부산 운수사 대웅전 목부재의 수종 식별 (Species Identification of Wooden Elements Used for Daewungjeon Hall in the Woonsoosa Temple, Busan)

  • 부산 운수사 대웅전에 쓰인 주요 목부재의 수종을 식별하기 위하여 기둥, 보, 장여, 도리, 주두, 소로, 포재, 대공, 반자 등 총 72점에 대하여 조사하였다. 수종을 분석한 결과, 소나무속의 소나무류 67점, 서어나무속 4점, 참나무속의 상수리나무류 1점 등 3수종이 식별되었다. 기둥에서는 소나무류 14점, 서어나무속 4점, 도리에서는 소나무류 4점, 상수리나무류 1점이 식별되었다. 그 외의 부재는 모두 소나무류로 식별되었다. 기둥의 부재중 동바리에 사용된 서어나무속 3개는 수리 시에 사용된 것으로 생각된다. 불단의 조각재 1점은 오리나무속으로 식별되었다. 결론적으로 후대에 교체된 것으로 보이는 서어나무를 제외하면 기본적으로 운수사 대웅전 건축 가구부재에 거의 대부분 소나무류가 사용되었음을 알 수 있었다.

  • 1. The plant communities on Mt. Sulak, a typical alpine mountain in central Korea was researched from the standpoint of ecological survey. 2. The plant on this mountain can largely be classified into 2 categories, the warm zone (below 1100m) and the frigid zone. The former is again classified into a broad-leaved tree formation and a needle-leved tree formation, and the latter is calssified into a broad-leaved tree formation and needle-leaved tree formation, and shrub formation. 3. The broad-leaved tree formation in the warm zone can largely be classified into a Carpinus laxiflora-syringa Palibiniana var. kanibayashi association(1), a Carpinus erosa-Acer Pseudo-sieboldianun association(2), and a Quercus mongolica- Tripterygium Regelii association(3). 4. The needle-leaved tree formation in the warm zone can also largely ben classified into a Pinus densiflora- Miscanthus sinensis association(4), Abies holophylla- Tripterygium regelii association(5), Pinus kordiensis- Rhododendron schlippenbachii association(6) In the association(4), the canopy of the Pinus densiflora is gradually being occupied by the Quercus mongolica which is one kind of broad-leaved trees. 5. We can fully see a DryoPteris type of the flour layer plant type in the association (3) and also see a Sasamorpha type in the association (5). 6. The broad-leaved tree formation in the frigid zone can largely be classified into Quercus mongolica- Rhododendron schlippenbachii association (8). The constituents of tree and subtree layers in (7) and (8) associations are similar to those association (3), but the constituents of the shrub layer are different from those of associtation (3) due to the difference in height. 7. The needle-leaved formation in the frigid zone can largely be classified into Thuja koraiensisRodgersia podophylla var. viritis association (9) and Abies nephrolepis-Acer tschonoskii var. rubripes asscciation (10). The species of alpine plants or subalpine plants are gradually increased in this formation. 8. The shrub formation can also largely be classified into a Rhododendron mucronulatum- Patrinia saniculaefolia association (11) and Pinus pumila associatio (12). Association (12) has largely developed on the windy place. Association (12) was burned due to the bombing during the Korean war, but now we recoginized there occurred an invasion of Tripterygium Regelii in such a place. 9. The herb layer species which constitute the shrub formation are mostly alpine or subalpine plants of small size, and their kinds are also very few. 10. The growth of the Moss layer is especially good because of the varied conditions of the habitat. The kinds which can easily be seen are the genus of Thunidium, Haplocladium, Brachythecium, Macromitrium, Holonitrium, Atrichum, Schwstchkeopsis, Grimmia, Hedwigia, Rhynchostegium and Mnium. 11. The genus of the Sphagnum densly grows like a mat at the acid and moisturous place above 1100m. The authors should express their thanks to father. Identification of Molecular Markers for Population Diagnosis of Korean Fir (Abies koreana) Vulnerable to Climate Change

