Changes in Oxygen Consumption Rates of Embryos in Korean Cattle (한우 수정란의 발달 단계별 산소 소비량 변화)

  • Oxygen consumption has been regarded as a useful indicator for assessment of mammalian embryo quality. However, there was no standard criterion to measure the oxygen consumption of embryos. Here, we measured oxygen consumption of bovine embryos at various developmental stages was measured using a scanning electrochemical microscopy (SECM). We found that the oxygen consumption significantly increased in blastocyst-stage embryos compared to other stage embryos (from 2-cell-stage to morula-stage), indicating that oxygen consumption reflects the cell number ($5.2{\sim}7.6{\times}10^{14}/mol\;s^{-1}$ versus $1.2{\sim}2.4{\times}10^{14}/mol\;s^{-1}$, p<0.05). In the morula-stage embryos, the oxygen consumption of in vivo derived embryos was significantly higher than that of in vitro produced embryos ($4.0{\times}10^{14}/mol\;s^{-1}$ versus $2.4{\times}10^{14}/mol\;s^{-1}$, p<0.05). However, there was no significant difference in consumption of oxygen by in vivo and in vitro-derived bovine blastocyst-stage embryos (p>0.05). In the frozen-thawed blastocyst-stage embryos, live embryos showed significantly higher oxygen consumption than dead embryos ($4.7{\times}10^{14}/mol\;s^{-1}$ versus $1.0{\times}10^{14}/mol\;s^{-1}$, p<0.05). These results indicate that the measuring oxygen consumption by SECM can be used to evaluate bovine embryo quality.

Effect of Production In Vitro Embryo with Frozen-thawed Semen using AndroMed Extender in Korean Black Cow Semen (AndroMed를 이용한 흑우 동결 정액으로 체외수정란 생산 효과)

  • The aim of present experiment was to examine commercial synthetic extender(AndroMed) for semen cryopreservation of Korean Black Bull. Semen was collected from a Korean Black Bull using an artificial vagina and transported to the laboratory. The semen was diluted 1:1 by AndroMed. The pellect was diluted to final sperm concentration of $5{\times}10^5/ml$ by doubling in every 10 minutes at $4^{\circ}C$ cold chamber. The semen was equilibrated for 1 hr at cold chamber and packed to 0.5 ml straw. The semen straws were located above 5 cm of liquid nitrogen for 5 minutes, above 5 cm for 10 minutes and above 10 cm for 10 min. And then the frozen straw was plunged to $LN_2$. The presented straws were examined the viability and motility after thawed at $37^{\circ}C$ water bath. Hanwoo semen was used as KPN (Korea Proven Bull Number) in this experiment. The survival rates was significantly higher in fresh semen than frozen semen ($80{\pm}14%\;and\;43{\pm}11%$). However, the motility rates was similar (80.7% and 66.4%). The survival and motility rates were higher in 5cm, 10 min treatment group than the other two groups in straw-located height and duration above $LN_2$ ($50{\pm}14%$ and 70.7% vs, 33.18% and $65{\pm}7%$ vs, 30.14% and 65.7%, respectively). The development rates to cleavage was higher in Black Cow than Hanwoo semen (62.2%, 64.4%), However, The development rates to blastocyst was higher in Hanwoo than Black cow semen (25.9%, 23.0%). In conclusion. The present results that acceptable fertilization and cryopreservation could be obtained by in vitro fertilization with frozen-thawed semen using a synthetic semen extender (AndroMed).

Effect of Different Feeding Ratios of Whole Crop Barley Silage on the Embryo Production in Hanwoo Donors

