Nineteen tanniferous browse plants were collected from South Africa to investigate their digestibility, gas production (GP) characteristics and methane production. Fresh samples were collected, dried in forced oven, and ground and analyzed for nutrient composition. In vitro GP and in vitro organic matter digestibility (IVOMD) were determined using rumen fluid collected, strained and anaerobically prepared. A semi-automated system was used to measure GP by incubating the sample in a shaking incubator at $39^{\circ}C$. There was significant (p<0.05) variation in chemical composition of studied browses. Crude protein (CP) content of the species ranged from 86.9 to 305.0 g/kg dry matter (DM). The neutral detergent fiber (NDF) ranged from 292.8 to 517.5 g/kg DM while acid detergent fiber (ADF) ranged from 273.3 to 495.1 g/kg DM. The ash, ether extract, non-fibrous carbohydrate, neutral detergent insoluble nitrogen, and acid detergent insoluble nitrogen and CP were negatively correlated with methane production. Methane production was positively correlated with NDF, ADF, cellulose and hemi-cellulose. Tannin decreased GP, IVOMD, total volatile fatty acid and methane production. The observed low methanogenic potential and substantial ammonia generation of some of the browses might be potentially useful as rumen manipulating agents. However, a systematic evaluation is needed to determine optimum levels of supplementation in a mixed diet in order to attain a maximal depressing effect on enteric $CH_4$ production with a minimal detrimental effect on rumen fermentation of poor quality roughage based diet.

Moringa oleifera Lam. (Moringaceae) is widely consumed in tropical and subtropical regions for their valuable nutritional and medicinal characteristics. Recently, extensive research has been conducted on leaf extracts of M. oleifera to evaluate their potential cytotoxic effects. However, with the exception of antimicrobial and antioxidant activities, little information is present on the cytotoxic activity of the essential oil obtained from M. oleifera seeds. Therefore, the present investigation was designed to investigate the potential cytotoxic activity of seed essential oil obtained from M. oleifera on HeLa, HepG2, MCF-7, CACO-2 and L929 cell lines. The different cell lines were subjected to increasing oil concentrations ranging from 0.15 to 1 mg/mL for 24h, and the cytotoxicity was assessed using MTT assay. All treated cell lines showed a significant reduction in cell viability in response to the increasing oil concentration. Moreover, the reduction depended on the cell line as well as the oil concentration applied. Additionally, HeLa cells were the most affected cells followed by HepG2, MCF-7, L929 and CACO-2, where the percentages of cell toxicity recorded were 76.1, 65.1, 59.5, 57.0 and 49.7%, respectively. Furthermore, the $IC_{50}$ values obtained for MCF-7, HeLa and HepG2 cells were 226.1, 422.8 and $751.9{\mu}g/mL$, respectively. Conclusively, the present investigation provides preliminary results which suggest that seed essential oil from M. oleifera has potent cytotoxic activities against cancer cell lines.

