Effects of Hwanggeum-tang Aqueous Extracts on Streptozotocin-induced Rat's Diabetes and Related Complications (황금탕이 Streptozotocin으로 유발된 당뇨병 백서의 혈당 및 당뇨병합병증에 미치는 영향)

  • The object of this study was to observe the effects of Hwanggeum-tang (HGT) aqueous extracts on Streptozotocin-induced rat's diabetes and related complications. Three different dosages of HGT extracts were orally administered oncea day for 28 days from 3 weeks after Streptozotocin treatment (60 mg/kg, single intraperitoneally administered). All the rats were checked at 3 weeks after Streptozotocin treatment as follows. Changes on the body weight, blood glucose level, kidney weight, serum BUN and creatinine level, liver weight, serum AST and ALT level, serum LDL, HDL, triglyceride and total cholesterol level were observed with changes on the pancreatic MDA content and GSH content. The results were compared with a potent antioxidant silymarin 100 mg/kg in which the effects on Streptozotocin-induced diabetes and related complications were already confirmed. As results of Streptozotocin-injected diabetes and related complications, dramatical decreases on the body weight, increase of the kidney and liver weight, increase of serum BUN, creatinine, AST, ALT, LDL, triglyceride, total cholesterol level and decreases of serum HDL level were detected in streptozotocin control as compared with intact control. In addition, marked increases of pancreatic MDA content and decreases of GSH content were also detected in streptozotocin control as compared with intact control. However, these diabetes and related complications, and inhibition of antioxidant effects induced by Streptozotocin were inhibited by 28 days continuous treatment of 50, 100 and 200 mg/kg of HGT extracts in the present study. HGT have favorable effects on the diabetes and various diabetic complications. Therefore, more detail mechanism studies should be conducedin future with the efficacy tests of individual herbal composition of HGT and the screening of the biological active compounds in herbs.

Effects of Liriopis Tuber Herbal-Acupuncture on Diabetes Mellitus Induced by Streptozotocin in Rat (맥문동약침액(麥門冬藥鍼液)이 Streptozotocin으로 유발된 흰쥐의 당뇨병에 미치는 영향)

  • This study was carried out to investigate the effects of Liriopis Tuber Herbal-Acupuncture on Diabetes Mellitus induced by streptozotocin in rat. Body weight, serum insulin, glucose, triglyceride, HDL cholesterol, free cholesterol and total protein were measured in streptozotocin induced diabetic rat injected with Liriopis Tuber Herbal-Acupuncture solution at Pisu(BL20) respectively for 30 days. In order to study the effects of Liriopis Tuber Herbal-Acupuncture at Pisu(BL20) on Diabetes Mellitus induced by Streptozotocin in rat, The groups were divided into five groups; Normal group(non treated group), N.-Saline group(administration of injection with normal saline at BL20 in 30 days after streptozotocin injection), Control group(non treated group after streptozotocin injection), H. Acup. group (administration of Liriopis Tuber Herbal-Acupuncture at BL20 in 30 days after streptozotocin injection), and Acup. group (administration of acupuncture at BL20 in 30 days after streptozotocin injection). The results obtained were summarized as follows; 1. As compared with Control group, there wae no significant tendency to diminish the rate of weight loss in H. - Acup. group. 2. As compared with Control group, there wae signiticant increase of serum insulin level in H. -Acup. group. 3. As compared with Control group, there was decrease of glucose level in H.- Acup. group. 4. As compared with Control group, there was decrease of triglyceride level in H. - Acup. group. 5. As compared with Control group, trere wae increase of HDL cholesterol level in H. - Acup. group. 6. As compared with Control group, there was significant increase of free cholesterol level in H. - Acup. group. 7. As compared with Control group, there was Increase of total protein level in H. - Acup. group. According to above mentioned results, Liriopis Tuber Herbal-Acupuncture was expected to be effective in treatment of Diabetes Mellitus and its complications.

