Scarless Genomic Point Mutation to Construct a Bacillus subtilis Strain Displaying Increased Antibiotic Plipastatin Production

  • Bacillus strains produce various types of antibiotics, and random mutagenesis has traditionally been used to overproduce these natural metabolites. However, this method leads to the accumulation of unwanted mutations in the genome. Here, we rationally designed a single nucleotide substitution in the degU gene to generate a B. subtilis strain displaying increased plipastatin production in a foreign DNA-free manner. The mutant strain (BS1028u) showed improved antifungal activity against Pythium ultimum. Notably, pps operon deletion in BS1028u resulted in complete loss of antifungal activity, suggesting that the antifungal activity strongly depends on the expression of the pps operon. Quantitative real-time PCR and lacZ assays showed that the point mutation resulted in 2-fold increased pps operon expression, which caused the increase in antifungal activity. Likewise, commercial Bacillus strains can be improved to display higher antifungal activity by rationally designed simple modifications of their genome, rendering them more efficient biocontrol agents.

Thermogram analysis of Sasang constitutional Four Burners and primary cancer organ in cancer patients (암환자의 사상체질별 사초 부위와 원발암 부위의 체열 분석 연구)

  • 1. Objective This study was conducted to research a corelation between the Sasang constitutions and primary cancer organs in cancer patient with their thermography. 2. Method We Analyzed the thermography taken from cancer patients and compared it with their cancer origins and Sasang constitution. 3. Result It was found that there are some relationships between the cancer origins and abnormal skin temperature. Also according to the Sasang constitutions of each cancer patient, the lesion of abnormal skin temperature could be differentiated. 4. Conclusion Thermogram can be affected by both the constitution and the primary cancer organs of the patient. To utilize the thermography as a diagnostic or prognostic method of cancer, further studies are needed.

Design and Implementation of a Mobile Ubiquitous Healthcare System (모바일 유비쿼터스 헬스케어시스템 설계 및 구현)

  • Recently, owing to the development of ubiquitous sensor network and mobile communication technologies, many studies on healthcare system are being carried out. In this paper, we have designed and implemented a mobile u-Healthcare system based on sensor network. The u-Healthcare system is composed of three components: wireless sensor network at home, healthcare center located at remote site, and gateway which relays sensing physiological signals to healthcare center. In order to measure patient's physiological signal three sensors are used: three channel ECG sensor, pulse oximeter, and blood pressure sensor. Each sensor is mounted on a mote which can send gathered signal to the base node using Zigbee communication protocol. Once the base node receives physiological signal from each sensor, the client in the base node transfers the signal to the healthcare center. The received physiological signal at the healthcare center is analyzed and processed using various algorithms. The processed results are compared to the standard healthcare database and appropriate treatment including dietetics and exercise cure would be sent to the patient as feedback using SMS message or healthcare center web site. Each patient can check and manage one's health state every day using the healthcare system and gain a recovery under the treatments from minor health problems.

  • As the title signifies, the current study aims at the realization of u-City, which is short for the Ubiquitous City. For this purpose, firstly, the meaning of u-City realization is sought in the study based on future urban changes. Along with this, current issues in the course of u-City realization are gathered based on in-depth interviews with relevant experts. Thirdly, necessary for the comprehensive planning and management of u-City, several strategies are presented by classifying it into u-Infra and u-Service, and considering relevant laws and regulations. And the study divided the execution process into project, development and management levels, and then suggests some realization strategies that are level-dependent.

  • 최근 정보기술과 유비쿼터스기술을 바탕으로 도시를 미래화하는 유비쿼터스도시(U-City)구축이 범 국가적으로 추진되고 있다. 이에, 정부에서는 2008년 9월 25일 유비쿼터스도시의 건설 등에 관한 법률 시행령을 발표하였으며, 유비쿼터스도시서비스(U-City Service)를 크게 행정, 교통, 보건 의료 복지, 환경, 방범 방재, 시설물 관리, 교육, 문화 관광 스포츠, 물류, 근로 고용 등의 분야로 구분하였다. 이러한 유비쿼터스도시서비스를 제공하고 운영 및 관리하는 시설로 유비쿼터스도시 통합운영센터(U-City Management Center)는 다양한 유비쿼터스도시서비스를 효과적으로 연계하여 서비스에서 발생되는 다양한 도시정보를 통합, 분석하고 제공하는 유비쿼터스도시 핵심기능을 담당한다. 하지만 이러한 유비쿼터스도시 통합운영센터의 구축은 운영조직구성, 운영비용확보, 통합서비스 제공방안, 법 제도 정비 등의 해결과제를 가지고 있어 구현방안에 대한 구체적인 연구가 요구된다. 본 연구에서는 선행연구를 바탕으로 유비쿼터스도시 통합운영센터에서 기본적으로 제공되어야 할 유비쿼터스도시서비스 유형을 제시하고 정의한다.

