A 990 bp full-length gene (xynAHJ2) encoding a 329-residue polypeptide (XynAHJ2) with a calculated mass of 38.4 kDa was cloned from Bacillus sp. HJ2 harbored in a saline soil. XynAHJ2 showed no signal peptide, distinct amino acid stretches of glycoside hydrolase (GH) family 10 intracellular endoxylanases, and the highest amino acid sequence identity of 65.3% with the identified GH 10 intracellular mesophilic endoxylanase iM-KRICT PX1-Ps from Paenibacillus sp. HPL-001 (ACJ06666). The recombinant enzyme (rXynAHJ2) was expressed in Escherichia coli and displayed the typical characteristics of low-temperature-active enzyme (exhibiting optimum activity at $35^{\circ}C$, 62% at $20^{\circ}C$, and 38% at $10^{\circ}C$; thermolability at ${\geq}45^{\circ}C$). Compared with the reported GH 10 low-temperature-active endoxylanases, which are all extracellular, rXynAHJ2 showed low amino acid sequence identities (<45%), low homology (different phylogenetic cluster), and difference of structure (decreased amount of total accessible surface area and exposed nonpolar accessible surface area). Compared with the reported GH 10 intracellular endoxylanases, which are all mesophilic and thermophilic, rXynAHJ2 has decreased numbers of arginine residues and salt bridges, and showed resistance to $Ni^{2+}$, $Ca^{2+}$, or EDTA at 10 mM final concentration. The above mechanism of structural adaptation for low-temperature activity of intracellular endoxylanase rXynAHJ2 is different from that of GH 10 extracellular low-temperature-active endoxylanases. This is the first report of the molecular and biochemical characterizations of a novel intracellular low-temperature-active xylanase.

Corrugated steel plate structure, which is built by assembling corrugated steel plate segments with bolts on site and filling the surroundings with quality soil, is widely used for buried structures as a eco-corridors, small bridges, and closed conduits. This experimental study is dealt with the static and fatigue performance of bolt connected corrugated steel plates under flexural loading. The experimental variables to verify the fatigue performance are bolt diameters and detailing of connection such as washer and the corrugation dimension of specimens has a $400{\times}150$ mm. The experimental ultimate strength of specimens under static loading was higher than the theoretical strength and all specimen failed by a bearing and tearing failure of bolt hole of upper plate. Therefore, a fatigue tests of specimens had 6.0mm and 7.0mm thickness was conducted in which the load range was up to 209kN and 516kN, respectively. From the fatigue test, failure patterns are changed from plate bearing and tearing which is a typical failure pattern of static failure to a bearing failure of plate and shear failure of bolt, and experimental fatigue limit at $2{\times}10^6$cycles is about 85MPa.

Although many studies have been performed for the structural details of orthotropic steel deck, most of them were focused on the trough rib of standard type, but not for orthotropic steel deck with longitudinal rib of open section used at beginning of the deck. In order to investigate the cause of fatigue crack for orthotropic steel deck bridge serviced 31 years with longitudinal rib of open section, in this study, the behavior characteristics of target structural details were analyzed based on measurement data under real traffic condition. Also the typical loading truck passing the target bridges was estimated with the structural analysis detailed, and the stresses and deformation patterns of target structural details were analyzed by performing the detailed structural analysis. Based upon the analysis, retrofitting methods of the fatigue crack were suggested and its validation was examined. From this study, it was clarified that fatigue crack of longitudinal rib with open section were affected with the stress increment by shear deformation in the rib and the occurrence of alternative stress due to moving vehicle. In addition, it was known that it is important to perform fatigue design reflected the local behavior of the structural details.

