3배체 무지개송어의 대량 생산에 대한 연구 (A Report of Triploid Rainbow Trout Production in Korea)

  • Triploid rainbow trout (Salmo gairdneri) were produced on a large scale practice by the application of moderate heat treatment in 1986. Triploid treatment of the fertilized eggs was carried out with heat shock at $27^{\circ}C$ for 10 to 14 minutes starting 10 minutes after insemination at $10^{\circ}C$. Estimated triploidy ranged from $45\%\;to\;57\%$ ana the incidence of triploidy slightly increased with the treatment time. No significant differences of growth rate were observed between triploidy and diploidy at the early stage of growth. This observation was consistent with results of other fish species and other reports on this species. This heat shock technique, expecting further improvement, should be useful in the mass production of sterile triploid rainbow trout.

  • Triploid abalone, Haliotis discus hannai, was induced by the cold $(0^{\circ}C\;and\;3^{\circ}C)$ or the heat $(35^{\circ}C\;and\;40^{\circ}C)$ shock procedure with fertilized eggs, 12 min. or 32 min. post fertilization with the various time intervals of shock duration. Fertilization rate of each experimental group was not significantly different from that of corresponding diploid control (P>0.05). However hatching rates and normality rates of triploid larvae were significantly different from those of corresponding diploid control (P <0.05). In heat shock groups at $40^{\circ}C$, fertilization rate of eggs was extremely low $(0\~2.7\%)$ and hatched larvae were not detected in these treatment groups. Incidence of triploidy was confirmed by chromosome count and the highest rates of triploid $(84.0\%)$ revealed in cold $(3^{\circ}C)$ shock with 15 min. treatment duration 12 min. after fertilization. The number of diploid chromosome was 2n: 36, and that of tiploid was 3n=54.

Genetic Identification on Natural Population of Triploid Crucian Carp, Carassius auraus in Korea (자연산 3배체 붕어 (Carassius auratus) 클론 집단에 대한 유전학적 동정)

  • Natural clonal stock of triploid crucian carp, Carassius aurahs was identified and its cytogenetic, molecular genetic and morphological traits were studied. Cytogenetic analysis of the clonal crucian carp revealed that they were natural triploidy, evidenced by 1.5-fold increases of cell size, DNA content, and chromosome number. Multi-locus DNA fingerprinting using $(GATA)_4$ probe showed that they had an identical fingerprint profile, indicating the clonal propagation of the population. External morphology and morphometric characteristics of triploid individuals were much uniform compared to those of diploids. Natural triploid crucian carp was proven to be all-female in this study.

Production of Triploid Somatic Hybrids Between Mandarin and Grapefruit through Electrofusion

  • Somatic hybrids were produced by electrofusion between embryogenic callus protoplasts of 'Syougun' mandarin and leaf protoplasts of grapefruit. Hybridity of the two plants was confirmed by leaf morphological characteristics and random amplified polymorphic DNA (RAPD) analysis. The cpDNA analysis using PCR-RFLP could not distinguish those of both parents. These plants showed normal growth and had chromosome number of 27. These unexpected triploid somatic hybrids might be derived from fused cells between diaploid protoplast of embryogenic calli and diploid protoplast of leaf, because polysomaty, a mixture of haploid cells and diploid cells was observed in the lactose medium-pretreated embryogenic calli of 'Syougun' by flow cytomehy analysis.

  • Induction of triploid cherry salmon, Oncorhynchus ma.sou was performed by the heat shock procedure. The triploids were induced by 15 and 20 min heat shock treatments after 10 min fertilization at $28^{\circ}C$. Incidences of the triploidy were $91.4\%\;and\;89.8\%$, respectively. Nucleus and erythrocyte from the induced triploids larger than those of the diploids in major axis, minor axis, major axis/minor axis, surface area and volume. The chromosome number of diploid was 2n= 66 (17 pairs of metacentrics or submetacentrics, 16 pairs of acrocentrics or telocentrics), and that of triploid was 3n=99. Satellites were observed in the short arm of the largest telocentrics, and these may be useful marker to determine the polyploidization. Diploid have outgrown to triploid, and female have also outgrown to male in the triploid group during the 22 months of observation period after hatching. During the spawning season, 22 to 26 months after hatching, diploid were observed the growth retardation because of their sexual maturation, however the triploid showed continuous growth.