  • The purpose of this study was to establish a model of the use-utility of cooking equipments and kitchen centers using survey data and to compare U.S.A. with Korea. The survey was conducted to identify attributes that were related to the efficiency of kitchen centers as well as equipments. Indices for the most important attributes were developed and used to compare different selections of equipment. The impact of adding an equipment item was found to vary with the attributes of the item, the degree of importance ascribed to the attributes of the work center and specific functions performed there. Similarities and differences between the U.S.A. and Korea are discussed, and implications for those who have interest in the use-utility model of cooking equipment and kitchen centers are provided.

Dexamethasone Interferes with Autophagy and Affects Cell Survival in Irradiated Malignant Glioma Cells

  • Objective : Radiation is known to induce autophagy in malignant glioma cells whether it is cytocidal or cytoprotective. Dexamethasone is frequently used to reduce tumor-associated brain edema, especially during radiation therapy. The purpose of the study was to determine whether and how dexamethasone affects autophagy in irradiated malignant glioma cells and to identify possible intervening molecular pathways. Methods : We prepared p53 mutant U373 and LN229 glioma cell lines, which varied by phosphatase and tensin homolog (PTEN) mutational status and were used to make U373 stable transfected cells expressing GFP-LC3 protein. After performing cell survival assay after irradiation, the IC50 radiation dose was determined. Dexamethasone dose (10 μM) was determined from the literature and added to the glioma cells 24 hours before the irradiation. The effect of adding dexamethasone was evaluated by cell survival assay or clonogenic assay and cell cycle analysis. Measurement of autophagy was visualized by western blot of LC3-I/LC3-II and quantified by the GFP-LC3 punctuated pattern under fluorescence microscopy and acridine orange staining for acidic vesicle organelles by flow cytometry. Results : Dexamethasone increased cell survival in both U373 and LN229 cells after irradiation. It interfered with autophagy after irradiation differently depending on the PTEN mutational status : the autophagy decreased in U373 (PTEN-mutated) cells but increased in LN229 (PTEN wild-type) cells. Inhibition of protein kinase B (AKT) phosphorylation after irradiation by LY294002 reversed the dexamethasone-induced decrease of autophagy and cell death in U373 cells but provoked no effect on both autophagy and cell survival in LN229 cells. After ATG5 knockdown, radiation-induced autophagy decreased and the effect of dexamethasone also diminished in both cell lines. The diminished autophagy resulted in a partial reversal of dexamethasone protection from cell death after irradiation in U373 cells; however, no significant change was observed in surviving fraction LN229 cells. Conclusion : Dexamethasone increased cell survival in p53 mutated malignant glioma cells and increased autophagy in PTEN-mutant malignant glioma cell but not in PTEN-wildtype cell. The difference of autophagy response could be mediated though the phosphatidylinositol 3-kinase/AKT/mammalian target of rapamycin signaling pathway.

  • u-City 통합운영센터는 u-City 운영의 가장 기본이면서 기존 도시와 u-City를 차별화 하는 중요한 요소이다. 이에 따라 도시통합운영센터에서 효과적으로 제공해야 하는 u-City 서비스 선정을 위한 분류가 마련되어야 한다. 본 연구에서는 u-City 도시통합운영센터의 중요 기능인 관제에 해당하는 서비스들을 분류하고 향후 u-City 도시통합운영센터를 구축할 때 관제 관점에서 고려되어야 할 서비스들에 대하여 제언하고자 한다.

  • 유비쿼터스 사회가 도래함에 따라 유비쿼터스 기술을 도시에 적용한 u-City가 개발되고 있다. u-City는 다양한 정보화 기기들이 존재하며 정보화 기기 사이를 연결하는 다양한 네트워크 기술이 상존한다. u-City의 핵심 요소인 u-City 통합운영센터는 u-City 내의 모든 서비스를 처리하도록 설계되었으며, 도시를 통제 관리하는 중요한 업무를 수행한다. 이 중에서 보안관리 업무는 시스템에서 수집, 가공, 산출하는 데이터 및 정보의 보안유지를 위해 중요한 부분이다. 따라서 본 논문에서는 u-City 통합운영센터의 보안유지를 위한 기술적, 물리적, 관리적인 측면에서 보안관리 대책을 제시하였다.