Recently, concerns relating to the maintenance of large structures have been increased. In addition, the number of large structures that need to be evaluated for their structural safety due to natural disasters and structural deterioration has been rapidly increasing. It is common for the structural characteristics of an older large structure to differ from the characteristics in the initial design stage, and changes in dynamic characteristics may result from a reduction in stiffness due to cracks on the materials. The process of deterioration of such structures enables the detection of damaged locations, as well as a quantitative evaluation. One of the typical measuring instruments used for the monitoring of bridges and buildings is the dynamic measurement system. Conventional dynamic measurement systems require considerable cabling to facilitate a direct connection between sensor and DAQ logger. For this reason, a method of measuring structural responses from a remote distance without the mounted sensors is needed. In terms of non-contact methods that are applicable to dynamic response measurement, the methods using the doppler effect of a laser or a GPS are commonly used. However, such methods could not be generally applied to bridge structures because of their costs and inaccuracies. Alternatively, a method using a visual image can be economical as well as feasible for measuring vibration signals of inaccessible bridge structures and extracting their dynamic characteristics. Many studies have been conducted using camera visual signals instead of conventional mounted sensors. However, these studies have been focused on measuring displacement response by an image processing technique after recording a position of the target mounted on the structure, in which the number of measurement targets may be limited. Therefore, in this study, a model experiment was carried out to verify the measurement algorithm for measuring multi-point displacement responses by using a DIC (Digital Image Correlation) technique.
Integral and rigid frame bridges have advantages in bridge maintenance and structural efficiency by eliminating expansion joints and bridge supports. However, the detail of typical girder-abutment connection is rather complex and increases construction cost depending on construction detail. For the purpose of compensating disadvantages such as complexity and additional cost, a new type of bridge is proposed in this study, which improves the efficiency of construction by simplifying the construction detail of girder-abutment connection. The proposed bridge has the connection detail of steel girder and abutment integrated by prestressed PS bar installed in the connection. In this study, finite element analysis and fatigue load test are conducted to evaluate the fatigue capacity of the proposed girder-abutment connection. The results of the finite element analysis revealed that the possibility of the fatigue damage in the girder-abutment connection is very low. The results of the fatigue load test verified that the integrity of the girder and abutment connection is maintained after 2,000,000 cycles of fatigue loading.

This paper presents LRB-based hybrid base isolation systems employing additional active/semiactive control devices for mitigating earthquake-induced vibration of a cable-stayed 29 bridge. Hybrid base isolation systems could improve the control performance compared with the passive type-base isolation system such as LRB-installed bridge system due to multiple control devices are operating. In this paper, the additional response reduction by the two typical additional control devices, such as active type hydraulic actuators controlled by LQG algorithm and semiactive-type magnetorheological dampers controlled by clipped-optimal algorithm, have been evaluated bypreliminarily investigating the slightly modified version of the ASCE phase I benchmark cable-stayed bridge problem (i.e., the installation of LRBs to the nominal cable-stayed bridge model of the problem). It shows from the numerical simulation results that all the LRB based hybrid seismic isolation systems considered are quite effective to mitigate the structural responses. In addition, the numerical results demonstrate that the LRB based hybrid seismic isolation systems employing MR dampers have the robustness to some degree of the stiffness uncertainty of in the structure, whereas the hybrid system employing hydraulic actuators does not. Therefore, the feasibility of the hybrid base isolation systems employing semiactive additional control devices could be more appropriate in realfor full-scale civil infrastructure applications is clearly verified due to their efficacy and robustness.

