Preparation of Mulberry Leaf Extract by Adding Mugwort and Pine Needle and Effects on Lipid Composition in Rats Fed High Cholesterol Diets

  • This study investigated the effects of feeding mulberry leaf extracts on lipid composition in rats fed high cholesterol diets. An initial 30-person sensory evaluation of preparations containing various concentrations of mulberry leaf extract showed that a preparation containing 9% mulberry leaf extracts was the most highly preferred. In addition, subsidiary materials of pine needle extracts and mugwort extracts were added to weaken the unpleasant smell of mulberry leaf extract A preparation containing 9% mulberry leaf extract with 3% mugwort extract and 7% pine needle extract was given highest preference scores by the 30-person panel. When comparing the functional ingredients contents of the various preparations of mulberry leaf extracts, such as GABA, DNJ and flavonoids, no significant differences were found as a result of adding subsidiary materials (pine needle and mugwort extracts). Sprague-Dawley male rats weighing l00±10g were randomly assigned to one normal diet group, and to four high cholesterol diet groups containing 1% cholesterol, to elucidate the functionality of the mulberry leaf extract The four high cholesterol diet groups were classified into: a mulberry leaf extract diet group free of subsidiary materials (EB group); a mulberry extract diet group with pine needle extracts (EP group); a mulberry leaf extract diet group with mugwort extracts (EM group); and a control group (HC group). The mulberry leaf extracts were provided as drinking water; the diet and water were fed ad libitum. Hepatic cholesterol and triglyceride levels were higher, by 279% to 475%, in the high cholesterol groups compared to the normal diet groups, but were significantly lower in the three groups supplied with mulberry leaf extracts, compared with the high cholesterol control. There were no changes in functionality of the mulberry leaf extract preparations due to the addition of subsidiary materials. In conclusion, preparations of mulberry leaf extracts were shown to improve lipid metabolism in rats fed a high cholesterol diet, by reducing hepatic and plasma triglyceride and cholesterol levels. Also human palatability of the mulberry leaf preparation was improved by adding subsidiary materials such as pine needle and mugwort extracts.

Health Status by Anthropometrie Values and Serum Biochemical Indices of Preschool Children in Day-Care Centers in Gumi (구미지역 보육시설 유아들의 신체계측과 혈액생화학적 지표에 따른 건강상태)

  • This study was conducted to investigate health status of preschool children utilizing anthropometric values and serum biochemical indices. The subjects were 1,159 boys and 1,019 girls in Gumi day-care centers aged 4 to 6 years. The mean height and weight of 4, 5 and 6 year old children were 103.0 cm and 17.1 kg, 107.2 cm and 18.3 kg and 113.4 cm and 20.6 kg, respectively. By the obesity index criteria, 5% of boys and 4% of girls were obese whereas 10% of boys and 9% of girls were underweight. By the Kaup index criteria, 7% of boys and 4% of girls were obese. By the percentage of body fat criteria, 5% of boys and girls were obese. The mean concentrations of serum total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, Atherogenic Index and serum glucose were 154.1-156.5 mg/dL, 80.1-81.9 mg/dL, 46.8-47.4 mg/dL, 90.9-92.8 mg/dL, 2.34-2.39 and 76.4-77.8 mg/dL, respectively. Serum triglyceride level, AI and body fat percentage of obese children were significantly higher than other children and were significantly lower in HDL-cholesterol level (p < 0.05). In addition there were significant correlation between anthropometric values and serum biochemical indices : Kaup index and obesity index were positively correlated to the systolic blood pressure and diastolic blood pressure in boys and girls (p < 0.01) and to the triglyceride concentration in boys (p < 0.05). These results indicate the need of developing and implementation of nutrition education program for preschool children to prevent further development of childhood obesity and to maintain normal serum lipid profiles.

