Epicuticular Waxes and Stomata of Adult Scale Leaves of the Chinese Juniper Juniperus chinensis

  • Leaf surface structures were investigated in the Chinese juniper Juniperus chinensis by scanning electron microscopy. Adult scale leaves were collected from the tree, air-dried at room temperature, and sputter-coated with gold without further specimen preparation. Approximately fi ve stomata were locally distributed and arranged in clusters on the leaf surface. Stomata were ovoid and ca. 40 ${\mu}m$ long. The epicuticular wax structures of J. chinensis leaves were tubules and platelets. Numerous tubules were evident on the leaf regions where stomata were found. The tubules were cylindrical, straight, and ca. 1 ${\mu}m$ in length. They almost clothed the stomatal guard cells, and occluded the slit-shaped stomatal apertures. Moreover, the wax ridges were flat crystalloids that were connected to the surface by their narrow side. They did not have distinct edges, and their width/height ratio varied. In particular, the wax ridges could be discerned on the leaf regions where stomata were not present nearby. Since the wax ridges did not have distinct edges on their margin, they were identified as platelets. Instances were noted where platelets were oriented either parallel to each other or perpendicular to the cuticle surface. These results can be used in biomimetics to design the hierarchical structures for mimicking the plant innate properties such as hydrophobicity and self-cleaning effects of the leaf surface.

Changes in Cell Proliferation During the Development of Renal Cell Tumors Induced by N-Nitrosomorpholine in Rats (흰쥐에서 신장암이 발생하는 동안 세포분열속도의 변화)

  • Sequential changes in cell proliferation during the development of epitherial kidney tumors induced in rats were investigated by autoradiographic determination of the $^3H$-thymidine-labeling index. Renal cell tumors were induced in male Sprague-Dawley rats by oral administration of N-nitrosomorpholine at the concentration of 120 mg/l in the drinking water for 7 weeks. At different times between 12 and 34 weeks after withdrawal of the carcinogen (stop model) animals were sacrificed. According to cytological criteria, neoplastic lesions were classified into clear cell, acidophilic cell, basophilic cell and oncocytic tumors. The labeling index was found to be increased in all types of preneoplastic tubules as compared to their corresponding original tubules. A much stronger elevation of cell proliferation was ocurred during the development of renal cell tumors from preneoplastic tubules. Of four tumor types, acidophilic cell tumor showed the highest labeling index while oncocytoma exhibited the lowest proliferative activity. These findings are in good accordance with the clinical observations that acidophilic cell tumors have a worse prognosis than oncocytoma. The data presented in this study suggest that the individual proliferation rates may be an objective biological marker of kidney tumor aggressiveness.

THE EFFECT OF ND : YAG LASER ON DENTINAL TUBULE SEAL AFTER ROOT END RESECTION (치근단절제시 노출된 상아세관의 밀폐효과에 대한 Nd : YAG 레이저의 효과)

  • If root and resection is done during surgical endodontic treatment, newly exposed dentinal tubules form pathways between the canal and the peripheral tissue. Nd : YAG laser was used to block this phenomenon, and its effect was studied with dye penetration and SEM techniques. 40 intact single rooted teeth were divided into 4 groups(10 each) : control group and test groups, in which retrograde cavity surface, cutting surface, retrograde cavity surface & cutting surface were treated with laser(1 watt 15pps) and finally retrograde filling with IRM was conducted. After that, they were stained with 2 % methylene blue, sectioned and evaluated by the maximum infiltration depth. And to observe surface change, they were prepared for SEM. The results were as follows ; 1. All experimental groups showed microleakage with variation in amount. 2. The 2nd group which treated both the retrograde cavity and cutting surface showed significantly less microleakage than the other groups(p<0.05). There was no significant difference between groups treated on one side only. 3. As a result of SEM observation of dentin surface, obstruction of dentinal tubules with marble shaped granules, which were different from normal dentin could be seen. Cracks could be seen also. 4. In summary of this experiment, it is thought that effort to obstruct the exposed dentinal tubules as well as retrograde cavity after root end resection is needed.

