한우 수정란의 동결보존 및 쌍자생산에 관한 연구 I. 동결 수정란의 이식과 자우 생산 (Studies on Embryo Cryopreservation and Twinning by Embryo Transfer of Korean Native Cattle I. Transfer of Frozen-thawed Embryos and Production of Calves)

  • This study was carried out to establish the techniques for producing the calves of genetically superior Korean Native cattle by transfer of frozen-thawed embryos. The effects of some factors related to embryo recovery following superovulation and pregnancy rate following transfer of frozen-thawed embryos were evaluated. Also calving state was investigated. The results obtained were as follows ; The mean number of total and transferrable embryos recovered per superovulated cow was 8.72 and 4.90, respectively, from a total of 72 superovulations using 34 donor cows. There were no significant differences in the number of total or transferrable embryos recovered per superovulated cow between products of follicle stimulating hormone (FSH), years, seasons, and collection numbers. The pregnancy rate was found 44.44% following transfer of frozen-thawed embryos of Korean Native cattle to a total of 180 recipient cows including 82 Angus, 27 Charolais, 62 Hereford and 9 Korean Native cows. The pregnancy rate was significantly (P<0.05) higher in the transfer of excellent (42.99) and good embryos (40.17%), compared with fair (5.90%) grade embryos. And the pregnancy rate was significantly (P<0.05) higher in the transfer of embryos of morula stage (43.86%) than blastocyst stage (15.51%). But there were no significant differences in pregnancy rates between natural and induced estrus estrus asynchrony of 1 days, breeds, and parities of recipient cows. The normal calving rate of 80 pregnant cows following transfer of frozen4hawed em-bryos was 87.5% and the other 10 pregnant cows showed abortion during the period from pregnancy diagnosis at 50~60 days to calving. The average gestation length of normally delivered recipients was 288.50 days and the average birth weight of 70 calves born was 24.22 kg. The gestation length was significantly (P<0.05) shorter in the recipients delivering female calves (286.70 days) than males (289.39 days). But there were no significant differences in gestation tength and birth weight of calves born between the recipient breeds.

Laparoscopic Transabdominal Transfer of Blastocysts in Korean Black Goats

  • As a part of the effort to improve post-transfer survival rate of embryos in Korean black goats, a technique for laparoscopic uterine transfer of blastocysts was carried out. A total of 26 transferrable embryos (morula to expanded blastocysts) were transferred to 13 recipient goats via transabdominal laparoscopic method. In consequence of our hormone protocol, 65% of the recipients (13/20) were found to have synchronized estrus. After confirmation of corpus luteum in each recipient goat, a Babcock laparoscopic forceps was inserted into the lower abdominal cavity to hold a uterine horn and fasten it near the peritoneum without causing injury. Then 7.5cm long 16G IV catheter was inserted directly into the uterine lumen through the abdominal wall. After removal of the stylet of the IV catheter, the embryo transfer tube (identical in size to the stylet and loaded with blastocysts) was inserted into the uterine lumen through the catheter to unload the embryos. Of the 13 estrus synchronized recipients, 9 were transferred blastocysts and 4 were transferred molurae (2 embryos in each recipient) in uterine ipsilateral to the ovary with corpus luteum. Four of the 9 recipients which blastocysts were transferred using this method has been confirmed pregnant (44.4% pregnancy rate).

소 수정란이식(受精卵移植)에 있어서 수란우(受卵牛)와 수정란(受精卵)의 조건이 수태율(受胎率)에 미치는 영향 (Effect of embryo and recipient condition on pregnancy rate following bovine embryo transfer)

  • This study was carried out to determine suitable selection factors for recipients and embryos which could improve pregnancy rates following bovine embryo transfer. The experiment included 52 surgical transfers from February, 1985 through June, 1986 performed on Kyungpook Breeding Center in southern Korea. The pregnancy rate was highest when recipients were in estrus within 6 hours before the donor to 12 hours after the donor (78.3% versus 50% for recipients in estrus earlier or later). Pregnancy rates were acceptable following culture under field conditions for up to 17 hours. More recipients over 15 months of age (76.1%) remained pregnant than those under 15 months (66.7%). Embryos transferred during the months from February to July resulted in higher pregnancy rates than those transferred during the remaining 6 months (77.3% versus 57.1%). Transferrable embryos were classified A (best) to C (worst); those graded A or B resulted in significantly higher pregnancy rates than those graded C (81.8% and 73.3% versus 25.0%, p<.05). Pregnancy rates among recipients of the Korean native breed tended to be higher than among Holstein recipients (100% versus 71.1%). Similarly, when the embryo was transferred to the right uterine horn, pregnancy rates tended to be higher than when it was transferred to the left (81.3% versus 65%). Pregnancy rates did not differ according to the stage of development of the embryo; they were for morulae, tight morulae, blastocysts, and advanced blastocysts, respectively: 75.0%, 66.7%, 75.0%, and 77.4%.