The opposite correlation between calcium ion and cyclic-AMP regarding the activation of microsomal triglyceride transfer protein in rat liver

  • In this study, the effects of Ca2+ and cyclic adenosine monophosphate (cAMP) on microsomal triglyceride (TG) transfer protein (MTP) activity were investigated in rat liver. MTP activity was high when liver contained low levels of cAMP, which was induced by administration of glucose, or high levels of total Ca2+ and TG. However, MTP activity increased by high levels of Ca2+ and TG was reduced in a dose-dependent manner by treatment with dibutyryl-cAMP (db-cAMP), a cAMP analogue. Conversely, when homogenates of liver from normal rats, with low levels of total Ca2+ and high levels of cAMP, were incubated with thapsigargin, a Ca2+-inducer, MTP activity was increased in a dose-dependent manner compared to control. Therefore, our results suggest that high levels of Ca2+ cause hypertriglyceridemia through the elevation of MTP activity, as opposed to high levels of cAMP, which suppress MTP activity and inhibit hypertriglyceridemia.

A Study of Salmon Oil Type Analysis by FT-IR and Carbon Isotopes Ratio (FT-IR과 탄소동위원소 분석을 통한 연어유의 구분에 관한 연구)

  • This study analyzes the structure types of salmon oil to evaluate the purity of salmon oil products based on the 38 different types of imported salmon oil products distributed in the Republic of Korea. The major types of omega-3 foods in the salmon oil are ethyl ester (EE) and triglyceride (TG). If the salmon oil contained potential contaminants and was processed in order to remove it, EE type omega-3 fatty acids are found in concentration. This provides a good guide in assessing if products were made with EE type ingredients or re-esterified contaminated materials. The results of the FT-IR analysis showed significant difference in the C=O, C-O band positions in TG and EE. There were 19 TG type products and 19 EE type products. The analysis of carbon isotope ratio was performed on the types of TG and EE. There were different properties in the 19 TG type products. In one product, the carbon isotope ratio was -25.15 and the other 18 products showed -22.15~-23.96. The carbon isotope ratio of all 19 EE type products showed -21.91~-23.74. The results of the TLC analysis showed similar results with FR-IR. The re-esterified TG form was not detected in the TG type products, confirming that the TG type products contained natural salmon oil. This study aimed to provide the basic material in classifying the types of natural salmon oil and re-esterified salmon oil, by analyzing the pattern and proportion of FT-IR spectrum, carbon isotope ratio, and TLC.

Effects of Sedang-hwan on Experimental Diabetic Rats induced by Streptozotocin (세당환(世糖丸)이 Streptozotocin으로 유발된 흰쥐의 실험적 당뇨에 미치는 영향)

  • Objectives : We performed this study in order to investigate the effects of Sedang-hwan(世糖丸) on the diabetes mellitus. Methods : We injected a vein with 65mg/kg of streptozotocin(STZ) on the rats. And then administered Sedang-hwan; 18.7mg/kg/day to sample groups and observed the weight and glucose level, insulin, C-peptide, triglyceride, albumin, T-protein, BUN(Blood Urea Nitrogen) and creatinine in blood. Results : 1. The sample groups showed a high suppressive effect of weight loss(P=0.002). 2. The sample groups' glucose level showed a effective in lowering level(P=0.001). 3. The sample groups showed a higher insulin level than control groups. 4. The sample groups showed a higher C-peptide level than control groups(P=0.001). 5. The sample groups' triglyceride level showed a effective in lowering level. 6. The sample groups showed a higher HDL-cholesterol level than control groups(P<0.001). 7. The sample groups showed a higher T-protein level than control groups(P=0.04). 8. The sample groups showed a higher albumin level than control groups(P<0.001). 9. The sample groups' creatinine level showed a effective in lowering level(P=0.04). 10. The sample groups' BUN level showed a effective in lowering level(P=0.03). Conclusions : Conclusively, Sedang-hwan was recognized to have decrease effect of serum glucose, triglyceride, creatinine, BUN of the diabetic rats induced by streptozotocin. It was suggested that Sedang-hwan can be used for effective diabetes mellitus medical treatment. It is also required to study further about detailed principles of blood-sugar dropping effect by Sedang-hwan, and recovery by diabetes mellitus.

