This study investigated the effects of conjugated linoleic acid(CLA) on the fat deposition, triglyceride levels and the expression of stearoyl-CoA desaturase 1(SCD1) in the livers of male ICR mice that were fed with either soybean oil or beef tallow supplemented with CLA. Mice weighing $25{\sim}30$ g were divided into four groups; soybean oil(SBO), and SBO supplemented with 1% CLA(SBOC), beef tallow(BT) and BT supplemented with 1% CLA(BTC). Each group consisted of 10 mice that were fed the experimental diets for 4 weeks. The experimental diets consisted of 64% carbohydrate, 20% protein, and 16% fat in terms of their contributions to total calories. All other nutrients were identical in the diets. Triglyceride measurements were completed using a kit. Fatty acid compositions were analyzed in the liver using gas chromatography. The levels of SCD1 expression were analyzed by RT-PCR in the liver. No significant differences were found for food intake level, body weight and food efficiency among the experimental groups. However, the weights of epididymal fat pads and plasma triglyceride levels were significantly lower in SBOC and BTC(p<0.05) compared to the SBO and BT groups. These effects were similar in the CLA supplemented groups. The expression level of SCD1 gene and ${\Delta}9$ desaturase index were not significantly different, regardless of the fat used for CLA supplementation. Based on these results, addition of CLA showed decreasing effects on the fat depots weight and the concentration of triglyceride regardless of the fat sources. The SCD1 gene expression and ${\Delta}9$ desaturase index were not influenced by the types of fats with respect to the CLA effects.
The purpose of this study was to investigate the effects of L-carnitine administration on lipid metabolism in streptozotocin-induced diabetes. Diabetes was induced by a single intraperitoneal injection of streptozotocin (50 mg/kg b.w.) and was confirmed by determination of urinary glucose secretion. Diabetic rats in the three L-carnitine treated groups were given L-carnitine, 50(D5O), 100(D100) and 200 (D200) mg/kg body weight, by subcutaneously every other day for four weeks, while animals in normal (N) and diabetic (DM) groups for control received saline by the same method. The daily weight gain was not different between normal and diabetic rats, but daily dietary intake was significantly higher in diabetic rats than in normal rat. Diabetic rats had a significantly lower carnitine concentration in both serum and liver compared to normal rats. Total carnitine concentration in serum was increased dose dependently upon carnitine administration, but statistic significance was shown only in D200 group. Diabetic rats had significantly higher serum triglyceride and cholesterol concentrations compared to normal rats. However there were no significant differences in liver L-carnitine administration to diabetic rats significantly decreased serum triglyceride but not cholesterol concentrations. In liver, triglyceride and cholesterol concentrations were not attired by L-carnitine administration. These results indicated that streptozotocin induced-diabetic rats have decreased carnitine and increased lipid concentrations compared with normal rats. Also it indicated that L-carnitine administration has an effect on the normalization of serum triglyceride concentrations in diabetic rats.
We investigated the effect of tea fungus/kombucha(TF) beverage on serum and liver lipid metabolism in growing male rats. The rats(Sprague-Dawley) were divided into five groups; control(TF free water), 20 or 40% TF groups(20 or 40% TF in water) and 20 or 40% TFS groups(20 or 40% TF autoclaved in water) according to the level of TF supplementation. The rats were fed ad libitum each of the experimental beverage for 7 weeks. The body weight gain of all the TF and TFS groups were similar to that of the control group. The total lipid and triglyceride concentrations of serum in the 40% TF group were increased more than those in the control group, but those in the 20% TF and 20% TFS groups were similar to that of the control group. The total cholesterol, HDL-cholesterol concentrations, HDL-cholesterol / total cholesterol ratio and atherogenic index of serum in all the TF and TFS groups were similar to those of the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group, but those of the 40% TF, 20% TFS and 40% TFS groups were similar to that of the control group. The liver weights of all the TF and TFS groups were heavier than that of the control group. The concentrations of hepatic triglyceride in the 20% TF and 20% TFS groups were significantly decreased, compared with that in the control group. The concentrations of hepatic total cholesterol in all the TF and TFS groups were significantly more decreased than that in the control group. These results indicated that the liver weights of tea fungus/kombucha beverage feeding rats were increased, also the serum triglyceride and total cholesterol concentrations of 40% TF feeding rats were increased. but the serum LDL-cholesterol, hepatic triglyceride and cholesterol levels of 20% TF feeding rats were decreased.
Effects of kimchi on lipid metabolism and immune function were studied in experiments using 63mals SD rats fed 6 inds of Baechu-kimchi containing diet during 4 weeks. Three kinds of freeze dried kimchi differ in fermentation period (not fermented, 3-, 6-week-fermented at 4$^{\circ}C$) were added at 5%, 10% of the diet containing 15% lard. The levels of serum total lipid and triglyceride and the content of liver total lipid and triglyceride of all kimchi groups were lower than those of a control group. But the levels of serum and liver cholesterol is not affected by kimchi intake. The triglyceride concentration of epididymal fat pad and feces of kimchi groups were higher than those of a control group. The food efficiency ratio, epididymal fat pad weight of 3-, 6-week-fermented kimchi 10% groups were significantly lower than control and not-fermented kimchi groups. Especially 6-week-fermented kimchi groups showed adipocytes, less in number and larger in size than those of other groups. The blastogenesis of spleen lymphocytes to LPS was higher in rats fed fermented kimchi diets than rats fed control and not-fermented kimchi diet. These results suggest that kimchi stimulates lipid mobilization to epididymal fat pad and lipid excretion via feces, so lower serum and liver triglyceride concentration. The fermented kimchi stimulate the proliferation of B cell and lower the lipid accumulation in epididymal fat pad, especially kimchi fermented for 6 weeks at 4$^{\circ}C$ lower the adipose cell number.