Downy mildew (DM), caused by several species in the Peronosclerospora and Scleropthora genera, is a major maize (Zea mays L.) disease in tropical or subtropical regions. DM is an obligate parasite species in the higher plants and spreads by oospores, wind, and mycelium in seed surface, soil, and living hosts. Owing to its geographical distribution and destructive yield reduction, DM is one of the most severe maize diseases among the maize pathogens. Positional cloning in combination with phenotyping is a general approach to identify disease resistant gene candidates in plants; however, it requires several time-consuming steps including population or fine mapping. Therefore, in the present study, we suggest a new combination strategy to improve the identification of disease resistant gene candidates. Downy mildew (DM) resistant maize was selected from five cultivars using the spreader row technique. Positional cloning and bioinformatics tools identified the DM resistant QTL marker (bnlg1702) and 47 protein coding genes annotations. Eventually, 5 DM resistant gene candidates, including bZIP34, Bak1, and Ppr, were identified by quantitative RT-PCR without fine mapping of the bnlg1702 locus. Specifically, we provided DM resistant gene candidates with our new strategy, including field selection by the spreader row technique without population preparation, the DM resistance region identification by positional cloning using bioinformatics tools, and expression level profiling by quantitative RT-PCR without fine mapping. As whole genome information is available for other crops, we propose applying our novel protocol to other crops or for other diseases with suitable adjustment.
Objectives: Erigeron bonariensis is a type of Erigeron found throughout the tropical and subtropical areas as one of the perennial plants or pioneer plants. It is known to show detoxifying, antipyretic, and anticancer effects for various cancers. However, there are no reports on the anticancer effect of E. bonariensis on oral cancer cells. In the present study, we investigated the effects of the methanol extract of Erigeron bonariensis (MEEB) on the inhibition of cell growth and induction of apoptosis in mucoepidermoid carcinoma (MEC) cell lines, including the MC3 and YD15 oral cancer cells. Methods: MC3 Cells were treated by dimethyl sulfoxide (DMSO) or methanol extracts of 20 various natural products 20 ㎍/mL for 48 hours and cell viability were analyzed as Trypan blue exclusion assay. The effects of MEEB treatment on the cell viability of MC3 and YD15 cells, for 48 h, were analyzed by Trypan blue exclusion assay. The anticancer efficacy and apoptosis of oral cancer cell lines were analyzed by western blot analysis. The statistical significance of differences between groups was analyzed by Student's two-tailed t-test. A value of P<0.05 compared to the vehicle control was considered statistically significant. Results: Among 20 different naturally derived products, MEEB significantly inhibited cell viability and increased cleaved poly [ADP-ribose] polymerase 1 (PARP) protein in the MC3 and YD15 cells in a concentration-dependent manner. Conclusions: These results suggest that MEEB can be used as a natural anticancer drug for the treatment of human oral cancer.
Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.

Acute aquatic toxicity of effluents discharged from five dyeing plants in Gyeong-gi province were evaluated to assess whether the current Korean water quality standards(KWQS) could protect aquatic life. Chemical analyses of all parameters regulated under KWQS, except for E-coli, were also carried out to determine regulation compliance of the samples. All the effluent samples were satisfied with KWQS except for the color in only one sample. In acute Daphnia magna toxicity tests, significant mortality was observed in one of five samples and EC50 was 12.1%(95% confidence interval 9.1-16.2), which was in compliance with KWQS. The result of the Microtox assay indicated that acute microbial toxicity existed in effluents from three out of five plants, two of which were in compliance with KWQS. The agreement between regulation compliance of chemical concentrations of effluent and observed toxicity from various biological toxicity tests was very poor to fair (kappa = 0.194~0.250). The data presented suggest that exposure to dyeing wastewater which were in compliance with Korean water quality standards may not be safe to aquatic biota, and multiple tropical levels should be considered in aquatic toxicity monitoring of dyeing industry.
Hanyak, Korean herbal medicine is defined as the herbs understood and explained by traditional Korean medical theories. Considering this definition, there are broad prospective of every species being consilienced and utilized as Korean herbal medicine. Most varied plant species are in tropical regions, and its of these regions posses its own particular traditional medicine. Brazil posses the most varied and abundant plant species and also characteristic traditional medicine, formed by the combination of native indians and immigrants from Africa and Europe. Brazilian traditional medicine are practices by 'garrafeiros', 'raizeiros', or 'curandeiros' and in Caatinga uses herbs in ritual ceremonies. But threatened by modernization, these knowledge may be vanished in a prompt time. Therefore we carried this research in the propose of understanding as well as preserving their traditional medical knowledge. We revised publications about the medical plants and summarized 314 species of 94 families according to repetitive references. The most cited families were COMPOSITAE, LABIATAE, LEGUMINOSAE, RUBIACEAE, SOLANACEAE, UMBELLIFERAE, VERBENACEAE. Also cited major medical efficacies which further medical uses in combinations or modification of current traditional Korean medicine should be studied. This study provides overall prospect of the plants resources of Brazil and their uses. It may serve in the consilience and understanding of varied traditional medicine by traditional Korean Medicine.

