Plantlet Regeneration by Tissue Cultures of Cyclamen persicum Mill. (시클라멘 (Cyclamen persicum Mill.)의 조직배양에 의한 식물체 재분화)

  • This study was conducted to examine the effects of explant sources and plant growth regulators on mass propagation of Cyclamen persicum. Tuber, cotyledon, and petiole tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins and cytokinins. Shoots were not induced from calli on cotyledon and petiole explants cultured on MS medium containing various concentrations of 2,4-D or NAA. However, multiple shoots were formed directly from tuber explants cultured on the medium containing 0.5 and 1.0 mg/L 2,4-D or NAA. In MS medium with cytokinin alone, TDZ was more effective in shoot formation than BA or kinetin in all explants. The combinations of NAA and BA was found to be most effective in shoot formation from tuber, cotyledon and petiole explants. Especially, shoots were formed from all the tuber explants on the medium containing 0.5 mg/L of NAA and BA. Hormonal effects on root formation were examined by subculturing single shoots on MS medium containing NAA or IBA. The medium with 0.5 mg/L IBA was most effective in root induction and subsequent plantlet regeneration.

  • Hairy root induced by A. rhizogenes from potato tuber (Solanm tuberosum L.) synthesized the agropine and mannopine which were demonstrated with paper electrophoresis. And the starch contents in hairy root were increased gradually following the developmental stage. But protein contents were decreased. The activity of ${\beta}-glucan$ synthetase II(GSII) which is related to the cell wall biosynthesis was stimulated in hairy root on the developmental stage. And chloropromazine did not influence the activity of GS II while verapamil inhibited about 60% of the activity GS II. Therefore, these results showed $Ca^{2+}$ to be effective factor in the cell wall formation. Isozyme pattern of peroxidase was investigated in the callus and hairy root induced from potato tuber.

  • Perennial weeds which were grown in transplanted paddy field formed their tubers under shortday condition. Under short day treatment, the induction periods for tuber formation were different depending upon the weed species. 80% cut tubers of the weeds did not loose their developing ability perfectly. Emergence also affected by soil conditions and depths covered.

  • This experiment was conducted to understand the environmental factors affecting growth and tuber formation such as temperature, day length, tight intensity, water condition and cutting time of Eleocharis kuroguwai Ohwi. Plant height, shoot number and dry weight of E. kuroguwai were higher at high temperature, 25/$25^{\circ}C$ (day/night), while nitrogen content was higher at low temperature, 20/$15^{\circ}C$. Plant height was more affected by water temperature, while shoot number and dry weight were more affected by air temperature. Contents and absorption of nitrogen, phosphorus, and potassium in top parts of E. kuroguwai were higher under greater difference between air and water temperatures, i.e., 18/$28^{\circ}C$ and 28/$18^{\circ}C$. The number and weight of tubers were increased under greater difference between air and water temperatures, i.e, 18/$28^{\circ}C$ and 28/$18^{\circ}C$, while they were inhibited at low or high air/water temperatures (18/$18^{\circ}C$ or 28/$28^{\circ}C$). Tubers of E. kuroguwai were formed at 8-or 12-hour day length, however, no tuber was formed at l6-hour day length. Photoinductive period for tuber initiation of E. kuroguwai was between 30 and 45 days after emergence, and the induction period of short-day treatment was less than 10 days. Tuber number and weight were reduced by shading due to inhibition of the growth of top and underground parts. Number of days from planting to tuber initiation was shortned as planting time was delayed and plant height, dry weight, and tuber number were also reduced by delayed planting. Tuber number at l0 to 15cm water depth was decreased 63 to 75% as compared with 1 to 5cm water depth. Tuber number and dry weight were not affected by the size of tubers at planting. Due to the reduced growth of top and underground parts, tuber number and dry weight of E. kuroguwai were decreased by delayed shoot cutting. The critical cutting time to inhibit the growth of E. kuroguwai was about 70 days after emergence.

  • Protopasts were isolated from cultured cells of potato (Solanum tuberosum L.) tuber tissue. The ability of callus formation from the culture cells was higher in cultivars Dejima and Superior than in Shimabara and Irish Cobbler on Lam's medium. Therefore, the former was used as sources for protoplast isolation. Friable calli were transferred to liquid media and cells in exponential phase were used for protoplast isolation. In both of Dejima and Superior, the yield of protoplasts was high in the enzyme solution of 2% Onozuka cellulase and 1% macerozyme. Also, viability of isolated protoplasts was very good. Thus, it seems that these protoplasts would be applicable to various aims of research.

A Case Report of Imports Morphological Variation of Pinelliae Tuber Based on the Genetic Analysis (유전자 분석 기반 수입산 형태 변이 반하 유통 사례 보고)

  • Objectives : The purpose of this study is to report that applying the genetic discrimination method to Pinelliae Tuber is suitable as a countermeasure for the limitations of morphological identification announced publicly in the Ministry of Food and Drug Safety(MFDS). Methods : Randomly selected fifty samples in Pinelliae Tuber imported from China were used for morphological and genetic identification. The morphological identification was applied method announced publicly by the MFDS. The traits of morphological identification were classified as Pinellia ternata, P. tripartita, Pinellia pedatisecta, and Typhonium flagelliforme, according to the formation of tuberous root and tuber morphology. The genetic identifications were conducted by Sequence Characterized Amplified Region(SCAR) marker and DNA barcoding analysis for cross-validation, respectively. SCAR marker was verified according to the presence or absence of amplicon through PCR amplification using species-specific primers. DNA barcoding analysis used sequence information of the matK region. Results : As a result of the morphological identification, 27 out of 50 samples were identified as original species 'P. ternata' of genuine 'Pinelliae Tuber', and 23 were identified as adulterant species 'P. pedatisecta'. Unlike this, the genetic identification was identified as the original species 'P. ternata' in all 50 samples in the SCAR marker and matK regional sequence analysis. Conclusions : Pinelliae Tuber of morphological mutant that can not be classified by morphological identification is imported from China. The SCAR marker would be used as accurate and efficient assays for species identification of the morphological mutant.