  • Korean fir (Abies koreana) is an evergreen coniferous tree species that is unique to South Korea. A. koreana is found in a limited sub-alpine habitat and is considered particularly vulnerable to climate change. Identification of populations vulnerable to climate change is an important component of conservation programs. In this study, a heat stress-induced transcriptome RNA-seq dataset was used to identify a subset of six genes for assessment as candidate marker genes for ecologically vulnerable populations. Samples of A. koreana were isolated from ecologically stable and vulnerable regions of the Halla and Jiri mountains, and the expression levels of the six candidate markers were assessed using quantitative real-time polymerase chain reaction. All six of the candidate genes exhibited higher expression levels in samples from vulnerable regions compared with stable regions. These results confirm that the six high temperature-induced genes can be used as diagnostic markers for the identification of populations of A. koreana that are experiencing stress due to the effects of climate change.

신생대 제4기 울릉도 나리칼데라 부석층에서 산출된 탄화목의 연륜연대 및 수종식별 연구 (A Study on Tree-ring Dating and Speciation of Charcoal found in Pumiceous Deposit of the Quaternary Nari Caldera, Ulleung Island, Korea)

  • 본 연구는 울릉도 나리칼데라에서 산출된 탄화목의 수종식별, 연륜분석, 방사성탄소 연대를 통해 고식생/고환경, 분출연대 및 화산쇄설물의 정치온도에 대한 정보를 제공하기 위해 수행되었다. 탄화목은 울릉도 최후기 화산쇄설층을 대표하는 나리테프라층의 최하부(Member N-5) 부석질 퇴적층 내에서 채취되었으며, 나이테의 원형(횡단면)이 잘 보존된 나무의 가지 부분이다. 분석결과, 수종은 소나무과 중 가문비나무속(Picea spp.)으로 식별되었으며 최외각 나이테의 연륜은 $263+{\alpha}$ 년으로 확인되었다. 나이테는 중심(pith)에서 바깥쪽을 향해 그 나이가 점차 젊어지며 $20,260{\pm}230$, $19,995{\pm}245$$19,975{\pm}265cal\;BP$ 의 보정연대를 가진다. 가장 젊은 연대인 19,710 cal BP 는 Member N-5의 분출과 관련된 연대일 가능성이 높다. 이 결과는 후기 플라이스토세 동안 울릉도 나리칼데라는 가문비나무가 번성하였으며, 한랭 습윤한 기후조건하에 있었음을 지시한다. 광택, 취성, 색 및 조흔색 등의 특성에 기초하면, 탄화를 야기한 화산쇄설물의 최소 온도는 약 $350{\sim}500^{\circ}C$ 정도일 것으로 추정된다.

구 조선총독부 청사 (구 중앙청) 기초말뚝 통나무의 수종과 재질 (Species and Macroscopic Analysis of Log Piles Used for the ex-Government General of Chosun Building)

  • 구 조선총독부 청사(구 중앙청 건물)는 1916 년에 착공되었으며 건물기초를 다지기 위해 백두산 압록강변에서 벌채한 낙엽송(잎갈나무) 말뚝 약 만개를 박았다는 기록이 있다. 1996 년에 철거된 중앙청 자리에 경복궁 흥례문을 중건하는 공사 중 발견된 구 조선총독부 청사 말뚝 100 여 개를 채집하여 그 중 15 개에 대한 수종과 나이테를 분석하였다. 말뚝의 직경은 평균 27cm이었다. 목재조직관찰을 위하여 프레파라아트를 만들어 현미경으로 조사한 결과 모두 잎갈나무임이 확인되었다. 나이테로 관찰한 수령은 평균적으로 103년이었으며 가장 오래된 것이 151 년생이었다. 나이테 너비를 측정하여 151년 나이테 그래프(연륜연대기)를 작성하여 중국쪽 장백산(백두산) 잎갈나무 나이테 그래프(1602-1994)와 유사성올 비교하였으나 서로 일치하지 않았다. 이는 총독부청사 말뚝 채취지역이 중국측과 고도가 차이가 있는 것으로 생각된다. 백두산 압록강변에 자라고 있는 잎갈나무에 대해 나이테 자료를 얻을 수 있다면 화산활동과 같은 과거 백두산지역의 역사와 고환경을 나이테를 통하여 알아낼 수 있을 것이다.