  • The purpose of this study was to determine the effect of different feeding ratios of whole crop barley silage on the embryo production in Hanwoo donors. All donors were basically fed 2.5 kg concentrate daily. Donors were divided into three groups according to the different feeding of forage; hay 70% and rice straw 30% (control, n = 21), whole crop barley silage 80% and rice straw 20% (T1, n = 25), and whole crop barley silage 60% and rice straw 40% (T2, n = 23) fed based on TDN 6.70/ BW 500 kg. All Hanwoo donors received a CIDR together with injections of 1 mg estradiol benzoate and 50 mg progesterone ($P_4$, Day 0). Four days later, they were superovulated with 28 mg FSH twice daily IM in decreasing doses over 4 days. Then donors received 2 doses of $PGF_2{\alpha}$ (25 and 15 mg) with the 5th and 6th injections of FSH on Day 6. CIDR were withdrawn at the 6th FSH injection and the donors received $100\;{\mu}g$ GnRH 36 h after the second $PGF_2{\alpha}$ injection. The donors were artificially inseminated twice, at 8 and 24 h after GnRH, and embryos were recovered 7 or 8 days after the 1st insemination. The flush rate of the donors following positive superovulation responses did not differ among groups (76.2~96.0%, p>0.05). The number of corpus luteum (CL) at embryo recovery also did not differ among groups (10.6~14.0, p>0.05). Furthermore, the mean numbers of total ova (9.4, 10.5 and 12.0) and transferable embryos (5.3, 12.0 and 6.5) did not significantly differ among the control, T1 and T2 groups, respectively (p>0.05). However, mean concentrations of serum $P_4$ of the T1 (64.2 ng/ml) and T2 groups (55.7 ng/ml) were higher than that of control group (43.3 ng/ml, p<0.01), while serum cholesterol concentrations in the control (105.8 mg/dl) and T2 groups ($96.9\;{\pm}\;mg/dl$) were significantly lower than in the T1 group (121.1 mg/dl, p<0.05). Conclusively, whole crop barley silage can be fed a good substitute for hay forage for Hanwoo donors. Furthermore the ratios of whole crop barley silage 60% and rice straw 40% might be more worthful for embryo production.

  • Urban railway are the most widely used means of transportation in the metropolitan area. It is difficult to draw out problems through the quantitative analysis technique because most of the research results show that most of the guide signs are installed with many or few guide signs. In this study, we propose a method to evaluate the information load and proper installation location at the main decision point in the transfer passageway for Suseo Station by applying Positive Guidance. As a result of analyzing the information load using this technique, it is suggested that the necessary information should be provided centered on the copper node because it provides confusion of the users by providing the same information in the transfer line. It is necessary to obtain guidance on how to remove the sign (advertising, high ceilings, etc). The study will be regarded as a key for the basis and placement of the guidelines for determining the appropriateness of the in the future urban railway station sign guide.

The Imprinted Messenger RNA Expression in Cloned Porcine Pre-implantation Embryos

  • The objective of this study was to determine the mRNA expression patterns of several putative imprinted genes in in vivo and in vitro fertilized, parthenogenetic, and cloned porcine preimplantation embryos. Both maternally (Dlk1, IGF2, Peg1/Mest and Ndn) and paternally (IGF2r, H19 and Xist) imprinted genes were selected. We have used reverse transcription polymerase chain reaction (RT-PCR) to investigate gene expression patterns in the porcine embryos. IGF2 transcripts were detected in the most of embryos. In nuclear transfer (NT), Peg1/MEST transcripts showed fluctuating pattern. Dlk1 was only expressed partially from the morula and blastocyst stage of NT embryos. Ndn gene expression was started somewhat early for in vivo embryos. However, the expressions of maternally imprinted genes were similar in all types of blastocysts (NT, in vivo and in vitro fertilized, and parthenogenetic embryos). The IGF2R gene expression level was somewhat irregular and varied among samples. However, for the majority samples of all types of embryos, IGF2R expression was diminished after one- to two-cell stages and reappeared at the morulae or blastocyst stage embryos. H19 gene was only expressed early in parthenogenetic and in vivo embryos. For NT embryos, H19 was only expressed in blastocysts. Xist expression was detected in all blastocysts with the earliest being in vivo 8-cell stage embryos and the last one being NT blastocysts. These putative imprinted genes appeared to have stage specific expression patterns with a fluctuating pattern for some genes (Peg/Mest, IGF2r, H19). These results suggest that stage specific presence of imprinted genes can affect the embryo implantation and fetal development.

