중부지방(中部地方) 주요활엽수(主要闊葉樹)의 직경성장(直徑成長)에 관한 조사연구(調査硏究) (A Study on the Diameter Increment of Major Hardwood in Middle Area of Korea)

  • 본(本) 연구(硏究)는 우리나라 중부지방(中部地方)에셔 자생(自生)하고 있는 활엽수(濶葉樹) 천연임분(天然林分)을 대상(對象)으로 몇 개의 주요(主要)한 수종(樹種)을 선택(選澤)하여 그들에 대(對)한 직경(直徑) 성장량(成長量)을 추정(推定) 비교(比較)하였는데 그 결과(結果)를 요약(要約)하면 다음과 같다. 1) 흉고직경급(胸高直徑級)에 따른 연년직경(連年直徑) 생장량(生長量)은 가래나무, 층층나무, 자작나무, 피나무, 들메나무, 사시나무, 느릅나무 순(順)으로 나타났다. 2) 수령급(樹令級)에 따른 흉고직경(胸高直徑) 총성장량(總成長量)은 가래나무, 층층나무, 자작나무, 피나무, 들메나무, 사시나무, 느릅나무 순(順)으로 나타났다. 3) 타(他) 수종(樹種)에 비(比)하여 가래나무는 성장(成長)이 월등(越等)히 우수(優秀)하였으며, 특(特)히 느릅나무와 사시나무는 성장(成長)의 둔화현상(鈍化現象)이 빨리 나타났다.

유근피(楡根皮)의 선천 면역 활성화에 의한 암 전이 억제 효과 (Experimental Studies on Antimetastatic and Immunomodulating Effects of Ulmus davidiana)

  • Purpose: This study was designed to investigate the antimetastatic and immunomodulating effects of extracts of Ulmus davidiana extracts(U. D. Ex.). Methods: Antimetastatic experiments were conducted in vitro and in vivo by using colon 26-M3.1 carcinoma, L5178Y-R lymphoma cell and Hela cell. To observe the immunomodulating effects of U. D. Ex., we measured IL-6, IL-10, IL-12 and TNF-$\alpha$ from peritoneal macrophages. And we evaluated the activation of NK cell by using anti-asialo-GM1 serum. Results: We found that the administration of U. D. Ex. significantly inhibited tumor metastasis in vivo. In vitro cytotoxicity analysis, cell growth are closer to 100% in case of Colon 26-M3.1 carcinoma, L5178Y-R lymphoma cell and Hela cell at low concentration. In case of macrophage, cell proliferation is closer to 100% less than $250{\mu}g/ml$ of U. D. Ex.. The level of cytokine such as IL-6, IL-10, IL-12 which stimulates U. D. Ex. was increased in dose-dependent manner compared to the control group. In case of TNF-$\alpha$, the level was increased at concentration of $1,000{\mu}g/ml$. The depletion of NK cells by anti-asialo GM1 serum partly abolished the inhibitory effect of U. D. Ex. on tumor metastasis. Conclusion: Ulmus davidiana appears to have considerable activity on the anti-metastasis by activation the immune system.

Effect of Ulmus davidiana Planch (Ulmaceae) on T-lymphocyte-producing cytokines such as IL-2, IL-6, and $IFN-{\gamma}$ production in collagen-induced arthritis of rats

  • Objective : The effect of Ulmus davidiana Planch(UD), which has long been known to have anti-inflammation and protective effects on damaged tissue, inflammation and bone among other functions, on the development of type II collagen (CII)-induced arthritis (CIA) in rats was studied. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA)or saline during the experiment. When compared with rats treated with saline as control, UDHA at doses of more than $20{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-2, interleukin-6, $IFN-{\gamma}$ when the cells were obtained from rats 14 days after immunization and cultured in vitro with CII. Results : When rats were injected intraperitoneally, UD -treated group and control group rats did not differ significantly when low doses of UD was given to rats. Conclusion : The recommended dose of UD in the management and treatment of rat CIA will be more than $20{\mu}g/100\;g$, which is two-firth of human therapeutic dose. From the results, it was concluded that the effect of UDHA is dependent of dosage.

