Single-node stem pieces ca. 1 cm in length containing a axillary bud were obtained from in vitro plants of potato (Solanum tuberosum L.). The influences by a position of the node and the existence of a leaf at the node were observed in the single-node culture on the 8% sucrose MS medium. The effect of CCC was also investigated for the microtuberization. The apical part node was excellent in the tuberization not to mention shoot length, fresh weight, diameter, the number of node on the in vitro culture of a single-node than the lower part. The differences in the diameter of a tuber formed in the part of the axillary bud on all treatments including the cultivation of the apical part node were not recognized. However, the fresh weight of the tuber showed high value in the tuber formed at the axillary bud of shoot apex part. At 20 days after cultivation, tuberization was promoted in the new stolen that developed from the bud of node with a leaf under SD condition of 8 hours at $20^{\circ}C$. The tuberization from axillary bud of the single-node without leaf was inhibited at high temperature of $28^{\circ}C$ regardless of daylength. Whereas, tuberization at $20^{\circ}C$ and $28^{\circ}C$ was similar without the difference under SD condition but the tuber formation ratio were low. CCC 500 mg/L promoted tuberization and the effect was also showed even under LD condition at $28^{\circ}C$. The inhibiton of tuberization under LD and high temperature condition could be solved by treatment with CCC.
Effect of various dosages of gamma ray ranging from 0 to 16 krad on wound periderm formation was investigated with aging potato tuber slices $(1cm{\times}2mm)$ under aseptic condition. Cell division was gradually inhibited with increasing dosage, and completely prevented with 16 krad treatment. Solanine content was not significantly different due to dosages. Gas chromatographic separation of 2,4-dinitrophenylhydrazone precipitate from radiation-induced carbonyl compounds in potato tubers showed that formaldehyde and acetone tended to be increased only with high dosages.
Polygoni Multiflori Radix (PMR) a dried root tuber of Polygonum multiflorum Thunberg with bioactivities in bone metabolism is one of the most famous tonic traditional medicines in China, Japan and Korea.To observe the anti-osteoporotic effect of PMR, we observed the effects on proliferation and alkaline phosphatase (ALP) activity of primary osteoblasts, bone nodule formation, pit formation of osteoclasts and osteoclastogenesis in vitro. As the result of that, although PMR extracts have no meaningful changes on the the proliferation of primary osteoblasts, significant (p < 0.01) increases of ALP acitivity of osteoblasts were dose-dependently observed in all PMR extracts treated groups tested compared to that of vehicle control. It means that it enhanced the differentiation (ALP activity) and bone nodule formation of osteoblast. In addition, PMR extracts dose-dependently inhibits the pit formation and the number of multinucleated osteoclast-like cells (OCLs), osteoclastogenesis in vitro. Therefore these results show the possibility of PMR extracts as another anti-osteoporotic agents.
The effect of flooding and soil salinity on the growth of yam (Dioscorea batatas) were studied on the experimantal site temporally established in the south-eastern part of Saemangeum Reclaimed Tidal Land (near Gwanghwal myun, Gimjae-gun, Jellabukdo, Korea). Yam seedlings planted by using aerial bulblet as alternative of sliced tubers, were grown for 20-days and transplanted in black-vinyl mulched ridges (about 20 cm in height) at 70cm interval by $20{\times}60cm$ spacing in the $4^{th}$ of May, 2010. Soil salinity was maintained at lower than 1.2 ds $m^{-1}$ during the growing period and did not result to salt injury in all plants. However, flooding injury very seriously led to plant death and plant mortality rates at $67{\pm}21$ and $82{\pm}9%$ of yam plants in the compost and no compost treatment, respectively, died by heavy flooding during the rainy summer season. The main reasons of the flooding injury included the decreased rainfall acceptable capacity (RAC) after the rising of water table and a slowdown of water infiltration rate after the formation of an impermeable soil crust in the furrow bottom with continuous and heavy downpour during the rainy summer season. The effect of compost treatment was not statistically observed because of the severe spatial difference caused by wet injury, although yam tuber yield was higher at 30 kg $10^{-1}$ in the compost treatment than in the no-compost treatment at 20 kg $10^{-1}$. However, the size of tuber ranged at 1.23 to 1.60 cm in diameter and 3.7 to 5.0 cm in length in all both treatment, which means they are still reproducible for the next cropping season. Conclusively, proper counter-flooding measure and soil salinity control critically important for successful yam production in Saemangeum Reclaimed Tidal Land.
As many crude drugs are used in the oriental medical field problems on the side effects of these drugs come to the front. To conduct delayed-type hypersensitivity we selected 29 kinds of drugs used frequently for therapeutic agents in oriental medical hospitals (Table I). The cell-mediated immune response was evaluated by measuring the foot pad swelling reaction and humoral immune response by measuring the antibody formation to these crude drugs. Mice were given these drugs intraperioneally for sensitization and challenged with same drug as used for sensitization respectively by intral dermal injection on the left and righ hind foot pad 4 days after senstization and then the foot pads were measured with the dial micrometer. The results were as follow; 1) Gentianae Scabrae Radix, Arecae Semen, Corydalis Tuber, and Paeoniae Radix were significant as delayed-type hypersensitivity inducers. 2) None of the crude drugs tested had effect on the induction of humoral immune response.