The Relationship between Pulse Wave Velocity (PWV) and Risk Factors of Cardiovascular Disease including Serum Lipid Values (Pulse Wave Velocity(PWV)와 혈청지질수치를 포함한 심혈관 질환 위험 요인들과의 상관관계 연구)

  • Objectives : There are lots of reports that cardiovascular disease, including hypertension, cerebro-vascular accident, and coronary heart disease, is related to atherosclerotic changes. Increased serum levels of lipids could play a role in these changes. This study aimed to investigate the relationship between PWV and risk factors of cardiovascular disease, including serum lipid values. Methods : This study included 261 subjects (49.32±11.79 years, 112 male) who underwent PWV and serum lipid evaluation. We investigated the correlation between serum lipid values, blood pressure, body mass index (EMI) and PWV. Pearson's correlation and partial correlation analysis were applied to examine the relationship between PWV and risk factors of cardiovascular disease. Results : Serum triglyceride, total cholesterol, systolic blood pressure, and diastolic blood pressure were significantly correlated with PWV. Partial correlation coefficient adjusted by age yielded significant correlation between serum triglyceride, systolic blood pressure, diastolic blood pressure and PWV. Conclusion : In this study, it seems that there are significant relationships between PWV, triglyceride and blood pressure. We could suggest that PWV might have some relationships with Dam-eum and blood stasis in oriental theory.

Effect of Capsaicin on the Body fat and Adipocyte in the Diet induced-obese Mice

  • As the obesity has been known to be related with the hyperlipidemia, cardiovascular disease, cerebral apoplexy, fatty liver, and other chronic diseases, recent researches have focused on the functional food materials and their anti-obesity activities. This study was performed to study the effects of vanilloid family capsaicin, major pungent ingredient of hot chillies and peppers, on anti-obesity activities. ICR male mice were fed one of the pellet diet, basal diet, and high fat diet with capsaicin (45 μg/day) solution for 5 days. Mice in the corresponding control groups were given water for 5 days. In results, capsaicin reduced body weights in any diet groups. Percent weight and cell size of the abdominal white adipose tissue in mice on the high fat diet with capcaicin were significantly lower compared with those in mice on the high fat diet with water. However, percent brown adipose tissue weight per body weight in mice on the high fat diet was not affected by capsaicin. Capsaicin reduced the levels of s-triglyceride and s-total cholesterol in the pellet diet or high fat diet groups. There was no difference in the s-protein levels between the capsaicin group and the control water group. These data indicate that 1) orally administered capsaicin has a reducing effect on the blood triglyceride and total cholesterol levels, and 2) capsaicin has lowering effects on the body weight, percent weight and cell size of the abdominal white adipose tissue.

Drug research and development tend to hyperlipidemia (이상지질혈증과 치료제 연구개발 경향)