Vanda is an aerial tropical orchid native to Thailand and nitrogen (N) fertilizer is mainly used to promote its growth and quality. However, little is known about the characteristics of N absorption and assimilation in Vanda. The objective of this study was to determine the appropriate source of N for Vanda cultivation. In this experiment, shoots and roots of Vanda 'Ratchaburi Fuchs-Katsura' were sprayed weekly with 100 ml of $^{15}N$ tracer solution (1) 10 mM of $^{15}NO_3{^-}$, (2) 5 mM of $^{15}NO_3{^-}$ plus 5 mM of $NH_4{^+}$, (3) 5 mM of $NO_3{^-}$ plus 5 mM of $^{15}NH_4{^+}$ and (4) 10 mM of $^{15}NH_4{^+}$. The results indicated that plants fed with a combined N fertilizer gave the highest of $^{15}N$ use efficiency ( $^{15}NUE$) of about 21.8%, 30 days after the first feeding (DAF), compared with those fed sole sources of $^{15}NO_3{^-}$ (21.0%) and $^{15}NH_4{^+}$ (16.6%). However, a sole nitrate fertilizer or combination fertilizer did not significantly affect the total N and labelled N content. Alanine was a major amino acid found in leaves and roots at 7 DAF, whereas glutamine was mainly found in stems. At 30 DAF, tyrosine and alanine became major components in the leaves, and glutamine decreased in stems when plants were fed with a single $^{15}NH_4{^+}$ source.
Studies that relate the structure of tropical regrowth vegetation to soil properties are generally lacking in the literature. This study proposes three indices for assessing the structural regeneration of secondary forests. They are: (1) the tree diameter class, (2) the plant life form and (3) the woody/herbaceous plants ratio indices. They were applied to assess the regeneration status of forest regrowth vegetation (aged 1-10 years), derived savanna regrowth vegetation in south western Nigeria, and to secondary forests in different stages of succession in Columbia and Venezuela, Bolivia, Mexico in South and Central America and semi-arid savanna in Ethiopia and seasonal deciduous forest successional stages in India. In all the cases, the indices increased with increasing age of regrowth vegetation and hence, with increasing structural complexity of regenerating vegetation. The tree diameter class index increased from 32.1% in a 9-year secondary forest to 69.0% in an 80-year-old secondary forest in Columbia and Venezuela and from 0.4% in a 1-year fallow to 20.9% in 10-year regrowth vegetation in southwestern Nigeria. In semi-arid savanna in northern Ethiopia, the woody/herbaceous plants ratio index increased from 18.1% in a 5-year protected grazing enclosure to 75.1% in 15-year protected enclosure, relative to the status of 20-year enclosure. The indices generally had correlations of 0.6-0.90 with species richness and Simpson's/Margalef's species diversity, implying that they are appropriate measures of ecosystem development over time. The proposed indices also had strong and positive correlations with soil organic carbon and nutrients. They are therefore, significant indicators of fertility status.

Brazzein is the smallest sweet protein and was isolated from the fruit pulp of Pentadiplandra brazzeana Baillon, native to tropical Africa. From ancient times, the indigenous people used this fruit in their diet to add sweetness to their daily food. Brazzein is 500 to 2000 times sweeter than sucrose on a weight basis and 9500 times sweeter on a molar basis. This unique property has led to increasing interest in this protein. However, it is expensive and difficult to produce brazzein other than in its native growing conditions which limits its availability for use as a food additive. In this study, we report high production yields of, brazzein protein in transgenic rice plants. An ORF region encoding brazzein and driven by the $2{\times}CaMV\;35S$ promoter was introduced into rice genome (Oryza sativa Japonica) via Agrobacterium-mediated transformation. After transformation, 17 regenerated plant lines were obtained and these transgene-containing plants were confirmed by PCR analysis. In addition, the selected plant lines were analyzed by Taqman PCR and results showed that 9 T0 lines were found to have a single copy out of 17 transgenic plants. Moreover, high and genetically stable expression of brazzein was confirmed by western blot analysis. These results demonstrate that recombinant brazzein was efficiently expressed in transgenic rice plants, and that we have developed a new rice variety with a natural sweetener.