The bleaching effect of potato lipoxygenase Isoenzymes on ${\beta}-carotene$ was studied. Two lipoxygenase Isoenzymes(LOX-1, LOX-2) from potato tuber were purified by CM-cellulose, DEAE-cellulose ion exchange chromatography. LOX-1 and LOX-2 seemed to have bleaching effect on ${\beta}-carotene$ in the presence of linoleic acid, which the decrease in the formation of conjugated dienes. LOX-2 was founded to have a greater pigment bleaching activity than that of LOX-1.
This study was carried out to investigate the expression patterns of foreign gene that controlled by tuber-specific patatin promoter in transgenic potatoes. Potato leaf disc cultured in vitro were transformed by the Agrobacterium strain LBA4404 containing pBl121 or pATGUS from potato cv. Desiree. In order to select the transgenic lines, gene-specific primers deduced from the NPTII were synthesized and used for polymerase chain reaction. The down part of the putative transgenic potatoes was transplanted weekly onto sucrose-enriched medium to accelerate the microtuber formation. RNA gel blot analysis was performed on the total RNAs obtained from tuber that had been harvested at a week interval. Also, histochemical assay was observed in the explants transformed with either pBI121 or pATGUS. Results showed that the transgenic plant containing pATGUS expressed GUS transcripts mainly at the tuber, not in stem, with the highest expression level in 5 weeks-grown microtubers. In contrast to pATGUS plants, the transformed plants with pBI121 showed an equal expression pattern throughout the whole developing stages. Consistent with RNA gel blot analysis, histochemical GUS staining and enzyme activity exhibited pATGUS transcripts were at the highest level in 5 weeks cultures. From these results, we suggest that the best stage to analyze the foreign gene introduced by patatin promoter into potato plants is at 5 weeks cultures after tuber formation.
Herbicide mixtures, butachlor + naproanilide, CG113 + naproanilide and benthiocarb + naproanilide were tested in order to control Sagittaria pygmaea Miq., one of the most serious perennial weeds in paddy field of Honam area. 92% of tuber of Sagittaria pygmaea Miq. was distributed within 9cm from the soil surface and number of emerged Sagittaria pygmaea Miq. increased until 40 days after rice transplanting but decreased thereafter. Number and dry weight of survived Sagittaria pygmaea Miq. were much less in butachlor + naproanilide, CG 113 + naproanilide and benthiocarb + naproanilide treatments than perfluidone and it was controlled by up to 95% by mixture treatments. Number of rhizome of Sagittaria pygmaea Miq. was decreased by all herbicide treatments and especially mixture treatment significantly reduced rhizome formation compared to perfluidone and 2.4 -D treatments. CG113 + naproanilide treatment caused phytotoxicity that tip of leaf sticked to leaf sheath curvedly in Indica ${\times}$ Japonica rice, Iri 358, but it was recovered within 10 days after herbicide treatment.
Epicoccosorus nematosporus was detected in all 22 counties surveyed, but the frequency of occurrence of fingerprint stem blight disease (FSBD) on Eleocharis kuroguwai caused by the fungus varied with fields and regions. The disease occurred more frequently in mountain areas than in plain areas. E. nematosporus was detected in less than 20% of plain areas such as Cimjae and Milyang, whereas, it occurred in 40-60% of mountain areas such as Sangiu and Jangsoo. In Milyang, mean temperature in July to August 1993 ranged from 17 to $27^{\circ}C$ with 14.3 h of dew period. Meanwhile, in the mountain area such as Sangiu, Gyeongbook, temperature ranged from 17 to $24^{\circ}C$ with 16.1 h of dew period. Plant mortality was 61% in Milyang and 82% in Sangju. Underground tuber formation was highly suppressed at 16 and 33 tubers per plot in Sangju and Milyang, respectively. In 1992 and 1995, plants infected ranged from 40 to 78% in July to September. This sharply decreased to an average of 15% in 1999.

Theobroxide, a novel compound isolated from a fungus Lasiodiplodia theobromae, stimulates potato tuber formation and induces flowering of morning glory by initiating the jasmonic acid synthesis pathway. To elucidate the effect of theobroxide on pathogen resistance in plants, Nicotiana benthamiana plants treated with theobroxide were immediately infiltrated with Pseudomonas syringae pv. tabaci. Exogenous application of theobroxide inhibited development of lesion symptoms, and growth of the bacterial cells was significantly retarded. Semiquantitative RT-PCRs using the primers of 18 defense-related genes were performed to investigate the molecular mechanisms of resistance. Among the genes, the theobroxide treatment increased the expression of patho-genesis-related protein 1a (PR1a), pathogenesis-related protein 1b (PR1b), glutathione S-transferase (GST), allen oxide cyclase (AOC), and lipoxyganase (LOX). All these data strongly indicate that theobroxide treatment inhibits disease development by faster induction of defense responses, which can be possible by the induction of defense-related genes including PR1a, PR1b, and GST triggered by the elevated jasmonic acid.