  • Most of the cholesterol is synthesized by liver in the body while about one of third is taken via dietary. The main functions of cholesterol is to protect membranes in cell surface, avoid the arterial bleeding by hypertension, and prolong the life of erythrocytes, and so on. However, overload of cholesterol leads to arteriosclerosis associated with leading death cause. Lack of physical activity, emotional and environmental stress, and low intake of protein or vitamin E induce the unbalance between HDL- and LDL-cholesterol so become a basis of ischemic disorders in heart, brain and elsewhere in the body. So far, four major classes of medications for hyperlipidemia are HMG-CoA reductase inhibitors (statins), bile acid sequestrants, nicotinic acid, and fibric acids. The statins can lower LDL and levels triglyceride, but may induce myopathy and an elevation of liver enzyme levels. The bile acid sequestrants lower LDL levels and raise HDL levels with no effect on triglyceride levels but side effects of gastrointestinal (GI) distress, constipation, and a decrease in the absorption of other drugs. Nicotinic acid and fibric acids lower LDL and triglyceride levels with showing flushing, hyperglycemia, hyperuricemia, GI distress, and hepatotoxicity dyspepsia, gallstones, myopathy, and unexplained noncardiac death as adverse effects. Above western drugs lower cholesterol by 15 to 30% while all have notable adverse effects. In traditional medicine, hyperlipidemia is regarded as retention of phlegm and fluid disease. Etiology and pathogenesis of hyperlipidemia is basically based on Spleen-Deficiency and Phlegm-Stagnation, accumulation and stasis of -heat, and Qi & blood stagnation induced by Phlegm-damp, water-dampness, and blood stasis. Thereby, strengthening Spleen and dissolving Phlegm, clearing away heat and diuresis, and supplementing Qi and activating blood circulation are commonly used therapeutic methods for hyperlipidemia. The traditional herbal medicine, have been used for patients with CVA, hypertension or hyperlipidemia in Oriental hospital or Oriental clinic. The lock and key theory is used to develop most of western medicine, however many diseases are caused by mixed factors in body-complex system. We expect that Oriental pharmacological theory could be newborn as a novel drug showing high advantage of blood levels of lipidsand QOL of performance without side effects.

Production of Lentinus edodes Mycelia in Submerged Culture and It's Hypoglycemic Effect in Diabetic Rats (표고버섯 균사체의 생산과 당뇨 유발 흰쥐에 대한 혈당강하 효과)

  • The optimum conditions for the production of Lentinus edodes mycelia and it's hypoglycemic effect was studied. Optimum pH and temperature for the production of mycelia in shaken flask culture were 5 and 30C, respectively, for 24 days cultivation. Culture period for maximum production of mycelia (8.13 g/l) in 5-l jar fermenter cultivation was shortened as much as 6 days compared to shaken flask culture. The mycelial dose of 5 % proved almost equally effective in lowering the plasma glucose, total cholesterol, and triglyceride level as much as 23.0, 20.7, and 27.1%, respectively. The values of alanine transaminase and aspartate transaminase were also estimated and exhibited a substantial reduction (29.6 and 34.3%, respectively) in activity after the administration of dried mycelia.

'The effects of Chungsimgangwhawhan (淸心降火丸) on blood pressure and hyperlipidemia' (청심강화환(淸心降火丸)이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響))

  • In order to study the effects of Chungsimgangwhawhan on blood pressure in normal rats and rabbits, and hyperlipidemia induced by 1% cholesterol diet, triton WR-1339, and 75% fructose in mice and rats, the amount of total cholesterol, triglyceride, phospholipid and blood pressure were measured. The results were as follows : 1. After per os medication of Chungsimgangwhawhan, blood pressure in rats were increased but were not significant. 2. Chungsimgangwha-Whan showed strong effects on decreasing blood pressure, depending on the concentration in blood vessels of rabbits. 3. In the model of hyperlipidemia induced by 1% cholesterol diet in mice, Chungsimgangwhawhan showed decreasing effects on total cholesterol level in serum significantly (p<0.05) depending on the concentration, and on triglyceride (p<0.01) and phospholipid level (p<0.001) in serum. 4. In the model of hyperlipidemia induced by triton WR-1339 in rats, Chungsimgangwhawhan showed mild decreasing effects on total cholesterol level in serum, but was not significant. 5. In the model of hyperlipidemia induced by 75% fructose in rats, Chungsimgangwhawhan showed decreasing effects on total cholesterol and triglyceride level in serum significantly(p<0.01) depending on the concentration. According to the above results, Chungsimgangwhawhan showed significant decreasing effects on blood pressure and hyperlipidemia, and it is considered that it is appropriate to apply for hypertension and hyperlipidemia.