Isolation and Identification of Fusicoccum Species from Quercus dentata

  • An imperfect fungus Fusiococcum species was isolated from Quercus dentata. A naturally infected Daimyo oak tree was collected and showed elongate wounds on the stem. The fungal cultures were initially white and cottony, and later turned dark gray. Numerous solitary pycnidia were developed on the medium surface, and typically spherical. Yellowish conidial masses were exuded from pycnidia on the culture plates. Conidial masses were swollen and measured as approximately 100 to $300{\mu}m$ in length. It appeared that conidia were usually held together in globose to oval drops. Conidia were hyaline, single-celled (nonseptate), ellipsoid to fusoid, and measured as approximately $8.0{\times}2.7{\mu}m$. Based on these cultural and morphological characteristics, the fungal isolate was identified as a species of Fusicoccum Corda. To preserve and examine fungal spores exuded from pycnidia on the medium surface, a vapor fixation procedure for scanning electron microscopy was employed in this study. The specimens were exposed to the vapor of 2% (v/v) glutaraldehyde and 2% (w/v) osmium tetroxide each for 2 h. With the vapor fixation we obtained excellent retention of conidial masses in this study. The simple and versatile procedure for demonstrating fungal spores and their exudation from fruiting bodies would facilitate characterization of diverse pathological and environmental isolates as they are in native environments.

우리나라 황단나무의 신분포 및 분류학적 특징 (A new distribution of Dalbergia hupeana Hance (Fabaceae) in Korea and its taxonomic characteristics)

  • 목포시 유달산에서 발견된 황단나무(Dalbergia hupeana)의 신분포를 보고한다. 본 종은 중국에만 분포하는 것으로 알려져 있었으나 이번 연구를 통해서 한반도에도 자라는 것이 확인되었다. 종의 동정은 형태 형질과 DNA 염기서열을 통해 확인하였다. 황단나무속(Genus Dalbergia) 또한 한국산으로 처음 보고되는데, 다른 한국산 콩과 수종들과는 양체웅예(5+5)를 가지는 것으로 뚜렷이 구분된다. 본 속과 종의 형태형질에 대해 기재하였으며, 이들의 국내 분포기원이 자생 또는 도입에 의한 것인지 논의하였다.

Anaplasma marginale and A. platys Characterized from Dairy and Indigenous Cattle and Dogs in Northern Vietnam

  • Anaplasma marginale and A. platys were detected and characterized (16S rDNA sequence analysis) from dairy and indigenous cattle, and the latter in domestic dogs in Vietnam. A phylogenetic tree was inferred from 26 representative strains/species of Anaplasma spp. including 10 new sequences from Vietnam. Seven of our Vietnamese sequences fell into the clade of A. marginale and 3 into A. platys, with strong nodal support of 99 and 90%, respectively. Low genetic distances (0.2-0.4%) within each species supported the identification. Anaplasma platys is able to infect humans. Our discovery of this species in cattle and domestic dogs raises considerable concern about zoonotic transmission in Vietnam. Further systematic investigations are needed to gain data for Anaplasma spp. and members of Anaplasmataceae in animal hosts, vectors and humans across Vietnam.