Effects of Growth Regulators on the Tuberization of Perennial Weed, Cyperus serotinus Rottb. (생장조절제(生長調節劑)가 다년생잡초(多年生雜草) 너도방동사니(Cyperus serotinus Rottb.) 괴경형성(塊莖形成)에 미치는 영향(影響))

  • This study was conducted to determine the possible role of growth regulators and the effect of foliar part cutting on the tuber formation of C. serotinus. Foliar application of 6-benzylaminopurine (BA) increased tuber number and fresh weight of C. serotinus by an average of 24.7% and 32.4%, respectively, as compared with the untreated control. However, the tuberization of C. seratinus was inhibited by single treatment of abscisic acid (ABA) and gibberellic acid (GA) as the concentrations increased. As compared with the untreated control, BA at $10^{-4}$M and combination of BA $10^{-5}$ + GA $10^{-5}$M increased 31.8% and 53.7% of tuber number, respectively, but GA at $10^{-5}$M decreased tuber number by 21.9%. 15㎏/10a of nitrogen under three levels of BA resulted in the highest number of tuber formation of C. serotinus, an increase of 24.7% as compared with the untreated control. GA treated increased flowering of C. serotinus by an average of 4.8% based on the untreated control. Tuber number and fresh weight of C. serotinus was markedly inhibited as the number of cutting times increased. Three times cutting inhibited tuber number and fresh weight of C. serotinus by 76.1% and 96.6%, respectively, under nitrogen of 15kg per 10a as compared with the untreated control.

Inhibition of Cell Proliferation and Induction of Apoptosis by Methanolic Extract of Aconiti Tuber in Human Renal Cell Carcinoma Cells (부자의 메탄올 추출물에 의한 신세포암의 증식억제 및 세포사멸 유도)

  • Aconiti Tuber is a traditional medicinal plant generally used in Oriental medicine therapy. In this study, we investigated the biochemical mechanisms of anti-proliferative effects by the methanol extract of Aconiti tuber (MEBJ) in Caki-1 human renal cell carcinoma cells. It was found that MEBJ could inhibit, in a dose-dependent manner, cell growth which was associated with apoptotic cell death such as formation of apoptotic bodies, DNA fragmentation and increased populations of apoptotic-sub G1 phase. Apoptosis of Caki-1 cells by MEBJ was associated with an up-regulation of pro-apoptotic Bax expression, and a down-regulation of anti-apoptotic Bcl-2 in a dose-dependent manner; however, the levels of IAP family were not affected. MEBJ treatment also induced the proteolytic activation of caspase-3 and -8, and a inhibition of poly(ADP-ribose) polymerase (PARP) and $PLC{\gamma}1$ protein. Furthermore, MEBJ treatment caused a dose-dependent inhibition of iNOS and cyclooxygenase-2 (Cox-2). Though further studies will be needed to identify the active compounds that confer the anti-cancer activity of MEBJ, the present findings provide important new insights into the possible molecular mechanisms of the apoptotic activity of MEBJ in cancer cells.

Plant growth promoting rhizobacteria influence potato tuberization through enhancing lipoxygenase activity

  • Molecular insights on the role of plant growth promoting rhizobacteria (PGPR) in potato tuberization are reported in the present study. The PGPRwere isolated from the soil collected from potato fields of Highland Agricultural Research Centre, Pyeongchang, Korea and they were identified to the genus level based on the 16S rRNA sequence analysis. These PGPR were heat-killed, filtered and the filtrates were addedindividually at a concentration of $10^7\;cfu\;mL^{-1}$ in MS (Murashige and Skoog's) medium supplemented with 7% (w/v) sucrose to study their influence on in vitro potato tuberization. Tuber initiation occurred early in untreated control, while tuber growth was pronounced in case of PGPR treatments. The control explants showed tuber formation as a result of sub-apical swelling of stolons while several sessile tubers formed directly in the axils of nodal cuttings in case of PGPR treatments, which is an indication of strong induction for tuberization. Theexplants cultured on MS medium supplemented with bacterial isolate 6 (Bacillus firmus strain 40) showed highest average tuber yield (Ca. 12.56 g per treatment) after 30 days of culture, which was 3 folds increase over the untreated control. A significant increase in lipoxygenase (LOX1) mRNA expression and activity of LOX enzyme were also detected in the tubers induced on PGPR treatments as compared to untreated control. This LOX expression level correlated with increased tuber growth and tuber yield. Further studies focused on the role of bacteria cell wall components, growth regulators and signal molecules released by PGPR are under investigation to elicit clues for PGPR-mediated signal pathway controlling potato tuberization.

  • This study was conducted as part of a phased study aimed at closely examining the mutual ecosystem between creators and viewers through paying attention to the personal media environment that is in the center of rapid changes in the media industry and particularly exploring the areas of activity of the single-person or minority media creators who lead the mobile media environment that could be accessed, viewed and produced anywhere. In particular, this study aimed to not only pay attention to the situation in which the potential to expand professional YouTuber areas encounters the changing desire and demand of content service users who continue to become more evolved as time passes, but also examine the influence of the reputation of professional YouTubers on users' loyalty and subscription intention, examine the moderating effect of sincerity on the relationship between YouTubers and users, and demonstrate such relationship formation process based on concrete data. In the conclusion section, implications that can be drawn from the study results and suggestions for further studies in the future were proposed.
