2주와 4주 후에 희생시켜 실험한 결과 다음과 같은 결과를 얻었다. 1. 조직화학적으로 형태를 관찰 한 결과 대조 1주군부터 근속과 근섬유사이에 염증세포가 관찰되고 핵이 근섬유속에 위치하는 것도 자주 관찰되었다. 2. 대조군도 비슷한 양상을 보였으나 4주군은 근괴사와 염증세포가 더욱 증가하였다. 3. 당원 1주군에서 실험군, 대조군 모두 정상근과 비슷한 양상을 보이나 4주군에서는 섬유를 구별할 수 없는 형태로 관찰되었다. 4. NADH-TR반응에서 적색섬유가 2주군에서 약간 증가하였고 그 후로는 구별이 불가능하였다. 5. 미세구조적으로 양쪽군 모두 근섬유가 굽어있고 mitochondria의 파괴로 인한 공포가 많이 관찰되었으나 전기자극 2주군에서는 일부에서 mitochondria증가를 관찰 하였다.

Lyngbya C. Agardh ex Gomont is a nonheterocytous cyanobacterial genus whose evolutionary history is still poorly known. The traditionally defined Lyngbya has been demonstrated to be polyphyletic, including at least five distinct clades, some of which have been proposed as new genera. Intraspecific diversity is also clearly underestimated in Lyngbya due to the lack of unique morphological characters to differentiate species. In this study, we describe the new genus Capillus T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes from benthic marine environments, including two new Brazilian species (here described as C. salinus T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes, and C. tropicalis T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes), and two species yet to be described, one of them from India (Capillus sp. 2.1), and the other from United States of America, based on strain PCC 7419. Capillus species presented cross-wise diagonal fragmentation, assisted or not by necridic cells, which has not been previously mentioned for Lyngbya. Ultrastructural analyses showed that C. salinus and C. tropicalis have numerous gas vesicles, which are rarely described for benthic marine species. The new genus formed a well-supported clade, and the D1-D1' and Box B secondary structures of internal transcribed spacer also supported the proposal of its new species. These findings help to clarify the diversity of species in the Lyngbya complex and the taxonomy of the group, and highlight the need of further floristic surveys in tropical coastal environments, which remain poorly studied.

In Islamic dietary guidelines, Halal foods are allowed as edible blessed food. Most foods were categorized within halal for Muslims. The main point of Halal food is that foods are clean in every process and based on Halal standard which might be different in each country. Most pancreatic ${\beta}$ cells synthetize, store, and release insulin. Specific molecular, functional as well as ultrastructural traits of pancreatic ${\beta}$ cells could control their insulin secretion properties and survival phentoype. Insulin-secreting pancreatic ${\beta}$-cells are essential regulators of mammalian metabolism. In addition, the pancreatic ${\beta}$ cell plays an important role in the pathogenesis of type 1 and type 2 diabetes as improving glucose homeostasis by preserving, expanding and improving the function of this key cell type. However, the pharmacological effect of halal food has not been unclear yet, especially food habit-dependent diabetes. The aim of the this study was to determine the preventive effect of Iran plants extract (Almond, Garlic, Cumin, Ginkgo biloba, Holy basil, Psyllium, Satureja khuzistanica, Fenugreek, Green tea, Ipomoea betatas, Blueberry) on RINm5F cells and MIN6 cells as pancreatic ${\beta}$ cell line. The cytotoxicity of the extracts of Iran plants on RINm5F cells and MIN6 cells were measured by using MTT assays. The preventive effects of Iran plant extracts were measured by WST-8 cell proliferation assay on streptozotocin (STZ)-induced cell death in MIN6 cells. In presented result showed that all extract of Iran plants (0.01-10mg/ml) did not show cytotoxicity in RINm5F cells and MIN6 cells. Among non-cytotoxic extract, the protective effects could be detect in high dose concentration. These results suggest that the extract of Iran plants may serve as a potential therapy for diabetes.