Ophiostomatoid Fungi in Pine Wilt Disease and Oak Wilt Disease in Korea

  • Pinewood nematode (PWN, Bursaphelenchus xylophilus) is a serious pathogenic worm that quickly dry pine trees to death. Recently, PWN has been devastating huge amounts of conifer trees in Korea. As a first step to explore the association and ecological roles of fungi in PWN life cycle in Korea, in this study we first isolated and indentified fungi from PWN-infested Korean pine and Japanese black pine wood sampled in Jinju, Sacheon, Pocheon, Chuncheon, Gwangju, and Hoengseong in Korea. A total of 144 fungal isolates were obtained from Japanese black pine wood and 264 fungal isolates from Korean pine wood. Their morphology and nucleotide sequences of the ITS rDNA and ♌-tubulin gene were examined for species identification. Ophiostoma ips, Botrytis anthophila, Penicillium sp., Hypocrea lixii, Trichoderma atroviride, O. galeiforme, Fusarium proliferatum were identified from Japanese black pine wood. Leptographium koreanum, L. pini-densiflorae, Ophiostoma ips, Penicillium raistrick, Trichoderma sp. were isolated from Korean pine wood. O. ips and L. koreanum were the major species on the two different PWN-infected pine tree. The cultivation of PWN on fungal mat of the identified species did some enhance PWN reproduction. The ambrosia beetle, Platypus koryoensis, is a serious pest of oak trees in Korea. In this study we investigated filamentous fungi present in the body of the beetle. Fourteen genera of filamentous fungi belonging to Ascomycota and Basidiomycota were isolated. All the obtained genera were isolated in the mitosporic state. The identified fungi were classified in 11 distinct orders including the Ascomycota (Eurotiales, Hypocreales, Microascales, Ophiostomatales, Pleosporales, and Sordiales) and Basidiomycota (Agaricales, Corticiales, Polyporales, and Russulales Xylariales). Within Ascomycota, 13 species were found. Meanwhile five species were found within Basidiomycota. The results showed the presence of diverse fungi in P. koryoensis. Among the isolated fungi, some were able to produce wood degrading enzymes. Further fungal isolation was performed with P. koryoensis infested Quercus mongolica trees sampled at Kumdan mountain in Hanam-Si, Gyeonggi province from June of 2009 to June of 2010. Penicillin spp. and Trichoderma spp. were the major species of mold fungi group. Pichia guilliermondii was the major species of mold yeast group. Raffaelea quercus-mongolicae was also isolated, but its isolation frequency was not high. Other species identified were Ambrosiella xylebori, Fusarium solani, Cryphonectria nitschke, Chaetomium globosum, and Gliocladium viride, Candida kashinagacola, C. maritima, C. vanderkliftii, Saccharomycopsis crataegensis.

A highly efficient computational discrimination among Streptococcal species of periodontitis patients using 16S rRNA amplicons

  • Due to the major role played by several species of Streptococcus in the etiology of periodontitis, it is important to assess the pattern of Streptococcus pathogenic pathways within the infected subgingival pockets using a bacterial specific 16S rRNA fragment. From the total of 50 patients with periodontitis included in the study, only 23 Streptococcal isolates were considered for further analyses, in which their 16S rRNA fragments were amplified and sequenced. Then, a comprehensive phylogenetic tree was constructed and in silico prediction was performed for the observed Streptococcal species. The phylogenetic analysis of the subgingival Streptococcal species revealed a high discrimination power of the 16S rRNA fragment to accurately identify three groups of Streptococcus on the species level, including S. salivarius (14 isolates), S. anginosus (5 isolates), and S. gordonii (4 isolates). The employment of state-of-art in silico tools indicated that each Streptococcal species group was characterized with particular transcription factors that bound exclusively with a different 16S rRNA-based secondary structure. In conclusion, the observed data of the present study provided in-depth insights into the mechanism of each Streptococcal species in its pathogenesis, which differ in each observed group, according to the differences in the 16S rRNA secondary structure it takes, and the consequent binding with its corresponding transcription factors. This study paves the way for further interventions of the in silico prediction, with the main conventional in vitro microbiota identification to present an interesting insight in terms of the gene expression pattern and the signaling pathway that each pathogenic species follows in the infected subgingival site.
