Effects of Pinelliae Tuber on the Surgical Induction of Endometriosis in the Rat (반하(半夏)가 자궁내막증 유발 흰쥐에 미치는 영향)

  • Purpose: Endometriosis has been thought to be related with blood stasis in uterus. Pinelliae Tuber is a herb which has effectiveness of dispelling phlegm which can be transformed into blood stasis, so is used in gynecology blood stasis-desease. Therefore in the present study, the effects of Pinelliae Tuber on endometriosis were investigated. Also retention enema therapy acts on the limited part directly, so is used frequently in gynecology clinic. This study was to examine the comparative effects on the surgical induction of endometriosis in the rat between orally and rectally administered with Pinelliae Tuber extract. Methods: The endometrial tissue was autografted to Rat's small intestine. Rats with surgically induced endometriosis were orally and rectally administerd with Pinelliae Tuber extract for 40 days. Size of ectopic uterine implants at the serosal wall and concentration of progesterone, estradiol, $TNF-{\alpha}$ and IL-2, IL-4, IL-6, IL-I0 in serum were examined and compared with the control group. Results: There was no difference in the size of ectopic uterine implants of experimental group compared with control group. The concentration of IL-10 level was significantly decreased in orally administered experimental group and the concentration of progesterone, IL-6 level was significantly decreased in rectally administered experimental group. There was no significant difference between orally and rectally administerd with Pinelliae Tuber. Conclusion: Judging from the above results, it can be suggested that Pinelliae Tuber should be not a useful agent for inhibiting the proliferation of uterine endometrial tissue. There is no difference between orally and rectally administerd with Pinelliae Tuber extract.

  • The experiment was carried out to study the bothanical characteristics of Aconitum charmichaeli Sieb. et Zucc in Korea.The results obtained are summarized as follows ; maximum flowering date was 15 Sep. and plantheight was 65cm, stem diameter was 8.7mm, No. of node was 25 and node of flowering initiation was15th.In flowering, No. of anothers were 52 and No. of flowers were 49.In leaf, length and width was same size, lower leaf size was 2-3 times than upper leaf, lower leafarea was 8.4 times than upper leaf.In tuber, No. of tuber per plant was 8, fresh weight per tuber was 25.4g, total fresh weight per plantwas about 200g.

Development of Recirculating Wick Hydroponic Techniques for Safe Seed Tuber Multiplication of Potatoe (심지 양액재배법에 의한 상위급 씨감자의 안정적 대량생산)

  • This study was conducted to establish the proper techniques of the recirculating wick hydroponics for safe seed tuber proliferation of potatoes (Solanum tuberosum L. CV. Dejima). To achieve these, several intact tubers (5, 10 and 20 g) and cut seed-pieces (two or four) were treated in wick hydroponic system beds. A polystyrene box (31cm in width, 20cm in height, 51cm in length, and $0.031m^3$ in volume) was placed on a styrofoam hox. Eight wicks $(width\;1.5cm\;{\times}\;length\;40cm) $ were put into each polystyrene hox and the boxes were filled with perlite + peatmoss (1 : 2, v/v) medium. Top fresh weight per plant increased with increasing the tuber size from 10 to 30g/tuber. As the tuber size increases from 10 to 30 g/tuher in case of uncut tuber, the marketable tuber (>5g) production per plant increased from 83.8 to 141.8 g/plant and the marketable tuber (>5g) production per plant of cut tuber was slightly higher than that of uncut treatment. Total tuber yield ranged from 4.16 to $6.56kg/m^2$ and the percentage of seed tubers was greater than $97\%$ for all treatments. These results indicate that seed tuber should be cut to produce large tuber $(\geq10g)$ in the recirculating wick hydroponic system.