Background: Massive rotator cuff tears (RCTs) are complicated by muscle atrophy, fibrosis, and intramuscular fatty degeneration, which are associated with postoperative tendon-to-bone healing failure and poor clinical outcomes. We evaluated muscle and enthesis changes in large tears with or without suprascapular nerve (SN) injury in a rat model. Methods: Sixty-two adult Sprague-Dawley rats were divided into SN injury (+) and SN injury (-) groups (n=31 each), comprising tendon (supraspinatus [SSP]/infraspinatus [ISP]) and nerve resection and tendon resection only cases, respectively. Muscle weight measurement, histological evaluation, and biomechanical testing were performed 4, 8, and 12 weeks postoperatively. Ultrastructural analysis with block face imaging was performed 8 weeks postoperatively. Results: SSP/ISP muscles in the SN injury (+) group appeared atrophic, with increased fatty tissue and decreased muscle weight, compared to those in the control and SN injury (-) groups. Immunoreactivity was only positive in the SN injury (+) group. Myofibril arrangement irregularity and mitochondrial swelling severity, along with number of fatty cells, were higher in the SN injury (+) group than in the SN injury (-) group. The bone-tendon junction enthesis was firm in the SN injury (-) group; this was atrophic and thinner in the SN injury (+) group, with decreased cell density and immature fibrocartilage. Mechanically, the tendon-bone insertion was significantly weaker in the SN injury (+) group than in the control and SN injury (+) groups. Conclusions: In clinical settings, SN injury may cause severe fatty changes and inhibition of postoperative tendon healing in large RCTs. The present study examined the possibility of long term storage, by cryopreservation in liquid nitrogen, of the sperm of Filefish (Thamnaconus septentrionalis), and the changes in motility, survival rate and ultrastructure of the sperm after freezing and thawing. The sperm was collected by stripping and stored on ice until experiments. For selection of the immobilizing solution, diluted artificial seawater (ASW) of 20, 30 and 40% were tested. The sperm motility was significantly inhibited in 30% ASW, and restored entirely after 100% ASW was added again. Two cryoprotectants, dimethyl sulfoxide ($Me_2$SO) and glycerol, were added to 30% ASW to formulate the extenders at the concentrations between 5 to 20% by volume for freezing. The sperm was diluted at the ratio of 1 :6 with the extenders, inserted into 0.5ml plastic straws and frozen at a freezing rate of $50^{\circ}C$/min to $-100^{\circ}C$ after equilibration for 10 min at room temperature, followed by plunging into liquid nitrogen. The straws were thawed in a $30^{\circ}C$ water bath for 15 sec. The highest post-thawed sperm motility and survival rate were obtained with 5% glycerol Afterward, the effect of different freezing rates was examined using 5% glycerol as a cryoprotectant, and the rate of $20^{circ}C/min to $-80^{\circ}C$ showed the best result Some ultrastructural changes of sperm, such as the detachment of plasmatic and nuclear membranes, destruction of mitochondria, were observed after cryopreservation. Morphological normality of the sperm in 5% glycerol frozen at the ratio of 1$0^{\circ}C$/min to $-80^{\circ}C$ was better than that of others.
Xanthomonas oryzae pv. oryzae (Xoo) causes a devastating bacterial leaf blight in rice. Here, the antimicrobial effects of ᴰ-limonene, ᴸ-limonene, and its oxidative derivative carveol against Xoo were investigated. We revealed that carveol treatment at ≥ 0.1 mM in liquid culture resulted in significant decrease in Xoo growth rate (> 40%) in a concentration-dependent manner, and over 1 mM, no growth was observed. The treatment with ᴰ-limonene and ᴸ-limonene also inhibited the Xoo growth but to a lesser extent compared to carveol. These results were further elaborated with the assays of motility, biofilm formation and xanthomonadin production. The carveol treatment over 1 mM caused no motilities, basal level of biofilm formation (< 10%), and significantly reduced xanthomonadin production. The biofilm formation after the treatment with two limonene isomers was decreased in a concentration-dependent manner, but the degree of the effect was not comparable to carveol. In addition, there was negligible effect on the xanthomonadin production mediated by the treatment of two limonene isomers. Field emission-scanning electron microscope (FE-SEM) unveiled that all three compounds used in this study cause severe ultrastructural morphological changes in Xoo cells, showing shrinking, shriveling, and holes on their surface. Moreover, quantitative real-time PCR revealed that carveol and ᴰ-limonene treatment significantly down-regulated the expression levels of genes involved in virulence and biofilm formation of Xoo, but not with ᴸ-limonene. Together, we suggest that limonenes and carveol will be the candidates of interest in the development of biological pesticides.