Suppressive effects of a water extract of Ulmus davidiana Planch (Ulmaceae) on collagen-induced arthritis in mice

  • Objective: Since Ulmus davidiana Planch (Ulmaceae) has been known to have anti-inflammatory and protective effects on damaged tissue, inflammation and bone among other functions, this study was undertaken to address whether the water extract of the bark of Ulmus davidiana Planch (Ulmaceae) (UD) could modulate the expression of inducible inflammatory cytokines in mice. The present study was also done in order to assess the therapeutic effects of UD in collagen-induced arthritis (CIA) in mice. Methods : DBA/1 mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with UD orally at 100 mg/kg once a day for 3 weeks. Paws were evaluated macroscopically for redness, swelling and deformities. The levels of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the ankle were examined. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Results : Administration of UD significantly suppressed the progression of CIA and inhibited the production of $TNF-{\alpha}\;and\;IL-1{\beta}$ in the paws. The erosion of cartilage was dramatically reduced in mouse knees after treatment with UD. In the serum of UD-treated mice, the levels of IL-4 and IL-10, anti-inflammatory cytokines, were increased. Conclusion : From the results, it was concluded that administration of UD has therapeutic effects on CIA including protection of cartilage and RA for a potential therapy.

Anti-Obesity and Lipid Metabolism Effects of Ulmus davidiana var. japonica in Mice Fed a High-Fat Diet

  • The root bark of Ulmus davidiana var. japonica (Japanese elm) is used in Korea and other East Asian countries as a traditional herbal remedy to treat a variety of inflammatory diseases and ailments such as edema, gastric cancer and mastitis. For this study, we investigated the lipid metabolism and anti-obesity efficacy of ethyl alcohol extract of Ulmus davidiana var. japonica root bark (UDE). First, HPLC was performed to quantify the level of (+)-catechin, the active ingredient of UDE. In the following experiments, cultured 3T3-L1 pre-adipocytes and high-fat diet (HFD)-fed murine model were studied for anti-obesity efficacy by testing the lipid metabolism effects of UDE and (+)-catechin. In the test using 3T3-L1 pre-adipocytes, treatment with UDE inhibited adipocyte differentiation and significantly reduced the production of adipogenic genes and transcription factors PPARγ, C/EBPα and SREBP-1c. HFD-fed, obese mice were administered with UDE (200 mg/kg per day) and (+)-catechin (30 mg/kg per day) by oral gavage for 4 weeks. Weight gain, epididymal and abdominal adipose tissue mass were significantly reduced, and a change in adipocyte size was observed in the UDE and (+)-catechin treatment groups compared to the untreated control group (***p < 0.001). Significantly lower total cholesterol and triglyceride levels were detected in UDE-treated HFD mice compared to the control, revealing the efficacy of UDE. In addition, it was found that lipid accumulation in hepatocytes was also significantly reduced after administration of UDE. These results suggest that UDE has significant anti-obesity and lipid metabolism effects through inhibition of adipocyte differentiation and adipogenesis.

느티나무에서 단리한 카달렌 화합물에 관한 연구 II - 7-Hydroxy-3-methoxycadalene의 생물활성 측정 및 카달렌 동족체 분리 - (Study on Cadalene Compounds Purified from Zelkova serrata Wood II - Biological activities of 7-hydroxy-3-methoxycadalene and purification of cadalene homologues -)