The Application of Network Theory to Subway Transportation in Seoul, Korea

  • Network approach is used to find the shortest paths and transportation time between the subway stations in Seoul, Korea. Because of transfer stations, we reconstruct the subway network to compute the shortest routes and corresponding transportation times. The reconstructed network is useful to obtain desired information because it can handle the transfer time between tracks. Time and route information about the subway system is obtained and it will be displayed in the subway guide board at each station. Then, all passengers can have the information of shortest route to a destination and corresponding transportation time.

  • This study was carried out to compare farrowing rate and litter traits for European and American lines with boar sperm frozen in straws. Farrowing rate, litter size and mean pig weght at birth and 21 days were investigated. A total of 36 gilts Landrace, Large white and Duroc were investigated at the Chungnam Provincial Animal Breeding Station. We obtained higher farrowing rate and litter traits for European line boars compared to American line boars.

EFFECT OF MILK YIELD ON GROWTH OF MULTIPLE CALVES IN JAPANESE BLACK CATTLE (WAGYU)

  • An experiment was conducted to examine the feasibility of producing multiple calves using embryo transfer in Japanese Black cattle. Milk yield of cows and forage intake of calves were measured for 11 cows with single calves, 14 cows with twins and one cow with triplets. The means of 26 weeks cumulative milk yield were 854, 1028 and 1271 kg for cows having singles, twins and triplets, respectively. Male birth weights for single calves, twins and triplet were 34.9, 26.6 and 19.9 kg, and female ones were 31.7, 24.1 and 22.1 kg, respectively. Weight and daily gain of calves were affected by weeks (W), sex (S), the number of calves (N), parity, birth season, $W{\times}N$, $S{\times}N$ and regression on milk yield. Growth rate was higher for single calves than for twins until about 9 weeks of age, then weights increased at a similar rate. Male calf weaning weights for singles, twins and triplets were 207.0, 177.1 and 162.2 kg, and those for females were 185.4, 151.6 and 180.4 kg, respectively. Average regression coefficients of calf growth on milk yield were significant, and single calf was affected more than twin calves by increment of milk yield. As the number of calves per cow increased, hay intake of calves decreased and concentrate intake tended to increase between 6 and 13 weeks of age.

A Study on the Guidance Design for the Metro Station's Effective Sign Awareness (도시 철도역의 효과적인 안내사인 전달을 위한 디자인 연구)

  • Metro stations are public places used by many people. If they use the station frequently, there is no struggling to find a way. But the first tourists or foreigners to visit are facing finding difficulties because of errors of the complicated sign and information. This study is the guidance of the metro station sign to provide effective information system and the research on the design improvement of metro station. In the first survey for this study I proceeded research about the concept of sign systems in metro station and literature search. In the second survey, I researched needs of the problems of the metro station users. In the third survey, I investigated Busan Metro Station's problems and design to improve the guide sign. In the survey results, users had a lot of difficulties in finding information because systematically increasing amount of information and guidance of the metro station. People can't find information because of sign indiscreet commercial advertising. Also they can get false information from sign without unity. To deal with these problems the metro station needs to improve design for a second-class citizen and refrain commercial advertising and design for information transfer capability.

Extension Services in Agricultural Experiment Station (지역특화작목시험장의 기술전파활동 현황)

  • The objectives of this study were 1) to explore the present status of extension services of agricultural experiment station, and 2) to get some implications for ameliorating extension services of agricultural experiment station. The data were collected through the semi-constructed interviews with 11 agricultural researchers in 6 local agricultural experiment stations and 11 farmers who have ever made contact with the researchers. The major findings of this study were as follows: The agricultural experiment stations were providing extension services as expedients to accommodate to new markets and structural changes of organizations, though they are originally research-oriented agencies. The extension methods of agricultural experiment stations were similar with those of extension agencies, and the extension methods used mainly were 'experiment plot demonstration exhibition', 'consultation using telephone, internet, face to face meeting', 'visiting farms and consulting'. These were generally carried out by farmer's request, therefore the extension services of agricultural experiment stations were provided in a passive way so far. Based on the results of this study, the followings are recommended for further development of extension services of agricultural experiment station: The rural development organizations should set principles and support for extension services of agricultural experiment stations. The alternative plans to strengthen the linkage between research and extension have to be made, and these plans should include financial, personnel and systematic arrangements for further development of extension services of agricultural experiment stations.
