Kyungsan nursery complex which has a vast area for the production of various species of fruit tree stocks is in a high demand of virus-free saplings. Apple tree stocks, the most important products, urgently need more rapid and reliable viral diagnosis. In this study, a bead beater was tested because of convenience in dealing with large number of samples. Also, industrial glass bead abrasive (0.4 mm in diameter) at very low cost was used in a disposable way. For bead beater-aided RNA extraction from apple stem tissues, the guanidine thiocyanate method was confirmed to be very reliable. Silca membrane filter tube in connection to vacuum filtering device was strongly suggested for simplifying RNA capture and washing steps. Apple virus detection was confirmed by RT-PCR.

Degraded plantation forests are increasing because of unfavorable forestry conditions prevailing in Japan, including falling timber prices, increasing operational costs, and aging and declining forestry workforce. To remedy this situation, appropriate management strategy is required. This study introduces the challenges of Odai Town, Mie Prefecture that employed a new management strategy by evaluating site suitability for conifer forests and that proposes a new forest management regime of planting deciduous trees in unsuitable sites. The site suitability for conifer forests was evaluated from two aspects: the natural site conditions and the relationship among site conditions, growth, and damage by Anaglyptus subfasciatus Pic. in Cryptomeria japonica D. Don and Chamaecyparis obtusa Sieb. Et Zucc. forests. By analyzing the relationship among site conditions, growth, and insect damage based on field data obtained in plantation forests, growth evaluation and insect damage evaluation maps were developed. Based on the natural forest investigation, natural site condition maps for C. japonica and C. obtusa were established. Furthermore, by integrating these evaluation maps with the forest road maps showing the accessibility to the forest, the forest management regime for the whole plantation area of Odai Town was established. The forest management regime map indicates the sites suitable for forestry: suitable for long-rotation, short-rotation, and potential sites for short-rotation. The sites unsuitable for forestry were considered to be more suitable for broadleaved forests. Clear-cutting was conducted in a small area and different seral stage saplings (approximately 20 deciduous tree species) suitable to the site conditions were planted in an area of $80-120m^2$ protected by deer-fences. This might establish a forest composed of many species with a multilayer vertical forest structure in a short period. The planted saplings were distributed neither randomly nor uniformly to reflect the natural distribution of trees in the forest. A challenge to develop new products using the deciduous trees has started, such as wood chips for preparing smoked food, essential oil, and deodorizer. As these challenges have just begun, their effects on enhancing sustainable resource management are still being monitored. Even with the challenges, this regime can be of high value as a management strategy to remedy the situation of expansion of degraded forests in Japan.
Distribution of size class, population regeneration and changes in the population structure of Quercus mongolica were studied from 1994 to 1999 in Mt. Jumbong (128°27' E, 38° 04' N) of Mt. Sorak National Park and Biosphere Reserve in central Korea. Three 20m × 20m permanent quadrats were set up at the elevation of 900m. The vegetation of the study site was dominated by Quercus mongolica and Acer pseudosieboldianum, but little change was observed in the community structure from 1995 to 1999. Most mortality in the study site was observed in small trees of A. pseudosieboldianum and Tilia amurensis. Mean annual growth in dbh (diameter of breast height) of Q. mongolica for 4 years was only 0.09cm, and no ingrowth of saplings (dbh < 2.5cm) into tree class was observed during the study period. Among the 21 Q. mongolica trees studied in the permanent quadrats. all the smaller trees (dbh < 30cm) were established in 1920 ∼ 1950, while many bigger trees (with 40cm < dbh < 80cm) were established in 1750 ∼ 1800, indicating that its establishment was episodic. Distribution of dbh classes among Q. mongolica trees shows that smaller trees were poorly represented, and no saplings of Q. mongolica occurred in the permanent quadrats studied, indicating that currently Q. mongolica is not regenerating well in the study site. Total seed production of Q. mongolica in 1994 was estimated as 88 acorns per square meter in the study site. Rate of predation including caching of acorns was highest in 1994, then declined sharply thereafter. Most of the acorns which managed to survive in the first year were predated in the second year, and only 5% of the acorns produced in 1994 survived into the third year. No seeds produced in 1994 or seedlings germinated from them succeeded to survive to 5 years after seed production. However, seedling emergence rate and seedling survival were high in the early growing season in 1995. These results suggest that predation can be a significant factor in the regeneration of Q. mongolica. and that Q. mongolica is not regenerating well in Mt. Jumbong and needs large scale disturbances for its new recruitment.