Effect of harvesting time and night temperature on tuber production of calla (Zantedeschia) (수확시기와 야간온도가 유색칼라(Zantedeschia)의 구근생산에 미치는 영향)

  • This experiment was carried out to determine optimal culture conditions for the production of tubers of Calla (Zantedeschia elliottiana 'Golden Affair' and 'Black Magic') in Korea highland. In vitro produced plantlets and tuberlets of Calla 'Golden Affair' and 'Black Magic' were planted plastic film greenhouse and grown for 100, 120, 140 days, with different night temperature treatments ($0{\sim}10^{\circ}C$ : no heating, 10, $15^{\circ}C$). In both cultivars, tuber size(tuber diameter, tuber height) and tuber weight increased with increasing cultivation period when the night temperature was maintained at $10^{\circ}C$. The largest tuber diameter in vitro produced plantlets was 5.8cm in 'Black Magic' and 3.2cm in 'Golden Affair', and daily tuber growth rate was 1.110g in 'Black Magic' and 0.092g in 'Golden Affair' under the culture conditions. Consequently we think that tuber harvest date was Oct. 30 and night temperature was $10^{\circ}C$ and no heating that was proper method of tuber production. However we had selection of $10^{\circ}C$ treatment for tuber production because it appeared freezing damage occasionally in highland late in October.

Growth and Tkber Development of 'Black Magic' Calla Lily as Affected by the Rain Shelter and Tuber Size (유색칼라 생육 및 구근 비대에 미치는 비가림 재배와 구근 크기의 효과)

  • Growth and tuber development of ‘Black Magic’calla lily as affected by the rain shelter with 50% shading and the tuber size were investigated. Tubers of five grades in size were cultivated in the rain shelter or in the open field (control). Days to emergence of shoots in the rain shelter was accelerated by 4.2 days as compared to the open field. Emergence ratio was higher as the tuber was larger, tubers grown in the rain shelter showed the significance in the growth characteristics as compared to the control. There were no significant differences in the number of flowers per tuber between the rain shelter and the open field. Flower quality was 12.2 cm longer than that in the open field. flowering characteristics was improved with the increasing tuber size. Low infection of soft rot disease of 3∼22% was found in the rain shelter as compared to the that of 19∼83% in the open field. Thus tubers produced under the rain shelter showed improved quality, Even if the small tubers with diameters of 0.5∼1.0 cm were cultivated in the rain shelter, the weight and diameters of tuber harvested after approximately 7 months were 50.2 g and 5.7 cm, respectively.

Sequential sampling method for monitoring potato tuber moths (Phthorimaea operculella) in potato fields

  • An effective sampling method is necessary to monitor potato tuber moths (Phthorimaea operculella) because they are the biggest concern in potato-cultivating areas. In this study, a sequential sampling method was developed based on the results of field surveys of potato tuber moths in South Korea. Potato tuber moths were collected in fields cultivating potatoes at six sites, and their spatial distribution was investigated using the Taylor power law. The optimal sampling size and cumulative number of potato tuber moths in traps to stop sampling were determined based on the spatial distribution pattern and mean density of the collected potato tuber moths. Finally, the developed sampling method was applied to propose a control action, and its sampling efficiency was compared with that of the traditional sampling method using a binomial distribution. The potato tuber moths tended to aggregate; the optimal number was approximately 5 - 16 traps for sampling, and the number varied with the mean density of potato tuber moths according to the sampling sites. In addition, one, two, and three sites might require the following actions: Continued sampling, control, and no control, respectively. Sampling with the binomial distribution showed the minimum sample size was 12 when considering the economic threshold level. Here, we propose an effective sampling method that can be applied for future monitoring and field surveys of potato tuber moths in South Korea.

