Three-Dimensional Analysis of the Mesophyll Plastids Using Ultra High Voltage Electron Microscopy (초고압전자현미경에 의한 엽육세포 색소체 미세구조의 3차원적 분석)

  • Image processing by ultra high voltage electron microscopy (UHVEM) and tomography has offered major contributions to research in the field of cellular ultrastructure. Furthermore, such advancements also have enabled the improved analysis of three-dimensional cellular structures in botany. In the present study. using UHVEM and tomography, we attempted to reconstruct the three-dimensional images of plastid inclusions that probably differentiate during photosynthesis. The foliar tissues were studied Primarily with the TEM and further examined with UHVEM. The spatial relationship between tubular elements and the thylakoidal membrane and/or starch grains within plastids mainly have been investigated in CAM-performing Sedum as well as in $C_4$ Salsola species. The inclusion bodies were found to occur only in early development in the former, while they were found only in mesophyll cells in the latter. The specimens were tilted every two degrees to obtain two-dimensional images with UHVEM and subsequently comparison has been made between the two types. Digital image processing was performed on the elements of the inclusion body using tilting, tomography, and IMOD program to generate and reconstruct three-dimensional images on the cellular level. In Sedum plastids, the inclusion bodies consisted of tubular elements exhibiting about 20 nm distance between elements. However, in Salsola, plastid inclusion bodies demonstrated quite different element structure, displaying pattern, and origin relative to those of the Sedum. The inclusion bodies had an integrative relationship with the starch grains in both species.

Ovarian Structure and Oogenesis of the Spiny Top Shell, Batillus cornutus (Lightfoot, 1786) (Gastropoda: Turbinidae) (소라, Batillus cornutus의 난소구조 및 난자형성과정)

  • The ovarian structure and ultrastructural changes in germ cells during oogenesis of the spiny top shell, Batillus cornutus, have been investigated by light and electron microscopic observations. Specimens of the spiny top shell were collected from the subtidal zone of Wando, south coast of Korea. Spiny top shell was dioecious. The ovary was located on the surface of the hepatopancreas in the spiral posterior region. The ovary had greenish color in the gonadal mature season. The ovary comprised many ovarian lobules, and the ovarian lobule consisted of connective mesenchymal tissue and epithelial cells. Oogonium had a large nucleus with nucleolus of high electron density. In previtellogenic oocyte, small yolk granules of low electron density were scattered in the cytoplasm. Oocytes in the initial vitellogenic stage were connected with the ovarian lobule by egg stalk. The result of TEM observations showed that initial vitellogenic oocyte contained well-developed Golgi complex, endoplasmic reticula, tubular mitochondria and numerous yolk granules of various electron densities and sizes. The electron density, size and quantity of yolk granules which were distributed in the active vitellogenic oocyte were increased compared to the previous stage. Thickness of egg envelope in the late active vitellogenic oocyte was approximately 4.4 $[\mu}m$. Cytoplasm of ripe oocyte was filled with proteid yolk globules of high electron density and lipid yolk globules of low electron density. In this stage, the thickness of egg envelope was approximately 6.5 ${\mu}m$.

A Study on Impression Formation According to Design Elements of Wedding Dresses and Perceiver's Gender(Part I) -Emphasis on Silhouettes and Necklines of Wedding Dresses- (웨딩드레스의 디자인 요소와 지각자 성별에 따른 인상형성 연구(제1보) -실루엣과 네크라인을 중심으로-)

  • The objectives of this study were to identify the dimensional structure of female impression formation based on silhouettes and necklines of wedding dresses, and to analyze the effect of silhouettes, necklines, and perceiver's gender on impression formation. The experimental design was 3$\times$6$\times$2 (silhouettes$\times$necklines$\times$gender) factorial design by 3 independent variables. Samples were 432 males and females. Four factors derived to account for the dimensions of impression formation. These were attractiveness, neatness, feminity, and prettiness. Dome silhouette had a positive effect on impression of attractiveness. Bell silhouette increased the impression of femininity and prettiness. Tubular silhouette had a negative effect on the impressions of attractiveness, femininity, and prettiness. Neckline had a significant effect on impression of attractiveness and negative on prettiness. Sweet-heart neckline has a positive effect on impression of attractiveness and negative on prettiness. Off-shoulder neckline increased the impression of prettiness. Round neckline decreased the impression of attractiveness. On the interaction effect according to the neckline and perceiver's gender, the men perceived high-necklines to be more feminine than did women, while the women perceived off-shoulder necklines to be more feminine than did men. There was interaction effect between silhouette and neckline on prettiness. In dome silhouette, heart-shape neckline was perceived to be prettier than the others, while in bell silhouette, off-shoulder neckline was perceived as prettier than the others.

