A Report of an Unrecorded Slime Mold Isolated from a Twig of Chaenomeles sinesis in Korea (모과나무 잔가지에서 분리한 국내 미기록 점균류 Chaenomeles sinesis 보고)

  • Chaenomeles sinensis, called as Chinese quince, belongs to the family Rosaceae and is widely distributed in Korea, China, and Japan. A microorganism was isolated from part of a twig of C. sinensis that showed an abnormal appearance. The microorganism was identified as the slime mold Stemonaria longa of the division Myxomycota, which was previously unrecorded in Korea. The present study reports the morphological characteristics of the isolated fungus and a phylogenetic relationship based on the ${\beta}$-tubulin gene sequences.

Availability of Identification by RAPD of Aspergillus species from Sputum (객담에서 분리한 Aspergillus 속의 RAPD를 이용한 분자생물학적 동정의 유용성)

  • On the basis of morphological characteristics, of total 128 strains of from sputum of tuberculos inpatient were identified as A. fumigatus (61 strains), A. niger (37), A. flavus (26), A. versicolor (1), A. nidulans (1), A. clavatus (1) and Neosartorya fennelliae (1). These strains were re-identified according to recent Aspergillus classification system which is mainly based on molecular characteristics. The strains were grouped by randomly amplified polymorphic DNA (RAPD) techniques. The representative strains from each group were sequenced with partial ${\beta}$-tubulin gene and compared with those of reference strains in the Aspergillus and were identified by the sequence. The identification was confirmed by morphology examination. As the results, they are reidentified as A. fumigatus (58), A. niger (11), A. tubingensis (26), A. flavus (27), A. sydowii (3), A. nidulans (1), A. clavatus (1) and Neosatorya fennelliae (1). This is the first report of A. tubuingensis in clinical field in Korea.

First Report of Diaporthe tectonae Isolated from Soil in Korea (토양에서 분리한 Diaporthe tectonae에 대한 보고)

  • An unrecorded fungal species in Korea, Diaporthe tectonae was isolated from soil in Jeon-ju of Korea. The isolate was characterized morphologically, and a phylogenetic analysis using a combined dataset of internal transcribed spacer, ${\beta}-tubulin$, and elongation factor $1-{\alpha}$ sequences indicated its similarity to D. tectonae strains reported previously. To our knowledge, this is the first report of D. tectonae in Korea.

  • Cadmium is an important industrial and environmental pollutant and has adverse effects on cell growth and metabolism, although the mechanisms of its cellular toxicity are still unclear. This study was performed to elucidate the cytotoxic mechanism of cadmium in the viewpoint of oxidative stress and cytoskeleton alterations in WB-F344 rat liver epithelial cells. 200 $\mu\textrm{M}$ $CdCl_2$ caused a severe disassembling of microtubule and micro filament and an apparent cell retraction under an observation with fluorescence micoscope. (equation omitted)-tubulin and F-actin protein were highly thiolated at 20 min and then disappeared from 1 hour after the treatment of 200 $\mu$M CdCl$_2$in the immunoblot analysis. Intracellular GSH was decreased from 1hr to 24 hrs by 66.6 or 200 $\mu\textrm{M}$ of $CdCl_2$. Intracellular protein thiol was also decreased by 22.2, 66.6 and 200 $\mu\textrm{M}$ of $CdCl_2$ at 1 hour after its treatment. The product of lipid peroxidation (malondialdehyde) was increased from 4 hrs by 66.6 and 200$\mu\textrm{M}$ of $CdCl_2$. These data indicate that cadmium induces oxidative stress involving disassembling of microtubule and micro filament, thiolation of (equation omitted)-tubulin and actin protein, depletion of GSH and protein thiol, and increase of lipid peroxidation.