Expression Pattern of Major Heat Shock Protein Genes in Diploid and Triploid Abalone Haliotis discus hannai Juveniles (이배체 및 삼배체 전복(Haliotis discus hannai) 치패에서 주요 열충격 단백질 유전자들(heat shock protein genes)의 발현 특징)

  • Basal and heat shock-induced mRNA expression patterns of major heat shock protein (HSP) genes, including those encoding heat shock protein (HSP) 90, HSP70, HSP70-12A, heat shock inducible protein 70 (HSIP70), heat shock binding protein 1 (HSPBP1), HSP60, and HSP40 were examined in the gill and hepatopancreas of 1-year-old diploid and triploid abalone Haliotis discus hannai juveniles. Under non-stimulated conditions at 19℃, triploid abalones displayed, in general, higher mRNA levels of various HSPs (HSP70, HSIP70, HSPBP1, HSP70-12A, and HSP60 in the gill and HSIP70, HSPBP1, and HSP60 in the hepatopancreas) than did communally cultured diploids. Conversely, only the hepatopancreatic expression of HSP70-12A was higher in diploids than in triploids. However, the fold changes in gene expression in response to an acute thermal challenge (elevation from 19 to 30℃) were generally greater in diploids than in triploids, such that the difference in basal expression was diminished, weakened, or even reversed after heat shock treatment. However, unlike other HSP genes, the basal expression of HSP60 (higher in 3N) was more pronounced after heat shock treatment. Collectively, the results of this study suggest that triploid abalones have different capacities for not only basal expression but also the heat-induced expression of HSPs in an HSP member-dependent manner.

  • The chromosome numbers and karyotypes were investigated in four Korean native species of the genus Hosta. The chromosome complements were diploid of 2n=60 in H. japonica var. lancifolia Nakai and H. capitata Nakai, aneuploid of 2n=59 in H. minor (Bak.) Nakai, and modified triploid of 2n=92 in H. longipes (Fr. et Sav.) Matsumura. All the species carried four sets of distinctly large chromosomes of which the chromosome types were telocentrics or subtelocentrics with $4.4{\sim}7.2\;{\mu}m$ in length. The other chromosomes were meta-, submeta, subtelo-, or telocentric types and showed gradual length degradation in the range of $1.0{\sim}3.0\;{\mu}m$. The satellites appeared vestigially in a pair or a triplet set of chromosomes which depends on the species. New chromosome number and karyotype in H. longipes were the first report in this species. The structural rearrangement was suggested to explain the modified triploid composition of 2n=92.

Triploidy Induction in Olive Flounder, Paralichthys olivaceus (3배체 넙치 (Paralichthys olivaceus)의 유도에 관한 연구)

  • Triploid fish were induced successfully in olive flounder (Paralichthys olivaceus) by cold shocking fertilized eggs 3 minutes post fertilization at $2^{\circ}C$ for 45 minutes. Percent incidence of triploid was $92.6\%$ in this treatment. Floating rate and fertilization rate of eggs were not significanlty different from that of diploid controls (P> 0.05). However, hatchability and abnormal larvae of triploids were significantly different from that of diploid controls (P< 0.05). Incidence of triploidy was confirmed by erythrocyte measurements and chromosome counts. The surface area of triploid erythrocytes and nucleus was 1.6 times larger than that of diploids. Diploids had 48 acrocentric chromosomes, while triploids had 72.

  • Triploid cherry salmon (Oncorhynchus masou) were induced by the heat shocking treatment of fertilized eggs 10 minutes post fertilization at $28^{\circ}C$ for 15 minutes. Haematological and physiological characteristics of diploid and the induced triploid fish were examined just before the spawning period. Triploids had significantly lower concentrations of circulating red blood cells than that of diploids (P<0.01). However, oxygen consumption, hematocrit, hemoglobin, RBC, and mean concentration of hemoglobin within the RBC of triploids were higher than those of diploids. The concentrations of total protein, albumin and cholesterol from triploids were also higher than those from diploids.

  • For a locus with two alleles (IA and IB), the frequencies of the alleles are represented by $$p=f(I^A)={\frac{2N_{AA}+N_{AB}}{2N}},\;q=f(I^B)={\frac{2N_{BB}+N_{AB}}{2N}}$$ where NAA, NAB and NBB are the numbers of IAIA, IAIB and IBIB respectively and N is the total number of populations. The frequencies of the genotypes expected are calculated by using p2, 2pq and q2. Choi defined the density and operator for the value of the frequency of one gene and found the adapted partial differential equation as a follow-up for the frequency of alleles and applied this adapted partial differential equation to several diploid model [1]. In this paper, we find adapted equations for the model for selection against recessive homozygotes and in case that the alley frequency changes after one generation of selection when there is no dominance. Also we consider the triploid model with three alleles IA, IB and i and determine whether six genotypes observed are in Hardy-Weinburg for equilibrium.