A Study on the Preference and Wearing Sensation of Spring/Summer Knitwears of Atopic Dermatitis (아토피 피부염환자의 봄.여름용 니트웨어 선호도 및 착용감 연구)

  • This study conducted a survey to find out atopic dermatitis patients' required performance and preference in design factors to develop spring/summer(S/S) knitwears preferred by atopic patients. On the platform of achieved information from statistical analysis, S/S knitwear samples for atopic patients were designed and constructed with cotton/Seacell(R) blended yarn, And subjective wearing sensation were evaluated comparing the samples and the commercial products made with cotton/chitosan blended yarn, trying to help producing more comfortable S/S knitwears for atopic patients. SPSS 12.0 program was used for frequency analysis, Chi-square analysis, ANOVA, and Duncan test. The result was as follows; For comfort of S/S knitwears, atopic patients most considered skin care property. Atopic patients preferred simple and basic knitwear in light color with thin and soft fabric such as plain knit. The preferred style was that 'appropriately fit with suitable ease'. They also preferred 'closed front'; 'boat neckline' and 'V neckline'; 'raglan sleeve' and 'set-in sleeve'. For hem trim, they preferred 'rib' structure. Male preferred simple and plain style with 'round collar' and 'tubular or rib trim", while female preferred sophisticated and characteristic style with 'set-in sleeve' with 'rib trim'. Under the circumstances with insufficient functional knitwears developed, this study tried to develop comfort S/S knitwears for atopic dermatitis patients and the developed knitwears showed as good as or better performance in wearing feeling, fitting, and appearance than the previously developed functional goods.

Combined Adenomatoid Odontogenic Tumor and Calcifying Epithelial Odontogenic Tumor in the Mandible: Case Report (하악 소구치 부위에 발생한 석회화상피성치성종양이 혼재된 선양치성종양: 증례보고)

  • Adenomatoid odontogenic tumors represent 3 to 7 percent of all odontogenic tumors. These tumors are more common in the maxilla than the mandible and usually include the anterior region. Clinically, the most common symptom is painless swelling and the tumor is associated with an unerupted tooth, typically a maxillary or mandibular cuspid. The adenomatoid odontogenic tumor appears radiographically as a unilocular radiolucency around the crown of an impacted tooth, resembling a dentigerous cyst. More often, it contains fine calcifications. Histopathologically, there is a thick wall cystic structure with a prominent intraluminal proliferation of the odontogenic epithelium. The most striking pattern is varying-sized solid nodules of spindle-shaped or cuboidal epithelial cells forming nests or rosette-like structures with minimal stromal connective tissues. Conspicuous within the cellular areas are structures of tubular or duct-like appearance. The duct-like spaces are lined with a single row of cuboidal or low columnar epithelial cells, of which the ovoid nuclei are polarized away from the luminal surface. Small foci of calcification may also be scattered throughout the tumor. These have been interpreted as abortive enamel formations. In some adenomatoid odontogenic tumors, the material has been interpreted as dentoid or cementum.

  • Constructions of buildings downtown are increasing as much as ever with a strong demand. Top-Down Method is suitable for its advantage in minimizing its disturbance to the neighborhood. Pre-founded when applied to CFT Column on-site welded is required for splicing. To complement the welded built-up square composite Column was developed. Top-down process will be pouring concrete in accordance with a step-by-step process. Thus, Pre-founded Column and cover concrete to determine the stress condition. Therefore, Concrete filled steel square tubular columns encased with precast concrete were studied. Five Centrally loaded Columns were tested to investigate the axial load carrying capacity. we analyzed the strength and behavior of CET Column by Loading conditions and concrete strength, thickness of cover concrete through structure experiments.

Histopathological Studies on the Effect of Korean Safflower (Carthamus tinctorious L.) Seed Powder on Cadmium Poisoning in Mice (홍화씨분말이 카드뮴 중독에 미치는 병리조직학적 관찰)

  • This study was undertaken to find out the effect of Korean safflower seed powder on histopathological changes of cadmium toxicity in mice. Fifty BALB/c mice were divided into a control group(A) and four experimental groups(B, C, D, E) : group A received tap water and basal diet, group B received tap water and diet supplemented with 3% Korean safflower seed powder alone, group C received basal diet and 300 $\mu\textrm{g}$/g of cadmium, group D and E received basal diet supplemented with 3% and 10% Korean safflower seed powder and 300$\mu\textrm{g}$/g of cadmium respectively. Cadmium dissolved in tap water was used, and the Korean safflower seed powder were mixed with feed. All mice were dissected on the 56th day. Histopathological changes in liver, kidney, lung, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur were observed. Group B showed no significant changes compared with the control group. But group C showed the unclearness of specific cells in liver, the loss of architecture and focal necrosis of hepatocyte, the glomerular swelling, degeneration and necrosis of convoluted tubules, desquamation and vacuolization of the greater part of the renal tubular epithelium, the marked congestion and thickness of the wall of alveolus in lung, slightly thinning of the cortical osseous tissue in femoral shaft, reduction of cancellous bone volume and marked narrowness of bone trabecular, marked thinning of epiphyseal cartilage plate and irregular arrangement of columnar structure of cartilage cells. On the other hand, Korean safflower seed powder-treated group showed a little convalescent changes and maintained their normal architectures in liner, kidney, lung, cortical osseous tissue of femoral shaft, bone trabecular of femur and epiphyseal cartilage plate of femur.