한우 및 젖소에서 과배란 처리를 이용한 체내수정란 생산과 신선 및 동결 수정란 이식 결과 (Production of In-Vivo Embryos by Superovulation and Result of Transfer with Fresh or Frozen Embryos for Hanwoo and Holstein Cattle)

  • 가축에서 생산성을 높이기 위해 사용되는 수정란이식술의 실용성을 알아보기 위하여 한우 및 젖소 36두로부터 과배란 처리를 하여 신선수정란은 25두에, 동결수정란은 17두에, 쌍태 생산을 위한 수정란 두 개 이식은 13두 수란우에 이식되어 그 생산 결과를 조사하였다. 1 과배란을 위한 호르몬 처리후 반응을 나타낸 공란우는 75.0%이었다. 2. 황체수와 비교하여 채란된 수정란 율은 91.8% 이었다. 3. 비외과적으로 채란된 전체 난자 중 생존성이 있는 배반포는 66.4%이었고, 이식 가능한 배반포는 48.6%이었다. 4. 간이채란법으로 시도된 one-catheter method에 의한 양 자궁 관류성공률은 75.0%이었다. 5. 신선 수정란을 이식후 분만결과에 따른 수태율은 60.0%이었다. 6. 동결수정란을 융해하여 이식후 분만결과에 따른 수태율은 35.3%이었다. 7. 쌍태 생산을 위한 수정란 두 개 이식결과에서 신선수정란 사용시 28.6%, 동결수정란 사용시 16.7%의 쌍태분만을을 나타내었다. 8.수정란 이식 후 생산된 산자에서 수컷의 비율은 56.3%이었다. 9. 수정란 이식 계절에 따른 분만 결과에서 여름에 이식시 가장 낮은 수태율을 나타내었다.

Pregnancy and fertilization potential of immature oocytes retrieved in intracytoplasmic sperm injection cycles

  • Objective: The goal of this study was to evaluate the pregnancy potential of immature (metaphase I or germinal vesicle stage) oocytes retrieved in intracytoplasmic sperm injection (ICSI) cycles. Methods: A total of 1,871 couples with infertility underwent 2,984 ICSI cycles. Cycles in which three or fewer oocytes were retrieved were included in this study in order to evaluate the pregnancy potential of immature oocytes. Cycles were divided into five groups (group I-V), according to the maturation status of the oocytes at the time of cumulus cell removal and ICSI. The fertilization and pregnancy rates after ICSI were analyzed and compared among the study groups based on the maturation status of the retrieved oocytes. Results: The retrieval of only immature oocytes was associated with a significant decrease in the fertilization rate ($76.1%{\pm}37.3%$ vs. $49.0%{\pm}49.1%$, $66.7%{\pm}48.7%$; group I vs. group II, group III, respectively) and the average number of transferred embryos ($1.5{\pm}0.7$ vs. $1.1{\pm}0.4$, $1.1{\pm}0.6$). The cycle cancellation rate was significantly higher when only immature oocytes were retrieved. The clinical pregnancy rate decreased significantly when the transferred embryos had originated from immature oocytes (16.9% vs. 10.3%, 1.2%). Conclusion: In ICSI cycles, the fertilization potential and pregnancy potential of the immature oocytes retrieved in ICSI cycles were inferior to those of mature oocytes. Therefore, increasing the number of injectable oocytes and transferrable embryos by using immature oocytes after their spontaneous in vitro maturation does not necessarily improve pregnancy outcomes.

과배란 처리된 한우의 수정란 생산에 미치는 산차와 계절의 효과 (Effects of Parity and Season on Production of Embryos in Superovulated Hanwoo)

  • 발정주기와 상관없이 CIDR을 삽입하는 날에 50 mg progesterone, 2.5 mg estradiol benzoate를 근육주사하였다. CIDR 삽입 후 4, 5일에 28 AU FSH (Antorin R10)을 4일 동안 감량법으로 주사하였다. 6, 7회 FSH 주사 후 25 mg, 15 mg $PGF_2{\alpha}$를 각각 주사한 다음, CIDR는 7회 FSH 주사 후 제거하였다. 1회째 $PGF_2{\alpha}$ 주사 후 48시간에 $200{\mu}g$ GnRH를 주사하였다. 공란우는 발정확인 후 12시간 간격으로 3회 인공수정을 실시하였으며, 수정란의 채란은 발정확인 후 7일째 회수하였다. 과배란 처리된 공란우에서 회수된 총 난자수, 이식가능 수정란, 퇴행란 및 미수정란은 각각 $11.6{\pm}7.9$, $5.5{\pm}4.4$, $3.0{\pm}3.3$$2.6{\pm}4.1$개였다. 1~2산의 공란우($8.9{\pm}1.9$)보다 3~5산의 공란우($14.3{\pm}1.3$)에서 총 난자수가 유의적으로 많았다(P<0.05). 회수된 정상 수정란 수도 3~5산의 공란우($7.3{\pm}0.8$)에서 6산 이상의 공란우($3.7{\pm}1.5$)보다 유의적으로 많았다(P<0.05). 회수된 총 난자수와 정상수정란도 각각 가을($10.1{\pm}1.8$, $4.5{\pm}1.1$)과 겨울($6.3{\pm}1.8$, $3.3{\pm}1.1$)보다 여름($16.4{\pm}2.3$, $8.1{\pm}1.4$)에 유의적으로 많았다(P<0.05). 이식가능 수정란은 겨울($3.0{\pm}1.0$)보다 여름($7.6{\pm}1.3$)에서 유의적으로 많이 회수하였다(P<0.05). 결론적으로 공란우의 산차, 과배란 처리계절은 한우수정란 생산에 영향을 미치는 것으로 나타났다.