Growth and Yield of Potato after Transplanting of Potato Plug Seedlings Grown at Different Plug Cell Size and Photoperiod (다른 플러그 셀 크기와 일장에서 생산된 감자 플러그 묘의 정식 후 생육과 수량)

  • This experiment were conducted to investigate the response of growth and yield of potato after transplanting of plug seedlings 'Superior' and 'Dejima' produced at different plug cell size (105, 162, and 288) and photoperiod (8/16, 12/12, and 16/8, day/night) for 20 days in controlled plant growth system. Growth and relative growth rate of plug seedling 'Superior' was affected by plug cell size and photoperiod at 7weeks after transplanting. Tuber weight of 'Superior' was increased as cell size and photoperiod increased. That of 'Dejima' was highest in 105 cell and different with photoperiod. At 90 days after transplanting, tuber weight ($258.9{\sim}471.9\;g/plant) of 'Superior' was high in 105 and 162 cell size and 16/8 hr photoperiod. That ($278.2{\sim}428.0\;g/plant$) of 'Dejima' was high in 105 cell size, but was not different with photoperiod. The number of tuber per plant was $2.6{\sim}6.9$ of 'Superior' and $2.2{\sim}3.6$ 'Dejima'. Tuber number per plant was not significantly different with cell size and photoperiod. The large tuber over 80 g was $32.0{\sim}50.9%$ of 'Superior' and $41.0{\sim}56.7%$ of 'Dejima'. The large tuber in 'Superior' and 'Dejima' lowered as the cell size decreased. The large tuber of 'Superior' increased as photoperiod increased, but that of 'Dejima' was not differed. As the results, the optimal plug cell size and photoperiod of potato seedling is considered to be below 162 cell and over 12 hr of photoperiod.

The Daily Dose and Uses of Pinellia Tuber, Vinegar and Eggshell of Goju-tang in Treatise on Cold Damage Diseases (《상한론(傷寒論)》고주탕(苦酒湯)의 반하(半夏)와 고주(苦酒), 계자각(鷄子殼) 용량과 용도)

  • Objectives : The purpose of this study was to clarify the daily dose and uses of Goju-tang in Treatise on Cold Damage Diseases. Methods : We compared major editions of Treatise on Cold Damage Diseases with the original text of Supplement to the Essential Prescriptions Worth a Thousand Gold and chose the most appropriate text. If none of the existing texts seemed reasonable, We compared texts item by item. When there is nothing adequate even item by item, it was newly modified. These results were compared with currently distributed Pinellia Tuber and standards of eggs. Results : Goju-tang consisted of Goju(vinegar), Pinellia Tuber and an eggshell. Generally, a daily dose was extracted at once and taken 3 times a day, but in Goju-tang, 4 pieces of Pinellia Tuber and one-third Sheong of Goju(solvent) were used, which were one third of average volume. Pinellia Tuber was used cut in small pieces. And it was boiled gradually by low heat. Goju was mainly to reduce the pungency of Pinellia Tuber by cutting raphides of calcium oxalate. The Pot preparing medicines was eggshell which was chosen by its small size, because the volume of Pinellia Tuber and Goju was one-third of average volume. Since an egg shell is consisted of protein, it didn't react with vinegar(Goju). Conclusions : The 4 pieces of Pinellia Tuber and one-third Sheong of Goju(Vinegar) in the Goju-tang were one-third of average dose, and the eggshell was Pot preparing medicines.

Overexpression of jasmonic acid carboxyl methyltransferase increases tuber yield and size in transgenic potato

  • Jasmonates control diverse plant developmental processes, such as seed germination, flower, fruit and seed development, senescence and tuberization in potato. To understand the role of methyl jasmonate (MeJA) in potato tuberization, the Arabidopsis JMT gene encoding jasmonic acid carboxyl methyltransferase was constitutively overexpressed in transgenic potato plants. Increases in tuber yield and size as well as in vitro tuberization frequency were observed in transgenic plants. These were correlated with JMT mRNA level-- the higher expression level, the higher the tuber yield and size. The levels of jasmonic acid (JA), MeJA and tuberonic acid (TA) were also higher than those in control plants. Transgenic plants also exhibited higher expression of jasmonate-responsive genes such as those for allene oxide cyclase (AOC) and proteinase inhibitor II (PINII). These results indicate that JMT overexpression induces jasmonate biosynthesis genes and thus JA and TA pools in transgenic potatoes. This results in enhanced tuber yield and size in transgenic potato plants.