This study was carried out to provide basic information on efficient preservation and management of an Abies koreana forest through analyzing the ecological characteristics by slope in Seseok, Mt. Jirisan. Soil moisture content at southern and northern slopes was 29.9% and 21.7%, respectively, and there was no significant difference among soil properties between southern and northern slopes. The importance value of A. koreana in the southern and northern slopes was high for tree and subtree layers, respectively. It is noteworthy that many seedling and saplings of A. koreana were present on the southern slope. The species diversity was 0.413 for the tree layer, 0.632 for the subtree layer, and 0.609 for the shrub layer on the southern slope and 0.396 for the tree layer, 0.783 for the subtree layer, and 1.215 for the shrub layer on the northern slope. Evenness and dominance ranged from 0.371 to 0.609 and 0.629 to 0.391 on the southern slope and from 0.380 to 0.968 and 0.620 to 0.032 on the northern slope, respectively. The mortality of A. koreana was 9.6% on the southern slope and 24.4% on the northern slope, a distinct difference between the slopes. The mortality type at two slopes was the highest proportion of standing dead. Annual mean tree ring growth of A. koreana on the southern slope (1.76 mm/yr) was higher than that on the northern slope (1.64 mm/yr).
The results of survey on the nematodes associated with sapling from 8 Forestry Experiment Stations(F.E.S.) in Korea are reported herein. A total of 35 species under 20 genera, 11 families, and 2 orders were identified from the soils around the roots of 119 tree species and amongst them, Xiphinema insigne is unrecorded species for Korea and Geocenamus sp. is a new species. Tylenchorhynchus claytoni, which showed the highest detection rate (D.R.) from all F.E.S., except Nambu F.E.S, was the dominant species among the nematodes associated with saplings. The D.R. of T. claytoni was the highest in Ch’ungnam F.E.S. by 100% from 15 saplings sampled, followed by 87.5% in Kangwon, 66.6% in Kyongbuk, 58.3% in Cheonbuk, 48.1% in Ch’ungbuk, 41.3% in Kyongki, and 40% in Cheonnam F.E.S. Other important nematode species which showed very high densities by stations are; Criconemella morgensis (D.R.: 37.5%), Helicotylenchus digonicus (D.R. : 35%), Pararotylenchus pini (D.R.: 37%), Tylenchorhynchus nudus (D.R.: 37.9%), Criconemella informis (D.R.: 27.5%), and Meloidogyne sp. (D.R.: 12.5), from Chunbuk, Chunnam, Ch’ungbuk, Kyongki, and Kangwon F.E.S., respectively.
Habitat types, community structure and population characteristics of Koelreuteria paniculata were investigated in Mt. Wolak, Chungbuk and Naesokdong, Daegu, which are natural habitats of the species in inland region of Korea, and its origin was discussed. Habitats of Koelreuteria paniculata were classified to 3 types: sand bar formed by the sands flooded in the course of flow of the mountain stream (Type 1). crevice on the rock bed within the mountain stream (Type 2) and crevice of the rock around the edge of mountain stream (Type 3). Most Koelreuteria paniculata communities in Mt. Wolak site were composed of 3 layers of subtree layer, shrub layer and herb layer and that of Daegu site was 4 layers including tree layer. In the floristic composition of the Koelreuteria paniculata community, plants occurring frequently in the wet and open site, such as Zelkova serrata and Fraxinus rhynchoph-vlla showed high frequency. Frequency distribution of diameter at ground surface of Koelreutrria paniculata showed reversed J-shaped type. It was supposed that expansion of Koelreuteria paniculata community in Mt. Wolak site might be accomplished by the flow of the stream. Many saplings capable of becoming a successor of mature trees in Daegu site in near degenerating phase were established on the forest floor of the Koelreuteria paniculata community. From this result, it was supposed that these saplings originated from the seeds dispersed from a seed tree might form the Koelreuteria paniculata community of the next generation. On the other hand, the origin of Koelreutevia paniculata in inland sites was explained by two hypotheses: the one was that Koelreuteria paniculata might be transplanted by human and the other was that the present site might be native habitat of the community.