Increase of Production Ratio of Pre-selected Superior Dairy Female Offspring by Combination of OPU derived Oocytes and X-bearing Semen

  • This study was designed to evaluate the possibility of increase through dairy female offspring's ratio by transfer of pre-selected transferrable blastocyst that was produced by pre-selected X-bearing semen with OPU derived oocytes. Elite dairy female cow is demanded strongly compared with male, the so called, farmer wants to produce only an elite female dairy offspring as a candidate female dairy cow for producing milk. In our study, we selected 2 elite dairy bull semen from National Agricultural Cooperative Federation to pre-select X-bearing semen and 5 elite dairy female cows as donor for collecting of OPU derived oocytes. OPU derived embryo production system was carried out an aspiration of immature oocytes from 5 donor cows 2 times per week, total 200 times for 2 to 7 months by an ultrasonographic guided follicular aspiration system and then produced in vitro-produced blastocysts by in vitro maturation, fertilization and culture. Dairy donor semen selected H-319, 320 bull in National Agricultural Cooperative federation was sorted X-bearing semen by flow-cytometer and frozen for using IVF with OPU derived oocytes. Donor cows were selected 5 elite dairy cows from Gyeongju Dairy Cow Community and then disease tests such as 4 kinds of disease before selecting was checked. Oocyte proportion of grade 1 to 3 from total collected oocytes was significantly lower in donor A and B than those in donor C, D and E (82.16 and 70.03% vs. 90.0, 91.78 and 93.57%), respectively (p<0.05). However, number of oocytes per session in donor A, C and E was significantly higher than those in donor B and D ($7.77{\pm}3.26$, $5.85{\pm}2.10$ and $7.03{\pm}2.14$ vs. $4.68{\pm}2.61$ and $5.21{\pm}1.97$ oocytes), but donor A was significantly higher than donor C (p<0.05). Development to blastocyst in donor B, C and E was significantly higher than those in donor A and D (31.0, 25.0 and 25.0% vs. 14.3 and 4.5%), but donor A was not different in donor C and E (p<0.05). Nine out of 10 blastocysts (90.0%) derived from OPU blastocysts were confirmed male embryos that was induced with Y-bearing semen to confirm sex ratio only. Total 96 blastocysts derived from female bearing semen were transferred into synchronized recipients and then confirmed 42 recipients (43.8%) pregnancy rate, 36 offspring (37.5%) and 91.7% female sex ratio (33 female vs. 3 male offspring). Taken together all data, elite dairy female offspring could be produced effectively by in vitro production system between pre-selected x-bearing semen and OPU derived oocytes that would be influential breeder in the breeding of dairy farm to increase effectively elite dairy offspring ratio as well as net income in the dairy farmer.

The effectiveness of earlier oocyte retrieval in the case of a premature luteinizing hormone surge on hCG day in in vitro fertilization-embryo transfer cycles

  • Objective: To evaluate the efficacy of earlier oocyte retrieval in IVF patients with a premature LH surge on hCG day. Methods: One hundred forty IVF patients (164 cycles) with premature LH surge on hCG day were included, retrospectively. We divided them into 2 study groups: LH surge with timed ovum pick-up (OPU) 36 hours after hCG injection (group B, 129 premature cycles), and LH surge with earlier OPU within 36 hours after hCG injection (group C, 35 cycles). Control groups were tubal factor infertility without premature LH surge (group A, 143 cycles). Results: The mean age (year) was statistically higher in group C than in groups A or B ($38.2{\pm}5.4$ vs. $36.2{\pm}4.2$ vs. $36.8{\pm}4.9$, respectively; p=0.012). The serum LH levels (mIU/mL) on hCG day were significantly higher in group B and C than in group A ($22.7{\pm}14.9$ vs. $30.3{\pm}15.9$ vs. $3.2{\pm}2.9$, respectively; p>0.001). Among groups A, B, and C, 4.9%, 31.7%, and 51.4% of the cycles, respectively, had no oocytes, and the overall rates of cycle cancellation (OPU cancellation, no oocyte, or no embryos transferrable) were 15.4%, 65.9%, and 74.3%, respectively. The fertilization rate (%) was significantly higher in group B than in group C ($73.2{\pm}38.9$ vs. $47.8{\pm}42.9$, p=0.024). The clinical pregnancy rate was significantly higher in group C than in groups A and B (44.4% vs. 27.3% vs. 9.1%, respectively, p=0.021). However, the miscarriage rate was also higher in group C than in group B (22% vs. 0%, respectively, p=0.026). Conclusion: Earlier OPU may not be effective in reducing the risk of cycle cancellation in patients with premature LH surge on hCG day. A larger scale study will be required to reveal the effectiveness of earlier ovum retrieval with premature LH surge.