  • Renal pathways for excretion of sulfadiazine has been studied by standard clearance technique in the rabbit. 1. Large part of sulfadiazine filtered in the glomeruli is reabsorbed in the tubules, as visualized from the fact that Csd (clearance of sulfadiazine) amounts only a fraction of simultaneously measured Ccr (GFR). 2. Csd changed linearly with the rate of urine flow, whether it is increased by the duir etics or decreased by clamping u reter. 3. Csd remained unchanged until the plasma level of the Csdremained unchanged drug reached 10.0 mg%, and the amount transported in the tubules increased linearly with the increase in the load, exhibiting No maximum capacity for transport. 4. Csd was increased by 2,4-dinitrophenol which is an uncoupling agent of oxidative phosphorylation and decreased by probenecid which is on uricosuric agent. 5. During sodium bicarbonate infusion net secretion of sulfadiazine by tubules observed. All the evidences obtained in the rabbit indicated that sulfadiazine was reabsorbed by active, energy-requiring, or passive, simple diffusion, process, and secreted simultaneously by a probenecid-sensitive, active procss.

  • Sprague-Dawley rats received subcutaneous injections of gentamicin sulfate, 50 and 100mg/kg for 3, 7 and 10 days. The hematological and blood chemical values were determined. Kidneys were examined histologically and ultrastructurally. The results obtained were summarized as follows; 1. The serum values of aspartate transaminase, blood urea nitrogen and creatinine in the rats administered gentamicin, 50 and 100mg/kg/day were significantly increased than those in the control. 2. The ratio of kidney weight to body weight was significantly increased in the rats injected gentamicin, 100mg/kg for 10 days than those in the control. 3. The brush borders of proximal convoluted tubules in the kindneys received gentamicin, 50 and 100mg/kg/day were decreased or absent in periodic acid-Schiff staining. 4. The necrosis of proximal convoluted tubules was shown in the rats given gentamicin, 50 and 100mg/kg for 7 and 10 days. 5. The regeneration of the proximal tubular epithelia was observed in the rats treated gentamicin, 100mg/kg for 10 days. 6. The number and size of lysosomes were increased in the proximal convoluted tubules of the rats injected gentamicin, 50mg/kg for 7 days, many of which contained myeloid bodies.

Fumonisin $B_1$ induced renal toxicity in Sprague-Dawley rats

  • We investigated the effect of a single intravenous dose of Fumonisin $B_1(FB_1$) of rat kidney on the time sequence. Male Sprague-Dawley rats were intravenouslyin jected with FB$_1$at 1.25 mg/kg and were euthanized at 12 hrs, 1, 2, 4, and 6 days after the injection. In $FB_1$ treated rats, serum BUN and creatinine were elevated from 12 hrs. Microscopically, the initial target site was tubules of inner stripe, with mild degenerative and necrotic changes at 12 hrs, but the tubules recovered on day 4. In outer stripe, there were only a few scattered necrotic cells on day 1. These changes became more obvious over the time passed and most severe on day 4. On day 6, regeneration occurred, manifest as hypertrophic, basophilic tubular cells. The dying cells were proved to necrotic cells instead of apoptotic cells by TUNEL. Ultrastructural changes were cytoplasmic vacuole, dilated endoplasmic reticulum, swollen mitochondria, ballooned microvilli of the tubular cell in the outer stripe. These results showed that the renal tubules of outer medulla were the target to $FB_1$-induced nephrotoxicity in the rat. However, initial target was mner stnpe of medulla.

Co-Infection of Two Myxosporean Parasites - Parvicapsula anisocaudata and an Unidentified Myxosporean - in the Kidney of Cultured Olive Flounder, Paralichthys olivaceus

  • Two species of myxosporean parasites - Parvicapsula anisocaudata and an unidentified myxosporean were found in the lumina of renal tubules and the tubular epithelium, respectively, from cultured olive flounder, Paralichthys olivaceus in Korea. The latter was also seen in interstitial tissue of spleen and interrenal gland of the head kidney. Group of pseudoplasmodia of P. anisocaudata were firmly attached on the epithelium of renal tubules through pseudopodia. In the renal tubule epithelium, a group of unidentified myxosporean trophozoites, which were 2-3 times larger than intraluminal trophozoites of P. anisocaudata, was observed. The parasites being burst out into the lumen was occasionally encountered with partial break of the epithelium. Although infection of P. anisocaudata and unidentified myxosporean parasites did not induce any cellular reaction of the host, occlusion of renal tubules and rupture of renal epithelium would impact negatively on the renal functions of severely infected fish.