Pinewood nematode (PWN: Bursaphelenchus xylophilus) is known to kill pine tree species that are indigenous to countries where the pest was inadvertently imported, but some cultures from the extraction of dead pines do not damage trees. Experiments were conducted to examine the effect of pre-inoculation of these low-pathogenic pinewood nematode on resistance of pine trees against the pest species. The pre-inoculated pine saplings showed induced resistance which lasted for a year, and repeated inoculation of these low-pathogenic nematodes enhanced tree resistance. All nematode samples extracted from dying or dead pines that had been killed not more than three months before the extraction were pathogenic, and most of those extracted from pines that had been killed 2-3 years before were low-pathogenic. When inoculated in pine saplings, number of low-pathogenic nematodes settled, as studied two days after inoculation, was not different from that of pathogenic ones. However, as studied after 30 days of inoculation, rate of reproduction in low-pathogenic nematodes was far lower than that of pathogenic nematodes. The rate of reproduction of several nematode isolates growing on fungal mat media of Botrytis cinerea varied, but three of four low-pathogenic isolates showed same level of reproduction rates as pathogenic ones.
Classified into sub-tree of the growth-form, Acer pseudosieboldianum and Carpinus cordata hardly reach the uppermost canopy with limited height growth potentiality in the natural deciduous forest. However, the two species usually dominate lower stories of the forest, playing an important role in overall ecological characteristics in the forest. Based on the vegetation data of 106 $20m{\times}20m$ sample plots in Mt. Jumbong area, this study was carried out to evaluate ecological constitution by several quantitative analysis so as to understand the mechanism of the natural deciduous forest. The results indicated that individuals of A. pseudosieboldianum and C. cordata were absent or few in overstory, but emerged the most dominant species in midstory and understory, providing adequate proof of the ecological importance. The comparison of indices of succession between presented and predicted values in midstory did not make much difference, suggesting that the species composition would not change much and come close to steady state in midstory and understory. The pair combination of species association noted that A. pseudosieboldianum had significant positive association with C. cordata, Quercus mongolica, and Tilia amurensis had significant positive association with A. pseudosieboldianum, A. pictum subsp. mono and Fraxinus mandshurica but negative association with F. rhynchophylla. Being compared with other major canopy tree species in the study forest, the target species of A. pseudosieboldianum and C. cordata had strong regeneration strategies, partially characterized by large number of saplings and pole sized trees and high ratio of live crown, which indicated high shade tolerance to survive in the limited amount of light under the canopy. Even though A. pseudosieboldianum and C. cordata do not reach and occupy the canopy layer mainly due to the inherent growth form, they would have highest competitive potentiality to prosper and dominate in the midstory of the natural deciduous forest.
Juniperus chinensis (Cupressaceae) is a valuable woody plant resource, but this rare plant is only distributed in restricted regions in Korea. In this study, we surveyed the geographic distribution regions, variation of sex ratio and the dynamics of natural populations of J. chinensis. in order to establish conservation strategies for the sustainable conservation of this important resource tree. The 17 natural populations of J. chinensis are restrictively distributed in the shore cliffs of (1) Ulleungdo island, (2) Gyeongju-si to Gangneung-si of the eastern coast, the riverside cliff along (3) Donggang-river of Jungsun-gun and Pyeongchang-gun and (4) branch of Nakdonggang-river in Uiseong-gun. The populations in Ulleungdo island showed continuous population distribution of over 200 individuals, but populations in other regions had discontinuous population distribution of under 200. The population of J. chinensis surveyed in Korea was estimated in ca. 3,200 individuals. The sex ratio (male/female) among populations of J. chinensis was diverse with variation from 0.46~1.67 range, but showed no significance. The sex ratio of the total individuals by species level was 0.74, and the percentage of female tree was significantly higher than the male. The results of dynamics analysis within population among regions showed that seedlings and saplings of J. chinensis in three regions surveyed were continuously regenerating. The distribution of DBH class showed a stable population structure of inverse J shape graph, but fragmentation and decrease in populations because of invading broad-leaved shrubs and trees from the surroundings were concerned. Further studies, and in situ and ex situ conservation strategies for the sustainable conservation on natural populations of this rare resources woody plant, J. chinensis in Korea are recommended.
The first purpose of this study is the verification of hypothesis on Abies holophylla planted as a guardian for Japanese. Second, the purpose of this study is on Abies holophylla planted in the historic sites for anti-Japan fighter. Japan planted Abies holophylla to honor the god of Suwataisha that took care of the Jingu-Kogo's Conquest of Samhan(Three Kingdoms). The fact that Abies holophylla was planted in the historic sites for anti-Japan fighter such as General Gwon Yul, Yi Sun-Shin was investigated. The hypothesis that Abies holophylla was planted as a guardian for Japanese was verified through survey on the pictures of Abies holophylla in Japanese temple, public sites, company of nursing the saplings, commemorative tree of governor and the judgment of Abies holophylla damage incident. Further studies should focus on the intention of planting Abies holophylla by Japan. Researcher should discuss Abies holophylla planted in the historic sites for anti-Japan fighter because old big trees will be respected.