Unrecorded Fungal Species Isolated from Indoor Air in the Log Bed- and Sawdust Media-based Mushroom Cultivation Houses (원목 및 톱밥배지 버섯 재배사 내 실내 공기서 분리한 미기록 진균 보고)

  • Oak mushroom is cultivated using logs and sawdust media as substrates. In this study, fungi were isolated during a monitoring of indoor air in the oak mushroom cultivation houses located in Cheongyang-gun of Chungnam, Geoje-gun of Gyeongnam, Gumi-si of Gyeongbuk, Jangheung-gun of Jeonnam and Yeoju-si of Gyeongggi-do. Identification of the fungi based on morphology and molecular analysis of the internal transcribed spacer region and 28S rDNA, translation elongation factor translation elongation factor 1 a gene, and ${\beta}-tubulin$ gene revealed that six fungi, Cenangium acuum, Neopestalotiopsis surinamensis, Metarhizium marquandii, Periconia macrospinosa, Trichoderma petersenii, and Trichoderma paratroviride that have not been recorded previously in Korea.

  • Two new records of Trichocomaceae, namely Aspergillus allahabadii and Penicillium sizovae, were isolated in 2016 during a survey of fungal diversity in different crop fields locations in Gyeongnam, Korea. These species were identified based on morphological characters and phylogenetic analysis using internal transcribed spacer region and ${\beta}-tubulin$-encoding gene sequence data. A. allahabadii and P. sizovae have not yet been reported in Korea. Thus, this is the first report of these species in Korea, and their descriptions as well as details of their morphological characters are presented.

Characterization of Six Novel Endophytic Fungi Isolated from Leaves of Plants Inhabiting Jeju Island (제주도에 서식하는 식물 잎에서 분리된 6종의 국내 미기록 내생균 보고)

  • Endophytic fungi were isolated from the leaves of diverse plants inhabiting Jeju Island, Korea. The fungal isolates were identified through phylogenetic analyses incorporating nucleotide sequences derived from the internal transcribed spacer region, large subunit region of ribosomal DNA, and beta-tubulin gene. Our results identified six endophytic fungi previously unknown in Korea namely, Diaporthe goulteri, Diaporthe vaccini, Rhizosphaera pini, Valsa friesii, Xylaria primorskensis, and Zalerion arboricola were identified. Here, we present their cultural and morphological characteristics and phylogenetic relationship.

Three Novel Endophytic Fungal Species Isolated from Roots of Medicinal Crops in Korea (약용작물의 뿌리에서 분리된 3종의 국내 미기록 내생균)

  • We isolated endophytic fungal strains from the roots of three medicinal crops, Ligusticum chuanxiong, Angelica gigas, and Cnidium officinale, cultivated at Yeongju, Korea. The fungal strains were identified based on their morphological characteristics and molecular analyses of their internal transcribed spacer, large subunit rDNA, and beta-tubulin regions. Thereby, we identified three previously unrecorded endophytic fungal species, Dactylonectria pauciseptata, Rhizopycnis vagum, and Sistotrema sernanderi, in Korea. In this report, we describe the morphological characteristics and phylogenetic analyses of these three fungal species.

A new record of Trichocladium griseum in Korea: morphological and molecular characterization

  • A unrecorded species of Trichocladium, Trichocladium griseum, was isolated in 2017 during a survey of fungal diversity in Ulsan province, South Korea. This species was identified based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer (ITS) rDNA and ${\beta}-tubulin$ gene sequences. T. griseum has not yet been reported in South Korea. Thus, we report for the first time a new record of Trichocladium griseum in Korea, and we include the descriptions and morphological illustrations of this fungus.

  • We isolated endophytic fungi from the leaves of woody plants inhabiting the Ulleungdo Island and Yeosu in South Korea. The fungal strains were identified based on their morphological characteristics in conjunction with phylogenetic analysis, which was performed using sequences of the internal transcribed spacer, large subunit rDNA, and β-tubulin DNA. We confirmed the presence of Neofusicoccum mangiferae and Thyronectria cucurbitula, two endophytic fungal species that had not been previously recorded in Korea. In this report, we describe the morphological characteristics of these fungal strains and results of the DNA-based phylogenetic analysis.