Dentinal Tubules Occluding Effect Using Nonthermal Atmospheric Plasma

  • Nonthermal atmospheric plasma has been studied for its many biomedical effects, such as tooth bleaching, wound healing, and coagulation. In this study, the effects of dentinal tubules occlusion were investigated using fluoride-carboxymethyl cellulose (F-CMC) gel, nano-sized hydroxyapatite (n-HA), and nonthermal atmospheric plasma. Human dentin specimens were divided to 5 groups (group C, HA, HAF, HAP, and HAFP). Group HA was treated with n-HA, group HAF was treated with n-HA after a F-CMC gel application, group HAP was treated with n-HA after a plasma treatment and group HAFP was treated with n-HA after a plasma and F-CMC gel treatment. The occlusion of dentinal tubules was investigated using scanning electron microscopy (SEM) and energy dispersive x-ray spectroscopy (EDS), which shows Ca/P ratio. In the EDS results, a higher Ca/P ratio was shown in the groups including n-HA than in the control group. The specimens of group HAP and HAFP had a higher Ca/P ratio in retentivity. In the SEM results, there was not a significant difference in the amount of times applied. Therefore, this study suggests F-CMC gel and n-HA treatment using nonthermal atmospheric plasma will be a new treatment method for decreasing hypersensitivity.

INTERFACIAL MORPHOLOGY BETWEEN DENTIN AND ADHESIVES ACCORDING TO TREATMENT OF DENTIN SURFACE OF CERVICAL ABRASION LESION (치경부 마모병소의 상아질 표면처리 방식에 따른 상아질과 접착제 간의 계면 양상)

  • In order to know the effect of dentin bonding agents on the restoration of cervical abrasion, Scotchbond Multipurpose Single Bond and Clearfil Liner Bond 2 were used in 51 teeth with abrasion lesion and normal teeth. The surface structure and dentinal tubules of acid etched dentin and resin replica were examined using scanning electron microscopy. The interfacial morphology between dentin and adhesives was investigated by confocal laser scanning microscopy. Following results were obtained. 1. The hybrid layer and resin tag of the dentin showing cross-sectional surface of dentinal tubules are thicker and longer than those of dentin showing oblique surface of dentinal tubules. 2. The sclerotic cast was frequently observed in dentinal tubule, and the cast looked like cuboidal or rhomboidal-shaped crystals clumped from outer side to inner side. 3. The formation of hybrid layer and resin tag was the most prominent in Scotchbond Multipurpose group, whereas Clearfil Liner Bond 2 group showed very poor formation. The formation of hybrid layer and resin tag in Single Bond group was less than Scotchbond Multipurpose group.

Histamine-induced dilation of gastric glandular tubules of flounder, Paralichthys olivaceus (히스타민 유도 넙치(Paralichthys olivaceus) 위선부의 확장성 내강에 대한 조직학적 관찰)

  • In order to differentiate the gastric tubular dilatation of olive flounder, Paralichthys olivaceus, from an atrophic change, a series of histological studies was carried out for normal gastric gland, naturally occurring and histamine-induced dilated glandular tubules. Normal glandular tubules were histologically quite similar to those already reported from other fish species. Naturally occurring, dilated gastric glandular tubules had the epithelial characteristics suffering from atrophy under light microscope, which was also the case in the histamine-induced gastric gland. But on semi-thin sections the area corresponding to the empty space of dilated lumina were actually shadowed with relatively bright area. Under transmission electron microscope this brightly shadowed area turned out to be an area tightly packed with abundant, slender cytoplasmic processes in both cases. These results strongly suggested that the dilative gastric glandular tubules under light microscope might be a morphological change indicating increased glandular activity rather than a pathologic atrophic change.