In South Korea, U-type girder development was attempted as a means to increase the length of I-type girder, but due to the large self-weight according to the post-tension method, the application of rail bridges of 30m or less is typical. There are not many examples of application of pre-tension type girder. This study does not limit the post-tension method, but applies the pre-tension method to induce a reduction in self-weight and materials used due to the reduction of the cross-section. In addition, we intend to apply the on-site pre-tensioning method using the internal reaction arm of the U-type girder. The prestressed concrete U-type girder bridge is composed of a concrete deck slab and a composite section. Compared to the PSC I-type, which is an open cross-section because the cross section is closed, structural performance such as resistance and rigidity is improved, the safety of construction is increased during the manufacturing and erection stage, and the height ratio is reduced due to the reduction of its own weight. Therefore, it is possible to secure the aesthetic scenery and economical of the bridge. As a result, it is expected that efficient construction will be possible with high-quality factory-manufactured members and cast-in-place members. In this paper, the introduction of the pre-tension method on-site and the analytical performance verification of the anchoring block for tension are included.
Robert Rauschenberg's artistic career has often been regarded as having reached its culmination when the artist won the first prize at the 1964 Venice Biennale. With this victory, Rauschenberg triumphantly entered the pantheon of all-American artists and firmly secured his position in the history of American art. On the other hand, despite the artist's ongoing new experiments in his art, the seemingly precocious ripeness in his career has led the critical discourses on Rauschenberg's art to the artist's early works, most of which were done in the mid-1950s and the 1960s. The crux of Rauschenberg criticism lies not only in focusing on the artist's 50's and 60's works, but also in its large dismissal of the significance of the imagery that the artist employed in his works. As art historians Roger Cranshaw and Adrian Lewis point out, the critical discourse of Rauschenberg either focuses on the formalist concerns on the picture plane, or relies on the "culturalist" interpretation of Rauschenberg's imagery which emphasizes the artist's "Americanness." Recently, a group of art historians centered around October has applied Charles Sanders Peirce's semiotics as art historical methodology and illuminated the indexical aspects of Rauschenberg's work. The semantic inquiry into Rauschenberg's imagery has also been launched by some art historians who seek the clues in the artist's personal context. The first half of this essay will examine the previous criticism on Rauschenberg's art and the other half will discuss the artist's 1955 work Rebus, which I think intersects various critical concerns of Rauschenberg's work, and yet defies the closure of discourses in one direction. The categories of signs in the semiotics of Charles Sanders Peirce and the discourse of Jean-Francois Lyotard will be used in discussing the meanings of Rebus, not to search for the semantic readings of the work, hut to make an analogy in terms of the paradoxical structures of both the work and the theory. The definitions of rebus is as follows: Rebus 1. a representation or words or syllables by pictures of object or by symbols whose names resemble the intended words or syllables in sound; also: a riddle made up wholly or in part of such pictures or symbols. 2. a badge that suggests the name of the person to whom it belongs. Webster's Third New International Dictionary of the English Language Unabridged. Since its creation in 1955, Robert Rauschenberg's Rebus has been one of the most intriguing works in the artist's oeuvre. This monumental 'combine' painting($6feet{\times}10feet$ 10.5 inches) consists of three panels covered with fabric, paper, newspaper, and printed reproductions. On top of these, oil paints, pencil and crayon drawings connect each section into a whole. The layout of the images is overall horizontal. Starting from a torn election poster, which is partially read as "THAT REPRE," on the far left side of the painting. Rebus leads us to proceed from the left to the right, the typical direction of reading in a Western context. Along with its seemingly proper title. Rebus, the painting has triggered many art historians to seek some semantic readings of it. These art historians painstakingly reconstruct the iconography based on the artist's interviews, (auto)biography, and artistic context of his works. The interpretation of Rebus varies from a 'image-by-image' collation with a word to a more general commentary on Rauschenberg's work overall, such as a work that "bridges between art and life." Despite the title's allusion to the legitimate purpose of the painting as a decoding of the imagery into sound, Rebus, I argue, actually hinders a reading of it. By reading through Peirce to Rauschenberg, I will delve into the subtle anxiety between words and images in their works. And on this basis, I suggest Rauschenberg's strategy in playing Rebus is to hide the meaning of the imagery rather than to disclose it.
Iron (hydro)oxides in aqueous environments are primarily formed due to mining activities, and they are known to be typical colloidal particles disturbing surrounding environments. Among them, hematites are widespread in surface environments, and their behavior is controlled by diverse factors in aqueous environments. This study was conducted to elucidate the effect of environmental factors, such as ionic composition and strength, pH, and natural organic matter (NOM) on the behavior of colloidal hematite particles. In particular, two analytical methods, such as dynamic light scattering (DLS) and single-particle ICP-MS (spICP-MS), were compared to quantify and characterize the behavior of colloidal hematites. According to the variation of ionic composition and strength, the aggregation/dispersion characteristics of the hematite particles were affected as a result of the change in the thickness of the diffuse double layer as well as the total force of electrostatic repulsion and van der Walls attraction. Besides, the more dispersed the particles were, the farther away the aqueous pH was from their point of zero charge (PZC). The results indicate that the electrostatic and steric (structural) stabilization of the particles was enhanced by the functional groups of the natural organic matter, such as carboxyl and phenolic, as the NOM coated the surface of colloidal hematite particles in aqueous environments. Furthermore, such coating effects seemed to increase with decreasing molar mass of NOM. On the contrary, these stabilization (dispersion) effects of NOM were much more diminished by divalent cations such as Ca2+ than monovalent ones (Na+), and it could be attributed to the fact that the former acted as bridges much more strongly between the NOM-coated hematite particles than the latter because of the relatively larger ionic potential of the former. Consequently, it was quantitatively confirmed that the behavior of colloidal hematites in aqueous environments was significantly affected by diverse factors, such as ionic composition and strength, pH, and NOM. Among them, the NOM seemed to be the primary and dominant one controlling the behavior of hematite colloids. Meanwhile, the results of the comparative study on DLS and spICPMS suggest that the analyses combining both methods are likely to improve the effectiveness on the quantitative characterization of colloidal behavior in aqueous environments because they showed different strengths: the main advantage of the DLS method is the speed and ease of the operation, while the outstanding merit of the spICP-MS are to consider the shape of particles and the type of aggregation.