These studies were undertaken to investigate changes of neutral lipid content and fatty acid composition were determined. Also accumulation process of monoglyceride, diglyceride and triglyceride content, and fatty acid composition were investigated during the development. The results were summarized as follows ; Changes of lipid during development sesame seeds, glycolipid contents which showed the highest in the early ripening stage and after that rapidly decreased, and phospholipid contents showed a similar pattern as glycolipid occurred. In contrast, the content of neutral lipid was rapidly increased by 29.21% 10 days after flowering(DAF), and showed the highest value by 91.84% at 40th day after flower. The neutral lipid, triglyceride content was rapidly increased as the seeds developed, and consisted of over 60% of the neutral lipid since 30 DAF. In the changes of neutral lipid, phospholipid and glycolipid, stearic acid and palmitic acid decreased during the seed ripening. However, oleic acid and linoleic acid increased during the same periods. Linolenic acid, which showed relatively higher value in the early ripening stage, but rapidly decreased as much as 1% at the later ripening stage.
This study was designed to obseve the effect of defatted perilla as a dietary fiber on lipid components of serum feces and liver in rats. Serum cholesterol and triglyceride were signfican-ly decreased in defatty perilla added groups. total lipid cholesterol and triglyceride of feces were significantly increased in defatted perilla added groups. Liver phospholipid content was higher in the defatted perilla added groups. The other lipid components of the liver were not affected by the defatted perilla administration. These results suggest the possibitlity that defatted perilla have a reducing effect of serum cholesterol and triglyceride through the incre-ment of feacal excretion of lipid component and phospholipid in liver.
Male mice (ddY strain) were fed a laboratory chow diet containing the water extract from stem bark of Rhus verniciflua Stokes (RVS) for 14 days. There were no significant difference in body weight gain, feed intake, the hepatic lipid profile and serum total cholesterol and phodpholipid concentrations. The concentration of triglyceride in serum was significantly lower in the RVS group than that in the control group. The concentration of high-density-lipoprotein cholestrol in serum was significantly higher in the RVS group than that in the control group. The methanol extract from RVS stem bark effectively inhibited the formation of thiobarbituric acid-reactive substances as a marker of lipid peroxidation of liver microsomes in a concentration-dependent manner. This study showed that the water extract from stem bark of RVS decreased the serum triglyceride concentration and methanol extract has an antioxidative activity.
The effects of the MeOH and water extracts from the marine algae Pelvetia siliquosa were evaluated on hyperlipidemic rats induced by cholesterol rich diet or poloxamer-407. The MeOH and water extracts, when administered orally for 3 consecutive days in hyperlipidemic rats induced by poloxamer-407 (1 ml of 30%), were found to cause a significant decrease in plasma cholesterol and triglyceride concentrations. Both extracts also showed a significant inhibition of serum total cholesterol and triglyceride level in rats treated with cholesterol rich diet. HDL-cholesterol, however, was increased significantly.
The effect of habitual alcohol consumption on the nutrient intake. body weight and serum values were studied in 194 upper middle class men aged 35-55yr. living in Seoul. Each subject was interviewed to get information of alcohol and nutrients intake. Blood samples were taken. Mean values for each variable were compared by fertile of alcohol intake. The average alcohol consumption of the subject was 32.2g/day. Most of times foods were taken along with drinks. The subjects preferred food items such as meat. fish, nuts. dry fishes when they drink. High alcohol consumers had a significantly higher intake of calorie. protein, fat. Fe, and riboflavin compared to nondrinkers and moderate drinkers. Body weight was significantly higher in high alcohol drinkers followed by moderate and nondrinker groups. They also showed the higher serum values of triglyceride and r-GTP. High alcohol consumption resulted in an increased intakes of most nutrients especially calorie and fat which leads to higher body weight and higher serum concentrations of triglyceride and r-GTP. Therefore it seems that heavy alcohol drinkers are more prone to obesity and related chronic diseases.
Rhodosporidium toruloides, an oleaginous yeast, can be used as a fast and reliable evaluation tool to screen new natural lipid-lowering agents. Herein, we showed that triglyceride (TG) accumulation was inhibited by 42.6% in 0.1% red radish coral sprout extract (RRSE)-treated R. toruloides. We also evaluated the anti-obesity effect of the RRSE in a mouse model. The body weight gain of mice fed a high-fat diet (HFD) with 0.1% RRSE (HFD-RRSE) was significantly decreased by 60% compared with that mice fed the HFD alone after the 8-week experimental period. Body fat of the HFD-RRSE-fed group was dramatically reduced by 38.3% compared with that of the HFD-fed group.