  • 본 연구에서는 느티나무의 에탄올 추출액에서 7-hydroxy-3-methoxycadalene 외에 3,7-dimethoxycadalene와 7-hydroxycadalene-5,8-quinone (keyakinone A) 등 2종의 카달렌 동족체를 분리하였고, 이 중에서 7-hydroxy-3-methoxycadalene을 대상으로 hydroxyl 라디칼 소거활성과 MTT assay에 의한 세포독성을 평가하였다. 7-Hydroxy-3-methoxycadalene의 hydroxyl 라디칼 소거활성은 농도에 의존적이며, 100 ppm에서는 hydroxy 라디칼이 거의 100% 제거되는 우수한 효과를 나타냈다. 그러나 7-hydroxy-3-methoxycadalene은 낮은 농도(1 ppm 이하)에서는 세포 생존율이 약 90% 가량으로 세포독성이 거의 나타나지 않았지만 농도가 높아질수록 세포 생존율에 크게 영향을 미치는 것으로 나타났다. 이러한 7-hydroxy-3-methoxycadalene은 4종의 느릅나무과 수종(느릅나무(Ulmus davidiana), 참느릅나무(Ulmus parvifolia), 큰잎느릅나무(Ulmus crophylla), 당느릅나무(Ulmus macrocarpa))에서는 전혀 발견되지 않았다. 느릅나무과 수종에서는 공통적으로 7-hydroxycadalene (분자량 214)으로 추정되는 화합물이 발견되었으며, 이는 느티나무에서 단리한 7-hydroxy-3-methoxycadalene에서 3번 위치의 메톡실기($OCH_3$)가 이탈된 구조의 화합물이다.

Inhibitory Effects of the Ethanol Extract of Ulmus davidiana on Apoptosis Induced by Glucose-glucose Oxidase and Cytokine Production in Cultured Mouse Primary Immune Cells

  • The bark of Ulmus darvidiana var. japonica Nakai (UDN) has been used for a long time to cure inflammation in oriental medicine. In the present study, two types of extracts, Ulmus water-eluted fraction (UWF) and Ulmus ethanol-eluted fraction (UEF), were prepared from the UDN stem bark, and employed to test the extracts to see if they had anti-oxidative properties against hydroxyl radicals that could alter immune reactivity in mouse immune cells. Deoxyribose assay, DNA nicking assay, and glucose/glucose oxidase assay showed that both fractions had scavenging activity against oxygen free radicals at 50 mg/ml. In addition, hydroxyl radical-mediated apoptosis in mouse thymocytes was not protected by UEF treatment, but the apoptosis was protected by UWF at the same concentration. DNA synthesis and cytokine production that were induced in splenocytes by mitogens (Concanavalin A and lipopolysaccharide) were reduced by the addition of both fractions. These results indicate that both extracts that were prepared from the UDN stem bark have anti-oxidative activities, anti-apoptotic effects, and inhibitory effects on DNA synthesis and cytokine production in mouse immune cell cultures.

A Sesquiterpene ortho-Naphthoquinone from the Root Bark of Ulmus davidiana Planch

  • A sesquiterpene ortho-naphtpoquinone was isolated from the 80% aqueous methanolic extract of root bark of Ulmus davidiana Planch whose stem and root bark have been used as an oriental medicine for the treatment of edema, mastitis, gastric cancer and inflammation. On the basis of spectral data obtained from UV-vis, IR, HR-EIMS and NMR spectrometry, including the pulse field gradient (PFG) HMQC and HMBC techniques, the structure of this compound was determined as 2,3-dihydro-3-hydroxy-3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione which has been known as mansonone 1. This compound has been isolated only from the heart-wood of Mansonia altissima Chev, a Sterculiacea from West Africa. Thus, this compound was isolated for the second time from natural source in this study. This is the first report to present the carbon chemical shift assignment of mansonone I.

Investigations into the immunomodulatory activity of Ulmus davidiana Planch extracts

  • Objective: Although the effect of Ulmus davidiana Planch (UD) extracts on collagen-induced-arthritis (CIA) and bone metabolism has been studies, research on its effect on human immunomodulatory activity is further a due. The objective of the present study was to investigate the immunomodulatory activity of UD on cellular and humoral immunity. Methods : Oral administration of the ethanolic and water extracts of UD, at doses of 20, 100 and 200 mg/kg in mice, dose dependently potentiated the delayed type hypersensitivity reaction induced both by sheep red blood cells (SRBC) and oxazolone. Results : It significantly enhanced the production of circulating antibody titre in response to SRBC in mice. Extracts of UD failed to show any effect on macrophage phagocytosis. Chronic administration of UD extracts significantly ameliorated the total white blood cell count and also restored the myelosuppressive effects induced by cyclophosphamide. Conclusion : The present investigation reveals that UD extracts possesses immunomodulatory activity.