Effect of Grape Seed Proanthocyanidins on Tumor Vasculogenic Mimicry in Human Triple-negative Breast Cancer Cells

  • Vasculogenic mimicry (VM) refers to the unique ability of highly aggressive tumor cells to mimic the pattern of embryonic vasculogenesis, which was associated with invasion and metastasis. The grape seed proanthocyanidins (GSPs) had attracted much attention as a potential bioactive anti-carcinogenic agent. However, GSPs regulation of VM and its possible mechanisms in a triple-negative breast cancer cells (TNBCs) remain not clear. Therefore, we examined the effect of GSPs on VM information in HCC1937 cell model. In this study, we identified the VM structure via the three-dimensional (3D) matrix in vitro. Cell viability was measured using the CCK8 assay. The effects of GSPs on human triple-negative breast cancer cells (TNBCs) HCC1937 in terms of related proteins of VM information were determined using western blot analysis. In vitro, the tubular networks were found in highly invasive HCC1937 cells but not in the non-invasive MCF-7 cells when plated on matrigel. The number of vascular channels was significantly reduced when cells were exposed in GSPs ($100{\mu}g$/ml) and GSPs ($200{\mu}g/mL$) groups (all p<0.001). Furthermore, we found that treatment with GSPs promoted transition of the mesenchymal state to the epithelial state in HCC1937 cells as well as reducing the expression of Twist1 protein, a master EMT regulator.GSPs has the ability to inhibit VM information by the suppression of Twist1 protein that could be related to the reversal of epithelial-to-mesenchymal (EMT) process. It is firstly concluded that GSPs may be an p otential anti-VM botanical agent for human TNBCs.

Reproductive Biology of the Slimy, Leiognathus nuchalis(Teleostei: Leiognathidae) (주둥치 Leiognathus nuchalis의 생식생물학적 연구)

  • Reproductive biology of the slimy, Leiognathus nuchalis was investigated by means of histological methods. Sex ratio was 1.81 : 1 in female to male. Developmental pattern of oocytes was group-synchronous. Testicular structure was restricted spermatogonial testis-type of tubular testis. The size of first group maturity is 7.5 cm in total length. Gonadosomatic index(GSI) of female was the highest in July(12.83) and the lowest in September(1.08). GSI of male was the highest in June(19.0) and the lowest in October(0.24). Hepatosomatic index(HSI) of female showed to be positively correlated with GSI. Thoracic spot index(TSI) showed to be the minimum value from May to July when the maturation and ripe season of gonad. Reproductive cycle of female could be classified into the growing(March~May), maturation(May~June), ripe and spent(June~August), recovery(August~November), and resting stage(November~March). Reproductive cycle of male could be classified into the multiplicative and growing(January~April), maturation(April~May), ripe and spent(June~August), recovery(August~October), and resting stage(October~December).

A comparative evaluation of $CO_2$ and erbium-doped yttrium aluminium garnet laser therapy in the management of dentin hypersensitivity and assessment of mineral content

  • Purpose: Dentin hypersensitivity is a potential threat to oral health. Laser irradiation may provide reliable and reproducible treatment but remains controversial. The present study aimed to evaluate the effects of $CO_2$ or erbium-doped yttrium aluminium garnet (Er:YAG) laser therapy, and to assess mineral content. Methods: Eighteen human single-rooted teeth affected with advanced periodontitis were obtained. Buccal and lingual surfaces were planed to form 36 specimens. Ethylenediaminetetraacetic acid gel (24%) was applied to remove the smear layer and simulate hypersensitive teeth. The experimental groups were: group 1, control (no irradiation); group 2, $CO_2$ laser (repetitive pulsed mode, 2 W, $2.7J/cm^2$); and group 3, Er:YAG laser (slight contact mode, 40 mJ/pulse and 10 Hz). To evaluate dentinal tubule occlusion, six specimens per group (2-mm thickness) were prepared and observed using scanning electron microscopy (SEM) for calculation of the occlusion percentage. To evaluate the mineral content, six specimens per group (0.6-mm thickness) were used, and then the levels of Ca, K, Mg, Na, and P were measured by inductively coupled plasma-atomic emission spectrometry. In addition, the surface temperature of the specimens during laser irradiation was analyzed by a thermograph. Results: The SEM photomicrographs indicated melted areas around exposed dentinal tubules and a significantly greater percentage of tubular occlusion in the $CO_2$ and Er:YAG laser groups than the control, and in the Er:YAG group than the $CO_2$ laser group. In addition, no significant differences were noted among the experimental groups for the mineral elements analyzed. The $CO_2$ laser group showed an evident thermal effect compared to the Er:YAG group. Conclusions: $CO_2$ and Er:YAG laser are effective in treating dentin hypersensitivity and reducing its symptoms. However, the Er:YAG laser has a more significant effect; thus, it may constitute a useful conditioning item. Furthermore, neither $CO_2$ nor Er:YAG lasers affected the compositional structure of the mineral content.