Gambigyeongsinhwan(2) Reduces Blood Triglyceride Levels and Improves Visceral Fat in High Fat Diet-Fed Obese Male C57BL/6N Mice (고지방식이 마우스 비만모델에서 감비경신환(減肥輕身丸)(2)에 의한 혈중 중성지방 농도와 내장지방의 변화)

  • Objectives : We investigated the effects of gambigyeongsinhwan2(GGH(2)) on body weight and examined whether blood triglyceride levels and visceral fat are inhibited by it in high fat diet-fed obese male mice. Methods : 8 weeks old, high fat diet-fed obese male mice were divided into 5 groups: C57BL/6N normal, control, GGH(2)-1, GGH(2)-2 and GGH(2)-3. After mice were treated with GGH(2) for 8 weeks, we measured body weight gain, food intake, feeding efficiency ratio, fat weight, plasma leptin and lipid levels. We also did histological analysis for liver and fat on the mice. Results : 1. Compared with controls, GGH(2)-treated mice had lower body weight gain and adipose tissue weight, the magnitudes of which were prominent in GGH(2)-3. 2. Compared with controls, GGH(2)-treated mice had lower feeding efficiency ratio, the magnitude of which was prominent in GGH(2)-3. 3. Compared with controls, GGH(2)-treated mice had lower blood plasma triglyceride level. 4. Blood plasma AST and ALT concentrations were not changed by GGH(2), indicating GGH(2) do not show any toxic effects. 5. Consistent with their effects on body weight gain, the size of adipocytes were significantly decreased by GGH(2), whereas the adipocyte number per unit area was significantly increased, suggesting that GGH(2) decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGH(2). Conclusions : These results demonstrate that GGH(2) effectively reduces body weight gain, feeding efficiency ratio, blood plasma triglyceride level and improves abdominal fat.

Effects of Dietary P/S Ratio on Lipid Content of Plasma and Tissues in Rats (식이 지방의 P/S비율이 흰쥐의 혈장 및 조직의 지질함량에 미치는 영향)

  • This study was designed to observe the effects of dietary P/S ratio on lipid component in plasma and tissues. Changes in plasma total cholesterol and triglyceride concentration and also cholesterol and triglyceride concentrations in liver, small intestine and aorta were determined in adult rats fed experimental diets providing different dietary P/S ratios as 0.05, 0.5, 1.0, 2.0, 3.0, and 4.0, respectively. The results obtained were summarized as follows; 1. Feed efficiency ratio was not significantly different among six groups at 2nd week. But at 4th week, the higher the dietary P/S ratio the higher the feed efficiency ratio. 2. Plasma cholesterol level was getting higher as increased dietary P/S ratio at 2nd week, But that was significantly decreased as P/S ratio increased at 4th week. 3. Plasma triglyceride level was significantly decreased as increased dietary P/S ratio at 4th week. 4. Cholesterol concentrations in liver and small intestines were getting higher as increased dietary P/S ratio at 4th week, But aortas cholesterol concentration was not influenced by P/S ratio. 5. Triglyceride concentrations in liver and small intestines were significantly increased as increased dietary P/S ratio. On the contrary, triglyceride concentration in aortas was not influenced by P/S ratio.

Relationship between Eating Attitude in Female Adolescent and Psychological, Biological Factors ; Depression, Aggression, Impulsiveness and Blood Cholesterol Level (여고생 식이태도와 심리적 및 생리적 요인과의 상관관계 ; 우울감, 공격성, 충동성 및 혈중 콜레스테롤에 대하여)

  • The purpose of this study is to evaluate correlation of adolescent eating attitude and psychological factors which might influence the eating attitude such as depression, aggression and impulsiveness. The author also tried to identify relationship between adolescent eating attitude and biological factors such as serum cholesterol level and triglyceride level. Two hundred and forty one female high school students without physical disorder were selected as study subjects. For the study, Eating Attitude Test-26KA (EAT -26KA), Beck Depression Inventory (BDI) -Korean version, Barrat Impulsiveness scale (BIS) -Korean version, and Buss-DurKee Hostility Inventory (BDHI) -Korean version were used. Serum cholesterol level and triglyceride level were also investigated. For the purpose of identifying factors which could influence eating attitude of adolescence, stepwise multiple linear regression analysis was done. Total score of BDHI could be most significant factor to explain eating attitude, and BDI, serum cholesterol/triglyceride level be second, third position respectively. But, BIS was not significant predicting parameter. The contributing portion of these factors to explain eating behavior was 25.9%.

Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat (어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

A Randomized, Double-blind, Placebo-controlled Study to the efficacy and Safety of NMED-01 and NMED-02 in Mild Alcoholic Liver Subjects (NMED-01과 NMED-02의 간기능 개선에 대한 인체 효능 평가)

  • Objectives : The purpose of this study was to determine whether NMED-01 or NMED-02 improves laboratory test results in participants with liver function disorder. Methods : This is a randomized, placebo-controlled trial in which participants, treating physicians and data management staff were blinded to treatment group. The study was conducted at Semyung university oriental medicine hospital in Jecheon where participants with high level of serum ${\gamma}$-GTP (60-350 U/L) were enrolled. The intervention consisted of three times daily ingestion of either two capsules of placebo, NMED-01 (NeuMed. co. ltd., Seoul), or NMED-02 (NeuMed. co. ltd., Seoul) for twelve weeks. To evaluate the efficacy and safety of NMED-01 and NMED-02, we primarily evaluated the degree of decrement of serum ${\gamma}$-GPT level among three groups. Secondarily the decrement of serum ALT, AST, and triglyceride level in each group were also evaluated. Adverse effects were monitored during the twelve weeks treatment. Results : The change of ${\gamma}$-GTP level of NMED-01 group was lower than that of placebo group at the end of 12-week administration ($28.1{\pm}38.7U/L$ vs. $9.3{\pm}27.0U/L$, p=0.046). Other variables including AST, ALT, and triglyceride level were not significantly reduced. The decrement of ${\gamma}$-GPT, AST, ALT, and triglyceride level of NMED-02 group was not significant. There were no significant adverse effects or toxicities during treatment period. Conclusions : Participants receiving NMED-01 had improvement in laboratory test results. Despite a modest sample size, our results suggest that NMED-01 are safe and may be potentially effective in improving liver function. However, NMED-02 have lack of a detectable effect in this study.

Effect of Molokhia (Corchorus olitorius) and Its Mucilage on Cholesterol Metabolism in High Cholesterol Fed Rats (몰로키아가 고 콜레스테롤 식이를 투여한 흰쥐에서의 콜레스테롤 대사에 미치는 영향)

  • Effects of molokhia (Corchorus olitorius) and its mucilage on in vitro bile acid adsorption capacity and lipid composition using cholesterol-fed SD rats were investigated. Mucilage showed stronger affinity toward bile acid than hot water extract of molokhia powder. As the extracting temperature increased from 50 to $80^{\circ}C$, bile acid binding capacity of mucilage also increased from 41 to 83%. When molokhia powder or its mucilage added into cholesterol diet at 5 or 10% levels (as fiber source) was compared with cellulose-added group, total cholesterol and triglyceride levels of plasma showed no significant differences, whereas, HDL-cholesterol level of cellulose group significantly increased. Accumulation of cholesterol and triglyceride in liver were significantly inhibited in molokhia and mucilage groups. Molokhia and mucilage lowered the liver weights significantly. As the concentration of molokhia or mucilage increased, cholesterol and triglyceride levels in the liver was lowered. Cholesterol and triglyceride excreted through feces were significant increased in molokhia- or mucilage-fed group, with excretion of cholesterol by molokhia-fed group being mere distinct.

Effects of Cereal and Red Ginseng Flour on Blood Glucose and Lipid Level in Streptozotocin - Induced Diabetic Rats (곡물류와 홍삼분말 급여가 당뇨성 흰쥐의 혈당 및 지질대사에 미치는 영향)

  • This study was conducted to examine the effects of feeding diets containing bioorganic power (BP) flour, wheat flour and rice flour on blood glucose and lipid level in streptozotocin-induced diabetic rats. BP flour was composed with the mixture of defatted flour (16 g soybean, 12 g wheat, 8 g barley), 2 g red ginseng and 2 g whole wheat flour per 100 g diet. Experimental groups were divided into non-diabetic normal and 4 diabetic groups containing control, 40% BP, 41.6% wheat and 41.6% rice flours with basal diet. BP group was significantly increased body weight gain and decreased plasma glucose compared with the diabetic control, wheat and rice flour groups (p<0.05). Food efficiency ratio of diabetic rats were significantly lower than that of the normal rats. The concentrations of total cholesterol and triglyceride in plasma and atherogenic index were significantly decreased in BP, wheat and rice diabetic groups than diabetic control (p<0.05). The concentration of total cholesterol and triglyceride in liver was significantly lower in BP group as compared with the diabetic control, wheat and rice flour groups (p<0.05). In conclusion, the results indicated that BP flour feeding improved plasma glucose level, total cholesterol and triglyceride in diabetic rats.

  • Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.
